1Serum levels of mi RNA in patients with hepatitis B virus-associated acute-on-chronic liver failure显示文摘Background: Hepatitis B virus(HBV)-associated acute-on-chronic liver failure(HBV-ACLF) is a lifethreatening condition and its exact pathophysiology and progression remain unclear. The present study aimed to assess the role of serum mi RNAs in the evaluation of HBV-ACLF and to develop a model to predict the outcomes for ACLF.Methods: Serum was collected from 41 chronic hepatitis B and 55 HBV-ACLF patients in addition to30 chronic asymptomatic HBV carriers as controls. The mi RNAs expressions were measured by real-time quantitative PCR(q-PCR). Statistical analyses were conducted to assess the ability of differentially expressed mi RNAs and other prognostic factors in identifying ACLF prognosis and to develop a new predictive model.Results: Real-time q-PCR indicated that serum miR-146 a-5 p, mi R-122-3 p and mi R-328-3 p levels were significantly upregulated in ACLF patients compared to chronic hepatitis B and chronic asymptomatic HBV carriers patients. In addition, multivariate regression analyses indicated that Na+, INR, gastrointestinal bleeding and mi R-122-3 p are all independent factors that are reliable and sensitive to the prognosis of HBV-ACLF. Therefore, we developed a new model for the prediction of HBV-ACLF disease state: Y = 0.402 × Na+-1.72 × INR-4.963 × gastrointestinal bleeding(Yes = 0; No = 1)-0.278 ×(mi R-122-3 p) + 50.449. 2Molecular Characterization of a Leaf Senescence-Related Transcription Factor BrWRKY75 of Chinese Flowering Cabbage显示文摘WRKY is a plant-specific transcription factor(TF) involved in the regulation of many biological processes; however, its role in leaf senescence of leafy vegetables remains unknown. In the present work, a WRKY TF, termed Br WRKY75 was isolated from Chinese flowering cabbage [Brassica rapa L. ssp. chinensis(L.) Mokino var. utilis Tsen et Lee]. Analysis of deduced amino acid sequence and the phylogenetic tree showed that Br WRKY75 has high homology with WRKY75 from Brassica oleracea and Arabidopsis thaliana, and belongs to the II c sub-group. Sub-cellular localization and transcriptional activity analysis revealed that Br WRKY75 is a nuclear protein with transcriptional repression activity, and was up-regulated during leaf senescence. Electrophoretic mobility shift assay confirmed that Br WRKY75 directly bound to the W-box(TTGAC) cis-element. 3Behavioral Abnormality along with NMDAR-related CREB Suppression in Rat Hippocampus after Shortwave Exposure显示文摘Objective To estimate the detrimental effects of shortwave exposure on rat hippocampal structure and function and explore the underlying mechanisms. Methods One hundred Wistar rats were randomly divided into four groups(25 rats per group) and exposed to 27 MHz continuous shortwave at a power density of 5, 10, or 30 m W/cm^2 for 6 min once only or underwent sham exposure for the control. The spatial learning and memory, electroencephalogram(EEG), hippocampal structure and Nissl bodies were analysed. Furthermore, the expressions of N-methyl-D-aspartate receptor(NMDAR) subunits(NR1, NR2 A, and NR2 B), c AMP responsive element-binding protein(CREB) and phosphorylated CREB(p-CREB) in hippocampal tissue were analysed on 1, 7, and 14 days after exposure. Results The rats in the 10 and 30 m W/cm^2 groups had poor learning and memory, disrupted EEG oscillations, and injured hippocampal structures, including hippocampal neurons degeneration, mitochondria cavitation and blood capillaries swelling. The Nissl body content was also reduced in the exposure groups. Moreover, the hippocampal tissue in the 30 m W/cm^2 group had increased expressions of NR2 A and NR2 B and decreased levels of CREB and p-CREB. Conclusion Shortwave exposure(27 MHz, with an average power density of 10 and 30 m W/cm^2) impaired rats' spatial learning and memory and caused a series of dose-dependent pathophysiological changes. 4Tuning the selectivity of photoreduction of CO2 to syngas over Pd/layered double hydroxide nanosheets under visible light up to 600 nm显示文摘Photocatalytic reduction of CO2 with H2 O to syngas is an effective way for producing high value-added chemical feedstocks such as methanol and light olefins in industry.Nevertheless,the precise control of CO/H2 ratio from photocatalytic CO2 reduction reaction still poses a great challenge for the further application.Herein,we prepared a series of highly efficient heterostructure based on highly dispersed palladium supported on ultrathin Co Al-layered double hydroxide(LDH).In conjunction with a Ru-complex sensitizer,the molar ratios of CO/H2 can be tuned from 1:0.74 to 1:3 under visible-light irradiation(λ>400 nm).More interestingly,the syngas can be obtained under light irradiation atλ>600 nm.Structure characterization and density functional theory calculations revealed that the remarkable catalytic activity can be due to the supported palladium,which improved the charge transfer efficiency.Meanwhile,more H atoms were used to generate H2 on the supported palladium for further tunable CO/H2 ratio.This work demonstrates a new strategy for harnessing abundant solar-energy to produce syngas from a CO2 feedstock.Xian Wang Zelin Wang Ya Bai Ling Tan Yanqi Xu Xiaojie Hao Jikang Wang Abdul Hanif Mahadi Yufei Zhao Lirong Zheng Yu-Fei Song 2020Journal of Energy Chemistry2020,29,7:5
5Prediction of dissolved oxygen in a fishery pond based on gated recurrent unit (GRU)显示文摘Dissolved oxygen(DO),an important water quality indicator in aquaculture,affects the survival rate of aquatic creatures and the yield of aquatic production.Therefore,it is important to predict DO in fishery ponds for applying artificial aeration with low energy and cost.Recently,deep learning models,such as recurrent neural network(RNN),long short-term memory(LSTM),and gated recurrent unit(GRU),are often used to predict the trend of time series,but it is unclear which one of them is more suitable for prediction of DO in fishery ponds.In this work,the RNN model,LSTM model,and GRU model were used to build three DO predicting models.The performance of the three models were compared by mean absolute error(MAE),mean square error(MSE),mean absolute percentage error(MAPE),and the coefficient of determination(R2).The performance of RNN is worse result than LSTM and GRU.The four evaluation indicators of GRU are 0.450 mg/L,0.411,0.054,and 0.994,and the four indicators of LSTM are 0.407 mg/L,0.294,0.059,and 0.970,which shows that the performance of GRU is similar to LSTM,but the time cost and number of parameters used for GRU is much lower than LSTM.It is concluded that the GRU has overall better performance and can be applied to practical applications.Wuyan Li Hao Wu Nanyang Zhu Yongnian Jiang Jinglu Tan Ya Guo 2021Information Processing in Agriculture2021,8,1:4
6Validation of the Simplified Chinese-character Version of the International Physical Activity Questionnaire-Long Form in Urban Community-dwelling Adults:a Cross-sectional Study in Hangzhou,China显示文摘Objective To assess the test-retest reliability and criterion validity of the Simplified Chinese-character version of the International Physical Activity Questionnaire Long form(IPAQ-L) in urban community-dwelling adults in Hanghzou,China.Methods A total of 158 eligible participants aged 25-59 years from 6 neighbourhoods in two central districts of Hangzhou completed the IPAQ-L questionnaire twice within a 7-day interval.Half of the subjects wore pedometers during the first 7 days.Test-retest reliability was examined by comparing the first(Day 1) and the second(Day 9) survey of IPAQ-L.Criterion validity was assessed by comparing IPAQ-L with pedometer data.Results Modest to good test-retest reliability was found with intraclass correlation coefficients of 0.67 for total PA,0.37 to 0.73 for specific dimensions,and 0.56 to 0.71 for different intensities of PA.Total PA measured by IPAQ-L was moderately correlated with exercise levels(partial r = 0.27,P = 0.020) and walking distance(partial r = 0.31,P = 0.007),which were measured by a pedometer,after adjusting for gender,age,educational attainment and employment status.Conclusion Our results indicate that the IPAQ-L is a reliable and validated measure for assessing physical activity levels in this population and possibly the adult population in other mainland Chinese cities.REN Yan Jun SU Meng LIU Qing Min TAN Ya Yun DU Yu Kun LI Li Ming LYU Jun 2017Biomedical and Environmental Sciences2017,30,4:4

12Allotransplantation of adult spinal cord tissues after complete transected spinal cord injury: long-term survival and functional recovery in canines显示文摘Spinal cord injury(SCI), especially complete transected SCI, leads to loss of cells and extracellular matrix and functional impairments. In a previous study, we transplanted adult spinal cord tissues(aSCTs) to replace lost tissues and facilitate recovery in a rat SCI model. However, rodents display considerable differences from human patients in the scale, anatomy and functions of spinal cord systems, and responses after injury. Thus, use of a large animal SCI model is required to examine the repair efficiency of potential therapeutic approaches. In this study, we transplanted allogenic aSCTs from adult dogs to the lesion area of canines after complete transection of the thoracic spinal cord, and investigated the long-term cell survival and functional recovery. To enhance repair efficiency, a growth factor cocktail was added during aSCT transplantation, providing a favorable microenvironment. The results showed that transplantation of a SCTs, in particular with the addition of growth factors, significantly improves locomotor function restoration and increases the number of neurofilament-, microtubule-associated protein2-, 5-hydroxytryptamine-, choline acetyltransferase-and tyrosine hydroxylase-positive neurons in the lesion area at 6 months post-surgery. In addition, we demonstrated that donor neurons in a SCTs can survive for a long period after transplantation. 13High-performance polarization management devices based on thin-film lithium niobate显示文摘High-speed polarization management is highly desirable for many applications,such as remote sensing,telecommunication,and medical diagnosis.However,most of the approaches for polarization management rely on bulky optical components that are slow to respond,cumbersome to use,and sometimes with high drive voltages.Here,we overcome these limitations by harnessing photonic integrated circuits based on thin-film lithium niobate platform.We successfully realize a portfolio of thin-film lithium niobate devices for essential polarization management functionalities,including arbitrary polarization generation,fast polarization measurement,polarization scrambling,and automatic polarization control.The present devices feature ultra-fast control speeds,low drive voltages,low optical losses and compact footprints.Using these devices,we achieve high fidelity polarization generation with a polarization extinction ratio up to 41.9 dB and fast polarization scrambling with a scrambling rate up to 65 Mrad s−1,both of which are best results in integrated optics.We also demonstrate the endless polarization state tracking operation in our devices.The demonstrated devices unlock a drastically new level of performance and scales in polarization management devices,leading to a paradigm shift in polarization management.Zhongjin Lin Yanmei Lin Hao Li Mengyue Xu Mingbo He Wei Ke Heyun Tan Ya Han Zhaohui Li Dawei Wang X.Steve Yao Songnian Fu Siyuan Yu Xinlun Cai 2022Light(Science & Applications)2022,11,5:3
14T cells expressing CD5/CD7 bispecific chimeric antigen receptors with fully human heavy-chain-only domains mitigate tumor antigen escape显示文摘Bispecific chimeric antigen receptor T-cell(CAR-T)therapies have shown promising results in clinical trials for advanced B-cell malignancies.However,it is challenging to broaden the success of bispecific CAR-T therapies to treat refractory/relapse(r/r)T-cell leukemia/lymphoma because targeting multiple T-cell-expressing antigens leads to exacerbated CAR-T cell fratricide and potential safety concerns.Fully human heavy chain variable(FHVH)antibodies that specifically target CD5 or CD7 were screened and constructed to CD5/CD7 bispecific CARs.A truncated Epidermal growth factor receptor were integrated into CAR constructs to address safety concerns.To tackle the fratricidal issue of CAR-T cells targeting T-cell-pan marker(s),CRISPR/Cas9-based CD5 and CD7 genes knockout were performed before lentiviral transduction of bispecific CARs.Functional comparison between different bispecific CAR structures:tandem CARs and dual CAR were performed in vitro and in vivo to determine the optimal construct suitable for addressing T-cell malignancy antigen escape in clinical setting.Knockout of CD5 and CD7 prevents fratricide of CD5/CD7 bispecific CAR-T cells,and FHVH-derived CD5/CD7 bispecific CAR-T cells demonstrate potent antitumor activity in vitro and in vivo.The fratricide-resistant FHVH-derived CD5/CD7 bispecific CAR-T cells have potent antitumor activity against T-cell malignancies,and tandem CARs are more effective than dual CAR in preventing tumor escape in heterogeneous leukemic cells.The meaningful clinical efficacy and safety of tandem CD5/CD7 CAR-T cells deserve to be explored urgently.Zhenyu Dai Wei Mu Ya Zhao Jiali Cheng Haolong Lin Kedong Ouyang Xiangyin Jia Jianwei Liu Qiaoe Wei Meng Wang Chaohong Liu Taochao Tan Jianfeng Zhou 2022Signal Transduction and Targeted Therapy2022,7,4:3
15MRI and Transvaginal Ultrasound Findings of Atypical Polypoid Adenomyoma: A Case Report显示文摘Atypical polypoid adenomyoma(APA)is an uncommon type of polypoid characterized by fibroid stroma and endometrial glands.It occurs mostly in premenopausal women and rarely in postmenopausal women with irregular vaginal bleeding.In our current case,a 76-year-old woman presented with irregular vaginal bleeding.The final pathological diagnosis of the mass was APA.APA is not easy to diagnose before surgery.On the one hand,there was no obvious particularity in imaging features and clinical features,especially for uncomfortably identifying endometrial cancer.On the other hand,APA has a pedicle,attaching to any part of the uterine cavity,which can cause pseudocoel between the mass with the uterine cavity wall.So,when it comes to getting the pathological tissue in the absence of hysteroscopy,it is easy to access to the pseudocoel and obtain endometrial tissue rather than the pathological tissue of the mass.Therefore,preoperative imaging examination is of great significance diagnosis way of thinking to clinicians for APA.In the meantime,pathological tissue of APA can be obtained by hysteroscopy in visual conditions.Ya Tan Xue Hu Xin Song Wenjun Zhang 2022Chinese Medical Sciences Journal2022,37,1:3
16A new gain-of-function OsGS2/GRF4 allele generated by CRISPR/Cas9 genome editing increases rice grain size and yield显示文摘Grain size is one of the most important factors affecting rice grain quality and yield,and attracts great attention from molecular biologists and breeders.In this study,we engineered a CRISPR/Cas9 system targeting the miR396 recognition site of the rice GS2 gene,which encodes growth-regulating factor 4(OsGRF4)and regulates multiple agronomic traits including grain size,grain quality,nitrogen use efficiency,abiotic stress response,and seed shattering.In contrast to most previous genome editing efforts in which indel mutations were chosen to obtain null mutants,a mutant named GS2^(E) carrying an in-frame 6-bp deletion and 1-bp substitution within the miR396-targeted sequence was identified.GS2^(E) plants showed increased expression of GS2 in consistent with impaired repression by miR396.As expected,the gain-of-function GS2^(E) mutant exhibited multiple beneficial traits including increased grain size and yield and bigger grain length/width ratio.Thousand grain weight and grain yield per plant of GS2^(E) plants were increased by 23.5%and 10.4%,respectively.These improved traits were passed to hybrids in a semidominant way,suggesting that the new GS2^(E) allele has great potential in rice improvement.Taken together,we report new GS2 germplasm and describe a novel gene-editing strategy that can be widely employed to improve grain size and yield in rice.This trait-improvement strategy could be applied to other genes containing miRNA target sites,in particular the conserved miR396-GRF/GIF module that governs plant growth,development and environmental response.Wenshu Wang Weipeng Wang Yanlin Pan Chao Tan Hongjing Li Ya Chen Xingdan Liu Jing Wei Nian Xu Yu Han Han Gu Rongjian Ye Qi Ding Chonglie Ma 2022The Crop Journal2022,10,4:3
17Offspring of rats with cerebral hypoxia-ischemia manifest cognitive dysfunction in learning and memory abilities显示文摘Neonatal hypoxic-ischemic encephalopathy is a serious neurological disease,often resulting in long-term neurodevelopmental disorders among surviving children.However,whether these neurodevelopmental issues can be passed to offspring remains unclear.The right common carotid artery of 7-day-old parental-generation rats was subjected to permanent ligation using a vessel electrocoagulator.Neonatal hypoxic-ischemic rat models were established by subjecting the rats to 8%O2–92%N2 for 2 hours.The results showed that 24 hours after hypoxia and ischemia,pathological damage,cerebral atrophy,liquefaction,and impairment were found,and Zea-Longa scores were significantly increased.The parental-generation rats were propagated at 3 months old,and offspring were obtained.No changes in the overall brain structures of these offspring rats were identified by magnetic resonance imaging.However,the escape latency was longer and the number of platform crossings was reduced among these offspring compared with normal rats.These results indicated that the offspring of hypoxic-ischemic encephalopathy model rats displayed cognitive impairments in learning and memory.This study was approved by the Animal Care&Welfare Committee of Kunming Medical University,China in 2018(approval No.kmmu2019072).Lu-Lu Xue Fang Wang Rui-Ze Niu Ya-Xin Tan Jia Liu Yuan Jin Zheng Ma Zi-Bin Zhang Ya Jiang Li Chen Qing-Jie Xia Jun-Jie Chen Ting-Hua Wang Liu-Lin Xiong 2020Neural Regeneration Research2020,15,9:3

Light hydrocarbons, including methane, ethane, ethylene, acetylene, propane etc, were found to be released continuously from the oil into headspace within a closed vial placed in an ultrasonic field.
