| 1 | In vitro assessment of thyroid hormone receptor activity of four organophosphate esters显示文摘Previous animal experiments have implied that organophosphate esters(OPEs) have a disruption effect on the thyroid endocrine system. However, knowledge of the toxicological mechanism remains limited. In this study, the activities of four OPEs have been characterized against the thyroid hormone(TH) nuclear receptor(TR) using two in vitro models, with the aim of evaluating their toxicity mechanisms towards the TR. The results of a TH-dependent cell proliferation assay showed that tris(2-chloro-1-(chloromethyl)ethyl)phosphate(TDCPP) could induce cell growth, while the other three OPEs had no effect. The results of a luciferase reporter gene assay revealed that all four of the OPEs tested in the current study showed agonistic activity towards TRβ, with TDCPP being the most potent one. Moreover, molecular docking revealed that all the tested OPEs could fit into the ligand binding pocket of TRβ, with TDCPP binding more effectively than the other three OPEs. | 2 | Combination of a biopharmaceutic classification system and physiologically based pharmacokinetic models to predict absorption properties of baicalein in vitro and in vivo显示文摘Objective: To determine the in vitro and in vivo absorption properties of active ingredients of the Chinese medicine, baicalein, to enrich mechanistic understanding of oral drug absorption.Methods: The Biopharmaceutic Classification System(BCS) category was determined using equilibrium solubility, intrinsic dissolution rate, and intestinal permeability to evaluate intestinal absorption mechanisms of baicalein in rats in vitro. Physiologically based pharmacokinetic(PBPK) model commercial software GastroPlus~(TM) was used to predict oral absorption of baicalein in vivo.Results: Based on equilibrium solubility, intrinsic dissolution rate, and permeability values of main absorptive segments in the duodenum, jejunum, and ileum, baicalein was classified as a drug with low solubility and high permeability. Intestinal perfusion with venous sampling(IPVS) revealed that baicalein was extensively metabolized in the body, which corresponded to the low bioavailability predicted by the PBPK model. Further, the PBPK model predicted the key indicators of BCS, leading to reclassification as BCS-II. Predicted values of peak plasma concentration of the drug(C_(max)) and area under the curve(AUC)fell within two times of the error of the measured results, highlighting the superior prediction of absorption of baicalein in rats, beagles, and humans. The PBPK model supported in vitro and in vivo evidence and provided excellent prediction for this BCS class II drug.Conclusion: BCS and PBPK are complementary methods that enable comprehensive research of BCS parameters, intestinal absorption rate, metabolism, prediction of human absorption fraction and bioavailability, simulation of PK, and drug absorption in various intestinal segments across species. | 5 | Potential Impacts of Exogenous Nitrogen Enrichment on Distribution and Transfer of Nitrogen in Plant-Soil System of Suaeda salsa Marsh in the Yellow River Estuary, China显示文摘To determine the potential impacts of exogenous nitrogen(N)enrichment on distribution and transfer of N in Suaeda salsa marsh in the Yellow River Estuary,the variations of N in plant-soil system during the growing season were investigated by field N addition experiment.The experiment included four treatments:NN(no N input treatment,0gNm^(−2) yr^(−1)),LN(low N input treatment,3.0 gNm^(−2) yr^(−1)),MN(medium N input treatment,6 gNm^(−2) yr^(−1))and HN(high N input treatment,12 gNm^(−2) yr^(−1)).Results showed that N additions generally increased the contents of total nitrogen(TN),ammonia nitrogen(NH_(4)^(+)-N)and nitrate nitrogen(NO_(3)^(−)-N)in different soil layers and the increasing trend was particularly evident in topsoil.Compared with the NN treatment,the average contents of TN in topsoil in the LN,MN and HN treatments during the growing season increased by 10.85%,30.14%and 43.98%,the mean contents of NH_(4)^(+)-N increased by 8.56%,6.96%and 14.34%,and the average contents of NO_(3)^(−)-N increased by 35.73%,45.99%and 46.66%,respectively.Although exogenous N import did not alter the temporal variation patterns of TN contents in organs,the N transfer and accumulation differed among tissues in different treatments.With increasing N import,both the N stocks in soil and plant showed increasing trend and the values in N addition treatments increased by 9.43%–38.22%and 13.40%–62.20%,respectively.It was worth noting that,compared with other treatments,the S.salsa in the MN treatments was very likely to have special response to N enrichment since not only the period of peak growth was prolonged by about 20 days but also the maximum of TN content in leaves was advanced by approximately one month.This paper found that,as N loading reached MN level in future,the growth rhythm of S.salsa and the accumulation and transference of N in its tissues would be altered significantly,which might generate great impact on the stability and health of S.salsa marsh ecosystem. | HU Xingyun SUN Zhigao YU Linying CHEN Bingbing | 2023 | Journal of Ocean University of China2023,22,1: | 0 |

| 10 | The crucial role of deep-sourced methane in maintaining the subseafloor sulfate budget显示文摘Methane(CH4)is a powerful greenhouse gas and its largest reservoir on Earth is held in marine sediments.CH4 in marine sediments is mainly stored in gas-hydrate reservoirs and deep sedimentary strata along continental margins,where large amounts of deep-sourced CH4 ascend to different degrees toward the seafloor.However,the amount of deep-sourced CH4 and its role in subseafloor carbon and sulfur cycling remains poorly constrained.We analyzed sulfate(SO_(4)^(2-))profiles of 157 sites along with previous published 85 sites to determine the regional distribution and amount of SO_(4)^(2-) reduction for an area of 1.23×10^(5) km^(2) of the northern South China Sea.Then we compared these obtained results with estimates based on sedimentation rates from the same area.Significantly higher regional SO_(4)^(2-) flux estimates based on SO_(4)^(2-) profiles(4.26×10^(-3)Tmol a^(-1)),compared to lower estimates based on sedimentation rates(1.23×10^(-3)Tmol a^(-1)),reflect abundant ascending deep-sourced CH4.The difference of the regional SO_(4)^(2-) flux estimates(3.03×10^(-3)Tmol a^(-1))represents the amount of SO_(4)^(2-) reduced by CH_(4) through the anaerobic oxidation of CH_(4)(AOM).Deep-sourced CH_(4) contributes 71%to total SO_(4)^(2-) consumption in the study area,largely exceeding SO_(4)^(2-) consumption by organoclastic sulfate reduction.Our findings substantiate that deep-sourced CH4 governs subseafloor carbon and sulfur cycling to a previously underrated extent,fueling extensive chemosynthesis-based ecosystems along continental slope and rise. | Yu Hu Dong Feng Jörn Peckmann Xinxin Zhang Linying Chen Junxi Feng Hongbin Wang Duofu Chen | 2023 | Geoscience Frontiers2023,14,3: | 0 |
| 11 | Network pharmacology and subsequent experimental validation reveal the synergistic myocardial protection mechanism of Salvia miltiorrhiza Bge.and Carthamus tinctorius L.显示文摘Objective:To reveal the molecular mechanism underlying the compatibility of Salvia miltiorrhiza Bge(S.miltiorrhiza,Dan Shen)and C.tinctorius L.(C.tinctorius,Hong Hua)as an herb pair through network pharmacology and subsequent experimental validation.Methods:Network pharmacology was applied to construct an active ingredient-efficacy target-disease protein network to reveal the unique regulation pattern of s.miltiorrhiza and C.tinctorius as herb pair.Molecular docking was used to verify the binding of the components of these herbs and their potential targets.An H9c2 glucose hypoxia model was used to evaluate the efficacy of the components and their synergistic effects,which were evaluated using the combination index.Western blot was performed to detect the protein expression of these targets.Results:Network pharmacology analysis revealed 5 pathways and 8 core targets of s.miltiorrhiza and C.tinctorius in myocardial protection.Five of the core targets were enriched in the hypoxia-inducible factor-1(HIF-1)signaling pathway.S.miltiorrhiza-C.tinctorius achieved vascular tone mainly by regulating the target genes of the HIF-1 pathway.As an upstream gene of the HIF-1 pathway,STAT3 can be activated by the active ingredients cryptotanshinone(Ctan),salvianolic acid B(Sal.B),and myricetin(Myric).Cell experiments revealed that Myric,Sal.B,and Ctan also exhibited synergistic myocardial protective activity.Molecular docking verified the strong binding of Myric,Sal.B,and Ctan to STAT3.Western blot further showed that the active ingredients synergistically upregulated the protein expressionof STAT3.Conclusion:The pharmacodynamic transmission analysis revealed that the active ingredients of S.miltiorrhiza and C.tinctorius can synergistically resist ischemia through various targets and pathways.This study provides a methodological reference for interpreting traditional Chinese medicine compatibility. | Linying Zhong Ling Dong Jing Sun Jie Yang Zhiying Yu Ping He Bo Zhu Yuxin Zhu Siyuan Li Wenjuan Xu | 2024 | Journal of Traditional Chinese Medical Sciences2024,11,1: | 0 |
