| 1 | Inhibiting autophagy enhances sulforaphane-induced apoptosis via targeting NRF2 in esophageal squamous cell carcinoma显示文摘Sulforaphane(SFN), a natural anti-tumor compound from cruciferous vegetables, has been reported to induce protective autophagy to cancer cells, which might impair the anti-tumor efficiency of SFN. However, the accurate function and mechanism of SFN inducing autophagy in cancers are still obscure, especially in esophageal squamous cell carcinoma(ESCC), one of malignancies with high incidence in North China. Here, we mainly explored the potential function of autophagy upon SFN treatment in ESCC and molecular mechanism. We demonstrated that SFN could inhibit cell proliferation and induce apoptosis by activating caspase pathway. Moreover, we found activation of NRF2 pathway by SFN was responsible for the induction of autophagy and also a disadvantage element to the anti-tumor effects of SFN on ESCC, indicating that SFN might induce protective autophagy in ESCC. We, therefore,investigated effects of autophagy inhibition on sensitivity of ESCC cells to SFN and found that chloroquine(CQ) could neutralize the activation of SFN on NRF2 and enhance the activation of SFN on caspase pathway, thus improved the anti-tumor efficiency of SFN on ESCC in vitro and in vivo. | 3 | Overexpressed Cyclin D1 and CDK4 proteins are responsible for the resistance to CDK4/6 inhibitor in breast cancer that can be reversed by PI3K/mTOR inhibitors显示文摘CDK4/6 inhibitors are the standard treatment in advanced HR+/HER2-breast cancer patients.Nevertheless,the resistance to CDK4/6 inhibitors is inevitable and the strategies to overcome resistance are of great interest.Here,we show that the palbociclibresistant breast cancer cells expressed significantly higher levels of Cyclin D1 and CDK4 proteins because of upregulated protein synthesis.Silencing Cyclin D1 or CDK4 led to cell cycle arrest while silencing Cyclin E1 or CDK2 restored the sensitivity to palbociclib.Furthermore,PI3K/mTOR pathway was hyper-activated in palbociclib-resistant cells,leading to more phosphorylated 4E-BP1 and higher levels of Cyclin D1 and CDK4 translation.Targeting PI3K/mTOR pathway with a specific PI3Kαinhibitor(BYL719)or an mTOR inhibitor(everolimus)reduced the protein levels of Cyclin D1 and CDK4,and restored the sensitivity to palbociclib.The tumor samples expressed significantly higher levels of Cyclin D1,CDK4,p-AKT and p-4E-BP1 after progression on palbociclib treatment.In conclusion,our findings suggest that overexpressed Cyclin D1 and CDK4 proteins lead to the resistance to CDK4/6 inhibitor and PI3K/mTOR inhibitors are able to restore the sensitivity to CDK4/6 inhibitors,which provides the biomarker and rationale for the combinational use of CDK4/6 inhibitors and PI3K/mTOR inhibitors after CDK4/6 inhibitor resistance in breast cancer. | Zijie Cai Jingru Wang Yudong Li Qianfeng Shi Liang Jin Shunying Li Mengdi Zhu Qi Wang Lok Lam Wong Wang Yang Hongna Lai Chang Gong Yandan Yao Yujie Liu Jun Zhang Herui Yao Qiang Liu | 2023 | Science China(Life Sciences)2023,66,1: | 4 |
| 4 | Real-time 3-D space numerical shake prediction for earthquake early warning显示文摘In earthquake early warning systems, real-time shake prediction through wave propagation simulation is a promising approach. Compared with traditional methods, it does not suffer from the inaccurate estimation of source parameters. For computation efficiency, wave direction is assumed to propagate on the 2-D surface of the earth in these methods. In fact, since the seismic wave propagates in the 3-D sphere of the earth, the 2-D space modeling of wave direction results in inaccurate wave estimation. | 5 | Diurnal Circadian Lighting Accumulation Model:A Predictor of the Human Circadian Phase Shift Phenotype显示文摘Light is an important external factor that affects human circadian rhythms.This study aimed to explore the effects of different dimensions of diurnal light exposure on the physiological circadian phase shift(CPS)of the human body.A strict light exposure experiment with different timing schemes(8:00-12:00,13:00-17:00,18:00-22:00),durations(4 h,8 h)and effective circadian stimulus levels(circadian stimulus:0.35,0.55)was performed in an enclosed laboratory.Fourteen participants,including seven males and seven females,with a mean age of 24.29±2.43(mean±standard deviation),participated in this experiment and experienced all six lighting schemes.The results showed that both time factor(F_(3,40)=29.079,p<0.001,the power of the sample size=0.98)and circadian stimulus levels(T_(20)=−2.415,p=0.025,the power of sample size=0.76)sig-nificantly affect the CPS.On this basis,a diurnal circadian lighting accumulation(DCLA)-CPS model was proposed in the form of the Boltzmann function,and was validated by experimental data with high correlation(R^(2)=0.9320,RSS=0.1184),which provides strong support for rationally arranging the light level at different times of the day. | Dandan Hou Caixin Lin Yandan Lin | 2022 | Phenomics2022,2,1: | 1 |

| 7 | Decay resistance and dimensional stability improvement of wood by low melting point alloy heat treatment显示文摘Wood has no inherent natural resistance against agents of biodegradation. We evaluated the effects of metal bath heat treatment(MBHT) on decay resistance and dimensional stability of Chinese parasol(Firmiana simplex) and Chinese fir(Cunninghamia lanceolata) wood. A low melting point alloy was used as heating medium in the treatment of the wood samples at 150, 180, and 210 °C for2, 4, and 8 h. Heat-treated and control samples were exposed to brown rot fungus(Poria placenta) and white rot fungus(Coriolus versicolor) for decay resistance testing and anti-swelling efficiency(ASE). The improved decay resistance with increase temperatures with low mass losses of 7.3 and 7.3% for F. simplex, and 3.9 and 3.6% for C.lanceolata at 210 °C for 8 h against Coriolus versicolor and Poria placenta, respectively. ASE indicated that MBHT contributed to improved dimensional stability of both wood species after treatment. Scanning electron micrograph results indicate that metal bath-treated samples showed strong decay resistance. | 8 | Tin-based metal bath heat treatment: an efficient and recyclable green approach for Pinus massoniana wood modification显示文摘Pinus massoniana L. was thermally treated with low melting point alloy as heating medium to investigate the strength properties changes. Contact angle, color and scanning electron microscopy were recorded to assess the effectiveness of the treatment. Samples were pre-treated in a micro-wave for 5 min followed by metal bath heat treatment at 150, 180, and 210 °C for 2, 4, and 8 h,respectively. Strength properties of metal bath treated wood were decreased with increase temperature and time.Density, modulus of rupture, impact bending, modulus of elasticity were reduced for all treatments. Maximum compressive strength slightly increased at 150 °C for 4 h followed by gradual reduction. The Janka hardness was reduced in the tangential and radial directions. Treatment of the wood at 210 °C for 8 h caused the wood to become brittle and rupture. The contact angle was considerably higher after thermal treatment. The color of the wood became darker with increasing temperature of thermal treatment. Micrographs of the heat-treated samples showed damage to the cell wall with increase in temperature.
