| 1 | N-phosphoryl amino acid models for P–N bonds in prebiotic chemical evolution显示文摘Post-translational modification of proteins by N-phosphorylation of the basic amino acid residues plays important roles in biological processes. The high-energy P–N bond might have contributed to the evolution of prebiotic chemistry. N-phosphoryl amino acids(PAAs) can serve as interesting small molecular models for the study of P–N bonds in prebiotic chemical evolution. PAAs are capable of simultaneously producing several important biomolecules such as polypeptides and oligonucleotides under mild reaction conditions. In this review, we describe the chemistry of PAAs, discusse their likely prebiotic origins and their reactivity and how they relate to biological P–N bond species. | 4 | Self-induced Secretion Expression of Trehalose Synthase in Bacillus subtilis WB800n显示文摘Bacillus subtilis is a non-pathogenic Gram-positive bacterium that has been widely used to produce industrially and pharmaceutically relevant proteins. Trehalose, a non reducing disaccharide used as protective agent and additive in foodstuffs and pharmaceutical products, can be prepared by trehalose synthase(TreS). The present work aims to construct a robust recombinant B. subtilis to achieve the secretory expression of TreS. In this study, the treS gene from Pseudomonas putida ATCC47054 was amplified by PCR and further cloned and expressed in B. subtilis WB800 N using pHT01 as expression vector. For avoiding the use of inducer, promoter P_(srfA) was used to replace the promoter P_(grac) in pHT01 and verify the activity of recombinant trehalose synthase. The TreS activity assay was employed to evaluate the performance of recombinant B. subtilis W800 N under different phosphate concentrations, carbon sources, carbon source concentrations, nitrogen sources and pH. The results showed that the P_(srfA) promoter had a good regulation effect under pH 8.0 condition, and the enzyme activity reached 6 000 U/L. Using the PhoD as the secretory signal peptide, TreS was effectively secreted, and the extracellular enzyme activity reached 2 100 U/L, accounting for 35% of the total enzyme activity. By optimizing the medium and fermentation conditions, the extracellular enzyme activity reached 6 900 U/L in 5 L of fermentor, and the proportion reached 48%. | 7 | Analysis of Buoyancy Module Auxiliary Installation Technology Based on Numerical Simulation显示文摘To reduce the requirement for lifting capacity and decrease the hoist cable force during the descending and laying process of a subsea production system(SPS), a buoyancy module auxiliary installation technology was proposed by loading buoyancy modules on the SPS to reduce the lifting weight. Two models are established, namely, the SPS lowering-down model and the buoyancy module floating-up model. The main study results are the following: 1) When the buoyancy module enters the water under wave condition, the amplitude of tension fluctuation is twice that when SPS enters water; 2) Under current condition, the displacement of SPS becomes three times larger because of the existence of the buoyancy module; 3) After being released, the velocity of the buoyancy module increases to a large speed rapidly and then reaches a balancing speed gradually. The buoyancy module floats up at a balancing speed and rushes out from the water at a pop-up distance; 4) In deep water, the floating-up velocity of the buoyancy module is related to its mass density and shape, and it is not related to water depth; 5) A drag parachute can reduce floating-up velocity and pop-up distance effectively.
