2Macrophage-mediated tumor-targeted delivery of engineered Salmonella typhimurium VNP20009 in anti-PD1 therapy against melanoma显示文摘Bacterial antitumor therapy has great application potential given its unique characteristics,including genetic manipulation, tumor targeting specificity and immune system modulation. However,the nonnegligible side effects and limited efficacy of clinical treatment limit their biomedical applications. Engineered bacteria for therapeutic applications ideally need to avoid their accumulation in normal organs and possess potent antitumor activity. Here, we show that macrophage-mediated tumor-targeted delivery of Salmonella typhimurium VNP20009 can effectively reduce the toxicity caused by administrating VNP20009 alone in a melanoma mouse model. This benefits from tumor-induced chemotaxis for macrophages combined with their slow release of loaded strains. Inspired by changes in the tumor microenvironment, including a decrease in intratumoral dysfunctional CD8+T cells and an increase in PDL1 on the tumor cell surface, macrophages were loaded with the engineered strain VNP-PD1nb, which can express and secrete anti-PD1 nanoantibodies after they are released from macrophages. This novel triple-combined immunotherapy significantly inhibited melanoma tumors by reactivating the tumor microenvironment by increasing immune cell infiltration, inhibiting tumor cell proliferation, remodeling TAMs to an M1-like phenotype and prominently activating CD8+T cells. 3Metal-organic framework membranes: From synthesis to electrocatalytic applications显示文摘Metal-orga nic frameworks(MOFs),as an emerging family of porous inorga nic-organic crystal materials,exhibit widely applications in gas storage and separation,drug release,sensing,and catalysis,owing to easily adjustable pore sizes,uniformly distributed metal centers,high surface areas,and tunable functionalities.However,MOF crystal powders are usually difficult to be directly applied into specific devices because of their brittleness,insolubility and low compatibility.Therefore,to expand versatile MOF membranes with robustness and operational flexibility is urgent to satisfy practical applications.Although numerous reports have reviewed the synthesis and applications of MOF membranes,relatively few reports the electrocatalytic properties based on MOF membranes.Herein,this mini-review provides an overview of preparation of MOF membranes,including directed synthesis,secondary growth and electrochemical deposition method.Meanwhile,fabrication of ultrathin 2D MOF nanosheets those can be also defined as a kind of nanoscale MOF membranes is also mentioned.Electrocatalytic performance of oxygen reduction reaction(ORR),oxygen evolution reaction(OER),hydrogen evolution reaction(HER)and CO2 reduction reaction(CO2RR) for diverse MOF membranes/nanosheets and their derivatives are introduced.Xiaobang Liu Ting Yue Kai Qi Yubing Qiu Bao Yu Xia Xingpeng Guo 2020Chinese Chemical Letters2020,31,9:3
4LncRNA NORAD promotes the progression of myocardial infarction by targeting the miR-22-3p/PTEN axis显示文摘NORAD is a newly identified long non-coding RNA(lncRNA)that plays an important role in cancers.NORAD has been found to be highly expressed in the mouse model of acute myocardial infarction(AMI).However,the role of NORAD in the regulation of AMI remains unknown.In the present study,we aimed to investigate the function of NORAD in AMI and explore the potential regulatory mechanisms.A mouse model of AMI was established and NORAD was knocked-down.The infarcted size of heart tissues and the cardiac function were evaluated.In addition,two cardiomyocyte cell lines were treated with hypoxia/re-oxygenation(H/R)to mimic AMI in vitro.Luciferase reporter assay,RNA pull-down assay,fluorescence in situ hybridization,qRT-PCR,and western blot analysis were performed.Apoptotic cells and the levels of L-lactate dehydrogenase(LDH)and malondialdehyde(MDA)were detected.Our results show that downregulation of NORAD efficiently attenuates heart damage in the AMI mouse model.NORAD interacts with miR-22-3p.Knock-down of NORAD inhibits H/R-induced cell apoptosis and reduces LDH and MDA levels,while its effects are abolished by miR-22-3p inhibitor.MiR-22-3p interacts with PTEN and inhibits its expression.Overexpression of miR-22-3p inhibits H/R-induced cell apoptosis and reduces LDH and MDA levels,while its effects are abolished by overexpression of PTEN.Finally,overexpression of NORAD inhibits the AKT/mTOR signaling pathway,and its effects are attenuated by overexpression of miR-22-3p.Taken together,our study reveals that NORAD promotes the progression of AMI by regulating the miR-22-3p/PTEN axis,and the AKT/mTOR signaling may also be involved in the regulatory processes.Chunxia Li Lihui Zhang Xingpeng Bu Guofang Chu Xiaofang Zhao Yaru Liu 2022Acta Biochimica et Biophysica Sinica2022,54,4:3
5p53-dependent upregulation of PIG3 transcription by γ-ray irradiation and its interaction with KAP1 in responding to DNA damage显示文摘PIG3 (p53-inducible gene 3), originally identified as one of a set of genes induced by p53 before the onset of apoptosis, was assumed to contribute to early cellular response to DNA damage. Here, we studied the relation between p53 status and the increased expression of PIG3 by ionizing radiation (IR), and the related clues regarding the involvement of PIG3 in the cellular response to IR-induced DNA damage signaling. We demonstrated that the pentanucleotide microsatellite sequence was responsible for the p53-dependent induction of PIG3 transcription after irradiation, while sequence upstream of PIG3 promoter could maintain the basal level of expression which was not inducible by irradiation. The interaction of PIG3 and the KRAB-ZFP-associated protein 1 (KAP1), a DNA damage response protein, was revealed. PIG3 nucleus foci were formed 15 min after γ-ray irradiation, and which were found to partially colocalize with the phospho-KAP-1 foci as well as γ-H2AX foci. Although the lac operator tagged EGFP based reporter system revealed that PIG3 does not remodel chromatin in large scale in the cells under normal growing condition, it indeed prompted the chromatin relaxation in the cellular response to DNA damage signaling. 6Direction-of-arrival estimation for uniform circular arrays under small sample size显示文摘With a small number of snapshots, performances of uniform circular array(UCA) root-MUSIC based methods for directionof-arrival(DOA) estimation suffer serious degradation, because the beamspace sample covariance matrix(BSCM) severely deviates from the true one. To solve this problem, an iterative technique is proposed. The proposed technique has two key points: transform array data into beamspace data and construct a BSCM; modify the BSCM repeatedly by removing the residual parts. Meanwhile,error and beamspace leakage associated with the BSCM are investigated. The proposed technique can significantly reduce the error and the beamspace leakage which in turn could improve the performance for DOA estimation. 7Population amount risk assessment of extreme precipitation-induced landslides based on integrated machine learning model and scenario simulation显示文摘In this study,the future landslide population amount risk(LPAR)is assessed based on integrated machine learning models(MLMs)and scenario simulation techniques in Shuicheng County,China.Firstly,multiple MLMs were selected and hyperparameters were optimized,and the generated 11 models were crossintegrated to select the best model to calculate landslide susceptibility;by calculating precipitation for different extreme precipitation recurrence periods and combining the susceptibility results to assess the landslide hazard.Using the town as the basic unit,the exposure and vulnerability of the future landslide population under different Shared Socioeconomic Pathways(SSPs)scenarios in each town were assessed,and then combined with the hazard to estimate the LPAR in 2050.The results showed that the integrated model with the optimized random forest model as the combination strategy had the best comprehensive performance in susceptibility assessment.The distribution of hazard classes is similar to susceptibility,and with an increase in precipitation,the low-hazard area and high-hazard decrease and shift to medium-hazard and very high-hazard classes.The high-risk areas for future landslide populations in Shuicheng County are mainly concentrated in the three southwestern towns with high vulnerability,whereas the northern towns of Baohua and Qinglin are at the lowest risk class.The LPAR increased with the intensity of extreme precipitation.The LPAR differs significantly among the SSPs scenarios,with the lowest in the“fossil-fueled development(SSP5)”scenario and the highest in the“regional rivalry(SSP3)”scenario.In summary,the landslide susceptibility model based on integrated machine learning proposed in this study has a high predictive capability.The results of future LPAR assessment can provide theoretical guidance for relevant departments to cope with future socioeconomic development challenges and make corresponding disaster prevention and mitigation plans to prevent landslide risks from a developmental perspective.Guangzhi Rong Kaiwei Li Zhijun Tong Xingpeng Liu Jiquan Zhang Yichen Zhang Tiantao Li 2023Geoscience Frontiers2023,14,3:2

9Genome-wide signatures of the geographic expansion and breeding of soybean显示文摘Soybean is a leguminous crop that provides oil and protein. Exploring the genomic signatures of soybean evolution is crucial for breeding varieties with improved adaptability to environmental extremes. We analyzed the genome sequences of 2,214 soybeans and proposed a soybean evolutionary route, i.e., the expansion of annual wild soybean(Glycine soja Sieb. & Zucc.) from southern China and its domestication in central China, followed by the expansion and local breeding selection of its landraces(G. max(L.) Merr.). We observed that the genetic introgression in soybean landraces was mostly derived from sympatric rather than allopatric wild populations during the geographic expansion. Soybean expansion and breeding were accompanied by the positive selection of flowering time genes, including GmSPA3c. 13Optimization design of fertilizer apparatus owned arc gears based on discrete element method显示文摘To improve the uniformity of the flow when fertilizer apparatus discharges fertilizer,a kind of fertilizer apparatus owned arc gears was designed.Also,the design and working principle of its general structure and key components were analyzed theoretically.Aiming at exploring the influence of arc structural parameters to the performance of arc gears discharging fertilizer,DEM simulation was used in this study.Fertilizer apparatus owned arc gears was taken as the research object to analyze the influence of two key components including the arc radius of gears’concave-grooves and the minimal length between two mutual meshing gears.The variation coefficient of fertilizer discharging amount stability and the linear determined coefficient of fertilizer discharging amount in unit time as evaluation index were selected.Two factors and five levels quadratic rotation orthogonal experiment was applied,and Design-Expert 8.0 software was used to achieve data processing and analysis.The results of experiment indicated that the arc radius of gears’concave-grooves had significant effect on the linear determined coefficient of fertilizer discharging amount in unit time,and the minimal length between two mutual meshing gears had significant effect on the variation coefficient of fertilizer discharging amount stability.The optimum structural parameters were 8.54 mm as the arc radius of gears’concave-grooves and 5.22 mm as the minimal length between two mutual meshing gears.Upon this circumstance,the variation coefficient was 0.28 and determination coefficient was 0.9972.The optimum apparatus was selected to conduct the bench test and field simulation test.The results indicated that the variation coefficient is 0.27,and the linear determined coefficient of is 0.9980.The results of simulation experiment were anastomotic basically with the real result,which showed that DEM simulation technology was usable to the experiment of testing fertilizer discharging.The results of this research can provide a reference to the improving of performance in discharging fertilizer.Guoqiang Dun Zhiyong Gao Yuxuan Liu Wenyi Ji Ning Mao Xingpeng Wu Wenhui Liu 2021International Journal of Agricultural and Biological Engineering2021,14,2:1

16Results of Atrial Fibrillation Ablation in Patients With or Without a History of Atrial Flutter显示文摘Objectives There are two kind of atrial flutter during circumferential ablation for atrial fibrillation (AF): new onset left atrial flutter (LAFL), with a history of atrial flutter (AFL). What is the relationship of AFL and AF? Whether there are some differences in clinical course and mechanism between the new onset LAFL and the with a history of AFL remained unclear. The aim of this study was to assess the impacts of circumferential ablation on the occurrence of arrhythmias in follow-up in 2 groups: (1) patients with a history of AFL and AF, and (2) patients with new onset LAFL. Methods Data from 465 patients who had circumferential pulmonary vein ablation (CPVA) or segmental pulmonary vein ablation (SPVA) were analyzed. Patients with a history of AFL ablation and patients who had concomitant AFL ablation were included from analysis. Forty-one patients constituted the history of AFL group (group 1, aged 57±13 years, 7 females) and twenty-eight patients constituted the new onset LAFL group (group 2, aged 55±12 years, 6 females). bipolar recordings were obtained from the tricuspid annulus, coronary sinus, interatrial septum and left atrium. Target sites were identified by early, fragmented or double potentials and by concealed entrainment. Linear lesions were created between target sites and nearby anatomical barriers (1) typical atrial flutter (cycle length, 242±39 ms). cavotricuspid isthmus ablation was performed. (2) new onset LAFL (cycle length, 282±153 ms). 20 episodes of AAFs were documented in 20/28 (71.4%) patients. Target sites were identified around pulmonary veins (n=10), gap in linear lesion (n=7), left atrial roof lines (1 case). For those cases the ablation line between PV and mitral annulus was performed. Patients in Group 2 had larger left atria, higher incidence of AFL pre-CPVA, and lower ejection fraction. Results There was no significant difference in post-CPVA AF recurrence between Groups 1 and 2, but AFL incidence after CPVA was higher in Group 2 (33% vs 4%, P < 0.0001). Ablation of AFL in group 1 patients resulted in an 88% acute success rate (group 2, 86%) and 12% (group 2,17%) recurrence over a mean follow-up of 287±101 days.Conclusions In patients with a history of atrial flutter, post-CPVA AF recurrence is similar to patients with new onset LAFL. However, LAFL is associated with a higher recurrence of AFL after PVAI. In a significant number of patients with LAFL, mitral Isthmus ablation is needed and can be performed effectively. 20Rationale and Study Design for Evaluating the Efficacy and Safety of Intracardiac Echocardiography-Guided Minimal-Fluoroscopy Ablation in Patients with Paroxysmal Atrial Fibrillation:A Non-Inferior,Multi-Center,Prospective Randomized Controlled Trial(PAF-ICE Trial)显示文摘The feasibility and safety of intracardiac echocardiography(ICE)-guided catheter ablation for atrial fibrillation(AF)using a minimal/zero-fluoroscopy approach have recently been reported.This approach helps to reduce ionizing radiation exposure and orthopedic complications resulting from using lead aprons.The objectives of this planned prospective,multicenter randomized controlled trial(RCT)(paroxysmal AF(PAF)-ICE trial;ChiCTR2000033624)are to evaluate the efficacy and safety of ICE-guided minimal-fluoroscopy ablation in patients with PAF and the impact on occupational hazards among lab staff.Patients will be randomized in a 1:1 ratio to 2 groups:minimal fluoroscopy group(n=216)and traditional approach group(n=216).In the minimal fluoroscopy group,an ICE catheter will be used for geometry/anatomic construction,transseptal puncture,catheter tracking,and effusion monitoring.Pulmonary vein isolation(PVI)will be performed using an open-irrigated radiofrequency SmartTouch Surround Flow or SmartTouch catheter(Biosense Webster,Diamond Bar,California,USA),and confirmed by a multipolar Lasso or PentaRay catheter(Biosense Webster).In the traditional approach group,an ICE catheter will not be used.Transseptal puncture will be performed under fluoroscopic guidance,with all geometries constructed by mapping the catheters.The primary efficacy endpoint is freedom from AF recurrence(without antiarrhythmic medications)at 12months after ablation.Other endpoints include duration of lead apron use,measures of intra-procedural efficiency,and peri-procedural complications.This RCT will evaluate the efficacy and safety of ICE-guided minimal-fluoroscopy ablation in patients with PAF,also evaluate the benefits to lab staff(regarding reducing occupational hazards)related to this“minimal/zero-fluoroscopy”and“leadless”mode.Ruhong Jiang Xingpeng Liu Jidong Zhang Yu Chen Rui Wang Mengzuo Wu Deyong Long Jia Li Haixiong Wang Jie Fan Weizhu Ju Weili Ge Xu Liu Hai Deng Weijian Wang Pingzhen Yang Ding Li Xiaobo Huang Xiongtao Liu Hailong Tao Paul CZei Roderick Tung Xunzhang Wang Chenyang Jiang 2021Cardiology Discovery2021,1,4:1
