| 2 | Biocompatibility and Immunotoxicology of the Preclinical Implantation of a Collagen-based Artificial Dermal Regeneration Matrix显示文摘Objective Graft rejection, with the possibility of a violent immune response, may be severe and life threatening. Our aim was to thoroughly investigate the biocompatibility and immunotoxicology of collagen-based dermal matrix(DM) before assessment in clinical trials. Methods DM was subcutaneously implanted in BALB/c mice in two doses to induce a potential immune response. The spleen and lymph nodes were assessed for shape, cell number, cell phenotype via flow cytometry, cell activation via CCK8 kit, Annexin V kit, and Ki67 immunostaining. Serum samples were used to measure antibody concentration by enzyme-linked immunosorbent assay. Local inflammation was analyzed by histology and immunohistochemistry staining. Data analysis was performed by one-way ANOVA and non-parametric tests. Results Our data illustrate that the spleen and lymph node sizes were similar between the negative control mice and mice implanted with DM. However, in the high-dose DM(DM-H) group, the total cell populations in the spleen and lymph nodes, T cells and B cells in the spleen had slight increases in prophase, and the low-dose DM(DM-L) group did not display gross abnormities. Moreover, DM-H initiated moderate cell activation and proliferation in the early phase post-immunization, whereas DM-L did not. Neither DM-H nor DM-L implantation noticeably increased IgM and IgG serum concentrations. Examination of the local cellular response revealed only benign cell infiltration and TNF-α expression in slides of DM in the early phase. Conclusion Overall, DM-H may have induced a benign temporary acute immune response post-implantation, whereas DM-L had quite low immunogenicity. | 4 | Biosynthesis of Triacylglycerol Molecules with a Tailored PUFA Profile in Industrial Microalgae显示文摘The composition of polyun saturated fatty acids (PUFAs) in triacylglycerols (TAGs) is key to health ben efits and for oil applications, yet the underlying genetic mechanism remains poorly understood. In this study, by in si Iico, ex vivo, and in vivo profili ng of type-2 diacylglycerol acyltransferases (DGAT2s) in Nannochloropsis oceanica we revealed two novel PUFA-preferring enzymes that discriminate individual PUFA species in TAG assembly, with NoDGAT2J for linoleic acid (LA) and NoDGAT2K for eicosapentaenoic acid (EPA). The LA and EPA composition of TAG molecules is mediated in vivo via the functional partitioning between NoDGAT2J and 2K, both of which are localized in the chloroplast envelope. By modulating transcript abundance of the DGAT2s, an N. oceanica strain bank was created, where proportions of LA and EPA in TAG vary by 18.7-fold (between 0.21% and 3.92% dry weight) and 34.7-fold (between 0.09% and 3.12% dry weight), respective!y. | 7 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 10 | Association of choroidal thickness with early stages of diabetic retinopathy in type 2 diabetes显示文摘AIM:To assess the correlation between choroidal thickness(CT) and the early stages of diabetic retinopathy(DR) in type 2 diabetic patients.METHODS:We divided 83 diabetic patients(51-80 years of age;50 females) into non diabetic retinopathy group(NDR) and mild/moderate nonproliferative diabetic retinopathy(NPDR) group,and compared them with 26 non-diabetic control subjects(51-78 years of age;16 females).Subfoveal choroidal thickness(SFCT) and parafoveal choroidal thickness(PFCT) were measured using enhanced depth imaging spectral-domain optical coherence tomography(EDI-OCT).Ocular health status,disease duration,body mass index,and hemoglobin A1c(Hb A1c) were recorded.RESULTS:The mean ages of the NDR,NPDR,and control groups were 68.0±6.9y,67.8±6.4y,and 65.1±6.3y,respectively(P=0.17).Pearson correlation of the right and left eyes for the control subjects was 0.95 and for the NDR subjects was 0.93.SFCT for the right eyes of the controls was 252.77± 41.10 μm,which was significantly thicker than that of the right eyes in NDR group(221.51±46.56 μm) and the worse eyes of the NPDR group(207.18±61.87 μm;ANOVA,P<0.01).In the diabetic patients pooled together,age was the only variable significantly associated with SFCT(multiple linear regression analysis,P=0.01).CONCLUSION:CT decreased significantly in the NDR and mild/moderate NPDR eyes compared with the control eyes.Age is significantly associated with SFCT in the diabetic patients.Diabetic choroidopathy may be present before clinical retinopathy. | Zhi-Jun Shen Xiu-Fen Yang Jun Xu Chong-Yang She Wen-Wen Wei Wan-Lu Zhu Ning-Pu Liu | 2017 | International Journal of Ophthalmology(English edition)2017,10,4: | 4 |

| 12 | CPI-17-mediated contraction of vascular smooth muscle is essential for the development of hypertension in obese mice显示文摘Several factors have been implicated in obesity-related hypertension, but the genesis of the hypertension is largely unknown. In this study, we found a significantly upregulated expression of CPI-17(C-kinasepotentiated protein phosphatase 1 inhibitor of 17 kDa) and protein kinase C(PKC) isoforms in the vascular smooth muscles of high-fat diet(HFD)-fed obese mice. The obese wild-type mice showed a significant elevation of blood pressure and enhanced calcium-sensitized contraction of vascular smooth muscles. However, the obese CPI-17-deficient mice showed a normotensive blood pressure, and the calcium-sensitized contraction was consistently reduced. In addition, the mutant muscle displayed an abolished responsive force to a PKC activator and a 30%-50% reduction in both the initial peak force and sustained force in response to various G protein-coupled receptor(GPCR) agonists. Our observations showed that CPI-17-mediated calcium sensitization is mediated through a GPCR/PKC/CPI-17/MLCP/RLC signaling pathway. | 13 | The Termination and Aftermath of the Lomagundi -Jatuli Carbon Isotope Excursions in the Paleoproterozoic Hutuo Group, North China显示文摘The Lomagundi-Jatuli Event(LJE) is one of the largest and earliest positive carbon isotope excursions preserving δ^(13)C_(carb) values between +5 and +16‰ in Paleoproterozoic carbonates worldwide. However, the duration, amplitude and patterns of these excursions remain poorly constrained. The 2.14–1.83 Ga Hutuo Group in the North China Craton is a >10 km thick volcano-sedimentary sequence, including >5 km thick well-preserved carbonates that were deposited in supra-tidal to sub-tidal environments. C-O isotopic and elemental analyses of 152 least altered samples of the carbonates revealed a three-stage δ^(13)C evolution. It began with an exclusively positive δ^(13)C_(carb)(+1.3 to + 3.4‰) stage in the ~2.1 Ga carbonate in the Dashiling and Qingshicun Formations, followed by a transition from positive values to oscillating positive and negative values in ~3 000 m thick carbonates of the Wenshan, Hebiancun, Jianancun, and Daguandong Formations, and end with exclusively negative δ^(13)C_(carb) values preserved in > 500 m thick dolostones of the Huaiyincun and Beidaxing Formations. It appears that much of the LJE, particularly those extremely positive δ^(13)C_(carb) signals, was not recorded in the Hutuo carbonates. The exclusively positive δ^(13)C_(carb) values(+1.3 to + 3.4‰) preserved in the lower formations likely correspond to the end of the LJE, whereas the subsequent two stages reflect the aftermath of the LJE and the onset of Shunga-Francevillian event(SFE). | 15 | Characterization of Surface Hardness and Microstructure of High Performance Concrete显示文摘The relationship between compressive strength obtained by universal testing machine and rebound value obtained by the hammer of high performance concrete was systematically investigated at the macro level. And a model of high performance concrete strength curve was established from them. At the micro level, the microstructure, hydration products and pore structure of concrete surface were analyzed by scanning electron microscopy(SEM), comprehensive thermal analysis(TG-DSC) and mercury intrusion porosimetry(MIP), respectively. The effect of carbonation on surface strength was also investigated. The results showed that the concrete surface hardness layer grew rapidly at early stage and then stabilized at last with ongoing curing age; the rebound value and compressive strength of concrete with slag were higher than those of concrete with the same content of fly ash. In addition, the strength curve obtained by the least square method can satisfy the local standard requirements with an average relative error of 8.9% and a relative standard deviation of 11.3%. In particular, two types of chemical admixtures were tested, i e, defoamer(tributyl phosphate(TBP)) and polycarboxylate superplasticizer(PS). The results indicate that the addition of TBP and PS eliminates big bubbles and promotes small non-connected pores forming in matrix. Besides, an optimum dosage of TBP and PS may be determined with respect to the frost resistance, permeability and fatigue resistance of CA mortar.
