| 1 | The Chromosome-Level Genome Sequence of the Autotetraploid Alfalfa and Resequencing of Core Germplasms Provide Genomic Resources for Alfalfa Research显示文摘Alfalfa(Medicago sativa)is one of the most important forage crops in the world;however,its molecular genetics and breeding research are hindered due to the lack of a high-quality reference genome.Here,we report a de novo assembled 816-Mb high-quality,chromosome-level haploid genome sequence for‘Zhongmu No.1’alfalfa,a heterozygous autotetraploid.The contig N50 is 3.92 Mb,and 49165 genes are annotated in the genome.The alfalfa genome is estimated to have diverged from M.truncatula approximately 8 million years ago.Genomic population analysis of 162 alfalfa accessions revealed high genetic diversity,weak population structure,and extensive gene flow from wild to cultivated alfalfa.Genome-wide association studies identified many candidate genes associated with important agronomic traits.Furthermore,we showed that MsFTa2,a Flowering Locus T homolog,whose expression is upregulated in salt-resistant germplasms,may be associated with fall dormancy and salt resistance.Taken together,these genomic resources will facilitate alfalfa genetic research and agronomic improvement. | Chen Shen Huilong Du Zhuo Chen Hongwei Lu Fugui Zhu Hong Chen Xiangzhao Meng Qianwen Liu Peng Liu Lihua Zheng Xiuxiu Li Jiangli Dong Chengzhi Liang Tao Wang | 2020 | Molecular Plant2020,13,9: | 21 |
| 2 | Recent advances in joining of SiC-based materials(monolithic SiC and SiC_f/SiC composites):Joining processes,joint strength, and interfacial behavior显示文摘Silicon carbide(SiC) has been widely concerned for its excellent overall mechanical and physical properties, such as low density, good thermal-shock behavior, high temperature oxidation resistance, and radiation resistance; as a result, the SiC-based materials have been or are being widely used in most advanced fields involving aerospace, aviation, military, and nuclear power. Joining of SiC-based materials(monolithic SiC and SiCf/SiC composites) can resolve the problems on poor processing performance and difficulty of fabrication of large-sized and complex-shaped components to a certain extent, which are originated from their high inherent brittleness and low impact toughness.Starting from the introduction to SiC-based materials, joining of ceramics, and joint strength characterization, the joining of SiC-based materials is reviewed by classifying the as-received interlayer materials, involving no interlayer, metallic, glass-ceramic, and organic interlayers. The results showed that the as-cast eutectic carbide network tends to be broken after ferrovanadium inoculation, and the carbides are changed to rod-like or nodular shape. After heat treatment, the carbides in the inoculated HSS are spheroidized and distributed more uniformly in the matrix. The impact toughness of high speed steel with ferrovanadium inoculation is obviously improved. | 5 | Brazing of WC–8Co cemented carbide to steel using Cu–Ni–Al alloys as filler metal: Microstructures and joint mechanical behavior显示文摘Three novel Cu–Ni–Al brazing filler alloys with Cu/Ni weight ratio of 4:1 and 2.5–10 wt% Al were developed and characterized, and the wetting of three Cu–Ni–Al alloys on WC–8 Co cemented carbide were investigated at 1190–1210?C by the sessile drop technique. Vacuum brazing of the WC–8 Co cemented carbide to SAE1045 steel using the three Cu–Ni–Al alloys as filler metal was further carried out based on the wetting test results. The interfacial interactions and joint mechanical behaviors involving microhardness, shear strength and fracture were analyzed and discussed. The experimental results show that all the three wetting systems present excellent wettability with final contact angles of less than 5?and fast spreading. An obvious degeneration layer with continuous thin strip forms in the cemented carbide adjacent to the Cu–Ni–Al/WC–8 Co interface. The variation of microhardness in the joint cross-section is closely related to the interactions(such as diffusion and solid solution) of WC–8 Co/Cu–Ni–Al/steel system. Compared with the other two brazed joints, the WC–8 Co/Cu–19 Ni–5 Al/steel brazed joint presents more reliable interlayer microstructure and mechanical property while brazing at the corresponding wetting temperatures for 5 min, and its average shear strength is over 200 MPa after further optimizing the brazing temperature and holding time. | 6 | A special numerical method for fluid-structure interaction problems subjected to explosion and impact loading显示文摘A three-dimensional(3D) embedded Eulerian-Lagrangian method is proposed to simulate the 3D fluid-structure interaction(FSI) problems subjected to explosion and impact loading. This method achieves a high-quality calculation of fluid and structure deformation by adding Lagrangian particles to Eulerian grids. The overall computational domain is solved by the Eulerian method, and the Lagrangian particles with specified volume and influence domains are used to track structural deformations. The bidirectional mapping of physical quantities is achieved using the weighted average of the influence domain, which are based on the topological relationship between Eulerian grids and Lagrangian particles. Then, the data dependence solution and parallel algorithm realization are presented for the large-scale numerical calculations of explosion and impact problems. Additionally, the corresponding parallel program is developed based on the message passing interface(MPI) standard, and the parallel efficiency of parallel hydrocode are tested. The numerical results of typical explosion and impact problems are compared with corresponding experimental data to verify the effectiveness of the method. These comparisons show that the embedded EulerianLagrangian method successfully combine the advantages of both the Eulerian and Lagrangian methods to efficiently calculate the processes of large deformation and dynamic damage to the materials. | 7 | Development and Application of a Tyrosinase-Based Time-Temperature Indicator(TTI) for Determining the Quality of Turbot Sashimi显示文摘Time-temperature indicators(TTIs) are convenient intuitive devices that are widely used to predict food quality. The aim of this study is to develop a new simple device which can be attached to food packages as a quality indicator for turbot sashimi. In this study, a solid TTI based on the reaction between tyrosinase and tyrosine was developed. The Arrhenius behavior of this enzymatic TTI was studied. The kinetics of the tyrosinase-based TTI was investigated in the form of color change from colorless to dark black induced by the enzymatic reaction. The mathematical formula for the color alterations as a function of time and temperature was established. The longest indication time for the developed TTI was 50 hours at 4℃. The activation energy of the tyrosinase-based TTI was 0.409 k J mol^(-1). The suitability of the tyrosinase-based TTI was validated for turbot sashimi using total plate count. The feasibility of using this TTI as a quality indicator for turbot sashimi was assessed based on the activation energy and indication time. | 10 | Design and development of safety evaluation system of buildings on a seismic field based on the network platform显示文摘This paper describes a set of on-site earthquake safety evaluation systems for buildings, which were developed based on a network platform. The system embedded into the quantitative research results which were completed in accordance with the provisions from Post-earthquake Field Works, Part 2: Safety Assessment of Buildings, GB18208.2-2001, and was further developed into an easy-to-use software platform. The system is aimed at allowing engineering professionals, civil engineeing technicists or earthquake-affected victims on site to assess damaged buildings through a network after earthquakes. The authors studied the function structure, process design of the safety evaluation module, and hierarchical analysis algorithm module of the system in depth, and developed the general architecture design, development technology and database design of the system. Technologies such as hierarchical architecture design and Java EE were used in the system development, and My SQL5 was adopted in the database development. The result is a complete evaluation process of information collection, safety evaluation, and output of damage and safety degrees, as well as query and statistical analysis of identified buildings. | 12 | Recovery of Chitin and Protein from Shrimp Head Waste by Endogenous Enzyme Autolysis and Fermentation显示文摘The industrial processing of shrimp produces massive quantities of solid waste that is a notable source of animal protein, chitin, carotenoids, and other bioactive compounds that are not appropriately utilized. In the present study, chitin and protein extraction from shrimp head with autolysis and fermentation using Bacillus licheniformis were investigated. The results showed that when shrimp heads were autolyzed with a natural pH at 50℃ for 4 h, the total amino acid nitrogen in the supernatant was 5.01 mg mL^-1. Then, when a 50%(v/m) inoculum of the hydrolysate was incubated at 60℃ for 10 h, a deproteinization rate of 88.3% could be obtained. The fermented supernatant was processed into a dry protein powder, while the residues were demineralized by 10% citric acid for chitin. The recovered protein powder contained 5.5% moisture, 11.5% ash, and 66.7% protein, while the chitin contained 3.5% moisture, 2.1% ash, and 3.1% protein. In addition, amino acids, minerals, heavy metals, the degree of acetylation, microstructure, and Fourier-transform infrared(FT-IR) spectroscopy results were analyzed. Furthermore, the statistics of the large scale trial after treatment with 20 kg of shrimp heads were analyzed. | 14 | Development of a Label-Free Colorimetric Aptasensor with Rationally Utilized Aptamer for Rapid Detection of Okadaic Acid显示文摘Okadaic acid(OA)is a typical marine toxin with strong toxicity causing diarrheic shellfish poisoning(DSP).Aptamers show great advantages in toxin detection and attract increasing attentions in the field of food analysis.In this study,a label-free col-orimetric aptasensor was constructed for visual and rapid detection of OA in shellfish.To exploit the binding capability of the anti-OA aptamer,the inherent molecular recognition mechanism of aptamer and OA was studied,based on molecular docking,fluorescent assay,and biolayer interferometry.Consistent results showed that the stem-loop near the 3’terminal of the aptamer exhibit dominate binding capacity.Based on the revealed recognition information,the aptamer was thus rationally utilized and combined with AuNPs and cationic polymer polydiallyl dimethyl ammonium chloride(PDDA)for the development of the label-free colorimetric aptasensor,in which the 3’terminal was thoroughly exposed to OA.The aptasensor provided robust performance with a linear detection range of 100-1200 nmol L-1,a limit of detection of 41.30 nmol L-1,recovery rates of 91.6%-106.2%,as well as a high selectivity towards OA in shellfish samples.The whole detection process can be completed within 1 h.To our best knowledge,this is the first time that the anti-OA aptamer was thoroughly studied,and a label-free colorimetric aptasensor was rationally designed in this way.This study not only provides a rapid detection method for highly sensitive and specific detection of OA,but also serves as a reference for the design of efficient aptasensors in the future. | YAN Xiaochen QI Xiaoyan ZHAO Yinglin LI Ling MA Rui WANG Lele WANG Sai MAO Xiangzhao | 2022 | Journal of Ocean University of China2022,21,2: | 2 |
