1Ubiquitination of Rheb governs growth factor-induced mTORC1 activation显示文摘Mechanistic target of rapamycin mTOR complex 1 (mTORC1)plays a key role in the integration of various environmental signals to regulate cell growth and metabolism,mTORC1 is recruited to the lysosome where it is activated by its interaction with GTP-bound Rheb GTPase.However,the regulatory mechanism of Rheb activity remains largely unknown.Here,we show that ubiquitination governs the nucleotide-bound status of Rheb.Lysosome-anchored E3 ligase RNF152 catalyzes Rheb ubiquitination and promotes its binding to the TSC complex.EGF enhances the deubiquitination of Rheb through AKT-dependent USP4 phosphorylation,leading to the release of Rheb from the TSC complex.Functionally,ubiquitination of Rheb is linked to mTORC1-mediated signaling and consequently regulates tumor growth.Thus,we propose a mechanistic model whereby Rheb-mediated mTORC1 activation is dictated by a dynamic opposing act between Rheb ubiquitination and deubiquitination that are catalyzed by RNF152 and USP4 respectively.Lu Deng Lei Chen Linlin Zhao Yan Xu Xiaoping Peng Xinbo Wang Lin Ding Jiali Jin Hongqi Teng Yanming Wang Weijuan Pan Fei Yu Lujian Liao Li Li Xin Ge Ping Wang 2019Cell Research2019,29,2:16
2Inhalation treatment of primary lung cancer using liposomal curcumin dry powder inhalers显示文摘Lung cancer is the leading cause of cancer-related deaths. Traditional chemotherapy causes serious toxicity due to the wide bodily distribution of these drugs. Curcumin is a potential anticancer agent but its low water solubility, poor bioavailability and rapid metabolism significantly limits clinical applications. Here we developed a liposomal curcumin dry powder inhaler(LCD) for inhalation treatment of primary lung cancer. LCDs were obtained from curcumin liposomes after freeze-drying. The LCDs had a mass mean aerodynamic diameter of 5.81 μm and a fine particle fraction of 46.71%, suitable for pulmonary delivery. The uptake of curcumin liposomes by human lung cancer A549 cells was markedly greater and faster than that of free curcumin. The high cytotoxicity on A549 cells and the low cytotoxicity of curcumin liposomes on normal human bronchial BEAS-2B epithelial cells yielded a high selection index partly due to increased cell apoptosis. Curcumin powders, LCDs and gemcitabine were directly sprayed into the lungs of rats with lung cancer through the trachea. LCDs showed higher anticancer effects than the other two medications with regard to pathology and the expression of many cancer-related markers including VEGF, malondialdehyde, TNF-α, caspase-3 and BCL-2. 3Influence of magma intrusion on gas outburst in a low rank coal mine显示文摘The effect of magma intrusion on gas outburst is illustrated by a case study of the exposed magma intrusion in the 313 mining area,upper coal seam Number 3,in the Qiwu Mine located in Shandong province.Vitrinite reflectance,mercury injection,and maceral statistical analysis are used to characterize the coal.The aspects of coal metamorphism include changes in micro-components as well as in coal structure,the formation of new substances,and changes in gas absorption and storage.The results show that vitrinite reflectance increases within the region influenced by magma intrusion.The metamorphosed region may be divided into a weakly affected belt,a medium affected belt,a strongly affected belt,and a completely affected belt.Compared to the unaffected coal the total pore volume,as well as the amount of big and middle sized holes,increases while the number of transition holes and micro-pores decreases.This diminishes the absorption capacity of the matrix but enlarges the total gas storage space.Vitrinite content initially decreases slightly but then increases rapidly while the inertinite content increases at first but then decreases.Exinite content decreases,then increases,and finally drops to zero.Higher vitrinite,and a lower inertinite,content increase gas absorption ability.This balances reduced adsorption caused by changes to pore structure.Consequently,gas adsorption capacity is not substantially reduced as the coal rank increases.Thermal metamorphism of the coal produces CH4 and other hydrocarbons that increase the total gas content in the coal seam.Asphaltene migrates into the medium and weakly affected regions filling in the pores and fractures there.This plugs the pathway for gas transport.A barrier is formed that hinders gas flow.CO2,H2S,N2,and other gases carried in by the magma react to produce CO2,which increases in relative concentration and enhances the risk of gas outburst.The two barriers,magma intrusion on one side and the medium and weakly affected belts on the other,as well as the unaffected coal seam itself,trap a large amount of gas during the thermal activity.This is the basic reason for gas outburst.These conclusions can enlighten activities related to gas prevention and control in a low rank coal mine affected by magma intrusion.Chen Shangbin Zhu Yanming Li Wu Wang Hui 2012International Journal of Mining Science and Technology2012,22,2:11
4Effect of initial material bulk density and easily-degraded organic matter content on temperature changes during composting of cucumber stalk显示文摘To inactivate the potentially pathogenic microorganisms and safely utilize vegetable waste compost, ultra-high temperatures(> 70°C) should be maintained during the composting without having an inhibitory effect on maturity. This study investigated the influence of bulk density(part 1) and easily-degraded organic matter content(EDOMC, part 2) on temperature evolution during vegetable waste composting: Part 1: corn straw with different particle sizes was used to achieve different bulk densities in the composting material(BD1–BD3); Part 2: partial or total substitution of the corn straw by corn starch was carried out to obtain different EDOMC(ED1–ED4). The composting experiments were conducted in a labscale reactor(1.75 kg material) and lasted for 30 d. Temperature and CO2 emission were recorded daily, and the organic matter, lignocellulose, microbial activity, germination index(GI) and C/N of the samples were measured at different stages. The highest temperature(65.7°C) in part 1 occurred in the treatment with the bulk density of 0.35 g/cm3, which also had the longest thermophilic phase. Bulk density was found to seriously influence the utilization efficiency of O2 and heat transfer through materials, rather than heat production from organic matter degradation. In experiment part 2, the highest temperature was obtained with EDOMC of 45%(71.4°C). 5Stepwise-Nanocavity-Assisted Transmissive Color Filter Array Microprints显示文摘Visible-light color flters using patterned nanostructures have attracted much interest due to their various advantages such as compactness,enhanced stability,and environmental friendliness compared with traditional pigment or dye-based optical flters.While most existing studies are based on planar nanostructures with lateral variation in size,shape,and arrangement,the vertical dimension of structures is a long-ignored degree of freedom for the structural colors.Herein,we demonstrate a synthetic platform for transmissive color flter array by coordinated manipulations between height-varying nanocavities and their lateral flling fractions.Te thickness variation of those nanocavities has been fully deployed as an alternative degree of freedom,yielding vivid colors with wide gamut and excellent saturation.Experimental results show that the color-rendering capability of the pixelated nanocavities can be still retained as pixels are miniaturized to 500 nm.Crosstalk between closely spaced pixels of a Bayer color flter arrangement was calculated,showing minimal crosstalk for 1μm2 square subpixels.Our work provides an approach to designing and fabricating ultracompact color flter arrays for various potential applications including stained-glass microprints,microspectrometers,and high-resolution image sensing systems.Yasi Wang Mengjie Zheng Qifeng Ruan Yanming Zhou Yiqin Chen Peng Dai Zhengmei Yang Zihao Lin Yuxiang Long Ying Li Na Liu Cheng-Wei Qiu Joel K.W.Yang Huigao Duan 2018Research2018,,1:8
6TSdb:A database of transporter substrates linking metabolic pathways and transporter systems on a genome scale via their shared substrates显示文摘TSdb (http://gffzzf860537ad99741d5hvxpcuquoocfk6v0b.ffgz.tsg.suse.edu.cn) is the first manually curated central repository that stores formatted information on the substrates of transporters. In total, 37608 transporters with 15075 substrates from 884 organisms were curated from UniProt functional annotation. A unique feature of TSdb is that all the substrates are mapped to identifiers from the KEGG Ligand com- pound database. Thus, TSdb links current metabolic pathway schema with compound transporter systems via the shared compounds in the pathways. Furthermore, all the transporter substrates in TSdb are classified according to their biochemical properties, biological roles and subcellular localizations. In addition to the functional annotation of transporters, extensive compound annotation that includes inhibitor information from the KEGG Ligand and BRENDA databases has been integrated, making TSdb a useful source for the discovery of potential inhibitory mechanisms linking transporter substrates and metabolic enzymes. User-friendly web interfaces are designed for easy access, query and download of the data. Text and BLAST searches against all transporters in the database are provided. 7Pore Structure Heterogeneity of the Xiamaling Formation Shale Gas Reservoir in the Yanshan Area of China: Evaluation of Geological Controlling Factors显示文摘Micro-heterogeneity is an integral parameter of the pore structure of shale gas reservoir and it forms an essential basis for setting and adjusting development parameters.In this study,scanning electron microscopy,high-pressure mercury intrusion and low-temperature nitrogen adsorption experiments were used to qualitatively and quantitatively characterize the pore structure of black shale from the third member of the Xiamaling Formation in the Yanshan area.The pore heterogeneity was studied using fractal theory,and the controlling factors of pore development and heterogeneity were evaluated in combination with geochemical parameters,mineral composition,and geological evolution history.The results show that the pore structure of the reservoir was intricate and complicated.Moreover,various types of micro-nano scale pores such as dissolution pores,intergranular pores,interlayer pores,and micro-cracks are well developed in member 3 of the Xiamaling Formation.The average porosity was found to be 6.30%,and the mean value of the average pore size was 4.78 nm.Micropores and transition pores provided most of the storage space.Pore development was significantly affected by the region and was mainly related to the total organic carbon content,vitrinite reflectance and mineral composition.The fractal dimension,which characterizes the heterogeneity,is 2.66 on average,indicating that the pore structure is highly heterogeneous.Fractal dimension is positively correlated with maturity and clay mineral content,while it is negatively correlated with brittle mineral content and average pore size.These results indicate that pore heterogeneity is closely related to thermal history and material composition.Combined with the geological background of this area,it was found that the pore heterogeneity was mainly controlled by the Jurassic magmatism.The more intense the magma intrusion,the stronger the pore heterogeneity.The pore structure and its heterogeneity characteristics present today are a general reflection of the superimposed geological processes of sedimentary-diagenetic-late transformation.The influence of magmatic intrusion on the reservoir is the main geological factor that should be considered for detailed evaluation of the Xiamaling Formation shale gas reservoir in the Yanshan area.LI Xueyuan CHEN Shangbin WANG Xiaoqi ZHU Yanming CHANG Minghua Clementine UWAMAHORO 2019Acta Geologica Sinica(English Edition)2019,93,3:5

9Multi-objective clustering analysis via combinatorial pigeon inspired optimization显示文摘Multi-objective data clustering is an important issue in data mining, and the realization of data clustering using the multiobjective optimization technique is a significant topic. A combinatorial multi-objective pigeon inspired optimization(CMOPIO)with ring topology is proposed to solve the clustering problem in this paper. In the CMOPIO, a delta-locus based coding approach is employed to encode the pigeons. Thus, the length of pigeon representation and the dimension of the search space are significantly reduced. Thereby, the computational load can be effectively depressed. In this way, the pigeon inspired optimization(PIO) algorithm can be discretized with an auxiliary vector to address data clustering. Moreover, an index-based ring topology with the ability of contributing to maintain flock diversity is adopted to improve the CMOPIO performance. 10cDNA cloning and expression of erythropoietin in the plateau zokor(Myospalax baileyi) from the Qinghai-Tibet Plateau显示文摘All organisms respond to variation in their environments and manage environmental stress through metabolic adjustments.The plateau zokor(Myospalax baileyi) is an endemic and keystone subterranean rodent species that inhabits the Qinghai-Tibet Plateau between 2800 and 4200 m above sea level.It is a hypoxic-tolerant mammal with a high ratio of oxygen utilization that enables it to cope with its harsh surroundings.To explore the molecular mechanism of altitude acclimatization of the plateau zokor,we cloned the zokor erythropoietin(Epo) gene and used real-time PCR to compare Epo mRNA levels in zokors inhabiting 16 different altitudes.The full-length zokor Epo open reading frame was 579 bp that encoded a precursor peptide of 192 amino acids with a signal peptide of 26 residues.The Epo gene of the plateau zokor was 81%-95% homologous to that of human,mouse,rat,root vole and the Golan Heights blind mole rat,with the highest homology(95%) to species of the genus Spalax.Epo mRNA was detected mainly in the zokor kidney and spleen among 8 selected tissues.The level of Epo mRNA increased in the liver and kidney with increases in altitude.The increase in the kidney was 5 times that in the liver.Remarkably,expression of Epo mRNA in the kidney of zokors living at the highest altitude(4268 m) was 12-fold higher than that of zokors living at the lowest(2492 m) altitude.These findings provide essential information for understanding the possible role of Epo in adaptation to hypoxia in the plateau zokor.WANG ZhenLong CHEN Yan YANG Jie CHEN WeiJun ZHANG YanMing ZHAO XinQuan 2012Chinese Science Bulletin2012,57,9:4

12Using big data to track marine oil transportation along the 21st-century Maritime Silk Road显示文摘China's designation of the '21 st-century Maritime Silk Road'(MSR) region is of extraordinary significance to its maritime rights, transportation security, and socio-economic development. We developed a technical framework allowing the use of 'big data' derived from the Automatic Identification System(AIS, an automatic ship-tracking network) for two purposes: the accurate mapping of oil tanker trajectories and the creation of heat maps showing the relative use of oil tanker routes and marine shipping chokepoints. We then applied these methods to 1.5 billion AIS records collected within the MSR in 2014 to statistically identify and analyze busy routes, areas, and chokepoints in this strategic region. 13Tendency for Evolution of High Myopia in 308 Chinese School Children from Xi'an City显示文摘Purpose: To observe the refractive status, especially the tendency for evolution of high myopia, in eyes of Chinese school children from Xi’an city.Methods: The study was conducted in 11514 eyes of the 5757students aged between 7~18 years in Xi’an city primary and high schools. The inclusion criterion was >-6D of the spherical equivalent refraction. The object ophthalmic examinations were done, included non-cycloplegic objective refraction, visual acuity, intraocular pressure, fundus evaluation by ophthalmologists and nurses with professional training. Specially designed questionnaires were filled in and the data were statistically analyzed with SPSS10.0.Results: Of 11514 eyes, the detection rate was 81.4%(9376eyes) for myopia and 5.3%(615 eyes) for high myopia. High myopia was found in 2.6%.(300 eyes).of right eyes, 2.7%(315 eyes) in left eyes, and 2.4%(275 eyes) were in boys and 2.9%(340 eyes) in girls. Among 12 school grades, the detection rates of high myopia increased significantly with student age, with 0.9% in the 1stgrade of primary school and12.5% in the 3rd year of senior middle school. The average refractive error of spherical equivalent refraction was(-7.43±1.29) D with 95% confidence interval(-7.54,-7.33). No significant differences were found between the right and left eyes or both genders. The distribution of myopic severity was lowest(-6D).in primary school students younger than 12 years,was higher(-8D) in junior middle school students older than13 years, and highest(-13D) in senior middle school students.Conclusion: A continuous growth was evident in the severity of high myopia throughout 12 years of primary and middle school except for the first year of primary school, with growth occurring in two transitional stages between the senior primary and junior middle school years, and between the junior and senior middle school years. The distribution of high myopia was lowest, at-6D, in primary school, increased to-8D in junior middle school, and progressed to-10D in senior middle school, indicating a high risk of development of pathologic myopia during the students’ later lifespan. 14Induced regulatory T cells suppress Tel cells through TGF-β signaling to ameliorate STZ-induced type 1 diabetes mellitus显示文摘Type 1 diabetes mellitus(T1D)is a chronic autoimmune condition in which the immune system destroys insulin-producing pancreatic β cells.In addition to well-established pathogenic effector T cells,regulatory T cells(Tregs)have also been shown to be defective in T1D.Thus,an increasing number of therapeutic approaches are being developed to target Tregs.However,the role and mechanisms of TGF-β-induced Tregs(iTregs)in T1D remain poorly understood.Here,using a streptozotocin(STZ)-induced preclinical T1D mouse model,we found that iTregs could ameliorate the development of T1D and preserve β cell function.The preventive effect was associated with the inhibition of type 1 cytotoxic T(Tel)cell function and rebalancing the Treg/Tc1 cell ratio in recipients.Furthermore,we showed that the underlying mechanisms were due to the TGF-β-mediated combinatorial actions of mTOR and TCF1.In addition to the preventive role,the therapeutic effects of iTregs on the established STZ-T1D and nonobese diabetic(NOD)mouse models were tested,which revealed improved β cell function.Our findings therefore provide key new insights into the basic mechanisms involved in the therapeutic role of iTregs in T1D.Li Zhou Xuemin He Peihong Cai Ting Li Rongdong Peng Junlong Dang Yue Li Haicheng Li Feng Huang Guojun Shi Chichu Xie Yan Lu Yanming Chen 2021Cellular & Molecular Immunology2021,18,3:3
15Influence of Si Content on Properties of Ti1-xSixN Coatings显示文摘To investigate the evolution of microstructure and wear behavior of TiSiN coatings with the variation of Si in targets and lays the foundation for its controllable mass production, Ti1-xSixN composite coatings were deposited onto Si(100) and cemented carbide substrates using TiSi targets with different Si content by cathodic arc ion plating. The influences of Si on the microstructure and mechanical properties were studied. Nano-amorphous composite structure appeared in the Ti1-xSixN coatings when Si content in TiSi target was higher than 5 at%. However, further increase of Si content in TiSi target exhibited a negligible effect on the microstructure of Ti1-xSixN coatings. Hardness and deformation resistance were correlated to the content of Si in TiSi targets. Maximum hardness was obtained as the Si content in target increased up to 20 at%. 17Estimating the Age of Oil-tea Camellia Trees显示文摘To determine the age of oil-tea camellia trees,regression equations including Logistic,Mitscherlich,Gompertz,Korf,and Richards were used to calculate accumulative growth rate using basal trunk disc and investigate the relations between the age of oil-tea camellia trees and their growth rate of secondary trunk.The Gompertz equation Y = 71. 296 1 exp(-3. 874 4 exp(-0. 006 4 t)) was the most optimal equation to simulate the accumulative growth rate of basal trunk disc.This equation could be used to estimate the age of oil-tea camellia trees that grow under similar environmental conditions. The Korf equation Y = 576. 900 1 exp(-4. 153 0 x^(-0. 314 2)) was the best equation to describe the relation between the age and growth rate of different secondary trunks. With the adjustment coefficient and average growth of different secondary trunk discs,it is possible to predict the age of ancient oil-tea camellia trees that grow under similar environmental conditions. In addition,taking three or more discs from the same diameter group and calculating their average growth rate could lead to more accurate results. 18Transport of black carbon aerosols from non-local sources:A case study in Shanghai显示文摘Black carbon(BC) aerosol mass carried by winds of varying directions from non-local sources was estimated based on hourly measured data of BC mass concentration(C_(BC)) and meteorological parameters from January 2008 to December 2012 in Shanghai,and the relationship between annual average C_(BC) and wind speed was analyzed.The results show that the annual average C_(BC) decreased with wind speed for speeds exceeding 0.3 m/s.The relationship between the two was determined by a linear fit with correlation coefficient 0.88.Assuming BC aerosol mass of non-local sources transported by a southeast wind was zero,annual average BC concentrations(μg/m^3) carried by winds of variable direction were 1.99(southwest),1.95(west),1.15(northwest),0.54(south),0.39(north),0.01(northeast),and 0.01(east).BC aerosol mass of non-local sources transported by wind to Shanghai was about 6404.051 per year,among which the total contribution of southwest,west,and northwest winds was nearly 84%.The aerosol mass transported to Shanghai in winter accounted for 35%that of the entire year,and was greater than that of the other seasons.Hongqiang Wang Yanming Kang Qianshan He Yonghang Chen 2015Particuology2015,13,3:2
19Seasonal variation in particle contribution and aerosol types in Shanghai based on satellite data from MODIS and CALIOP显示文摘Although the concentration of atmospheric particulate matter in Shanghai has declined in recent years,aerosols remain one of the major pollutants affecting air quality.Herein,spatio-seasonal variation in aerosol optical properties and aerosol types were studied overa 10-year period(2006-2015)in Shanghai,China,using satellite data from Moderate Resolution Imaging Spectroradiometer(MODIS)and CloudAerosol Lidar with Orthogonal Polarization(CALIOP).The average aerosol optical depth values for central urban and suburban areas of Shanghai were between 0.9 and 1.0.Clear seasonal variation in aerosol concentrations occurred,causing strongest attenuation in summer and weakest attenuation in autumn.Polluted dust,polluted continental(urban/industrial)and smoke aerosols were the main aerosol types.Desert dust aerosols occurred in the Shanghai area at higher altitudes(greater than 3 km)in spring,related to dusty weather in the north;while in winter,smoke aerosols occurred at high altitudes,related to haze pollution in the north.The aerosols detected in autumn were mainly from local sources,comprising polluted dust,polluted continental,and smoke aerosols.Aerosols in Shanghai clearly reflect both local and regional sources at different times.Qiong Liu Xin Liu Tongqiang Liu Yanming Kang Yonghang Chen Jiming Li Hua Zhang 2020Particuology2020,18,4:2
20Bacteroides ovatus-mediated CD27^(−)MAIT cell activation is associated with obesity-related T2D progression显示文摘Type 2 diabetes(T2D)is highly associated with obesity.However,the factors that drive the transition from excessive weight gain to glucose metabolism disruption are still uncertain and seem to revolve around systemic immune disorder.Mucosal-associated invariant T(MAIT)cells,which are innate-like T cells that recognize bacterial metabolites,have been reported to be altered in obese people and to lead to metabolic dysfunction during obesity.By studying the immunophenotypes of blood MAIT cells from a cross-sectional cohort of obese participants with/without T2D,we found an elevation in CD27^(-)negative(CD27−)MAIT cells producing a high level of IL-17 under T2D obese conditions,which could be positively correlated with impaired glucose metabolism in obese people.We further explored microbial translocation caused by gut barrier dysfunction in obese people as a triggering factor of MAIT cell abnormalities.Specifically,accumulation of the bacterial strain Bacteroides ovatus in the peripheral blood drove IL-17^(-)producing CD27−MAIT cell expansion and could be associated with T2D risk in obese individuals.Overall,these results suggest that an aberrant gut microbiota–immune axis in obese people may drive or exacerbate T2D.Importantly,CD27−MAIT cell subsets and Bacteroides ovatus could represent targets for novel interventional strategies.Our findings extend current knowledge regarding the clinical relevance of body mass index(BMI)-associated variation in circulating MAIT cells to reveal the role of these cells in obesity-related T2D progression and the underlying cellular mechanisms.Yue Li Yi Yang Jin Wang Peihong Cai Mei Li Xixiang Tang Ying Tan Yuchan Wang Fan Zhang Xiaofeng Wen Qiaoxing Liang Yuanpeng Nie Tufeng Chen Xiang Peng Xuemin He Yanhua Zhu Guojun Shi Wai WCheung Lai Wei Yanming Chen Yan Lu 2022Cellular & Molecular Immunology2022,19,7:2
