1Inuence of oxytetracycline on the structure and activity of microbial community in wheat rhizosphere soil显示文摘The microbial community composition in wheat rhizosphere was analyzed by detecting colony forming units (CFUs) in agar plates. The total CFUs in rhizosphere were 1.04 × 109/g soil with 9.0 × 108/g bacteria, 1.37 × 108/g actinomyces and 3.6 × 106/g fungi. The 10 dominant bacteria were isolated from wheat rhizosphere and were grouped into genus Bacillus according to their full length 16S rRNA gene sequences. Although belonging to the same genus, the isolated strains exhibited dierent sensitivities to oxytetracycline. When a series of the rhizosphere soil was exposed under various concentrations of oxytetracycline, the microbial community structure was highly aected with significant decline of CFUs of bacteria and actinomyces (22.2% and 31.7% at 10 mg/kg antibiotic, respectively). This inhibition was clearly enhanced with the increase exposure dosage of antibiotic and could not be eliminated during 30 d incubation. There was no obvious inuence of this treatment on fungi population. 5Two hydroxypyruvate reductases encoded by OsHPR1 and OsHPR2 are involved in photorespiratory metabolism in rice显示文摘Mutations in the photorespiration pathway display a lethal phenotype in atmospheric air,which can be fully recovered by elevated CO2.An exception is that mutants of peroxisomal hydroxypyruvate reductase(HPR1)do not have this phenotype,indicating the presence of cytosolic bypass in the photorespiration pathway.In this study,we constructed overexpression of the OsHPR1 gene and RNA interference plants of OsHPR1 and OsHPR2 genes in rice(Oryza sativa L.cv.Zhonghua 11).Results from reverse transcription-polymerase chain reaction(RT-PCR),Western blot,and enzyme assays showed that HPR1 activity changed significantly in corresponding transgenic lines without any effect on HPR2 activity,which is the same for HPR2.However,metabolite analysis and the serine glyoxylate aminotransferase(SGAT)activity assay showed that the metabolitefiux of photorespiration was disturbed in RNAi lines of both HPR genes.Furthermore,HPR1 and HPR2proteins were located to the peroxisome and cytosol,respectively,by transient expression experiment.Double Research mutant hpr1 hpr2 was generated by crossing individual mutant of hpr1 and hpr2.The phenotypes of all transgenic lines were determined in ambient air and CO2-elevated air.The phenotype typical of photorespiration mutants was observed only where activity of both HPR1 and HPR2 were downregulated in the same line.Thesefindings demonstrate that two hydroxypyruvate reductases encoded by OsHPR1and OsHPR2 are involved in photorespiratory metabolism in rice.Nenghui Ye Guozhen Yang Yan Chen Chan Zhang Jianhua Zhang Xinxiang Peng 2014Journal of Integrative Plant Biology2014,56,2:2

8Synthesis of three-dimensional hierarchical cobalt hydroxide microstructures显示文摘β-Co(OH)2 with three-dimensional (3-D) structures was prepared by a simple hydrothermal method. It was found that the amount of cetyltrimethylammonium bromide (CTAB), the pH value, and the reaction time all had an important influence on the formation of this morphology. 9Partition airflow varying features of chaos-theory-based coalmine ventilation system and related safety forecasting and forewarning system显示文摘To realize real-time monitoring and short-term forecasting and forewarning of coalmine ventilation systems(CVS), in this paper, we first established a joint surface and underground CVS safety management system consisting of main ventilation fan, safety-partition linked passageways, and air-required locations. We then applied chaos theory to identify the air quantity and gas concentration of underground partition boundaries, and adopted a fixed data quantity, multi-step progressive, weighted first-order local-domain method to setup a chaos prediction model and a CVS safety forecasting and forewarning system formed by the normal change level, orange forewarning level, and red alarm level. We next conduct the on-field application of the system in a coalmine in Jining, Shandong, China. The results showed that (1) in the statistical scale of 5 min, the changes in both air quantity and gas concentration along CVS partition airflow boundaries were characteristic of chaos and could be used for short-term chaos prediction, and the latter was more chaotic than the former;(2) the setup chaos prediction model had a higher prediction precision and the established safety prediction system could not only predict the variation in CVS stability but also reflect the rationality of underground mining intensity. 14circCAPRIN1 interacts with STAT2 to promote tumor progression and lipid synthesis via upregulating ACC1 expression in colorectal cancer显示文摘Background:Circular RNAs(circRNAs)generated by back-splicing of precursor mRNAs(pre-mRNAs)are often aberrantly expressed in cancer cells.Accumulating evidence has revealed that circRNAs play a critical role in the progression of several cancers,including colorectal cancer(CRC).However,the current understandings of the emerging functions of circRNAs in CRC lipid metabolism and the underlying molecular mechanisms are still limited.Here,we aimed to explore the role of circCAPRIN1 in regulating CRC lipid metabolism and tumorigenesis.Methods:circRNA microarray was performed with three pairs of tumor and non-tumor tissues from CRC patients.The expression of circRNAs were determined by quantitative PCR(qPCR)and in situ hybridization(ISH).The endogenous levels of circRNAs in CRC cells were manipulated by transfection with lentiviruses overexpressing or silencing circRNAs.The regulatory roles of circRNAs in the occurrence of CRC were investigated both in vitro and in vivo using gene expression array,RNA pull-down/mass spectrometry,RNA immunoprecipitation assay,luciferase reporter assay,chromatin immunoprecipitation analysis,and fluorescence in situ hybridization(FISH).Results:Among circRNAs,circCAPRIN1 was most significantly upregulated in CRC tissue specimens.circCAPRIN1 expression was positively correlated with the clinical stage and unfavorable prognosis of CRC patients.Downregulation of circCAPRIN1 suppressed proliferation,migration,and epithelial-mesenchymal transition of CRC cells,while circCAPRIN1 overexpression had opposite effects.RNA sequencing and gene ontology analysis indicated that circCAPRIN1 upregulated the expressions of genes involved in CRC lipid metabolism.Moreover,circCAPRIN1 promoted lipid synthesis by enhancing Acetyl-CoA carboxylase 1(ACC1)expression.Further mechanistic assays demonstrated that circCAPRIN1 directly bound signal transducer and activator of transcription 2(STAT2)to activate ACC1 transcription,thus regulating lipid metabolism and facilitating CRC tumorigenesis.Conclusions:These findings revealed the oncogenic role and mechanism of circCAPRIN1 in CRC.circCAPRIN1 interacted with STAT2 to promote CRC tumor progression and lipid synthesis by enhancing the expression of ACC1.circCAPRIN1 may be considered as a novel potential diagnostic and therapeutic target for CRC patients.Yufei Yang Dakui Luo Yang Shao Zezhi Shan Qi Liu Junyong Weng Weijing He Ruoxin Zhang Qingguo Li Ziliang Wang Xinxiang Li 2023Cancer Communications2023,43,1:0

16ppGpp is present in,and functions to regulate sleep of,Drosophila显示文摘Sleep is essential for animals,and receives inputs from circadian,homeostasis,and environment,yet the mechanisms of sleep regulation remain elusive.Discovery of molecules in living systems and demonstration of their functional roles are pivotal in furthering our understanding of the molecular basis of biology.Here,we report that guanosine-50-diphos-phate,30-diphosphate(ppGpp)is present in Drosophila,and plays an important role in regulation of sleep and starvation-induced sleep loss(SISL).ppGpp is detected in germ-free Drosophila and hydrolyzed by an enzyme encoded by the mesh1 gene in Drosophila.Nighttime sleep and SISL were defected in mesh1 mutant flies,and rescued by expression of wildtype Mesh1,but not the enzymatically defective mutant Mesh1E66A.Ectopic expression of RelA,the Escherichia coli synthetase for ppGpp,phenocopied mesh1 knockout mutants,whereas overexpression of Mesh1 resulted in the opposite phenotypes,supporting that ppGpp is both necessary and sufficient in sleep regulation.A chemo connectomic screen followed by genetic intersection experiments implicates the Dilp2 neurons in the pars intercerebralis(PI)brain region as the site of ppGpp function.Our results have thus validated the presence of ppGpp in Drosophila and revealed a physiological role of ppGpp in sleep regulation for the first time.Xihuimin Dai Wei Yang Xiaohui Zhang Enxing Zhou Renbo Mao Ying Liu Tao Wang Wenxia Zhang Xinxiang Zhang Yi Rao 2023hLife2023,1,2:0

20Performance of the suspension method in large cross-section shallow-buried tunnels显示文摘Large cross-section tunnel construction induces ground surface settlements, potentially endangering both subterranean projects and nearby above-ground structures. A novel tunnel construction method, known as the suspension method,is introduced in this paper to mitigate surface settlement. The suspension method employs vertical tie rods to establish a structural connection between the initial tunnel support system and the surface steel beam, thereby exerting effective control settlements. To analyze the performance of the proposed method, systematic numerical simulations were conducted based on the practical engineering of Harbin Subway Line 3. The surface settlement and vault settlement characteristics during construction are investigated. The results show a gradual increment in both surface and vault settlement throughout the construction process, culminating in a stabilized state upon the completion of construction.In addition, compared to the double-side drift method and the Cross Diaphragm Method(CRD) method, the suspension method can obviously reduce the surface settlement and vault settlement. Moreover, the surface settlements and the axial force of tie rods were continuously monitored during the construction process at the trial tunnel block.These specific monitoring measurements are illustrated in comparison to numerical analysis results. The monitored results show great agreement with the numerical predictions, confirming the success of the project.
