| 4 | Inulin Can Alleviate Metabolism Disorders in ob/ob Mice by Partially Restoring Leptinrelated Pathways Mediated by Gut Microbiota显示文摘Inulin has been used as a prebiotic to alleviate glucose and lipid metabolism disorders in mice and humans by modulating the gut microbiota. However, the mechanism underlying the alleviation of metabolic disorders by inulin through interactions between the gut microbiota and host cells is unclear. We use ob/ob mice as a model to study the effect of inulin on the cecal microbiota by16 S rRNA gene amplicon sequencing and its interaction with host cells by transcriptomics. The inulin-supplemented diet improved glucose and lipid metabolism disorder parameters in ob/ob mice,alleviating fat accumulation and glucose intolerance. The a diversity of gut microbial community of ob/ob mice was reduced after inulin treatment, while the b diversity tended to return to the level of wild type mice. Interestingly, Prevotellaceae UCG 001(family Prevotellaceae) was obviously enriched after inulin treatment. A comparative analysis of the gene expression profile showed that the cecal transcriptome was changed in leptin gene deficiency mice, whereas the inulin-supplemented diet partially reversed the changes in leptin gene-related signaling pathways, especially AMPK signaling pathway, where the levels of gene expression became comparable to those in wild type mice.Further analysis indicated that Prevotellaceae UCG 001 was positively correlated with the AMPK signaling pathway, which was negatively correlated with markers of glycolipid metabolism disorders. | 7 | Occurrence of Neoproterozoic low-grade metagranite in the western Beihuaiyang zone,the Dabie orogen显示文摘Based on detailed field investigations and petrographic observations,we discovered Neoproterozoic-emplaced granite from the metavolcanics of the Dingyuan Formation in the western Beihuaiyang zone,the Dabie orogen. This study reports the results of zircon U-Pb dating and preliminary petrographic observations on two metagranite samples. The studied rocks experienced epidote-amphibolite facies metamorphism and strong structural deformation. Their U-Pb ages are 726 ± 6 and 758 ± 12 Ma,respectively,similar to those for the Luzhenguan complex in the eastern segment of the Beihuaiyang zone. In combination with previously determined 635 ± 5 Ma low-grade metagabbro,this study suggests the occurrence of at least two types of Neoproterozoic intrusive rocks in the Beihuaiyang zone,the northern margin of the South China Block (SCB):726-758 Ma metagranite and 635 Ma metagabbro. These rocks occur within the metamorphosed Ordovician volcanic zone (originally named the Dingyuan Formation) and are in tectonic contact to each other,but they formed in different tectonic settings. The protolith ages for the Neoproterozoic low-grade metaigneous rocks are in good agreement not only with ages for two episodes of mid-and late-Neoproterozoic mafic and felsic magmatism in the Suizhou and Zaoyang area,Hubei Province,but also agree with protolith ages of ultrahigh-pressure metaigneous rocks in the Dabie-Sulu orogenic belt. In this work, a columnar Al film was deposited at the top of 7 wt% yttriastabilized zirconia(7YSZ) TBC by magnetron sputtering. A vacuum heat treatment was then carried out to improve the insulation property of Al-deposited TBC. Reaction mechanism of Al-Zr O2 system in Aldeposited TBC was studied by differential thermal analysis(DTA). The phase structures of the assprayed TBC, the Al-deposited and vacuum-treated TBC were characterized. The microstructure evolution of Al-deposited TBC was illustrated after vacuum heat treatment. And the insulation property of the assprayed TBC and treated TBC was compared. The results show that a multi-scaled layer, consisting of micron/nano structured α-Al2O3 and Al3 Zr grain was in situ synthesized at the top of 7YSZ coating via vacuum heat treatment. Simulated results show that the effects of atmospheric pressure fluctuations on coal fires and surface anomalies depend on two factors: the fluctuant amplitude and the pressure difference between inlet(s) and outlet(s) of the air ventilation system. | 19 | Crustal P-wave velocity structure in the northeastern margin of the Qinghai-Tibetan Plateau and insights into crustal deformation显示文摘The transitional area between the northeastern margin of the Qinghai-Tibetan Plateau, Ordos Block and Alxa Block,also being the northern segment of the North-South Seismic Belt, is characterized by considerably high seismicity level and high risk of strong earthquakes. In view of the special tectonic environment and deep tectonic setting in this area, this study used two seismic wide-angle reflection/refraction cross profiles for double constraining, so as to more reliably obtain the fine-scale velocity structure characteristics in both the shallow and deep crust of individual blocks and their boundaries in the study area,and further discuss the seismogenic environment in seismic zones with strong historical earthquakes. In this paper, the P-wave data from the two profiles are processed and interpreted, and two-dimensional crustal velocity structure models along the two profiles are constructed by travel time forward modeling. The results show that there are great differences in velocity structure,shape of intra-crustal interfaces and crustal thickness among different blocks sampled by the two seismic profiles. The crustal thickness along the Lanzhou-Huianbu-Yulin seismic sounding profile(L1) increases from ~43 km in the western margin of Ordos Block to ~56 km in the Qilian Block to the west. In the Ordos Block, the velocity contours vary gently, and the average velocity of the crust is about 6.30 km s-1; On the other hand, the velocity structures in the crust of the Qilian Block and the arclike tectonic zone vary dramatically, and the average crustal velocities in these areas are about 0.10 km s-1 lower than that of the Ordos Block. In addition, discontinuous low-velocity bodies(LVZ1 and LVZ2) are identified in the crust of the Qilian Block and the arc-like tectonic zone, the velocity of which is 0.10–0.20 km s-1 lower than that of the surroundings. The average crustal thickness of the Ordos Block is consistently estimated to be around 43 km along both Profile L2(Tongchuan-Huianbu-Alashan left banner seismic sounding profile) and Profile L1. In contrast to the gently varying intra-crustal interfaces and velocity contours in the Ordos Block along Profile L1, which is a typical structure characteristic of stable cratons, the crustal structure in the Ordos Block along Profile L2 exhibits rather complex variations. This indicates the presence of significant structural differences in the crust within the Ordos Block. The crustal structure of the Helan Mountain Qilian Block and the Yinchuan Basin is featured by 'uplift and depression' undulations, showing the characteristics of localized compressional deformation.Moreover, there are low-velocity zones with alternative high and low velocities in the middle and lower crust beneath the Helan Mountain, where the velocity is about 0.15–0.25 km s-1 lower than that of the surrounding areas. The crustal thickness of the Alxa Block is about 49 km, and the velocity contours in the upper and middle-lower crust of the block vary significantly. The complex crustal velocity structure images along the two seismic sounding profiles L1 and L2 reveal considerable structural differences among different tectonic blocks, their coupling relationships and velocity structural features in the seismic zones where strong historical earthquakes occurred. | 20 | Circ-Smad5 retards the G1/S transition of cell cycle via inhibiting the activity of wnt/lef/cyclind1 signaling in JB6 cells显示文摘Circular RNAs are a large class of noncoding RNAs.Smad5 functions in cell differentiation,cell proliferation and metastasis.It has been reported that lnc-Smad5 can inhibit the proliferation of diffuse large B cell lymphoma.However,the function of circ-Smad5 has not yet been reported.Lentivirus vectors were constructed to establish circ-Smad5 upregulated and circ-Smad5 downregulated cell models.A CCK-8 assay was used to detect the proliferation of JB6 cells.FACS was used to analyze the cell cycle in the cell models.Western blot,immunofluorescence staining and TOP/FOP flash dual luciferase activity assays were used to determine the activity of the Wnt signaling pathway.The results revealed that the expression level of circ-Smad5 in JB6 cells was significantly lower than the expression level of linearizedSmad5.Compared with the control group,the percentage of S phase cells and the expression level of cyclin D1 protein were significantly higher in the sh-circ-Smad5 group.In the sh-circSmad5 group,b-catenin and LEF-1 were significantly increased,p-b-catenin was significantly decreased,and the relative activity of the TOP/FOP reporter gene was higher compared to the control group levels.These phenomena could be reversed by treating with Wnt signaling inhibitor PNU-74654.We conclude that the circ-Smad5 retards the proliferation and the cell cycle progression of JB6 cells.Thus,circ-Smad5 may function by inhibiting the activation of Wnt/b-catenin/Lef 1 signaling,which inhibits the expression of cyclin D1.To the best of our knowledge,we are the first to report the function of circ-Smad5. | Xiaogen Ma Fei Xiang Zhuo Pei Jiafeng Miao Pan Wu Xiaofeng Song Yuhong Li Yiming Zhang | 2021 | Genes & Diseases2021,8,3: | 3 |
