| 1 | Relation between electricity structure of the crust and deformation of crustal blocks on the northeastern margin of Qinghai-Tibet Plateau显示文摘Study on the electricity structure along a magnetotelluric (MT) sounding profile on the northeastern margin of Qinghai-Tibet Plateau indicates that four crustal blocks can be de-termined from southwest to northeast, namely Bayan Har block (BH), Qin-Qi block (QQ), Hai-yuan block (HY) or the North-South seismotectonic belt and Ordos block (OD). The BH, QQ and OD blocks display a similar electricity structure of the crust. The upper crust represents a high- resistivity layer and the upper part of lower crust represents a low-resistivity layer with the resis-tivity increasing gradually with depth from the lower part of lower crust to the upper mantle. The electricity structure of the crust in these three blocks is similar to that in the complete blocks on the southern and eastern margins of the Qinghai-Tibet Plateau and belongs to normal electricity layering of the crust in slightly deformed or complete intracontinental blocks. The crust in HY block as a boundary zone has been significantly deformed, hence its electricity layering was de-stroyed and the structure was complex and the block became a recent tectonically active and great seismo-active region. The contact belts between the blocks on the northeastern margin of Qinghai-Tibet Plateau exhibit both upthrusting outward and strike-slip movement different from those on the southern and eastern margins of the plateau. | 2 | Neuroprotective effect of pretreatment with ganoderma lucidum in cerebral ischemia/reperfusion injury in rat hippocampus显示文摘Ganoderma lucidum is a traditional Chinese medicine,which has been shown to have both anti-oxidative and anti-inflammatory effects,and noticeably decreases both the infarct area and neuronal apoptosis of the ischemic cortex.This study aimed to investigate the protective effects and mechanisms of pretreatment with ganoderma lucidum(by intragastric administration)in cerebral ischemia/reperfusion injury in rats.Our results showed that pretreatment with ganoder-ma lucidum for 3 and 7 days reduced neuronal loss in the hippocampus,diminished the content of malondialdehyde in the hippocampus and serum,decreased the levels of tumor necrosis fac-tor-αand interleukin-8 in the hippocampus,and increased the activity of superoxide dismutase in the hippocampus and serum.These results suggest that pretreatment with ganoderma lucidum was protective against cerebral ischemia/reperfusion injury through its anti-oxidative and an-ti-inflammatory actions. | Wangxin Zhang Qiuling Zhang Wen Deng Yalu Li Guoqing Xing Xianjun Shi Yifeng Du | 2014 | Neural Regeneration Research2014,9,15: | 6 |

| 4 | Glutamine deprivation induces ferroptosis in pancreatic cancer cells显示文摘Ferroptosis is a type of programmed cell death closely related to amino acid metabolism.Pancreatic cancer cells have a strong dependence on glutamine,which serves as a carbon and nitrogen substrate to sustain rapid growth.Glutamine also aids in self-protection mechanisms.However,the effect of glutamine on ferroptosis in pancreatic cancer remains largely unknown.Here,we aim to explore the association between ferroptosis and glutamine deprivation in pancreatic cancer.The growth of pancreatic cancer cells in culture media with or without glutamine is evaluated using Cell Counting Kit-8.Reactive oxygen species(ROS)are measured by 2′,7′-dichlorodihydrofluorescein diacetate staining.Ferroptosis is assessed by BODIPY-C11 dye using confocal microscopy and flow cytometry.Amino acid concentrations are measured using ultrahigh-performance liquid chromatography-tandem mass spectrometry.Isotope-labelled metabolic flux analysis is performed to track the metabolic flow of glutamine.Additionally,RNA sequencing is performed to analyse the genetic alterations.Glutamine deprivation inhibits pancreatic cancer growth and induces ferroptosis both in vitro and in vivo.Additionally,glutamine decreases ROS formation via glutathione production in pancreatic cancer cells.Interestingly,glutamine inhibitors(diazooxonorleucine and azaserine)promotes ROS formation and ferroptosis in pancreatic cancer cells.Furthermore,ferrostatin,a ferroptosis inhibitor,rescues ferroptosis in pancreatic cancer cells.Glutamine deprivation leads to changes in molecular pathways,including cytokine-cytokine receptor interaction pathways(CCL5,CCR4,LTA,CXCR4,IL-6R,and IL-7R).Thus,exogenous glutamine is required for the detoxification of ROS in pancreatic cancer cells,thereby preventing ferroptosis. | Zhiwen Xiao Shengming Deng He Liu Ruijie Wang Yu Liu Zhengjie Dai Wenchao Gu Quanxing Ni Xianjun Yu Chen Liu Guopei Luo | 2023 | Acta Biochimica et Biophysica Sinica2023,55,8: | 0 |
| 5 | HNF-1a promotes pancreatic cancer growth and apoptosis resistance via its target gene PKLR显示文摘Pancreatic ductal adenocarcinoma is one of the deadliest malignant tumors,and many genes play important roles in its development.The hepatocyte nuclear factor-1a(HNF-1a)gene encodes HNF-1a,which is a transcriptional activator.HNF-1a regulates the tissue-specific expression of multiple genes,especially in pancreatic islet cells and in the liver.However,the role of the HNF-1a gene in the development of pancreatic cancer is still unclear.Here,we used immunohistochemical staining and real-time PCR to analyze HNF-1a expression in pancreatic cancer tissue.Stable cell lines with HNF-1a knockdown or overexpression were established to analyze the role of HNF-1a in pancreatic cancer cell proliferation and apoptosis by colony formation assay and flow cytometry.We also analyzed the L-type pyruvate kinase(PKLR)promoter sequence to identify the regulatory effect of HNF-1a on PKLR transcription and confirmed the HNF-1a binding site in the PKLR promoter via a chromatin immunoprecipitation assay.HNF-1a was found to be overexpressed in pancreatic cancer and promoted proliferation while inhibiting apoptosis in pancreatic cancer cells.PKLR was identified as the downstream target gene of HNF-1a and binding of HNF-1a at two sites in PKLR(-1931/-1926 and-966/-961)regulated PKLR transcription.In conclusion,HNF-1a is overexpressed in pancreatic cancer,and the transcription factor HNF-1a can promote pancreatic cancer growth and apoptosis resistance via its target gene PKLR. | Zhiyao Fan Kun Fan Shengming Deng Yitao Gong Yunzhen Qian Qiuyi Huang Chao Yang He Cheng Kaizhou Jin Guopei Luo Chen Liu Xianjun Yu | 2020 | Acta Biochimica et Biophysica Sinica2020,52,3: | 0 |
