3Clinical and genetic findings in a Chinese family with VDR-associated hereditary vitamin D-resistant rickets显示文摘Hereditary vitamin D-resistant rickets(HVDRR) is a rare autosomal recessive disorder characterized by severe rickets,hypocalcemia,hypophosphatemia,secondary hyperparathyroidism,and elevated alkaline phosphatase.This disorder is caused by homogeneous or heterogeneous mutations affecting the function of the vitamin D receptor(VDR),which lead to complete or partial target organ resistance to the action of 1,25-dihydroxy vitamin D.A non-consanguineous family of Chinese Han origin with one affected individual demonstrating HVDRR was recruited,with the proband evaluated clinically,biochemically and radiographically.To identify the presence of mutations in the VDR gene,all the exons and exon–intron junctions of the VDR gene from all family members were amplified using PCR and sequenced.The proband showed rickets,progressive alopecia,hypocalcemia,hypophosphatemia,secondary hyperparathyroidism,and elevated alkaline phosphatase.She also suffered from epilepsy,which is rarely seen in patients with HVDRR.Direct sequencing analysis revealed a homozygous missense mutation c.122G4A(p.C41Y) in the VDR gene of the proband,which is located in the first zinc finger of the DNA-binding domain.Both parents had a normal phenotype and were found to be heterozygous for this mutation.We report a Chinese Han family with one individual affected with HVDRR.A homozygous missense mutation c.122G4A(p.C41Y) in the VDR gene was found to be responsible for the patient's syndrome.In contrast to the results of treatment of HVDRR in other patients,our patient responded well to a supplement of oral calcium and a low dose of calcitriol.Qianqian Pang Xuan Qi Yan Jiang Ou Wang Mei Li Xiaoping Xing Jin Dong Weibo Xia 2016Bone Research2016,4,1:9
4FGF23 and Phosphate Wasting Disorders显示文摘A decade ago,only two hormones,parathyroid hormone and 1,25(OH)2D,were widely recognized to direct-ly affect phosphate homeostasis.Since the discovery of fibroblast growth factor 23(FGF23) in 2000(1),our understanding of the mechanisms of phosphate homeostasis and of bone mineralization has grown exponentially.FGF23 is the link between intestine,bone,and kidney together in phosphate regulation.However,we still do not know the complex mechanism of phosphate homeostasis and bone mineralization.The physiological role of FGF23 is to regulate serum phosphate.Secreted mainly by osteocytes and osteo-blasts in the skeleton(2-3),it modulates kidney handling of phosphate reabsorption and calcitriol produc-tion.Genetic and acquired abnormalities in FGF23 structure and metabolism cause conditions of either hyper-FGF23 or hypo-FGF23.Hyper-FGF23 is related to hypophosphatemia,while hypo-FGF23 is related to hyperphosphatemia.Both hyper-FGF23 and hypo-FGF23 are detrimentalto humans.In this review,we will discuss the pathophysiology of FGF23 and hyper-FGF23 related renal phosphate wasting disorders(4).Xianglan Huang Yan Jiang Weibo Xia 2013Bone Research2013,1,2:9

12Three-Dimensional Water Quality Model Based on FVCOM for Total Load Control Management in Guan River Estuary,Northern Jiangsu Province显示文摘Guan River Estuary and adjacent coastal area(GREC) suffer from serious pollution and eutrophicational problems over the recent years.Thus,reducing the land-based load through the national pollutant total load control program and developing hydrodynamic and water quality models that can simulate the complex circulation and water quality kinetics within the system,including longitudinal and lateral variations in nutrient and COD concentrations,is a matter of urgency.In this study,a three-dimensional,hydrodynamic,water quality model was developed in GREC,Northern Jiangsu Province.The complex three-dimensional hydrodynamics of GREC were modeled using the unstructured-grid,finite-volume,free-surface,primitive equation coastal ocean circulation model(FVCOM).The water quality model was adapted from the mesocosm nutrients dynamic model in the south Yellow Sea and considers eight compartments:dissolved inorganic nitrogen,soluble reactive phosphorus(SRP),phytoplankton,zooplankton,detritus,dissolved organic nitrogen(DON),dissolved organic phosphorus(DOP),and chemical oxygen demand.The hydrodynamic and water quality models were calibrated and confirmed for 2012 and 2013.A comparison of the model simulations with extensive dataset shows that the models accurately simulate the longitudinal distribution of the hydrodynamics and water quality.The model can be used for total load control management to improve water quality in this area.ZHANG Li LIN Weibo LI Keqiang SHENG Jianming WEI Aihong LUO Feng WANG Yan WANG Xiulin ZHANG Longjun 2016Journal of Ocean University of China2016,15,2:1
13From oral structure to molecular evidence:new insights into the evolutionary phylogeny of the ciliate order Sessilida(Protista,Ciliophora),with the establishment of two new families and new contributions to the poorly studied family Vaginicolidae显示文摘Ciliated protists represent one of the most primitive and diverse lineages of eukaryotes,with nuclear dimorphism,a distinctive sexual process(conjugation),and extensive genome rearrangements.Among divergent ciliate lineages,the peritrich order Sessilida includes members with a colonial lifestyle,which may hint to an independent evolutionary attempt for multicellularity,although they are still single-celled organisms.To date,the evolution and phylogeny of this group are still far from clear,in part due to the paucity of molecular and/or morphological data for many taxa.In this study,we extend taxon sampling of a loricate group of sessilids by obtaining 69 new r DNA(SSU r DNA,ITS1-5.8S rDNA-ITS2,and LSU r DNA)sequences from 20 wellcharacterized representative species and analyze the phylogenetic relationships within Sessilida.The main findings are:(i)the genera Rhabdostyla and Campanella each represents a unique taxon at family level,supporting the establishment of two new families,i.e.,Rhabdostylidae n.fam.and Campanellidae n.fam.,respectively,the former being sister to a morphologically heterogeneous clade comprising Astylozoidae and several incertae sedis species and the latter occupying the basal position within the Sessilida clade;(ii)the structure of infundibular polykinety 3 is likely to be a phylogenetically informative character for resolving evolutionary relationships among sessilids;(iii)differences between sparsely and the densely arranged silverline systems could be a suprageneric taxonomic character;(iv)the monophyly of Vaginicolidae is confirmed,which is consistent with its specialized morphology,i.e.,the possession of a typical peritrich lorica which might be an apomorphy for this group;(v)within Vaginicolidae,the monotypic Cothurniopsis sensu Stokes,1893 is a synonym of Cothurnia Ehrenberg,1831,and a new combination is created,i.e.,Cothurnia valvata nov.comb.;(vi)Vaginicola sensu lato comprises at least two distinctly divergent clades,one affiliated with Thuricola and the other with a systematically puzzling clade represented by Vaginicola tincta.Borong Lu Xiaozhong Hu Alan Warren Weibo Song Ying Yan 2023Science China(Life Sciences)2023,66,7:1

18Chromosome organization and gene expansion in the highly fragmented genome of the ciliate Strombidium stylifer显示文摘Chromosomes are well-organized carriers of genetic information in eukaryotes and are usually quite long,carrying hundreds and thousands of genes.Intriguingly,a clade of single-celled ciliates,Spirotrichea,feature nanochromosomes—also called“gene-sized chromosomes”.These chromosomes predominantly carry only one gene,flanked by short telomere sequences.However,the organization and copy number variation of the chromosomes in these highly fragmented genomes remain unexplored in many groups of Spirotrichea,including the marine Strombidium.Using deep genome sequencing,we assembled the macronuclear genome of Strombidium stylifer into more than 18,000 nanochromosomes(~2.4 Kb long on average).Our results show that S.stylifer occupies an intermediate position during the evolutionary history of Strombidium lineage and experienced significant expansions in several gene families related to guanyl ribonucleotide binding.Based on the nucleotide distribution bias analysis and conserved motifs search in non-genic regions,we found that the subtelomeric regions have a conserved adenine-thymine(AT)-rich sequence motif.We also found that the copy number of nanochromosomes lacks precise regulation.This work sheds light on the unique features of chromosome structure in eukaryotes with highly fragmented genomes and reveals that a rather specialized evolutionary strategy at the genomic level has resulted in great diversity within the ciliated lineages.Chao Li Xiao Chen Weibo Zheng Thomas G.Doak Guangyi Fan Weibo Song Ying Yan 2021Journal of Genetics and Genomics2021,48,10:1
