| 1 | Clinicopathologic features and treatment efficacy of Chinese patients with BRAF-mutated metastatic colorectal cancer: a retrospective observational study显示文摘Background: The prognostic role of the V600 E mutation of v-raf murine sarcoma viral oncogene homolog B1(BRAF)in metastatic colorectal cancer(mCRC) is well established, but the therapeutic regimen targeting this disease is lacking. This study aimed to analyze the clinicopathologic features of and treatment efficacy of commonly used regimens on BRAF-mutated mCRCs.Methods: We collected and reviewed the medical records of mCRC patients treated at Peking University Cancer Hospital & Institute(Beijing, China) between July 2011 and July 2016. Kirsten rat sarcoma viral oncogene homolog(KRAS),neuroblastoma RAS viral oncogene homolog(NRAS), and BRAF mutational status was assayed using direct sequencing. The details of clinicopathologic characteristics of patients and their responses to FOLFOXIRI regimen or standard therapy were obtained by reviewing the medical records. The progression-free survival(PFS) and overall survival(OS)were assessed using Kaplan-Meier analysis and compared using the log-rank test.Results: Of 1694 patients studied, 75 had BRAF exon 15 mutations. Of these 75 patients, 71 had V600 E mutation, 1 had D594 G mutation, 2 had K601 E mutation, and 1 had a novel T599_V600 insAGA alteration. No patients had KRAS or NRAS mutations. Of 63 patients with BRAFV600 E-mutated mCRC and sufficient clinical data, 27(42.9%) had right-sided colon tumors, 19(30.2%) had left-sided colon tumors, and 17(26.9%) had rectal tumors; 26(41.3%) had peritoneal metastases, and 50(79.4%) had distant lymph node metastases.The patients with BRAF K601 E-and T599_V600 insAGA-mutated tumors had similar clinicopathologic features to those with BRAFV600 E-mutated tumors. Patients with the BRAFV600 E mutation benefited more from FOLFOXIRI regimen compared with patients who underwent standard therapy(overall response rate 83.3% vs. 14.0%; median PFS 6.4 months vs. 2.8 months, P = 0.220; median OS11.0 months vs. 6.9 months, P = 0.048).Conclusions: BRAFV600 E mutations were commonly identified in right-sided tumors and showed a high incidence of peritoneal and distant lymph nodes metastases.This subtype of mCRC was characterized by short OS and unique patterns of metastasis. | 2 | Bake hardening behavior of TRIP and DP steels显示文摘The bake hardening (BH) behavior of transformation-induced plasticity (TRIP) and dual-phase (DP) steels after different prestrains was studied. The experimental results indicate that TRIP steel exhibits good BH ability while DP steel does not, and pre-strain displays a strong effect on the BH values of both steels. The comparison of microstructures of the two steels showed that the hard second phase in the matrix might be harmful to the BH ability. | 3 | Multi-scale finite element simulation on large deformation behavior of wood under axial and transverse compression conditions显示文摘Multi-scale finite element method is adopted to simulate wood compression behavior under axial and transverse loading. Representative volume elements (RVE) of wood microfibril and cell are proposed to analyze orthotropic mechanical behavior. Lignin, hemicellulose and crystalline-amorphous cellulose core of spruce are concerned in spruce nanoscale model. The equivalent elastic modulus and yield strength of the microfibril are gained by the RVE simulation. The anisotropism of the crystalline-amorphous cellulose core brings the microfibril buckling deformation during compression loading. The failure mechanism of the cell-wall under axial compression is related to the distribution of amorphous cellulose and crystalline cellulose. According to the spruce cell observation by scanning electron microscope, numerical model of spruce cell is established using simplified circular hole and regular hexagon arrangement respectively. Axial and transverse compression loadings are taken into account in the numerical simulations. It indicates that the compression stress-strain curves of the numerical simulation are consistent with the experimental results. The wood microstructure arrangement has an important effect on the stress plateau during compression process. Cell-wall buckling in axial compression induces the stress value drops rapidly. The wide stress plateau duration means wood is with large energy dissipation under a low stress level. The numerical results show that loading velocity affects greatly wood microstructure failure modes in axial loading. For low velocity axial compression, shear sliding is the main failure mode. For high velocity axial compression, wood occur fold and collapse. | 9 | QoS-awared replica placement techniques in data grid applications显示文摘Quality of service is one of the most important factors for successful grid applications. However, individual QoS constraint (IQC) and QoS-awared replica placement have not yet been well addressed in the existing data grid. Existing QoS-awared replica placement algorithms neither can support multiple QoS attributes, nor have good scalability. In this paper, a three-phrase QoS-awared replica placement algorithm based on matrix, namely TP-GABMAC, is presented to solve these problems. Furthermore, a replica ring mechanism is introduced to maintain replica consistency and manage update conflicting. | 15 | Alterations in DNA damage response and repair genes as potential biomarkers for immune checkpoint blockade in gastrointestinal cancer显示文摘Objective:Immune checkpoint inhibitors(ICIs)have achieved remarkable results in cancer treatments.However,there is no effective predictive biomarker for gastrointestinal(GI)cancer.Methods:We conducted integrative analyses of the genomic and survival data of ICI-treated GI cancer patients from the Memorial Sloan Kettering Cancer Center cohort(MSK-GI,n=227),the Janjigian cohort(n=40),and the Peking University Cancer Hospital&Institute cohort(PUCH,n=80)to determine the possible associations between DNA damage response and repair(DDR)gene mutations and clinical outcomes.Data from The Cancer Genome Atlas database were analyzed to determine the possible correlations between DDR gene mutations and the tumor microenvironment.Results:In the MSK cohort,the presence of≥2 DDR gene mutations was correlated with prolonged overall survival(OS).The Janjigian and PUCH cohorts further confirmed that subgroups with≥2 DDR gene mutations displayed a prolonged OS and a higher durable clinical benefit.Furthermore,the DDR gene mutation load could be considered as an independent prognostic factor,and exhibited a potential predictive value for survival in GI cancer patients treated with ICIs.Mechanistically,we showed that the presence of≥2 DDR gene mutations was correlated with higher levels of tumor mutation burden,neoantigen,and T cell infiltration.Conclusions:The DDR gene mutation status was correlated with favorable clinical outcomes in GI cancer patients receiving ICIs,which could serve as a potential biomarker to guide patient selection for immunotherapy. | Yujiao Wang Xi Jiao Shuang Li Huan Chen Xin Wei Chang Liu Jifang Gong Xiaotian Zhang Xicheng Wang Zhi Peng Changsong Qi Zhenghang Wang Yanni Wang Na Zhuo Jianling Zou Henghui Zhang Jian Li Lin Shen Zhihao Lu | 2022 | Cancer Biology & Medicine2022,19,8: | 0 |
