2Molecular Epidemiology of Echovirus 18 Circulating in China's Mainland from 2015 to 2016显示文摘Echovirus 18(E18), a serotype of Enterovirus B(EV-B) species, is an important pathogen in aseptic meningitis. E18 had rarely been detected in China's Mainland, but became the predominant pathogen associated with viral encephalitis(VE) and meningitis in Hebei province for the first time in 2015. To investigate the molecular epidemiology and genetic characteristics of E18 in China's Mainland, sixteen E18 strains from patient throat swabs with hand, foot, and mouth disease(HFMD) in six provinces in China collected between 2015 and 2016, and four E18 strains isolated from 18 patient cerebrospinal fluid specimens with VE in Hebei Province in 2015 were obtained and sequenced. Combined with the sequences from the GenBank database, we performed an extensive genetic analysis. Phylogenetic analysis of VP1 gene sequences revealed that all E18 strains from China's Mainland after 2015 belonged to subgenotype C2. There were no obvious specific differences in phylogenetic and variation analyses of E18 genome sequences between HFMD and VE/meningitis strains. Potential multiple recombination may have occurred in the 50-untranslated region and in the P2 and P3 nonstructural protein-encoding regions of E18 strains from China. The current E18 strains were potential multiplerecombinant viruses. 3Skp2 dictates cell cycle-dependent metabolic oscillation between glycolysis and TCA cycle显示文摘Whether glucose is predomi nantly metabolized via oxidative phosphorylati on or glycolysis differs between quiesce nt versus proliferating cells,iincluding tumor cells.However,how glucose metabolism is coordinated with cell cycle in mammalian cells remains elusive.Here,we report that mammalian cells predominantly utilize the tricarboxylic acid(TCA)cycle in G1 phase,but prefer glycolysis in S phase.Mechanistically,coupling cell cycle with metabolism is largely achieved by timely destruction of IDH1/2,key TCA cycle enzymes,in a Skp2-dependent manner.As such,depleting SKP2 abolishes cell cycle-dependent fluctuation of IDH1 protein abundance,leading to reduced glycolysis in S phase.Furthermore,elevated Skp2 abundance in prostate cancer cells destabilizes IDH1 to favor glycolysis and subsequent tumorigenesis.Therefore,our study reveals a mechanistic link between two cancer hallmarks,aberrant cell cycle and addiction to glycolysis,and provides the underlying mechanism for the coupling of metabolic fluctuation with periodic cell cycle in mammalian cells.Jing Liu Yunhua Peng Le Shi Lixin Wan Hiroyuki Inuzuka Jiangang Long Jianping Guo Jinfang Zhang Min Yuan Shuangxi Zhang Xun Wang Jing Gao Xiangpeng Dai Shozo Furumoto Lijun Jia Pier Paolo Pandolfic John M.Asara William G.Kaelin Jr. Jjankang Liu Wenyi Wei 2021Cell Research2021,31,1:2
