| 3 | Multi-layer Quasi Three-dimensional Equivalent Model of Axial-Flux Permanent Magnet Synchronous Machine显示文摘Axial-flux permanent magnet synchronous machine(AFPMSM)enjoys the merits of high torque density and high efficiency,which make it one good candidate in the direct-drive application.The AFPMSM is usually analyzed based on the three-dimensional finite element method(3D FEM)due to its three-dimensional magnetic field distribution.However,the 3D FEM suffers large amount of calculation,time-consuming and is not suitable for the optimization of AFPMSM.Addressing this issue,a multi-layer quasi three-dimensional equivalent model of the AFPMSM is investigated in this paper,which could take the end leakage into consideration.Firstly,the multi-layer quasi three-dimensional equivalent model of the AFPMSM with single stator and single rotor is derived in details,including the equivalent processes and conversions of structure dimensions,motion conditions and electromagnetic parameters.Then,to consider the influence of end leakage on the performance,a correction factor is introduced in the multi-layer quasi three-dimensional equivalent model.Finally,the proposed multi-layer quasi three-dimensional equivalent model is verified by the 3D FEM based on an AFPMSM under different structure parameters.It demonstrates that the errors of flux linkage and average torque obtained by the multi-layer quasi three-dimensional equivalent model and 3D FEM are only around 2%although the structure parameters of the AFPMSM are varied.Besides,the computation time of one case based on the multi-layer quasi three-dimensional equivalent model is only 6 min,which is much less than that of the 3D FEM,1.8 h,under the same conditions.Thus,the proposed multi-layer quasi three-dimensional equivalent model could be used to optimize the AFPMSM and much time could be saved by this method compared with the 3D FEM. | Mingjie He Weiye Li Jun Peng Jiangtao Yang | 2021 | CES Transactions on Electrical Machines and Systems2021,5,1: | 7 |

| 11 | Metabolic reprogramming and redox adaptation in sorafenib-resistant leukemia cells:detected by untargeted metabolomics and stable isotope tracing analysis显示文摘Background:Internal tandem duplications(ITD)within the juxtamembrane domain of FMS-like tyrosine kinase 3(FLT3)represent a poor prognostic indicator in acute myeloid leukemia(AML).Therapeutic benefits of tyrosine kinase inhibitors,such as sorafenib,are limited due to the emergence of drug resistance.While investigations have been con-ducted to improve the understanding of the molecular mechanisms underlying the resistance to this FLT3 inhibitor,a profile of cell functioning at the metabolite level and crosstalk between metabolic pathways has yet to be created.This study aimed to elucidate the alteration of metabolomic profile of leukemia cells resistant to the FLT3 inhibitor.Methods:We established two sorafenib-resistant cell lines carrying FLT3/ITD mutations,namely the murine BaF3/ITD-R and the human MV4-11-R cell lines.We performed a global untargeted metabolomics and stable isotope-labe-ling mass spectrometry analysis to identify the metabolic alterations relevant to the therapeutic resistance.Results:The resistant cells displayed fundamentally rewired metabolic profiles,characterized by a higher demand for glucose,accompanied by a reduction in glucose flux into the pentose phosphate pathway(PPP);and by an increase in oxidative stress,accompanied by an enhanced glutathione synthesis.We demonstrated that the highest scoring network of altered metabolites in resistant cells was related to nucleotide degradation.A stable isotope tracing experiment was performed and the results indicated a decrease in the quantity of glucose entering the PPP in resistant cells.Further experiment suggested that the inhibition of major enzymes in the PPP consist of glucose-6-phosphate dehydrogenase deficiency(G6PD)in the oxidative arm and transketolase(TKT)in the non-oxidative arm.In addition,we observed that chronic treatment with sorafenib resulted in an increased oxidative stress in FLT3/ITD-positive leu-kemia cells,which was accompanied by decreased cell proliferation and an enhanced antioxidant response.Conclusions:Our data regarding comparative metabolomics characterized a distinct metabolic and redox adaptation that may contribute to sorafenib resistance in FLT3/ITD-mutated leukemia cells. | Xin You Weiye Jiang Wenhua Lu Hui Zhang Tiantian Yu Jingyu Tian Shijun Wen Guillermo Garcia-Manero Peng Huang Yumin Hu | 2019 | Cancer Communications2019,39,1: | 1 |

| 17 | A dynamic risk-based early warning monitoring system for population-based management of cardiovascular disease显示文摘Risk prediction tools are crucial for population-based management of cardiovascular disease(CVD).However,most prediction models are currently used to assess long-term risk instead of the risk of short-term CVD onset.We developed a Dynamic Risk-based Early wAming Monitoring(DREAM)system using large-scale,real-time electronic health record data from 2010 to 2020 from the CHinese Electronic health Records Research in Yinzhou study.The dynamic risk scores were derived from a 1:5 matched nested case-control set comprising 70,470 individuals(11,745 CVD events)and then validated in a cohort of 81,205 individuals(5950 CVD events).The individuals were Chinese adults aged 40-79 years without a history of CVD at baseline.Eleven predictors related to vital signs,laboratory tests,and health service utilization were selected to establish the dynamic scores.The proposed scores were significantly associated with the subsequent CVD onset(adjusted odds ratio,1.21;95%confidence interval,1.20-1.23).The area under the receiver operating characteristic curves(AUCs)was 0.6010(0.5929-0.6092)and 0.6021(0.5937-0.6105)for the long-term 10-year CVD risk<10%and≥10%groups in the derivation set,respectively.In the long-term 10-year CVD risk>10%group in the validation set,the change in AUC in addition to the long-term risk was 0.0235(0.0155-0.0315).By increasing the risk threshold from 7 to 16 points,the proportion of true subsequent CVD cases among those given alerts increased from 40.61%to 85.31%.In terms of management efficiency,the number needed to manage per CVD case ranged from 2.46 to 1.17 using the risk scores.With the increasing popularity and integration of EHR systems with wearable technology,the DREAM scores can be incorporated into an early-warning system and applied in dynamic,real-time,EHR-based,automated management to support healthcare decision making for individuals,general practitioners,and policymakers. | Xiaofei Liu Qianqian Li Weiye Chen Peng Shen Yexiang Sun Qi Chen Jinguo Wu Jingyi Zhang Ping Lu Hongbo Lin Xun Tang Pei Gao | 2021 | Fundamental Research2021,1,5: | 1 |
