| 1 | Facile one-step synthesis of graphitic carbon nitride-modified biochar for the removal of reactive red 120 through adsorption and photocatalytic degradation显示文摘Converting waste biomass into value-added biochar has been considered as a green and sustainable strategy for resource management and pollution control.In this study,graphitic carbon nitride(g-C_(3)N_(4))modified biochars(BCs)were produced through one-pot pyrolysis of urea and hickory chips in differential ratios at 520℃.The resulting BC/g-C_(3)N_(4)composites were evaluated in laboratory for their physicochemical,adsorptive,and photocatalytic properties.The characterization tests showed the successful synthesis of the BC/g-C_(3)N_(4)composites that introduced g-C_(3)N_(4)structure,N-containing surface func-tional groups,reduced surface area,and better thermal stability to the biochar.After modification,the BC/g-C_(3)N_(4)composites showed better adsorption ability to reactive red 120(RR120)than the pristine BC,due to the strong electrostatic attrition between N-containing functional groups of g-C_(3)N_(4)on biochar surface and anionic RR120.The BC/g-C_(3)N_(4)composites also inherited g-C_(3)N_(4)’s photocatalytic activity,which is visible light responsive to generate free radicals for RR120 degradation.In addition,the composites with higher urea modification ratios were more effective in the degradation of RR120.Overall,this study demonstrates the feasibility and promising potential of combining biochar and photocatalyst for the removal of aqueous dye.Because of the synergistic adsorption and photodegradation ability,BC/g-C_(3)N_(4)composites present a novel and cost-effective solution for the removal of aqueous dye and other photodegradable contaminants under natural conditions. | Yulin Zheng Yichen Yang Yue Zhang Weixin Zou Yidan Luo Lin Dong Bin Gao | 2019 | Biochar2019,1,1: | 5 |

| 6 | trans-2-Enoyl-CoA Reductase Tecr-Driven Lipid Metabolism in Endothelial Cells Protects against Transcytosis to Maintain Blood-Brain Barrier Homeostasis显示文摘The transport and metabolism of lipids in cerebrovascular endothelial cells(ECs)have been hypothesized to regulate blood-brain barrier(BBB)maturation and homeostasis.Long-chain polyunsaturated fatty acids(LCPUFAs)as the important lipids components of cell membranes are essential for the development and function of BBB,but the direct links of lipid metabolism and ECs barrier function remain to be established.Here,we comprehensively characterize the transcriptomic phenotype of developmental cerebrovascular ECs in single-cell resolution and firstly find that trans-2-enoyl-CoA reductase(Tecr),a verylong-chain fatty acid synthesis,is highly expressed during barriergenesis and decreased after BBB maturation.EC-specific knockout of Tecr compromises angiogenesis due to delayed vascular sprouting.Importantly,EC-specific deletion of Tecr loss restrictive quality of vascular permeability from neonatal stages to adulthood,with high levels of transcytosis,but maintains the vascular tight junctions.Moreover,lipidomic analysis shows that the expression of Tecr in ECs is associated with the containing of omega-3 fatty acids,which directly suppresses caveolae vesicles formation.These results reveal a protective role for Tecr in BBB integrity and suggest that Tecr as a novel therapeutic target in the central nervous system(CNS)diseases associated with BBB dysfunction. | Jinxuan Wang Jianxiong Xu Guangchao Zang Tao Zhang Qi Wu Hongping Zhang Yidan Chen Yi Wang Weixi Qin Shuang Zhao Erdai Qin Juhui Qiu Xiaojuan Zhang Lin Wen Yeqi Wang Guixue Wang | 2022 | Research2022,,3: | 1 |

| 13 | Enhancement of CAR-T cell activity against cholangiocarcinoma by simultaneous knockdown of six inhibitory membrane proteins显示文摘Background:Existing treatments for cholangiocarcinoma have poor efficacy.However,chimeric antigen receptor-T(CAR-T)cells are emerging as a potential therapeutic strategy.Solid tumors possess multiple adverse factors in an immunosuppressive microenvironment that impair CAR-T cell infiltration and function.This study aimed to improve the function of CAR-T cells through knock down immune checkpoints and immunosuppressive molecular receptors.Methods:We evaluated the expression of epidermal growth factor receptor(EGFR)and B7 homolog 3 protein(B7H3)antigens in cholangiocarcinoma tissues using immunohistochemistry and screened specific immune checkpoints in the cholangiocarcinoma microenvironment via flow cytometry.Subsequently,we engineered CAR-T cells targeting EGFR and B7H3 antigens.We simultaneously knocked down immune checkpoints and immunosuppressive molecular receptors in CAR-T cells by constructing two clusters of small hairpin RNAs and evaluated the engineered CAR-T cells for antitumor activity both in vitro,using tumor cell lines and cholangiocarcinoma organoid models,and in vivo,using humanized mouse models.Results:We observed high expression of EGFR and B7H3 antigens in cholangiocarcinoma tissues.EGFR-CAR-T and B7H3-CAR-T cells demonstrated specific anti-tumor activity.We found an abundance of programmed cell death protein 1(PD-1),T cell immunoglobulin and mucin domain-containing protein 3(Tim-3),and T cell immunoglobulin and ITIM domain(Tigit)on infiltrated CD8^(+)T cells in the cholangiocarcinoma microenvironment.We then decreased the expression of these 3 proteins on the surface of CAR-T cells,named PTG-scFV-CAR-T cells.Furthermore,we knocked-down the expression of transforming growth factor beta receptor(TGFβR),interleukin-10 receptor(IL-10R),and interleukin-6 receptor(IL-6R)of PTG-scFV-CAR-T cells.Those cells,named PTG-T16R-scFVCAR-T cells,potently killed tumor cells in vitro and promoted apoptosis of tumor cells in a cholangiocarcinoma organoidmodel.Finally,the PTG-T16R-scFv-CART cells showed greater inhibitory effect on tumor growth in vivo,and were superior in prolonging the survival of mice.Conclusions:Our results revealed that PTG-T16R-scFV-CAR-T cells with knockdown of sextuplet inhibitory molecules exhibited strong immunity against cholangiocarcinoma and long-term efficacy both in vitro and in vivo.This strategy provides an effective and personalized immune cell therapy against cholangiocarcinoma. | Yidan Qiao Jie Chen Xuemei Wang Shumei Yan Jizhou Tan Baijin Xia Yongjian Chen Keming Lin Fan Zou Bingfeng Liu Xin He Yiwen Zhang Xu Zhang Hui Zhang Xiangyuan Wu Lijuan Lu | 2023 | Cancer Communications2023,43,7: | 0 |
