2Chemotherapy initiation with single-course methotrexate alone or combined with dactinomycin versus multi-course methotrexate for low-risk gestational trophoblastic neoplasia: a multi-centric randomized clinical trial显示文摘We aimed to evaluate the effectiveness and safety of single-course initial regimens in patients with low-risk gestational trophoblastic neoplasia(GTN).In this trial (NCT01823315),276 patients were analyzed.Patients were allocated to three initiated regimens:single-course methotrexate(MTX),single-course MTX+dactinomycin(ACTD),and multi-course MTX(control arm).The primary endpoint was the complete remission(CR)rate by initial drug(s).The primary CR rate was 64.4%with multi-course MTX in the control arm.For the single-course MTX arm,the CR rate was 35.8%by one course;it increased to 59.3%after subsequent multi-course MTX,with non-inferiority to the control(difference-5.1%,95%confidence interval(CI)-19.4%to 9.2%,P=0.014).After further treatment with multi-course ACTD,the CR rate(93.3%)was similar to that of the control(95.2%,P=0.577).For the single-course MTX+ACTD arm,the CR rate was 46.7%by one course,which increased to 89.1%after subsequent multi-course,with non-inferiority(difference 24.7%,95%CI 12.8%-36.6%,P<0.001)to the control.It was similar to the CR rate by MTX and further ACTD in the control arm(89.1%vs.95.2%,P=0.135).Four patients experienced recurrence,with no death,during the 2-year follow-up.We demonstrated that chemotherapy initiation with single-course MTX may be an alternative regimen for patients with low-risk GTN.Lili Chen Ling Xi Jie Jiang Rutie Yin Pengpeng Qu Xiuqin Li Xiaoyun Wan Yaxia Chen Dongxiao Hu Yuyan Mao Zimin Pan Xiaodong Cheng Xinyu Wang Qingli Li Danhui Weng Xi Zhang Hong Zhang Quanhong Ping Xiaomei Liu Xing Xie Beihua Kong Ding Ma Weiguo Lu 2022Frontiers of Medicine2022,16,2:2
3Vegetation and Soil Characteristics of Different Desertification Grasslands in Northwest Sichuan显示文摘[Objective]The paper was to study vegetation and soil characteristics of different desertification grasslands in northwest Sichuan. [Method]By taking different desertification grasslands as the research object,the characteristic factors of vegetation community,biomass,soil moisture content,volume weight and porosity were analyzed through scientific investigation,sampling and formula calculation to reveal the changes in vegetation and soil characteristics of different desertification grasslands in Northwest Sichuan. [Result]Community succession presented the pattern of ' hygrophyte-mesophyte-xerophyte' with the aggravation of grassland desertification. The height and coverage of community decreased,species richness was declined by 88%,and composition of dominant species also changed greatly. The diversity index of light-desertification grassland was the highest among tested grasslands. Total biomass was decreased by 90. 4%,and the underground biomass decreased far more than aboveground biomass. In desertification progress,both soil moisture content and water holding capacity decreased,while volume weight showed upward trend and porosity showed downward trend; soil characteristics had large variation in early stage of desertification,so restoration treatment of desertification grassland should be carried out in the early stage of desertification. 4Flexible,stretchable,and transparent InGaN/GaN multiple quantum wells/polyacrylamide hydrogel-based light emitting diodes显示文摘Visualization is a direct,efficient,and simple interface method to realize the interaction between human and machine,whereas the flexible display unit,as the major bottleneck,still deeply hinders the advances of wearable and virtual reality devices.To obtain flexible optoelectronic devices,one of the effective methods is to transfer a high-efficient and long-lifetime inorganic optoelectronic film from its rigid epitaxial substrate to a foreign flexible/soft substrate.Additionally,piezo-phototronic effect is a fundamental theory for guiding the design of flexible optoelectronic devices.Herein,we demonstrate a flexible,stretchable,and transparent InGaN/GaN multiple quantum wells(MQWs)/polyacrylamide(PAAM)hydrogel-based light emitting diode coupling with the piezo-phototronic effect.The quantum well energy band and integrated luminous intensity(increased by more than 31.3%)are significantly modulated by external mechanical stimuli in the device.Benefiting from the small Young's modulus of hydrogel and weak Van der Waals force,the composite film can endure an extreme tensile condition of about 21.1%stretching with negligible tensile strains transmitted to the InGaN/GaN MQWs.And the stable photoluminescence characteristics can be observed.Moreover,the hydrogen-bond adsorption and excellent transparency of the hydrogel substrate greatly facilitate the packaging and luminescence of the optoelectronic device.And thus,such a novel integration scheme of inorganic semiconductor materials and organic hydrogel materials would help to guide the robust stretchable optoelectronic devices,and show great potential in emerging wearable devices and virtual reality applications.Jiwei Chen Jiangwen Wang Keyu Ji Bing Jiang Xiao Cui Wei Sha Bingjun Wang Xinhuan Dai Qilin Hua Lingyu Wan Weiguo Hu 2022Nano Research2022,15,6:1

11Adjuvant chemotherapy versus adjuvant concurrent chemoradiotherapy after radical surgery for early-stage cervical cancer:a randomized,non-inferiority,multicenter trial显示文摘We conducted a prospective study to assess the non-inferiority of adjuvant chemotherapy alone versus adjuvant concurrent chemoradiotherapy (CCRT) as an alternative strategy for patients with early-stage (FIGO 2009 stage IB–IIA) cervical cancer having risk factors after surgery. The condition was assessed in terms of prognosis, adverse effects, and quality of life. This randomized trial involved nine centers across China. Eligible patients were randomized to receive adjuvant chemotherapy or CCRT after surgery. The primary end-point was progression-free survival (PFS). From December 2012 to December 2014, 337 patients were subjected to randomization. Final analysis included 329 patients, including 165 in the adjuvant chemotherapy group and 164 in the adjuvant CCRT group. The median follow-up was 72.1 months. The three-year PFS rates were both 91.9%, and the five-year OS was 90.6% versus 90.0% in adjuvant chemotherapy and CCRT groups, respectively. No significant differences were observed in the PFS or OS between groups. The adjusted HR for PFS was 0.854 (95% confidence interval 0.415–1.757;P = 0.667) favoring adjuvant chemotherapy, excluding the predefined non-inferiority boundary of 1.9. The chemotherapy group showed a tendency toward good quality of life. In comparison with post-operative adjuvant CCRT, adjuvant chemotherapy treatment showed non-inferior efficacy in patients with early-stage cervical cancer having pathological risk factors. 12SGK and 14-3-3 protein are involved in the serine/threonine phosphorylation mechanism for TPO/MPL signal transduction显示文摘Thrombopioetin (TPO), the critical regulator of platelet production, acts by binding to its cell surface receptor, c-Mpl. Yeast two-hybrid screening was performed to isolate the proteins interacting with the cytoplasmic domain of c-Mpl. 48 positive clones were isolated from 5 xxxxxxxxxx106 independent transformants. The results of sequence analysis demonstrate that they represent 13 different protein encoding sequences. Among them there are a partial coding sequence of serine/threonine protein kinase SGK (serum and glucocorticoid-inducible kinase ) and 14-3-3 theta protein partial coding sequence. GST-pull-down assay and co-im-munoprecipitation in mammal cells have confirmed the interaction between these two proteins and c-Mpl. By constructing a series of deleted c-Mpl cytoplasmic domain, the interaction region in c-Mpl cytoplasmic tail was localized in amino acids 523-554. At the same time, the directed interaction between SGK and 14-3-3 proteins also has been verified by yeast two-hybrid assay. 13A Common Functional Variant at the Enhancer of the Rheumatoid Arthritis Risk Gene ORMDL3 Regulates its Expression Through Allele-Specific JunD Binding显示文摘Genome-wide association studies(GWASs)have identified over 100 loci associated with rheumatoid arthritis(RA);how-ever,the functionally affected genes and the underlying molecular mechanisms contributing to these associations are often unknown.In this study,we conducted an integrative genomic analysis incorporating multiple“omics”data and identified a functional regulatory DNA variant,rs56199421,and a plausible mechanism by which it regulates the expression of a puta-tive RA risk gene,ORMDL Sphingolipid Biosynthesis Regulator 3(ORMDL3).The T allele of rs56199421,located in the enhancer region of ORMDL3,exhibited stronger direct binding ability than the other C allele of rs56199421 did in vitro with the transcription factor JunD and demonstrated higher transcriptional activity.Moreover,the T allele of rs56199421 is associated with elevated RA risk,and ORMDL3 expression is increased in RA patients.Thus,these findings suggest that the T allele of rs56199421 enhances JunD transcription factor binding,increases enhancer activity,and elevates the expression of the RA risk gene ORMDL3.Wenjing Ye Yiyun Yu Xiaoxia Zhu Weiguo Wan Yun Liu Hejian Zou Zaihua Zhu 2023Phenomics2023,3,5:0
