6A novel D(−)-lactic acid-inducible promoter regulated by the GntR-family protein D-LldR of Pseudomonas fluorescens显示文摘Lactic acid has two stereoisomers of D(−)-and L(+)-forms,both of which are important monomers of biodegradable plastic,poly-lactic acid.In this study,a novel D-lactate inducible system was identified in Pseudomonas fluorescens A506,partially characterized and tested as biosensor.The D-lactate catabolic operon(lldP-dld-II)was negatively regulated through the inversely transcribed D-lldR(encoding a GntR-type regulator),where the repression is relieved by addition of D-lactate.The derepression was specific to D-lactate and marginally affected by L-lactate.The D-LldR-responsive operator,showing dyad symmetry and separated by one base,was located between+11 and+27 from the transcription start site of the lldP-dld-II operon.By site-directed mutagenesis,a motif with a dyad symmetry(AATTGGTAtTACCAATT),present in the upstream region of lldP,was identified as essential for the binding of LldR.D-lactate biosensors were developed by connecting the upregulation by D-lactate to a green fluorescent readout.About^6.0-fold induction by 100mM D-lactate was observed compared to Llactate.Kalpana Singh Satish Kumar Ainala Yeonhee Kim Sunghoon Park 2019Synthetic and Systems Biotechnology2019,4,3:0
7Layer-by-layer heparinization of decellularized liver matrices to reduce thrombogenicity of tissue engineered grafts显示文摘Background: Tissue-engineered liver grafts may offer a viable alternative to orthotopic liver transplantation and help overcome the donor organ shortage. Decellularized liver matrices (DLM) have a preserved vasculature and sustain hepatocellular function in culture, but graft survival after transplantation remains limited due to thrombogenicity of the matrix. Aim: To evaluate the effect of heparin immobilization on DLM thrombogenicity. Methods: Heparin was immobilized on DLMs by means of layer-by-layer deposition. Grafts with 4 or 8 bilayers and 2 or 4 g/L of heparin were recellularized with primary rat hepatocytes and maintained in culture for 5 days. Hemocompatibility of the graft was assessed by ex vivo diluted whole-blood perfusion and heterotopic transplantation. Results: Heparin was deposited throughout the matrix and the heparin content in the graft was higher with increasing number of bilayers and concentration of heparin. Recellularization and in vitro albumin and urea production were unaffected by heparinization. Resistance to blood flow during ex vivo perfusion was lower with increased heparinization and, macroscopically, no clots were visible in grafts with 8 bilayers. Following transplantation, flow through the graft was limited in all groups. Histological evidence of thrombosis was lower in heparinized DLMs, but transplantation of DLM grafts was not improved. Conclusions: Layer-by-layer deposition of heparin on a DLM is an effective method of immobilizing heparin throughout the graft and does not impede recellularization or hepatocellular function in vitro. Thrombogenicity during ex vivo blood perfusion was reduced in heparinized grafts and optimal with 8 bilayers, but transplantation remained unsuccessful with this method. 8Analgesic efficacy of median nerve stimulation in mice with chemotherapy-induced peripheral neuropathy via modulation of brain-derived neurotrophic factor expression显示文摘OBJECTIVE:Chemotherapeutic agents such as docetaxel(DTX)can trigger chemotherapy-induced peripheral neuropathy(CIPN),which is characterized by unbearable pain.This study was designed to investigate the analgesic effect and related neuronal mechanism of low-frequency median nerve stimulation(LFMNS)on DTX-induced tactile hypersensitivity in mice.METHODS:To produce CIPN,DTX was administered intraperitoneally 4 times,once every 2 d,to male ICR mice.LFMNS was performed on the wrist area,and the pain response was measured using von Frey filaments on both hind paws.Western blot and immunofluorescence staining were performed using dorsal root ganglion and spinal cord samples to measure the expression of brainderived neurotrophic factor(BDNF).RESULTS:Repeated LFMNS significantly attenuated the DTX-induced abnormal sensory response and suppressed the enhanced expression of BDNF in the DRG neurons and spinal dorsal area.CONCLUSIONS:LFMNS might be an effective nonpharmaceutical option for treating patients suffering from CIPN via regulating the expression of peripheral and central BDNF.Dong-Wook Kang Jae-Gyun Choi Hee Ju Song Jaehyuk Kim Miae Lee Taehee Kim Suk-Yun Kang Yeonhee Ryu Hwa Seung Yoo Jin Sun Lee Jin Bong Park Sang Do Lee Hyun-Woo Kim 2023Journal of Traditional Chinese Medicine2023,43,4:0
