3Pesticides Could Alleviate the Symptoms of Sweet Potato Virus Disease(SPVD)显示文摘In recent years,sweet potato virus disease( SPVD) has severely affected the production of sweet potato in China. In order to select chemical agents to prevent sweet potato from being infected with SPVD,14 pesticides were sprayed on the plants of sweet potato infected with SPVD,and the relative mRNA level of the viruses in the leaves and physiological indicators of sweet potato plants were detected. The results showed that after the application of most of the pesticides,the relative mRNA level of the two viruses that caused SPVD decreased,and the chlorophyll content and biomass of sweet potato infected with SPVD increased significantly or extremely significantly. Among the pesticides,BASF virus liquid,0. 1% physcion and Aolike powder for cucumber virus were proved to be the best,as the chlorophyll content and stem length of sweet potato increased significantly. 5Ionic-microenvironment stabilizes the disulfide engineered lysine decarboxylase for efficient cadaverine production显示文摘Cadaverine is the key monomer for the synthesis of nylon 5X.Efficient and alkaline stable lysine decarboxylases are highly desirable for cadaverine production as the reaction pH increasing from 6.3 to 8.5.However,the most studied lysine decarboxylase CadA(E.coli)lost almost all activity at pH 8.0,which is the foremost challenge for the industrial-cadaverine production.In this study,we first found that the Na^(+)-microenvironment significantly improved the alkaline stability of the disulfide engineered lysine decarboxylaseΔLdcEt3(P233C/L628C)(half-life 362 h),compared to the conventional buffer(half-life 0.66 h)at pH 8.0.Meanwhile,the whole-cell conversion efficiency of the industrial-grade L-lysine withΔLdcEt3 could reach up to 99%in 2 h in the fermenter.Experi-mental investigation and molecular dynamics confirmed that Na^(+)-microenvironment could improve active-aggregation state and affect secondary structure ofΔLdcEt3.Therefore,Na^(+)-microenvironment stabilizesΔLdcEt3 providing a great potential industrial application for high-level cadaverine production.Zhuang Li Yaju Xue Xiuling Ji Yuhong Huang 2023Green Chemical Engineering2023,4,2:1
6Effects of Growth Period on Quality and Tuber Traits of New Sweetpotato Cultivar Xuzishu 8显示文摘Xuzishu 8 is a new cultivated sweetpotato with high anthocyanidin content.The growth period affects its tuber traits and quality.In this study,the changes in sweetpotato shape,number of tubers per plant,rate of big and medium tubers,yield,anthocyanin content and starch content of Xuzishu 8 were investigated under 12 growth periods from 78 to 188 d.The results showed that various traits of Xuzishu 8 were significantly different between different growth periods.The rate of large and medium tubers,the fresh sweetpotato yield,the starch content and the anthocyanin yield increased gradually with the extension of the growth period.The length/width ratio of sweetpotato tubers,the rate of small tubers and the contents of reducing sugar and soluble sugar decreased with the prolongation of the growth period.The correlation analysis showed that the fresh sweetpotato yield was in significant or extremely significant positive correlation with the number of tubers per plant,rate of big and medium tubers,dry matter content,dry yield and starch content.The 148-day growth period treatment exhibited the highest number of tubers per plant,the highest fresh sweetpotato yield and anthocyanin yield reaching 3 927 and 4.25 kg/667 m^2,respectively,as well as the rate of large and medium tubers up to 90%.Therefore,the tubers had good marketability and eating quality.Hui YAN Meng KOU Wei TANG Yaju LIU Xin WANG Qiang LI Daifu MA Yungang ZHANG 2019Agricultural Biotechnology2019,8,4:0
7Transcriptome Analysis of a Wild Eggplant Germplasm M239 in Response to Verticillium dahliae Infection显示文摘In this study,wild eggplant germplasm No.M239,which is highly susceptible to Verticillium wilt,was used as the experimental material.The physiological and biochemical indices(SOD,PAL,MDA and soluble protein)of M239 roots were measured at different times(0,12,24,36,48,60 and 72 h)post inoculation with Verticillium dahliae,and the key time points for the M239 response to Verticillium wilt infection were screened.Then,RNA-Seq technology was used to screen the differentially expressed genes(DEGs)in M239 roots at 0,12 and 48 h post-inoculation(hpi).The transcriptional results of M239 were also compared with those resistance genes from some reported wild relative Solanum species(S.sisymbriifolium and S.aculeatissimum).Then some DEGs were chosen for validation by qRT–PCR.The results showed that 12 and 48 hpi were the turning points in the changes in all physiological and biochemical indices.A total of 6,783 DEGs were identified by RNA-Seq,including 6,141 DEGs(3,046 upregulated and 3,095 downregulated)at the M_12 h vs.M_0 h,1,903 DEGs(792 upregulated and 1,111 downregulated)at M_48 h vs.M_12 h,and 1,261 DEGs that appeared simultaneously in both stages.KEGG enrichment analysis showed that there were 5 metabolic pathways enriched from DEGs,which were mostly related to primary metabolism,such as glycolysis,amino acid and ribosome biogenesis.Compared with the NCBI non-redundant protein(NR)database,one Ve2 homologous gene and 8 PR protein-related genes were screened.Transcription factor analysis showed that there were a large number of DEGs,such as MYB,AP2-EREBP,bHLH,NAC and Orphans in the two stages.Compared with the reported Verticillium wilt-resistant wild eggplant species,it was found that there were fewer genes and enriched metabolic pathways in the M239 response to Verticillium wilt infection,and it also lacked the response of some known key resistance genes.These results proved that the above resistance genes and metabolic pathways played a key role in the wild eggplant response to V.dahliae infection.Liyan Wu Jie Cheng Yaju Gong Rui Bao Zhibin Li Min Gui Guanghui Du 2023Phyton-International Journal of Experimental Botany2023,92,2:0
