| 1 | Circular HDAC9/microRNA-138/Sirtuin-1 Pathway Mediates Synaptic and Amyloid Precursor Protein Processing Deficits in Alzheimer’s Disease显示文摘Synaptic dysfunction and abnormal processing of amyloid precursor protein(APP) are early pathological features in Alzheimer’s disease(AD). Recently, noncoding RNAs such as micro RNAs(mi RNAs) and circular RNAs(circ RNAs) have been reported to contribute to the pathogenesis of AD. We found an age-dependent elevation of mi R-138 in APP/PS1(presenilin-1) mice. Mi R-138 inhibited the expression of ADAM10 [a disintegrin and metalloproteinase domain-containing protein 10], promoted amyloid beta(Ab) production, and induced synaptic and learning/memory deficits in APP/PS1 mice, while its suppression alleviated the AD-like phenotype in these mice. Overexpression of sirtuin 1(Sirt1), a target of mi R-138, ameliorated the mi R-138-induced inhibition of ADAM10 and elevation of Ab in vitro. The circ RNA HDAC9(circ HDAC9) was predicted to contain a mi R-138 binding site in several databases. Its expression was inversely correlated with mi R-138 in both Ab-oligomertreated N2 a cells and APP/PS1 mice, and it co-localized with mi R-138 in the cytoplasm of N2 a cells. Circ HDAC9 acted as a mi R-138 sponge, decreasing mi R-138 expression, and reversing the Sirt1 suppression and excessive Ab production induced by mi R-138 in vitro. | 3 | The cotton WRKY transcription factor GhWRKY70 negatively regulates the defense response against Verticillium dahliae显示文摘WRKY transcription factors (TFs) play important roles in the regulation of biotic and abiotic stresses. However, the functions of most WRKY TFs in upland cotton (Gossypium hirsutum) are still unknown. In this study, we functionally identified a group Ⅲ WRKY transcription factor, GhWRKY70, in upland cotton. Reverse transcription-quantitative PCR analysis showed that GhWRKY70 expression was induced by Verticillium dahliae, salicylic acid (SA) and methyl jasmonate. Virus-induced gene silencing of GhWRKY70 increased the resistance of cotton to V. dahliae. Specifically, jasmonic acid (JA) response-associated genes were upregulated and SA-related genes were downregulated in GhWRKY70-silenced cotton plants. Overexpression of GhWRKY70 reduced tolerance to V. dahliae in Arabidopsis thaliana. Transgenic Arabidopsis plants showed increased expression of SA-associated genes and reduced expression of JA response-associated genes. | 4 | Effect of heat treatment on microstructures and properties of a novel Al-Zn-Mg-Cu alloy for oil drilling显示文摘In view of the special requirements for strength, heat resistance and corrosion resistance of Al-Zn-Mg-Cu alloy for oil drilling, the Al-6.2 Zn-2.5 Mg-1.6 Cu alloy was prepared by increasing Cu content on basis of Russian Series 1953 alloy. The effect of heat treatment on the microstructures and properties of the alloy was characterized by optical microscope(OM), scanning electron microscope(SEM) and transmission electron microscope(TEM), and investigated by tensile test at room temperature, thermal exposure test and corrosion test. The results show that the strength after T6 aging treatment exhibit a decrease trend as an increase of the solution temperature from465 ℃ to 480 ℃. After the solution treated by the rate of 470 ℃/1 h, second phases dissolve into the matrix very well and the strength property reaches optimum. The alloy has better comprehensive properties treated by a solution treatment of 470 ℃/1 h and then followed by an aging treatment of 120 ℃/24 h + 170 ℃/1 h + 120 ℃/24 h. Under the aging state, the precipitated phases inside the grains are suitable in size, while on the grain boundary distribute discontinuously and the precipitate-free zone is obvious. Besides, the alloy still maintain high tensile properties. The yield strength, tensile strength and elongation are 650 MPa, 686 MPa,12.0%, respectively. The yield strength retention after heat exposure is 92%. The alloy has good corrosion resistance and the exfoliation corrosion degree. The average corrosion rate in the H2S and CO2 environment is 0.0024 mm/a, which is far less than the required 0.12 mm/a. | 19 | Branched core-shell a-TiO_(2)@N-TiO_(2) nanospheres with gradient-doped N for highly efficient photocatalytic applications显示文摘A branched core-shell nanosphere composed of an anatase TiO_(2)(a-TiO_(2)) core and a TiO_(2)nanobranch shell with gradient-doped N(a-TiO_(2)@N-TiO_(2)) is synthesized by a simple in situ doping method, in which mixed crystal anatase-rutile TiO_(2)(ar-TiO_(2)) nanosphere is first prepared by oxidizing Ti using H_(2)O_(2), and then is etched by NH_(3)·H_(2)O to form(NH_(4))2TiO_(3)nanobranches, which is converted into a-TiO_(2)@N-TiO_(2)following an ambient annealing process. The diameter of a-TiO_(2)core is ~500 nm, and the thickness of NTiO_(2)branched shell is ~100 nm with gradually increased N concentration from the bottom to the edge.Ultra-thin amorphous coating layers on the branches are also observed. The morphology of the composites could be further tuned by the amount of NH_(3)·H_(2)O, and its effect on the photocatalytic performance is also investigated. The optimized a-TiO_(2)@N-TiO_(2)shows an outstanding hydrogen evolution rate of 308.1 μmol g^(-1)h^(-1)under air mass(AM) 1.5 illumination, and also exhibits highly active in photocatalytic degradation of various refractory organic pollutants, including organic dyes, phenols, antibiotics,and personal care products, with removal ratios higher than 96% after 2 h operation.
