1PsyMuKB: An Integrative De Novo Variant Knowledge Base for Developmental Disorders显示文摘De novo variants(DNVs)are one of the most significant contributors to severe earlyonset genetic disorders such as autism spectrum disorder,intellectual disability,and other developmental and neuropsychiatric(DNP)disorders.Presently,a plethora of DNVs have been identified using next-generation sequencing,and many efforts have been made to understand their impact at the gene level.However,there has been little exploration of the effects at the isoform level.The brain contains a high level of alternative splicing and regulation,and exhibits a more divergent splicing program than other tissues.Therefore,it is crucial to explore variants at the transcriptional regulation level to better interpret the mechanisms underlying DNP disorders.To facilitate a better usage and improve the isoform-level interpretation of variants,we developed NeuroPsychiatric Mutation Knowledge Base(PsyMuKB).It contains a comprehensive,carefully curated list of DNVs with transcriptional and translational annotations to enable identification of isoformspecific mutations.PsyMuKB allows a flexible search of genes or variants and provides both table-based descriptions and associated visualizations,such as expression,transcript genomic structures,protein interactions,and the mutation sites mapped on the protein structures.It also provides an easy-to-use web interface,allowing users to rapidly visualize the locations and characteristics of mutations and the expression patterns of the impacted genes and isoforms.PsyMuKB thus constitutes a valuable resource for identifying tissue-specific DNVs for further functional studies of related disorders.PsyMuKB is freely accessible at http://gffzz08064908ab1147fdhunknpxfquu5c6pnv.ffgz.tsg.suse.edu.cn.Guan Ning Lin Sijia Guo Xian Tan Weidi Wang Wei Qian Weichen Song Jingru Wang Shunying Yu Zhen Wang Donghong Cui Han Wang 2019Genomics, Proteomics & Bioinformatics2019,17,4:2

3Existence and uniqueness of extreme point of total power rate functional for hot rolling problem with rigid-plastic SCM model显示文摘The total power rate functional for hot rolling problem with the rigid-plastic SCM model is considered. The gradient operator of the plastic deformation power rate functional is deduced. it is strictly monotone mapping. Further, it is proved that the frictional power rate functional is a convex functional and the tensional stress power rate functional is a linear one. Hence, the total power rate functional is a strictly convex functional. 4AvoidPwd: A Mnemonic Password Generation Strategy Based on Keyboard Transformation显示文摘Identity authentication is the first line of defense for network security.Passwords have been the most widely used authentication method in recent years.Although there are security risks in passwords,they will be the primary method in the future due to their simplicity and low cost.Considering the security and usability of passwords,we propose AvoidPwd,which is a novel mnemonic password generation strategy that is based on keyboard transformation.AvoidPwd helps users customize a“route”to bypass an“obstacle”and choose the characters on the“route”as the final password.The“obstacle”is a certain word using any language and the keys adjacent to the“obstacle”are typed with the“Shift”key.A two-part experiment was conducted to examine the memorability and security of the AvoidPwd strategy with other three password strategies and three leaked password sets.The results showed that the passwords generated by the AvoidPwd strategy were more secure than the other leaked password sets.Meanwhile,AvoidPwd outperformed the KbCg,SpIns,and Alphapwd in balancing security and usability.In addition,there are more symbols in the character distribution of AvoidPwd than the other strategies.AvoidPwd is hopeful to solve the security problem that people are difficult to remember symbols and they tend to input letters and digits when creating passwords.Shunying Lyu Qian Yao Jianhua Song 2022China Communications2022,19,10:0
5Galectin-7 overexpression destroys airway epithelial barrier in transgenic mice显示文摘When the integrity of airway epithelium is destroyed,the ordered airway barrier no longer exists and increases sensitivity to viral infections and allergens,leading to the occurrence of airway inflammation such as asthma.Here,we found that galectin-7 transgenic(+)mice exhibited abnormal airway structures as embryos and after birth.These abnormalities included absent or substantially reduced pseudostratified columnar ciliated epithelium and increased monolayer cells with irregular arrangement and widening of intercellular spaces.Moreover,airway tissue from galectin-7 transgenic(+)mice showed evidence of impaired cell–cell junctions and decreased expression of zonula occludens-1(ZO-1)and E-cadherin.When treated with respiratory syncytial virus(RSV)or ovalbumin(OVA),galectin-7 transgenic(+)mice developed substantially increased bronchial epithelial detachment and apoptosis,airway smooth muscle and basement membrane thickening,and enhanced airway responsiveness.We found that Galectin-7 localized in the cytoplasm and nucleus of bronchial epithelial cells,and that increased apoptosis was mediated through mitochondrial release of cytochrome c and upregulated JNK1 activation and expression of caspase-3 in galectin-7 Tg(+)mice.These findings suggested that Galectin-7 causes airway structural defects and destroys airway epithelium barrier,which predispose the airways to RSV or OVA-induced epithelial apoptosis,injury,and other asthma responses.Jing TIAN Ruxuan HE Yimu FAN Qianqian ZHANG Baolin TIAN Chunju ZHOU Chunyan LIU Mingjing SONG Shunying ZHAO 2021Integrative Zoology2021,16,2:0
