1The double face of miR-320:cardiomyocytes-derived miR-320 deteriorated while fibroblasts-derived miR-320 protected against heart failure induced by transverse aortic constriction显示文摘MicroRNAs(miRNAs)are aberrantly expressed in the pathophysiologic process of heart failure(HF).However,the functions of a certain miRNA in different cardiac cell types during HF are scarcely reported,which might be covered by the globe effects of it on the heart.In the current study,Langendorff system was applied to isolate card io myocytes(CMs)and cardiac fibroblasts(CFs)from transverse aortic constriction(TAC)-induced mice.Slight increase of miR-320 expression was observed in the whole heart tissue of TAC mice.Interestingly,miR-320 was significantly elevated in CMs but decreased in CFs from TAC mice at different time points.Then,recombinant adeno-associated virus 9 with cell-type-specific promoters were used to manipulate miR-320 expressions in vivo.Both in vitro and in vivo experiments showed the miR-320 overexpression in CMs exacerbated cardiac dysfunction,whereas overexpression of miR-320 in CFs alleviated cardiac fibrosis and hypertrophy.Mechanically,downstream signaling pathway analyses revealed that miR-320 might induce various effects via targeting PLEKHM3 and IFITM1 in CMs and CFs,respectively.Moreover,miR-320 mediated effects could be abolished by PLEKHM3 re-expression in CMs or IFITM1 re-expression in CFs.Interestingly,miR-320 treated CFs were able to indirectly affect CMs function,but not vice versa.Meanwhile,upstream signaling pathway analyses showed that miR-320 expression and decay rate were rigorously manipulated by Ago2,which was regulated by a cluster of cell-type-specific TFs distinctively expressed in CMs and CFs,respectively.Together,we demonstrated that miR-320 functioned differently in various cell types of the heart during the progression of HF.Xudong Zhang Shuai Yuan Huaping Li Jiabing Zhan Feng Wang Jiahui Fan Xiang Nie Yan Wang Zheng Wen Yanghui Chen Chen Chen Dao Wen Wang 2021Signal Transduction and Targeted Therapy2021,6,3:4
2EDTA-assisted hydrothermal synthesis of flower-like CoSe_2 nanorods as an efficient electrocatalyst for the hydrogen evolution reaction显示文摘Hydrogen evolution reaction(HER) is a prospective method to generate pure hydrogen. The development of superior electrocatalysts based on earth-abundant materials, plays a critical role in the future.CoSe_2, one of the earth-abundant electrocatalysts, has been proved to be a promising catalyst for hydrogen generation. In our work, flower-like CoSe_2 nanorods with high quality are successfully synthesized through a facile ethylenediaminetetraacetic acid ligand(EDTA)-assisted hydrothermal process. The flower-like CoSe_2 nanorods show the brilliant electrochemical HER performance with 100 mA cm^(-2) at overpotential of 273 m V, a small Tafel slope of 35 mV dec^(-1) and strong durability in acid solution. The sparkly HER catalytic activity of CoSe_2 can be ascribed to its particular structure with large surface area and abundant active sites. 3Flexible Oxide-Based Thin-Film Transistors on Plastic Substrates for Logic Applications显示文摘Phosphorus doped(P-doped) nanogranular SiO2 films have been deposited by plasma-enhanced chemical vapor deposition. A high proton conductivity of ~3.2x10-4S/cm and a large electric double layer(EDL) capacitance of ~3.2μF/cm2 have been obtained. Flexible coplanar-gate EDL thin film transistors(TFTs) gated by P-doped nanogranular SiO2 films are self-assembled on plastic substrates at room temperature. Due to the big EDL capacitance,such TFTs show ultra-low voltage operation of 1 V,a large field-effect mobility of 18.9 cm2/Vs,a small subthreshold swing of 85 m V/decade and a high current on/off ratio of 107. Furthermore,the EDL TFT could work in dual coplanar gate mode. AND logic operation is realized. 5Molecular Mechanisms of Spawning Habits for the Adaptive Radiation of Endemic East Asian Cyprinid Fishes显示文摘Despite the widespread recognition of adaptive radiation as a driver of speciation,the mechanisms by which natural selection generates new species are incompletely understood.The evolutionary radiation of endemic East Asian cyprinids has been proposed as evolving through a change in spawning habits,involving a transition from semibuoyant eggs to adhesive eggs in response to crosslinked river-lake system formation.Here,we investigated the molecular mechanisms that underpin this radiation,associated with egg hydration and adhesiveness.We demonstrated that semibuoyant eggs enhance hydration by increasing the degradation of yolk protein and accumulation of Ca2+and Mga+ions,while adhesive eggs improve adhesiveness and hardness of the egg envelope by producing an adhesive layer and a unique 4th layer to the egg envelope.Based on multiomics analyses and verification tests,we showed that during the process of adaptive radiation,adhesive eggs downregulated the'vitellogenin degradation pathway','zinc metalloprotease pathway,'and'ubiquitin-proteasome pathway'and the pathways of Ca^(2+)and Mg^(2+)active transport to reduce their hydration.At the same time,adhesive eggs upregulated the crosslinks of microfilament-associated proteins and adhesive-related proteins,the hardening-related proteins of the egg envelope,and the biosynthesis of glycosaminoglycan in the ovary to generate adhesiveness.These findings illustrate the novel molecular mechanisms associated with hydration and adhesiveness of freshwater fish eggs and identify critical molecular mechanisms involved in the adaptive radiation of endemic East Asian cyprinids.We propose that these key egg attributes may function as“magic traits”in this adaptive radiation.Feng Chen Yeke Wang Jun He Liang Chen Ge xue Yan Zhao Yanghui Peng Carl mith Jia Zhang Jun Chen Ping Xie 2023Research2023,,1:0
6NUMERICAL STUDY OF TURBULENT FLOW AROUND TWIN PLATE AT LARGE ATTACK ANGLE显示文摘The present work is mainly aimed to study the fluid dynamic force on twin plate in a turbulent flow by solving the governing equations numerically with the well known approach SIMPLE, in which the multiple time scale turbulent k-ε model has been employed to determine the eddy viscosity of turbulent flow field. The turbulence model is used with a restriction for the kinetic energy of smaller turbulent scale to achieve numerical stability. The lengths of recirculation zone for the turbulent flow around twin plate with the definite angle of attack AOA = 50° and constant flow blockage ratio 10% are in good agreement with experimental data, which indicates the wall functions used in the numerical investigation are acceptable as well. It is found that the drag force and distribution of wall pressure are intimately influenced by the arrangement of twin plate at constant flow blockage. 7Machine learning modeling identifies hypertrophic cardiomyopathy subtypes with genetic signature显示文摘Previous studies have revealed that patients with hypertrophic cardiomyopathy(HCM)exhibit differences in symptom severity and prognosis,indicating potential HCM subtypes among these patients.Here,793 patients with HCM were recruited at an average follow-up of 32.78±27.58 months to identify potential HCM subtypes by performing consensus clustering on the basis of their echocardiography features.Furthermore,we proposed a systematic method for illustrating the relationship between the phenotype and genotype of each HCM subtype by using machine learning modeling and interactome network detection techniques based on whole-exome sequencing data.Another independent cohort that consisted of 414 patients with HCM was recruited to replicate the findings.Consequently,two subtypes characterized by different clinical outcomes were identified in HCM.Patients with subtype 2 presented asymmetric septal hypertrophy associated with a stable course,while those with subtype 1 displayed left ventricular systolic dysfunction and aggressive progression.Machine learning modeling based on personal whole-exome data identified 46 genes with mutation burden that could accurately predict subtype propensities.Furthermore,the patients in another cohort predicted as subtype 1 by the 46-gene model presented increased left ventricular end-diastolic diameter and reduced left ventricular ejection fraction.By employing echocardiography and genetic screening for the 46 genes,HCM can be classified into two subtypes with distinct clinical outcomes.Jiaqi Dai Tao Wang Ke Xu Yang Sun Zongzhe Li Peng Chen Hong Wang Dongyang Wu Yanghui Chen Lei Xiao Hao Liu Haoran Wei Rui Li Liyuan Peng Ting Yu Yan Wang Zhongsheng Sun Dao Wen Wang 2023Frontiers of Medicine2023,17,4:0
8Identification of COL3A1 variants associated with sporadic thoracic aortic dissection: a case-control study显示文摘Thoracic aortic dissection(TAD)without familial clustering or syndromic features is known as sporadic TAD(STAD).So far,the genetic basis of STAD remains unknown.Whole exome sequencing was performed in 223 STAD patients and 414 healthy controls from the Chinese Han population(N=637).After population structure and genetic relationship and ancestry analyses,we used the optimal sequence kernel association test to identify the candidate genes or variants of STAD.We found that COL3A1 was significantly relevant to STAD(P=7.35×10^(−6))after 10000 times permutation test(P=2.49×10^(−3)).Moreover,another independent cohort,including 423 cases and 734 non-STAD subjects(N=1157),replicated our results(P=0.021).Further bioinformatics analysis showed that COL3A1 was highly expressed in dissected aortic tissues,and its expression was related to the extracellular matrix(ECM)pathway.Our study identified a profile of known heritable TAD genes in the Chinese STAD population and found that COL3A1 could increase the risk of STAD through the ECM pathway.We wanted to expand the knowledge of the genetic basis and pathology of STAD,which may further help in providing better genetic counseling to the patients.Yanghui Chen Yang Sun Zongzhe Li Chenze Li Lei Xiao Jiaqi Dai Shiyang Li Hao Liu Dong Hu Dongyang Wu Senlin Hu Bo Yu Peng Chen Ping Xu Wei Kong Dao Wen Wang 2021Frontiers of Medicine2021,15,3:0
9Whole-exome sequencing reveals genetic risks of early-onset sporadic dilated cardiomyopathy in the Chinese Han population显示文摘To reveal genetic risks of early-onset sporadic dilated cardiomyopathy(DCM) patients in the Chinese Han population, we enlisted 363 DCM cases and 414 healthy controls. Whole-exome sequencing and phenotypic characterization were conducted. In total, we identified 26 loss-of-function(LOF) candidates and 66 pathogenic variants from 33 genes, most of which were novel.The deleterious variants can account for 25.07%(91/363) of all patients. Furthermore, rare missense variants in 21 genes were found to be significantly associated with DCM in burden tests. Other than rare variants, twelve common SNPs were significantly associated with an increased risk of DCM in allele-based genetic model association analysis. Of note, in the cumulative risk model, high-risk subjects had a 3.113-fold higher risk of developing DCM than low-risk subjects. Also, DCM in the high-risk group had a younger age of onset than that in the low-risk group. 10Nutritional Value of Lippia nodiflora( L. ) Greene as Feed显示文摘To analyze the nutritional value as feed,the aerial stem and leaves of Lippia nodiflora( L.) Greene and Phyla nodiflora( L.) Greene were sampled to measure the nutritional indices. The nutritional indices were also compared with common forage grass,and the development value of L. nodiflora as feed was analyzed. Compared with P. nodiflora,L. nodiflora had obvious fast-growing and high-yield abilities. The contents of crude fat,crude fiber,crude ash,lysine,vitamin C,Ca,and Fe in L. nodiflora were obviously higher than that in P. nodiflora,but the content of crude protein and total P in L. nodiflora were significantly lower than that in P. nodiflora. Compared with common forage feed ryegrass,alfalfa,millet straw and corn straw,L. nodiflora had higher contents of crude fat and crude ash and lower contents of crude fiber and crude protein.
