9Generation of GGTA1 biallelic knockout pigs via zinc-finger nucleases and somatic cell nuclear transfer显示文摘Genetically modified pigs are valuable models of human disease and donors of xenotransplanted organs.Conventional gene targeting in pig somatic cells is extremely inefficient.Zinc-finger nuclease(ZFN)technology has been shown to be a powerful tool for efficiently inducing mutations in the genome.However,ZFN-mediated targeting in pigs has rarely been achieved.Here,we used ZFNs to knock out the porcineα-1,3-galactosyl-transferase(GGTA1)gene,which generates Gal epitopes that trigger hyperacute immune rejection in pig-to-human transplantation.Primary pig fibroblasts were transfected with ZFNs targeting the coding region of GGTA1.Eighteen mono-allelic and four biallelic knockout cell clones were obtained after drug selection with efficiencies of 23.4%and 5.2%,respectively.The biallelic cells were used to produce cloned pigs via somatic cell nuclear transfer(SCNT).Three GGTA1 null piglets were born,and one knockout primary fibroblast cell line was established from a cloned fetus.Gal epitopes on GGTA1 null pig cells were completely eliminated from the cell membrane.Functionally,GGTA1 knockout cells were protected from complement-mediated immune attacks when incubated with human serum.This study demonstrated that ZFN is an efficient tool in creating gene-modified pigs.GGTA1 null pigs and GGTA1 null fetal fibroblasts would benefit research and pig-to-human transplantation.BAO Lei CHEN HaiDe JONG UiMyong RIM CholHo LI WenLing LIN XiJuan ZHANG Dan LUO Qiong CUI Chun HUANG HeFeng ZHANG Yan XIAO Lei FU ZhiXin 2014Science China(Life Sciences)2014,57,2:9

13First record of Thalassiosira curviseriata Takano (Bacillariophyta) and its bloom in the East China Sea显示文摘A bloom caused by a diatom, Thalassiosira curviseriata Takano (Bacillariophyta), is recorded in the East China Sea for the first time in China during a red tide investigation cruise (MC2005-2) from 27 March to 12 April 2005. This bloom was developed with the competition of Chaetoceros debilis and Skeletonema spp. The highest cell density of T. curviseriata, which has reached 1.27×106 cells/dm3, was found in the surface and middle water layers of Stas ZD, ZB in the East China Sea (27.22°~29.48°N, 121.53°~122.98°E)in early spring in 2005. During the blooming period of T. curviseriata, the population with high cell density was found in the water area with temperature of 10~15 ℃ and salinity of 29.0~33.5. The percentage of the predominant species, T. curviseriata, has reached 95.8% of total diatom cells at one time in the middle water layer. The morphological characteristics of T. curviseriata, were observed with light microscope (LM) and transmission electronic microscope (TEM). The cells are 5.0~12.6 μm in diameter, connecting each other by mucilaginous thread to form spiral and curved chains. Description and LM and TEM images of T. curviseriata are presented. 14Natural selection maintains the transcribed LTR retrotransposons in Nosema bombycis显示文摘Eight intact LTR retrotransposons(Nbr1?Nbr8)have been previously characterized from the genome of Nosema bombycis,a eu-karyotic parasite with a compact and reduced genome.Here we describe six novel transcribed Nbr elements(Nbr9?Nbr14)identified through either cDNA library or RT-PCR.Like previously determined ones,all of them belong to the Ty3/Gypsy superfamily.Retrotransposon diversity and incomplete domains with insertions(Nbr12),deletions(Nbr11)and in-frame stop codons in coding regions(Nbr9)were detected,suggesting that both defective and loss events of LTR retrotransposon have happened in N.bombycis genome.Analysis of selection showed that strong purifying selection acts on all elements except Nbr11.This implies that selective pressure keeps both these Nbrs and their functions in genome.Interestingly,Nbr11 is under positive selection and some positively selected codons were identified,indicating that new functionality might have evolved in the Nbr11 retrotransposon.Unlike other transposable elements,Nbr11 has integrated into a conserved syntenic block and probably resulted in the inversion of both flanking regions.This demonstrates that transposable element is an important factor for the reshuffling and evolution of their host genomes,and may be maintained under natural selection.Heng Xiang Guoqing Pan Ruizhi Zhang Jinshan Xu Tian Li Wenle Li Zeyang Zhou Zhonghuai Xiang 2010Journal of Genetics and Genomics2010,37,5:7
15Development and effectiveness of pseudotyped SARS-CoV-2 system as determined by neutralizing efficiency and entry inhibition test in vitro显示文摘With the development of the COVID-19 epidemic,there is an urgent need to establish a system for determining the effectiveness and neutralizing activity of vaccine candidates in biosafety level 2(BSL-2)facilities.Previously,researchers had developed a pseudotyped virus systemfor SARS-CoV andMERS-CoV,based onHIV-1 core,bearing virus spike protein.During the development of a pseudotyped SARS-CoV-2 system,a eukaryotic expression plasmid expressing SARSCoV-2 spike(S)protein was constructed and then co-transfectedwith HIV-1 based plasmid which containing the firefly luciferase reporter gene,into HEK293T cells to prepare the pseudotyped SARS-CoV-2 virus(ppSARS-2).We have successfully established the pseudotyped SARS-CoV-2 system for neutralization and entry inhibition assays.Huh7.5 cell line was found to be the most susceptible to our pseudotyped virus model.Different levels of neutralizing antibodies were detected in convalescent serum samples of COVID-19 patients using ppSARS-2.The recombinant,soluble,angiotensin-converting enzyme 2 protein was found to inhibit the entry of ppSARS-2 in Huh7.5 cells effectively.Furthermore,the neutralization results for ppSARS-2 were consistent with those of live SARS-CoV-2 and determined using the serum samples fromconvalescent patients.In conclusion,we have developed an easily accessible and reliable tool for studying the neutralizing efficiency of antibodies against SARS-CoV-2 and the entry process of the virus in a BSL-2 laboratory.Ren Yang Baoying Huang Ruhan A Wenhui Li Wenling Wang Yao Deng Wenjie Tan 2020Biosafety and Health2020,2,4:5
16CMTM4 inhibits cell proliferation and migration via AKT, ERK1/2,and STAT3 pathway in colorectal cancer显示文摘CMTM4(CKLF-like MARVEL transmembrane domain containing 4),a potential tumor suppressor gene,is involved in several types of malignancies.It has been reported to be downregulated and exhibit anti-tumorigenic activities by regulating cell growth and cell cycle in clear cell renal cell carcinoma.It has also been identified as a tumor suppressor in hepatocellular carcinoma(HCC),and its negative expression is a risk factor for poor prognosis of HCC patients.In the present study,an integrated bioinformatics analysis based on The Cancer Genome Atlas(TCGA)database showed that CMTM4 was frequently reduced in colorectal cancer(CRC)and high expression of CMTM4 was associated with increased overall survival rates.Based on these findings,we adopted gain-of-function and lost-of-function strategies using SW480 and HT29 CRC cell lines which have relatively low and high endogenous CMTM4 levels,respectively.We observed impeded cell proliferation and migration upon overexpression of CMTM4 in SW480 cells,and the opposite effects were observed upon knockdown of CMTM4 in HT-29 cells.Cell signaling pathways essential for CRC progression were then examined,and the phosphorylation levels of AKT,ERK1/2,and STAT3 were found to be decreased by CMTM4 overexpression in SW480 cells and elevated by CMTM4 silencing in HT29 cells.Their inhibitors were used to validate that the three signaling pathways contributed to the inhibitory effects of CMTM4 on CRC cells.Taken together,our results suggest that CMTM4 plays a tumor suppressive role in CRC.Hui Xue Ting Li Pingzhang Wang Xiaoning Mo Hejun Zhang Shigang Ding Dalong Ma Wenping Lv Jing Zhang Wenling Han 2019Acta Biochimica et Biophysica Sinica2019,51,9:5
17Qualitative and quantitative assessment of related substances in the Compound Ketoconazole and Clobetasol Propionate Cream by HPLC-TOF-MS and HPLC显示文摘Related substances in pharmaceutical formulations are associated with their safety, efficacy and stability. However, there is no overall study already published on the assessment of related substances in the Compound Ketoconazole and Clobetasol Propionate Cream. In this work, a reliable HPLC-TOF-MS qualitative method was developed for the analysis of related substances in this preparation with a quick and easy extraction procedure. Besides the active pharmaceutical ingredients, two compounds named ketoconazole impurity B′ optical isomer and ketoconazole impurity E were identified. Furthermore, a new HPLC method for qualitative and quantitative assessment on related substances and degradation products, which were found in the stability test, was established and validated. The single standard to determine multi-components method was applied in the quantitative analysis, which was an effective way for reducing cost and improving accuracy. 18Tuning Atomically Dispersed Fe Sites in Metal–Organic Frameworks Boosts Peroxidase‑Like Activity for Sensitive Biosensing显示文摘Although nanozymes have been widely developed,accurate design of highly active sites at the atomic level to mimic the electronic and geometrical structure of enzymes and the exploration of underlying mechanisms still face significant challenges.Herein,two functional groups with opposite electron modulation abilities(nitro and amino)were introduced into the metal–organic frameworks(MIL-101(Fe))to tune the atomically dispersed metal sites and thus regulate the enzymelike activity.Notably,the functionalization of nitro can enhance the peroxidase(POD)-like activity of MIL-101(Fe),while the amino is poles apart.Theoretical calculations demonstrate that the introduction of nitro can not only regulate the geometry of adsorbed intermediates but also improve the electronic structure of metal active sites.Benefiting from both geometric and electronic effects,the nitro-functionalized MIL-101(Fe)with a low reaction energy barrier for the HO*formation exhibits a superior POD-like activity.As a concept of the application,a nitro-functionalized MIL-101(Fe)-based biosensor was elaborately applied for the sensitive detection of acetylcholinesterase activity in the range of 0.2–50 mU mL−1 with a limit of detection of 0.14 mU mL−1.Moreover,the detection of organophosphorus pesticides was also achieved.This work not only opens up new prospects for the rational design of highly active nanozymes at the atomic scale but also enhances the performance of nanozyme-based biosensors.Weiqing Xu Yikun Kang Lei Jiao Yu Wu Hongye Yan Jinli Li Wenling Gu Weiyu Song Chengzhou Zhu 2020Nano-Micro Letters2020,12,12:4

A three-dimensional linearly elastic body with a small thickness denoted by 2ε, which is clamped along its part of the lateral boundary and subjected to the regular loads. In the linear case, one can use the two-dimensional models of Ciarlet or Koiter to calculate the displacement for the shell. Some error estimates between the approximate solution of these models and the three-dimensional displacement vector field of a flexural or membrane shell have been obtained.
