1Characterization of miRNomes in Acute and Chronic Myeloid Leukemia Cell Lines显示文摘Myeloid leukemias are highly diverse diseases and have been shown to be associated with microRNA(miRNA) expression aberrations. The present study involved an in-depth miRNome analysis of two human acute myeloid leukemia(AML) cell lines, HL-60 and THP-1, and one human chronic myeloid leukemia(CML) cell line, K562, via massively parallel signature sequencing. mRNA expression profiles of these cell lines that were established previously in our lab facilitated an integrative analysis of miRNA and mRNA expression patterns. miRNA expression profiling followed by differential expression analysis and target prediction suggested numerous miRNA signatures in AML and CML cell lines. Some miRNAs may act as either tumor suppressors or oncomiRs in AML and CML by targeting key genes in AML and CML pathways. Expression patterns of cell type-specific miRNAs could partially reflect the characteristics of K562, HL-60 and THP-1 cell lines, such as actin filament-based processes, responsiveness to stimulus and phagocytic activity. miRNAs may also regulate myeloid differentiation, since they usually suppress differentiation regulators. Our study provides a resource to further investigate the employment of miRNAs in human leukemia subtyping, leukemogenesis and myeloid development. 2Preparation of SiC Porous Ceramics by Crystalline Silicon Cutting Waste显示文摘SiC porous ceramics were prepared at 1 400 ℃ for4 h with crystalline silicon cutting waste and activated carbon as main starting materials and NH_4HCO_3 as the pore-forming agent. Effects of NH_4HCO_3 additions( 0,20%,30%,40%,by mass) on the phase composition,microstructure,sintering properties,cold compressive strength and thermal shock resistance of as-prepared Si C porous ceramics were investigated. The results show that:( 1) addition of NH_4HCO_3 remarkably influences the apparent porosity and cold compressive strength of specimens. The apparent porosity achieves its maximum value( 63. 40%) when 40% NH_4HCO_3 is added,while the minimum cold compressive strength is 4. 77 MPa;( 2) the specimen with 40% NH_4HCO_3 has the best thermal shock resistance. 3Thrombin is a therapeutic target for non-small-cell lung cancer to inhibit vasculogenic mimicry formation显示文摘Tumor cells transform into endothelial cells by epithelial-to-mesenchymal transition,which is characterized by vasculogenic mimicry(VM).VM not only accelerates tumor progression but also increases drug-induced resistance.However,very little is currently known about the molecular determinants that enable VM.Targeting VM might bring a new breakthrough in cancer treatment.Thrombin is the key enzyme of the blood coagulation system and could contribute to tumor progression.Nevertheless,the association between thrombin and VM formation remains largely unknown.We found that VM was associated with the overall survival of non-small-cell lung cancer(NSCLC)patients,and that thrombin expression was closely related to VM formation.This research revealed that thrombin induced VM formation via PAR-1-mediated NF-κB signaling cascades.The novel thrombin inhibitors r-hirudin and DTIP inhibited VM formation and spontaneous metastases in subcutaneous tumors.Clinical pathological analysis confirmed that NSCLC patients with thrombin-positive/PAR-1-high expression had the poorest prognosis and were the most likely to form VM.The promotional activity of thrombin in VM formation and tumor metastasis was abolished in PAR-1-deficient NSCLC cells.The EGFR inhibitor gefitinib had no effect on VM and increased VEGF expression in tumors.The combination therapy of DTIP and gefitinib achieved a better therapeutic effect than either agent alone.This study is the first to illustrate that thrombin substantially contributes,together with PAR-1,to VM formation and to illustrate that VM might be a target of r-hirudin and DTIP to suppress tumor progression.The anticoagulants r-hirudin and DTIP could be employed for antitumor therapy.Combination therapy with DTIP with an EGFR inhibitor might achieve superior therapeutic effects.Bing Zhao Mengfang Wu Zhihuang Hu Yixin Ma Wang Qi Yanling Zhang Yaran Li Min Yu Huijie Wang Wei Mo 2020Signal Transduction and Targeted Therapy2020,5,1:2
4Effect of microstructure evolution and crystal structure on thermal properties for plasma-sprayed RE_(2)SiO_(5)(RE=Gd,Y,Er)environmental barrier coatings显示文摘In this work,the microstructure evolution,thermal expansion,thermal conductivity,and thermal shock resistance properties of the plasma-sprayed Xl-Gd_(2)SiO_(5),X2-Y_(2)SiO_(5),and X2-Er_(2)SiO_(5)coatings were evaluated and compared by experimental measurement and theoretical exploration.Results showed that significant microstructure evolution such as crystallization of amorphous phase,grain growth,and defects reduction was observed in the RE_(2)SiO_(5)coatings after thermal aging at 1400℃.The Xl-Gd_(2)SiO_(5)coating exhibited higher CTE values than the X2-Y_(2)SiO_(5)and X2-Er_(2)SiO_(5)coatings,which was related to their crystal structure.The thermal conductivity of thermal-aged RE_(2)SiO_(5)coating was much higher than that of the as-sprayed RE_(2)SiO_(5)coating,and thermal conductivity was determined not only by crystal structure but also mainly by the microstructure of the coatings.The X2-Y_(2)SiO_(5)and X2-Er_(2)SiO_(5)coatings with lower thermal mismatch stre s ses presented much better thermal shock resistance than the X1-Gd_(2)SiO_(5)coating.Xin Zhong Tao Zhu Yaran Niu Haijun Zhou Le Zhang Xiangyu Zhang Qilian Li Xuebin Zheng 2021Journal of Materials Science & Technology2021,,26:1
5Design and fabrication of r-hirudin loaded dissolving microneedle patch for minimally invasive and long-term treatment of thromboembolic disease显示文摘Cardiovascular disease is the leading cause of global mortality,with anticoagulant therapy being the main prevention and treatment strategy.Recombinant hirudin(r-hirudin)is a direct thrombin inhibitor that can potentially prevent thrombosis via subcutaneous(SC)and intravenous(IV)administration,but there is a risk of haemorrhage via SC and IV.Thus,microneedle(MN)provides painless and sanitary alternatives to syringes and oral administration.However,the current technological process for the micro mould is complicated and expensive.The micro mould obtained via three-dimensional(3D)printing is expected to save time and cost,as well as provide a diverse range of MNs.Therefore,we explored a method for MNs array model production based on 3D printing and translate it to micro mould that can be used for fabrication of dissolving MNs patch.The results show that r-hirudin-loaded and hyaluronic acid(HA)-based MNs can achieve transdermal drug delivery and exhibit significant potential in the prevention of thromboembolic disease without bleeding in animal models.These results indicate that based on 3D printing technology,MNs combined with r-hirudin are expected to achieve diverse customizableMNs and thus realize personalized transdermal anticoagulant delivery for minimally invasive and long-term treatment of thrombotic disease.Mengfang Wu Tian Xia Yaran Li Tianfa Wang Shijia Yang Jinchao Yu Qiaoyan Liang Teng Shen Min Yu Bing Zhao 2022Asian Journal of Pharmaceutical Sciences2022,17,2:1

8Dynamic covalent nano-networks comprising antibiotics and polyphenols orchestrate bacterial drug resistance reversal and inflammation alleviation显示文摘New antimicrobial strategies are urgently needed to meet the challenges posed by the emergence of drug-resistant bacteria and bacterial biofilms.This work reports the facile synthesis of antimicrobial dynamic covalent nano-networks(aDCNs)composing antibiotics bearing multiple primary amines,polyphenols,and a cross-linker acylphenylboronic acid.Mechanistically,the iminoboronate bond drives the formation of aDCNs,facilitates their stability,and renders them highly responsive to stimuli,such as low pH and high H2O2 levels.Besides,the representative A1B1C1 networks,composed of polymyxin B1(A1),2-formylphenylboronic acid(B1),and quercetin(C1),inhibit biofilm formation of drug-resistant Escherichia coli,eliminate the mature biofilms,alleviate macrophage inflammation,and minimize the side effects of free polymyxins.Excellent bacterial eradication and inflammation amelioration efficiency of A1B1C1 networks are also observed in a peritoneal infection model.The facile synthesis,excellent antimicrobial performance,and biocompatibility of these aDCNs potentiate them as a much-needed alternative in current antimicrobial pipelines.Yuanfeng Li Yin-Zi Piao Hua Chen Keqing Shi Juqin Dai Siran Wang Tieli Zhou Anh-Tuan Le Yaran Wang Fan Wu Rujiang Ma Linqi Shi Yong Liu 2023Bioactive Materials2023,,9:0
9Orderedisorder transition and thermal conductivities of the(NdSmEuGd)_((1-x)/2)Dy_(2x)Zr_(2)O_(7)series显示文摘Rare-earth zirconates with pyrochlore and fluorite structures have recently been identified as promising thermal barrier coating materials owing to their low thermal conductivities.In this study,six samples with the general formula(NdSmEuGd)_((1-x)/2)Dy_(2x)Zr_(2)O_(7)were synthesized to further reduce the thermal conductivity.X-ray diffraction and Raman spectroscopy showed that the transition from an ordered pyrochlore to a disordered fluorite structure is due to cation and anion disorder.Transmission electron microscopy showed that anion disorder occurred before cation disorder.A modified mass disorder parameter was introduced into this system,which can describe the change in thermal conductivity well.This parameter can be a basis for designing more complex materials with lower thermal conductivities.Mengyao Li Chucheng Lin Yaran Niu Jimei Zhang Yi Zeng Xuemei Song 2023Journal of Materiomics2023,9,1:0

12Adaptive Bird-like Genome Miniaturization During the Evolution of Scallop Swimming Lifestyle显示文摘Genome miniaturization drives key evolutionary innovations of adaptive traits in vertebrates,such as the flight evolution of birds.However,whether similar evolutionary processes exist in invertebrates remains poorly understood.Derived from the second-largest animal phylum,scallops are a special group of bivalve molluscs and acquire the evolutionary novelty of the swimming lifestyle,providing excellent models for investigating the coordinated genome and lifestyle evolution.Here,we show for the first time that genome sizes of scallops exhibit a generally negative correlation with locomotion activity.To elucidate the co-evolution of genome size and swimming lifestyle,we focus on the Asian moon scallop(Amusium pleuronectes)that possesses the smallest known scallop genome while being among scallops with the highest swimming activity.Whole-genome sequencing of A.pleuronectes reveals highly conserved chromosomal macrosynteny and microsynteny,suggestive of a highly contracted but not degenerated genome.Genome reduction of A.pleuronectes is facilitated by significant inactivation of transposable elements,leading to reduced gene length,elevated expression of genes involved in energy-producing pathways,and decreased copy numbers and expression levels of biomineralization-related genes.Similar evolutionary changes of relevant pathways are also observed for bird genome reduction with flight evolution.The striking mimicry of genome miniaturization underlying the evolution of bird flight and scallop swimming unveils the potentially common,pivotal role of genome size fluctuation in the evolution of novel lifestyles in the animal kingdom.Yuli Li Yaran Liu Hongwei Yu Fuyun Liu Wentao Han Qifan Zeng Yuehuan Zhang Lingling Zhang Jingjie Hu Zhenmin Bao Shi Wang 2022Genomics, Proteomics & Bioinformatics2022,20,6:0
