| 1 | Statistical design and evaluation of a propranolol HCl gastric floating tablet显示文摘The purpose of this research was to apply statistical design for the preparation of a gastric floating tablet(GFT)of propranolol HCl and to investigate the effect of formulation variables on drug release and the buoyancy properties of the delivery system.The contents of polyethylene oxide(PEO)WSR coagulant and sodium bicarbonate were used as independent variables in central composite design of the best formulation.Main effects and interaction terms of the formulation variables were evaluated quantitatively using a mathematical model approach showing that both independent variables have significant effects on floating lag time,%drug release at 1 h(D_(1h))and time required to release 90%of the drug(t_(90)).The desired function was used to optimize the response variables,each with a different target,and the observed responses were in good agreement with the experimental values.FTIR and DSC studies of the statistically optimized formulation revealed there was no chemical interaction between drug and polymer.The statistically optimized formulation released drug according to first order kinetics with a non-Fickian diffusion mechanism.Evaluation of the optimized formulation in vivo in human volunteers showed that the GFT was buoyant in gastric fluid and that its gastric residence time was enhanced in the fed but not the fasted state. | Meka Venkata Srikanth Nali Sreenivasa Rao Songa Ambedkar Sunil Battu Janaki Ram Venkata Ramana Murthy Kolapalli | 2012 | Acta Pharmaceutica Sinica B2012,2,1: | 6 |
| 2 | Life style modification improves insulin resistance and liver histology in patients with non-alcoholic fatty liver disease显示文摘AIM: To study the effect of regular aerobic exercise on insulin resistance, serum aminotransferase and liver histology in nonalcoholic fatty liver disease (NAFLD) patients. METHODS: Sixty (mean age 40.0 ± 8.5 years, 75% male) NAFLD patients were included in the study. After baseline anthropometric measurement i.e., body mass index (BMI), waist circumference (WC); all patients were advised regular aerobic exercise for 30 min/d, for at least 5 d/wk and trained to achieve around 70% of his maximal heart rate. In addition, moderately energy restricted diet was advised to patients with high BMI (> 25 kg/m 2 ). Monthly follow up was done by measuring BMI, WC, aspartate aminotransferase, and alanine aminotransferase (ALT). Insulin resistance was calculated using homeostasis model assessment (HOMA) of insulin resistance (HOMA-IR) model, at baseline and after 6mo. Insulin resistance was arbitrarily considered altered when it was ≥ 2. Liver biopsy was done in a section of patients at baseline and after 6 mo. RESULTS: Seventy percent (42/60) patients were overweight or obese; 95% (57/60) had central obesity (WC > 90 cm in men, > 80 cm in women). In the 45 exercise compliant patients insulin resistance decreased from 6.4 ± 6.1 to 1.3 ± 1.0, BMI from 26.7 ± 3.3 kg/m 2 to 25.0 ± 3.3 kg/m 2 , WC from 95.7 ± 8.9 cm to 90.8 ± 7.3 cm and ALT from 84.8 ± 43.5 U/L to 41.3 ± 18.2 U/L (P < 0.01). In 15 exercise noncompliant patient's insulin resistance, BMI, WC and ALT did not show significant change at 6 mo follow up. Six of 8 patients in compliant group on repeat liver biopsy showed significant change in steatosis and necroinflammation. Nonalcoholic steatohepatitis scores improved form 5.3 ± 1.5 to 3.35 ± 1.5. The decline in insulin resistance correlated with decline in ALT (P = 0.01, r s = 0.90) and liver histology (P = 0.03, r s = 0.73). | 4 | Physical and degradation properties of PLGA scaffolds fabricated by salt fusion technique显示文摘Tissue engineering scaffolds require a controlled pore size and interconnected pore structures to support the host tissue growth. In the present study, three dimensional (3D) hybrid scaffolds of poly lactic acid (PLA) and poly glycolic acid (PGA) were fabricated using solvent casting/particulate leaching. In this case, partially fused NaCl particles were used as porogen (200-300μ) to improve the overall porosity (≥90%) and internal texture of scaffolds. Differential scanning calorimeter (DSC) analysis of these porous scaffolds revealed a gradual reduction in glass transition temperature (Tg) (from 48°C to 42.5°C) with increase in hydrophilic PGA content. The potential applications of these scaffolds as implants were further tested for their biocompatibility and biodegradability in four simulated body fluid (SBF) types in vitro. Whereas, simulated body fluid (SBF) Type1 with the optimal amount of HCO 3 ions was found to be more appropriate and sensible for testing the bioactivity of scaffolds. Among three combinations of polymer scaffolds, sample B with a ratio of 75:25 of PLA: PGA showed greater stability in body fluids (pH 7.2) with an optimum degradation rate (9% to 12% approx). | 5 | Effect of Commiphora mukul gum resin on hepatic marker enzymes,lipid peroxidation and antioxidants status in pancreas and heart of streptozotocin induced diabetic rats显示文摘Objective:To study the antioxidant efficacy of Commiphora mukul(C.mukul) gum resin ethanolic extract in streptozotocin(STZ) induced diabetic rats.Methods:The male Wistar albino rats were randomly divided into four groups of eight animals each:Control group(C),CM-treated control group(C_+CMEE),Diabetic control group(D),CM- treated diabetic group(D_+CMEE).Diabetes was induced by intraperitoneal injection of STZ(55 mg/kg/ bwt).After being confirmed the diabetic rats were treated with C.mukul gum resin ethanolic extract(CMEE) for 60 days.The biochemical estimations like antioxidant,oxidative stress marker enzymes and hepatic marker enzymes of tissues were performed.Results:The diabetic rats showed increased level of enzymatic activities aspartate aminotransaminase(AST),alanine aminotransaminase(ALT) in liver and kidney and oxidative markers like lipid peroxidation(LPO) and protein oxidation(PO) in pancreas and heart.
