1Ulva prolifera green-tide outbreaks and their environmental impact in the Yellow Sea, China显示文摘The Ulva prolifera green tides in the Yellow Sea, China, which have been occurring since 2007, are a serious environmental problem attracting worldwide attention. Despite extensive research, the outbreak mechanisms have not been fully understood. Comprehensive analysis of anthropogenic and natural biotic and abiotic factors reveals that human activities, regional physicochemical conditions and algal physiological characteristics as well as ocean warming and biological interactions(with microorganism or other macroalgae) are closely related to the occurrence of green tides. Dynamics of these factors and their interactions could explain why green tides suddenly occurred in 2007 and decreased abruptly in 2017.Moreover, the consequence of green tides is serious. The decay of macroalgal biomass could result in hypoxia and acidification, possibly induce red tide and even have a long-lasting impact on coastal carbon cycles and the ecosystem. 2Response of phytoplankton to multispecies mariculture:a case study on the carrying capacity of shellfish in the Sanggou Bay in China显示文摘A multispecies model for shellfish polyculture in the Sanggou Bay in China used for large-scale long-line cultivation of the Chinese scallop Chlamys farreri,the Pacific oyster Crassostrea gigas and the kelp Laminaria japonica is presented. The model includes key physical processes which are the transports of matter at the system boundary,and the main biological process that is the primary production and nutrients release from the bottom. By the model,the seasonal fluctuations of phytoplankton biomass and dissolved inorganic nitrogen(DIN) in 1994 are simulated. Furthermore,if the kelp culture scale is kept constant and the Chinese scallop and the Pacific oyster culture scales are adjusted,virtual shellfish farms are funded and responses of phytoplankton to the large-scale shellfish culture are simulated. According to these simulated results,the room limitation,and the hypothesis that shellfish will not grow well if the phytoplankton biomass is less than 8.2 mg/m3,the expandable multiple of scallop culture k and that of oyster culture y are determined as k=-0.276 5y+4.690 5 and 0.133 3k+0.006 6y≤0.667 5,where,k (or y) is equal to 1,the culture scale of scallop (or oyster) is 8.8×109 individuals (or 66 ha,with a density of 59 ind./m2),and the kelp culture scale is 3 300 ha with a density of 12 ind./m2.GE Changzi FANG Jianguang SONG Xiefa MAO Yuze YE Naihao 2008Acta Oceanologica Sinica2008,27,1:11
3Biodiversity-based development and evolution:the emerging research systems in model and non-model organisms显示文摘Evolutionary developmental biology,or Evo-Devo for short,has become an established field that,broadly speaking,seeks to understand how changes in development drive major transitions and innovation in organismal evolution.It does so via integrating the principles and methods of many subdisciplines of biology.Although we have gained unprecedented knowledge from the studies on model organisms in the past decades,many fundamental and crucially essential processes remain a mystery.Considering the tremendous biodiversity of our planet,the current model organisms seem insufficient for us to understand the evolutionary and physiological processes of life and its adaptation to exterior environments.The currently increasing genomic data and the recently available gene-editing tools make it possible to extend our studies to non-model organisms.In this review,we review the recent work on the regulatory signaling of developmental and regeneration processes,environmental adaptation,and evolutionary mechanisms using both the existing model animals such as zebrafish and Drosophila,and the emerging nonstandard model organisms including amphioxus,ascidian,ciliates,single-celled phytoplankton,and marine nematode.In addition,the challenging questions and new directions in these systems are outlined as well.Long Zhao Feng Gao Shan Gao Yujun Liang Hongan Long Zhiyi Lv Ying Su Naihao Ye Liusuo Zhang Chengtian Zhao Xiaoyu Wang Weibo Song Shicui Zhang Bo Dong 2021Science China(Life Sciences)2021,64,8:9
4Activities of principal photosynthetic enzymes in green macroalga Ulva linza:functional implication of C_4 pathway in CO_2 assimilation显示文摘The green-tide-forming macroalga Ulva linza was profiled by transcriptome sequencing to ascertain whether the alga carries both C3 and C4 photosynthesis genes.The key enzymes involved in C4 metabolism including pyruvate orthophosphate dikinase(PPDK),phosphoenolpyruvate carboxylase(PEPC),and phosphoenolpyruvate carboxykinase(PCK) were found.When measured under normal and different stress conditions,expression of rbcL was higher under normal conditions and lower under the adverse conditions,whereas that of PPDK was higher under some adverse conditions,namely desiccation,high salinity,and low salinity.Both ribulose-1,5-biphosphate carboxylase(RuBPCase) and PPDK were found to play a role in carbon fixation,with significantly higher PPDK activity across the stress conditions.These results suggest that elevated PPDK activity alters carbon metabolism in U.linza leading to partial operation of the C4 carbon metabolism,a pathway that,under stress conditions,probably contributes to the hardy character of U.linza and thus to its wide distribution.XU JianFang ZHANG XiaoWen YE NaiHao ZHENG Zhou MOU ShanLi DONG MeiTao XU Dong MIAO JinLai 2013Science China(Life Sciences)2013,56,6:2
5Cloning and sequence analysis of the partial sequence of the rbcL from Bryopsis hypnoides显示文摘The partial sequence of the rbcL from Bryopsis hypnoides, including the sequences of the upstream, extron and partial intron, was amplified by PCR and their sequences were determined. With Spinacia oleracea as the outgroup, neighbor-joining method and maximum parsimony method were used respectively to build phylogenetic trees according to the rbcL exon sequence among 13 species that were the typical species of six phyla. Two kinds of trees showed clearly that there were two groups among those species, the green lineage and the non-green lineage. And the relationships of algae in the green lineage were similar in the two trees but those in the non-green lineage were not consistent. 7Gene duplication and functional divergence of new genes contributed to the polar acclimation of Antarctic green algae显示文摘Psychrophilic microalgae successfully survive in the extreme and highly variable polar ecosystems,which represent the energy base of most food webs and play a fundamental role in nutrient cycling.The success of microalgae is rooted in their adaptive evolution.Revealing how they have evolved to thrive in extreme polar environments will help us better understand the origin of life in polar ecosystems.We isolated a psychrophilic unicellular green alga,Microglena sp.YARC,from Antarctic sea ice which has a huge genome.Therefore,we predicted that gene replication may play an important role in its polar adaptive evolution.We found that its protein-coding gene number significantly increased and the duplication time was dated between 37 and 48 million years ago,which is consistent with the formation of the circumpolar Southern Ocean.Most duplicated paralogous genes were enriched in pathways related to photosynthesis,DNA repair,and fatty acid metabolism.Moreover,there were a total of 657 Microglena-specific families,including collagen-like proteins.The divergence in the expression patterns of the duplicated and species-specific genes reflects sub-and neo-functionalization during stress acclimation.Overall,key findings from this study provide new information on how gene duplication and their functional novelty contributed to polar algae adaptation to the highly variable polar environmental conditions.Xiaowen Zhang Wentao Han Xiao Fan Yitao Wang Dong Xu Ke Sun Wei Wang Yan Zhang Jian Ma Naihao Ye 2023Marine Life Science & Technology2023,5,4:1

11Biomass,nutrient uptake and fatty acid composition of Chlamydomonas sp.ICE-L in response to different nitrogen sources显示文摘Nitrogen removal from media by microalgae provides the potential benefit of producing lipids for biodiesel and biomass. However, research is limited on algal growth and biomass under different nitrogen sources and provides little insight in terms of biofuel production. We studied the influences of nitrogen sources on cell growth and lipid accumulation of Chlamydomonas sp. ICE-L, one of a promising oil rich micro algal species. Chlamydomonas sp.ICE-L grown in NH_4 Cl medium had maximum growth rate. While the highest dry biomass of 0.28 g/L was obtained in media containing NH_4NO_3, the highest lipid content of 0.21 g/g was achieved under nitrogendeficiency condition with a dry biomass of 0.24 g/L. 12Genetic variability of half-smooth tongue sole Cynoglossus semilaevis populations using microsatellite markers显示文摘Ten highly variable microsatellite loci were performed in order to evaluate the genetic variability of wild and hatchery samples of half-smooth tongue sole Cynoglossus semilaevis.A group of 200 genotypes belonging to four wild samples,Laizhou (LZ),Weihai (WH),Qingdao (QD),Rizhao (RZ) and one hatchery sample,Mingbo (MB),were screened.All of the ten microsatellite loci screened in this study showed marked polymorphism.A total of 70 different alleles were observed over all loci.The number of alleles per locus ranged from 4.2 to 10.6.The average of expected and observed heterozygosity ranged from 0.67 to 0.82, and from 0.77 to 0.87,respectively.A total of 10,16,10,11 and 5 unique alleles each were found in LZ,WH,QD,RZ and MB populations.The effective number of alleles varied from 2.87 for HSTS_7 to 6.83 for HSTS_h. The number of average genotypes ranged from 6.0 for HSTS_a to 15.4 for HSTS_h. 13Identification of genes under positive selection reveals evolutionary adaptation of Ulva mutabilis显示文摘Ulvophytes are attractive model systems for understanding the evolution of growth,development,and environmental stress responses.They are untapped resources for food,fuel,and high-value compounds.The rapid and abundant growth of Ulva species makes them key contributors to coastal biogeochemical cycles,which can cause significant environmental problems in the form of green tides and biofouling.Until now,the Ulva mutabilis genome is the only Ulva genome to have been sequenced.To obtain further insights into the evolutionary forces driving divergence in Ulva species,we analyzed 3905 single copy ortholog family from U.mutabilis,Chlamydomonas reinhardtii and Volvox carteri to identify genes under positive selection(GUPS)in U.mutabilis.We detected 63 orthologs in U.mutabilis that were considered to be under positive selection.Functional analyses revealed that several adaptive modifications in photosynthesis,amino acid and protein synthesis,signal transduction and stress-related processes might explain why this alga has evolved the ability to grow very rapidly and cope with the variable coastal ecosystem environments.Jian Zhang Xiaowen Zhang Wentao Han Xiao Fan Yitao Wang Dong Xu Yan Zhang Jian Ma Chengwei Liang Naihao Ye 2020Acta Oceanologica Sinica2020,39,10:0
14Characterization of a broad substrates specificity acyl-CoA:diacylglycerol acyltransferase 1 from the green tide alga Ulva prolifera显示文摘Triacylglycerols(triglycerides,TAGs)are the major carbon and energy storage forms in various organisms,and important components of cellular membranes and signaling molecules;they have essential functions in multiple physiological processes and stress regulation.Acyl-CoA:diacylglycerol acyltransferase(DGAT)catalyzes the final and only committed acylation step in the synthesis of TAGs in eukaryotes.The present work identified and isolated a novel gene,UpDGAT1,from the green tide alga Ulva prolifera.The activity of UpDGAT1 was confirmed by heterologous expression in a Saccharomyces cerevisiae TAG-deficient quadruple mutant.Results of thin-layer chromatography and BODIPY staining indicated that UpDGAT1 was able to restore TAG synthesis and lipid body formation in the yeast.Lipid analysis of yeast cells revealed that UpDGAT1 showed broad substrate specificity,accepting saturated as well as mono-and polyunsaturated acyl-CoAs as substrates.High salinity and high temperature stresses increased UpDGAT1 expression and TAG accumulation in U.prolifera.The present study provides clues to the functions of UpDGAT1 in TAG accumulation in,and stress adaptation of,U.prolifera.Xiaowen Zhang Xiaoyuan Chi Yitao Wang Jian Zhang Yan Zhang Dong Xu Xiao Fan Chengwei Liang Naihao Ye 2020Acta Oceanologica Sinica2020,39,10:0
15Glutathione S-Transferase(GST)Identified from Giant Kelp Macrocystis pyrifera Increases the Copper Tolerance of Synechococcus elongatus PCC 7942显示文摘The glutathione S-transferases gene family plays an important regulatory role in growth and development,and responses to environmental change.In this study,six complete GST genes(Mp GST1,Mp GST2,Mp GST3,MpGST4,Mp GST5,and Mp GST6)were cloned from the gametophytes of brown alga Macrocystis pyrifera.Subsequent bioinformatics analysis showed that these six genes encoded proteins with 202,216,288,201,205,and 201 aa,respectively.Moreover,Mp GST3 differs from the other GST genes.Phylogenetic analysis suggested that MpGST3 belongs to the Ure2p type GST.Domain analysis suggested that the other GSTs from M.pyrifera belong to the soluble GST family and form an independent branch with the GSTs found in the other macroalgae,suggesting that a new GST type was formed during macroalgal evolution.GST genes were upregulated in M.pyrifera when 2.5 mg L^(-1)Cu ions were added to the medium.Six GST genes were integrated into the genome of Synechococcus elongatus PCC 7942,and their functions were verified by measuring light absorbance,photosynthetic pigment content,and photosynthetic parameters of the transformed strains under 0.3 mg L^(-1)Cu ion stress.The results showed much higher levels of various parameters in the transformed strains than in the wild strain.The transformed strains(with the MpGST genes)showed significantly enhanced resistance to Cu ion stress,while the wild strain almost died.The results of this study lay a theoretical foundation for further research on the Cu ion stress resistance function of GSTs in M.pyrifera.GU Zipeng REN Yudong LIANG Chengwei ZHANG Xiaowen GENG Yilin XU Dong YE Naihao 2023Journal of Ocean University of China2023,22,3:0
