1New Algorithm Rules Out Acute-on-chronic Liver Failure Development within 28 Days from Acute Decompensation of Cirrhosis显示文摘Background and Aims:Approximately 10%of patients with acute decompensated(AD)cirrhosis develop acute-on-chronic liver failure(ACLF)within 28 days.Such cases have high mortality and are difficult to predict.Therefore,we aimed to establish and validate an algorithm to identify these patients on hospitalization.Methods:Hospitalized patients with AD who developed ACLF within 28 days were considered pre-ACLF.Organ dysfunction was defined accord-ing to the chronic liver failure-sequential organ failure as-sessment(CLIF-SOFA)criteria,and proven bacterial infec-tion was taken to indicate immune system dysfunction.A retrospective multicenter cohort and prospective one were used to derive and to validate the potential algorithm,re-spectively.A miss rate of<5%was acceptable for the calcu-lating algorithm to rule out pre-ACLF.Results:In the deri-vation cohort(n=673),46 patients developed ACLF within 28 days.Serum total bilirubin,creatinine,international normalized ratio,and present proven bacterial infection at admission were associated with the development of ACLF.AD patients with≥2 organ dysfunctions had a higher risk for pre-ACLF patients[odds ratio=16.58195%confidence interval:(4.271-64.363),p<0.001].In the derivation co-hort,67.5%of patients(454/673)had≤1 organ dysfunction and two patients(0.4%)were pre-ACLF,with a miss rate of 4.3%(missed/total,2/46).In the validation cohort,65.9%of patients(914/1388)had≤1 organ dysfunction,and four(0.3%)of them were pre-ACLF,with a miss rate of 3.4%(missed/total,4/117).Conclusions:AD patients with≤1 organ dysfunction had a significantly lower risk of developing ACLF within 28 days of admission and could be safely ruled out with a pre-ACLF miss rate of<5%.Xiaoting Tang Hai Li Guohong Deng Xin Zheng Xianbo Wang Yan Huang Yanhang Gao Zhongji Meng Zhiping Qian Feng Liu Xiaobo Lu Yu Shi Beiling Li Wenyi Gu Xiaomei Xiang Yan Xiong Yixin Hou Jun Chen Na Gao Sen Luo Liujuan Ji Jing Li Rongjiong Zheng Haotang Ren Jinjun Chen 2023Journal of Clinical and Translational Hepatology2023,11,3:1

3Cisplatin selects for CD133+cells in lung cancer cells显示文摘Objective Platinum-based chemotherapy is the first-line treatment for non-small cell lung cancer,but the chemoresistance of tumor cells continues to be a considerable challenge in the management of NSCLCs,leading to recurrence of most patients.CD133(prominin-1)is a five-transmembrane glycoprotein,and recent evidence suggests that CD133+cells are the cause of drug resistance and tumor recurrence.In this study,the correlation between cisplatin and CD133+cells was investigated systematically.Methods Four lung cancer cell lines,including A549,H460,801D and H1299,were treated with different concentrations of cisplatin.Cell viability was determined by MTT assay.Sphere-forming assay was performed to detect the capability of sphere-forming.CD133+cells was detected by BD FACScaliber flow cytometer.Results The results showed that cisplatin could increase the number of CD133+cells in both time-and dose-dependent manner.The enrichment would weaken but the proportion of CD133+cells was still higher than the basic level as incubation time extended after cisplatin was withdrawn.Compared with adherent culture,the proportion of CD133+cells was higher when the cells were maintained suspension culture.The proportion of CD133+cells significantly increased when cisplatin was provided in suspension culture.Conclusion These results revealed that cisplatin induces the enrichment of CD133+cells and CD133 is a new therapeutic target.Our data partially explained drug resistance to second-line chemotherapy in cisplatin-treated patients with NSCLCs.Jiaheng Li Mei Jiang Xiaoting Zhao Ziyu Wang Meng Gu Weiying Li 2020Oncology and Translational Medicine2020,6,1:1
4Generation mechanism and control of high-frequency vibration for tracked vehicles显示文摘A crawler system provides much larger ground contact,leading to excellent terrain adaptability.Due to its structural characteristics,high‐frequency vibration proportional to the vehicle speed is generated during the driving process.This is a result of the polygon and rolling effects between the track and the wheels.A field test of a tracked vehicle is performed to monitor movement signals of the chassis and a rocker arm.Their corresponding power spectral density distributions confirm the correctness of the frequency‐calculation equation.Then,a novel elastic track tensioning device with a damper is designed as a cushion between the idler and the chassis.Depending on its geometry,the equivalent damping coefficient for a dynamic model is evaluated.Subsequently,the damping is altered in response to different operating conditions by a hybrid damping fuzzy semiactive control system.The controller accounts for both chassis and track vibration.Based on the transfer matrix method for multibody systems,a dynamical model of the track system is developed.Control performances are evaluated using two numerical simulations of obstacle crossing and off‐road driving operations.Results indicate that the proposed semiactive tensioner is substantially better than the conventional one.This paper provides a novel feasible scheme for vibration reduction of tracked vehicles.Pingxin Wang Xiaoting Rui Feifei Liu Guoping Wang Hailong Yu Bin He Junjie Gu 2023International Journal of Mechanical System Dynamics2023,3,2:0
5Changes in corneal curvature and aberrations after cataract surgery显示文摘Background:Surgically induced astigmatism(SIA)and corneal high-order aberrations(HOAs)are the two main causes of poor visual quality after cataract surgery.Changes in the parameters of corneal HOAs after cataract surgery and their effects on and relationships with changes in corneal curvature have not yet been reported.This study aimed to explore changes in anterior,posterior and total corneal curvature,astigmatism and HOAs after microincision cataract surgery.Methods:Sixty-one age-related cataract patients(61 eyes)were included in this prospective study.The total,anterior and posterior corneal astigmatism and corneal HOAs were analyzed by anterior segment optical coherence tomography(AS-OCT)and iTrace before,one day,one week and three months after 2.2 mm temporal microincision coaxial phacoemulsification to evaluate the changes in anterior,posterior and total corneal curvature,astigmatism and corneal HOAs.Results:The mean J0 and J45 values of anterior,posterior and total corneal curvature obtained by ASOCT showed no statistically significant difference between preoperatively and any postoperative followup.SIA occurred on the anterior,posterior and total corneal surfaces and showed no statistically significant difference at any postoperative follow-up.No significant changes in 3rd-order oblique trefoil,vertical coma or 4th-order spherical aberrations were observed after surgery except for a significant increase in horizontal coma at postoperative day 1(POD1).Conclusions:There were no significant changes in corneal curvature after 2.2 mm temporal microincision coaxial phacoemulsification,and the corneal HOAs were not changed significantly except for the increase in horizontal coma at POD1,which may be one of the main reasons of poor visual quality at POD1 in some cataract patients who have good uncorrected or corrected distance vision.Ye Dai Xiaoting Ruan Wei Wang Xiaoyun Chen Guangming Jin Lanhua Wang Xiaoxun Gu Bo Qu Jianping Liu Xuhua Tan Enen Zhang Jun Fu Lixia Luo Zhenzhen Liu Yizhi Liu 2022Annals of Eye Science2022,,3:0
6Eigenvalue analysis of planar linear multibody system under conservative force based on the transfer matrix method显示文摘The linear multibody system transfer matrix method(LMSTMM)provides a powerful tool for analyzing the vibration characteristics of a mechanical system.However,the original LMSTMM cannot resolve the eigenvalues of the systems with ideal hinges(i.e.,revolute hinge,sliding hinge,spherical hinge,cylindrical hinge,etc.)or bodies under conservative forces due to the lack of the corresponding transfer matrices.This paper enables the LMSTMM to solve the eigenvalues of the planar multibody systems with ideal hinges or rigid bodies under conservative forces.For a rigid body,the transfer matrix can now consider coupling terms between forces and kinematic state perturbations.Also,conservative forces that contribute to the eigenvalues can be considered.Meanwhile,ideal hinges are introduced to LMSTMM,which enables the treatment of eigenvalues of general multibody systems using LMSTMM.Finally,the comparative analysis with ADAMS software and analytical solutions verifies the effectiveness of the proposed approach in this paper.Jinghong Wang Xiaoting Rui Xun Wang Jianshu Zhang Qinbo Zhou Junjie Gu 2023International Journal of Mechanical System Dynamics2023,3,1:0
7沉默COX-2抑制A549细胞的恶性增殖(英文)显示文摘Objective: The aim of this study was to explore the effects on malignant proliferation of A549 cell by silencing cy-clooxygenase (COX)-2. Methods: In the present study, we constructed three siRNA vectors producing small interference RNA. The siRNA vectors and the vacant vectors were transfected into A549 cell with lipofectamine respectively and the transfected cell strains were constructed. The change of COX-2 expression levels was examined by Western blot and RT-PCR. The effects on the proliferation of lung cancer cells were studied by cell growth curve, clonogenic assay and xenograft assays. Results: The siRNA expression vectors produced marked effects in A549 cell but the inhibited effects were different. The effect of psi-10 was best and the mRNA and protein levels of COX-2 reduced 61.2% and 56.2% respectively in A549-si10 cell in contrast to the control. The growth of A549 cell slowed and the colony formation rate reduced after silencing COX-2. In xenograft assays, the growth speeds of tumor became slow and the numbers of tumor reduced after silencing COX-2. 8New considerations on the phylogeny of Sessilida(Protista:Ciliophora:Peritrichia)based on multiple-gene information,with emphasis on colonial taxa显示文摘The subclass Peritrichia,containing two orders Sessilida and Mobilida,is a major group of ciliates with worldwide distribution and high species diversity.Several studies have investigated the phylogeny of peritrichs;however,the evolutionary relationships and classification of some families and genera within the Sessilida remain unclear.In the present study,we isolated and identified 22 peritrich populations representing four families and six genera and obtained 64 rDNA sequences to perform phylogenetic analyses and assess their systematic relationships.Ancestral character reconstruction was also carried out to infer evolutionary routes within the Sessilida.The results indicate:(1)family Vaginicolidae is monophyletic and acquisition of the typical peritrich lorica represents a single evolutionary divergence;(2)core epistylidids evolved from a Zoothamnium-like ancestor and experienced spasmoneme loss during evolution;(3)Campanella clusters with species in the basal clade and shows stable morphological differences with other epistylidids,supporting its assignment to a separate family;(4)the structure of the peristomial lip may be a genus-level character rather than a diagnostic character for discriminating Epistylididae and Operculariidae,thus a redefinition of Operculariidae should be carried out when more species have been investigated;(5)some characters,such as lifestyle(solitary or colonial),spasmoneme and living habit(sessile or free-swimming),evolved repeatedly among sessilids indicating that species with non-contractile stalks or that are free-swimming have multiple evolutionary routes and might derive from any sessilid lineage without a lorica.The close phylogenetic relationships of some morphologically distinct sessilids imply that the diagnoses of some genera and families should be improved.Zhe Wang Xiaoting Feng Lifang Li Khaled A.S.Al-Rasheid Alan Warren Chundi Wang Zemao Gu 2022Marine Life Science & Technology2022,4,4:0

10Changes in crystalline lens parameters during accommodation evaluated using swept source anterior segment optical coherence tomography显示文摘Backgrounds:To assess changes in anterior segment biometry during accommodation using a swept source anterior segment optical coherence tomography(SS-OCT).Methods:One hundred-forty participants were consecutively recruited in the current study.Each participant underwent SS-OCT scanning at 0 and−3 diopter(D)accommodative stress after refractive compensation,and ocular parameters including anterior chamber depth(ACD),anterior and posterior lens curvature,lens thickness(LT)and lens diameter were recorded.Anterior segment length(ASL)was defined as ACD plus LT.Lens central point(LCP)was defined as ACD plus half of the LT.The accommodative response was calculated as changes in total optical power during accommodation.Results:Compared to non-accommodative status,ACD(2.952±0.402 vs.2.904±0.382 mm,P<0.001),anterior(10.771±1.801 vs.10.086±1.571 mm,P<0.001)and posterior lens curvature(5.894±0.435 vs.5.767±0.420 mm,P<0.001),lens diameter(9.829±0.338 vs.9.695±0.358 mm,P<0.001)and LCP(4.925±0.274 vs.4.900±0.259 mm,P=0.010)tended to decreased and LT thickened(9.829±0.338 vs.9.695±0.358 mm,P<0.001),while ASL(6.903±0.279 vs.6.898±0.268 mm,P=0.568)did not change significantly during accommodation.Younger age(β=0.029,95%CI:0.020 to 0.038,P<0.001)and larger anterior lens curvature(β=−0.071,95%CI:−0.138 to−0.003,P=0.040)were associated with accommodation induced greater steeping amplitude of anterior lens curvature.The optical eye power at 0 and−3 D accommodative stress was 62.486±2.284 and 63.274±2.290 D,respectively(P<0.001).Age was an independent factor of accommodative response(β=−0.027,95%CI:−0.038 to−0.016,P<0.001).Conclusions:During−3 D accommodative stress,the anterior and posterior lens curvature steepened,followed by thickened LT,fronted LCP and shallowed ACD.The accommodative response of−3 D stimulus is age-dependent.Lanhua Wang Guangming Jin Xiaoting Ruan Xiaoxun Gu Xiaoyun Chen Wei Wang Ye Dai Zhenzhen Liu Lixia Luo 2022Annals of Eye Science2022,,4:0
11环氧化酶-2在人肺癌中的表达及其意义(英文)显示文摘Objective: We aimed to examine the expression of cyclooxygenase-2 and the relationship with the pathological types, TNM stage, lymph node metastasis, the degree of differentiation, smoking and the survival. Methods: Immunohistochemical staining method was used to examine the expression of cyclooxygenase-2 of 121 cases of lung cancer and three control groups. The data were statistically analyzed. Results: Compared with the health group, cyclooxygenase-2 was over expressed in the inflammatory tissue(P = 0.036), lung adenocarcinoma(P = 0.005) and squamous carcinoma(P = 0.047). Compared with patients without lymph node metastasis, cyclooxygenase-2 was over expressed(P = 0.033) in patients with lymph node metastasis. Compared with high differentiation group, cyclooxygenase-2 was over expressed(P = 0.004) in low differentiation group. Compared with non-smokers, the expression of cyclooxygenase-2 increased in smokers(P = 0.000). The median survival time of patients that the expression of cyclooxygenase-2 were negative was 9 months(95% CI, 5.6–12.4 months). The median survival time of patients that the expression of cyclooxygenase-2 were positive was 5 months(95% CI, 3–7 months). They was statistical difference(P = 0.001).
