1Development and utilization of a new chemically-induced soybean library with a high mutation density显示文摘Mutagenized populations have provided important materials for introducing variation and identifying gene function in plants. In this study, an ethyl methanesulfonate(EMS)-induced soybean(Glycine max) population,consisting of 21,600 independent M_2 lines, was developed.Over 1,000 M_(4(5))families, with diverse abnormal phenotypes for seed composition, seed shape, plant morphology and maturity that are stably expressed across different environments and generations were identified. Phenotypic analysis of the population led to the identification of a yellow pigmentation mutant, gyl, that displayed significantly decreased chlorophyll(Chl) content and abnormal chloroplast development. Sequence analysis showed that gyl is allelic to Minn Gold, where a different single nucleotide polymorphism variation in the Mg-chelatase subunit gene(ChlI1a) results in golden yellow leaves. A cleaved amplified polymorphic sequence marker was developed and may be applied to marker-assisted selection for the golden yellow phenotype in soybean breeding. 6UPLC-QTOF/MS Method for Screening and Determination of 59 Non-labeled Components in Veterinary Drug Preparations显示文摘[Objectives]This study was conducted to provide an accurate and reliable method for rapid high-throughput screening of veterinary drug preparations.[Methods]For the matrixes of veterinary drug preparations,high-performance liquid chromatography-high resolution quadrupole time-of-flight mass spectrometry(UPLC-QTOF/MS)was used to establish a fast screening method for 59 non-standard components in five categories of antiviral agents,aminoglycosides,quinolones,sulfonamides,and tetracyclines in veterinary drugs.The target drugs were separated by a Waters ACQUITY UPLC HSS T3 chromatographic column(50 mm×2.1 mm,1.8μm),and data were collected in the positive ion mode.Good separation of the 59 drugs was achieved within 7 min.[Results]In the concentration range of 0-100 ng/ml,each drug showed a good linear relationship,and the correlation coefficients were all greater than 0.999.The detection limits of the 59 drugs were in the range of 0.1-0.5 mg/ml,and the recovery under the addition concentration of 5 mg/ml was in the range of 85.2%-103.8%.[Conclusions]The method is fast,simple,accurate,and highly sensitive,and is suitable for high-throughput screening and qualitative identification of non-standard components in veterinary drug preparations.Xueliang PANG Lei WANG Biao QI Sining TANG Yanhua YAN Xuesong WANG Lixue DONG Yi ZHANG Aijun LI 2022Agricultural Biotechnology2022,11,1:1
7Immediate and legacy effects of snow exclusion on soil fungal diversity and community composition显示文摘Background:Soil fungi play crucial roles in ecosystem functions.However,how snow cover change associated with winter warming affects soil fungal communities remains unclear in the Tibetan forest.Methods:We conducted a snow manipulation experiment to explore immediate and legacy effects of snow exclusion on soil fungal community diversity and composition in a spruce forest on the eastern Tibetan Plateau.Soil fungal communities were performed by the high throughput sequencing of gene-fragments.Results:Ascomycota and Basidiomycota were the two dominant fungal phyla and Archaeorhizomyces,Aspergillus and Amanita were the three most common genera across seasons and snow manipulations.Snow exclusion did not affect the diversity and structure of soil fungal community in both snow-covered and snow-free seasons.However,the relative abundance of some fungal communities was different among seasons.Soil fungal groups were correlated with environmental factors(i.e.,temperature and moisture)and soil biochemical variables(i.e.,ammonium and enzyme).Conclusions:These results suggest that the season-driven variations had stronger impacts on soil fungal community than short-term snow cover change.Such findings may have important implications for soil microbial processes in Tibetan forests experiencing significant decreases in snowfall.Li Zhang Yuzhi Ren Kaijun Yang Zhijie Li Bo Tan Yang Liu Han Li Chengming You Sining Liu Lixia Wang Rui Yin Jian Zhang Zhenfeng Xu 2021Forest Ecosystems2021,8,2:1
8Nitrogen addition mediates the response of foliar stoichiometry to phosphorus addition: a meta-analysis显示文摘Background:Changes in foliar nitrogen(N)and phosphorus(P)stoichiometry play important roles in predicting the efects of global change on ecosystem structure and function.However,there is substantial debate on the efects of P addition on foliar N and P stoichiometry,particularly under diferent levels of N addition.Thus,we conducted a global meta-analysis to investigate how N addition alters the efects of P addition on foliar N and P stoichiometry across different rates and durations of P addition and plant growth types based on more than 1150 observations.Results:We found that P addition without N addition increased foliar N concentrations,whereas P addition with N addition had no efect.The positive efects of P addition on foliar P concentrations were greater without N addition than with N addition.Additionally,the efects of P addition on foliar N,P and N:P ratios varied with the rate and duration of P addition.In particular,short-term or low-dose P addition with and without N addition increased foliar N concentration,and the positive efects of short-term or low-dose P addition on foliar P concentrations were greater without N addition than with N addition.The responses of foliar N and P stoichiometry of evergreen plants to P addition were greater without N addition than with N addition.Moreover,regardless of N addition,soil P availability was more efective than P resorption efciency in predicting the changes in foliar N and P stoichiometry in response to P addition.Conclusions:Our results highlight that increasing N deposition might alter the response of foliar N and P stoichiometry to P addition and demonstrate the important efect of the experimental environment on the results.These results advance our understanding of the response of plant nutrient use efciency to P addition with increasing N deposition.Chengming You Changhui Peng Zhenfeng Xu Yang Liu Li Zhang Rui Yin Lin Liu Han Li Lixia Wang Sining Liu Bo Tan Paul Kardol 2021Ecological Processes2021,10,1:0

10Do root modules still exist after they die?显示文摘Background:The terminal branch orders of plant root systems are increasingly known as an ephemeral module.This concept is crucial to recognize belowground processes.However,it is unknown if root modules still exist after they die?Methods:The decomposition patterns of the first five root orders were observed for 3 years using a branch-order classification,a litter-bag method and sequential sampling in a common subalpine tree species(Picea asperata)of southwestern China.Results:Two root modules were observed during the 3-year incubation.Among the first five branch orders,the first three order roots exhibited temporal patterns of mass loss,nutrients and stoichiometry distinct from their woody mother roots throughout the experimental period.This study,for the first time,reported the decomposition pattern of each individual root order and found a similar decomposition dynamic among ephemeral root branches in a forest tree species.Conclusions:Results from this study suggest that root modules may also exist after death,while more data are needed for confirmation.The findings may further advance our understanding of architecture-associated functional heterogeneity in the fine-root system and also improve our ability to predict belowground processes.Jihong Li Chengming You Li Zhang Han Li Bo Tan Yang Liu Lixia Wang Sining Liu Zhenfeng Xu 2021Forest Ecosystems2021,8,2:0

15JUNB mediates oxaliplatin resistance via the MAPK signaling pathway in gastric cancer by chromatin accessibility and transcriptomic analysis显示文摘tients,and chemotherapy resistance is one of the main reasons for treatment failure.Thus,it is important to reveal the mechanism of oxaliplatin resistance and to seek effective intervention strategies to improve chemotherapy sensitivity,thereby improving the survival and prognosis of gastric cancer patients.To understand the molecular mechanisms of oxaliplatin resistance,we generate an oxaliplatin-resistant gastric cancer cell line and conduct assay for transposase-accessible chromatin sequencing(ATAC-seq)and RNA sequencing(RNA-seq)for both parental and oxaliplatin-resistant AGS cells.A total of 3232 genomic regions are identified to have higher accessibility in ox-aliplatin-resistant cells,and DNA-binding motif analysis identifies JUNB as the core transcription factor in the regulatory network.JUNB is overexpressed in oxaliplatin-resistant gastric cancer cells,and its upregulation is associated with poor prognosis in gastric cancer patients,which is validated by our tissue microarray data.Moreover,chromatin immunoprecipitation sequencing(ChIP-seq)analysis reveals that JUNB binds to the tran-scriptional start site of key genes involved in the MAPK signaling pathway.Knockdown of JUNB inhibits the MAPK signaling pathway and restores sensitivity to oxaliplatin.Combined treatment with the ERK inhibitor piperlongu-mine or MEK inhibitor trametinib effectively overcomes oxaliplatin resistance.This study provides evidence that JUNB mediates oxaliplatin resistance in gastric cancer by activating the MAPK pathway.The combination of MAPK inhibitors with oxaliplatin overcomes resistance to oxaliplatin,providing a promising treatment opportunity for oxaliplatin-resistant gastric cancer patients.Suyao Li Yichou Wei Xun Sun Mengling Liu Mengxuan Zhu Yitao Yuan Jiayu Zhang Yu Dong Keshu Hu Sining Ma Xiuping Zhang Bei Xu Hesheng Jiang Lu Gan Tianshu Liu 2023Acta Biochimica et Biophysica Sinica2023,55,11:0
16A Camouflaged Film Imitating the Chameleon Skin with Color-Changing Microfluidic Systems Based on the Color Information Identification of Background显示文摘To adapt to a complex and variable environment,self-adaptive camouflage technology is becoming more and more important in all kinds of military applications by overcoming the weakness of the static camouflage.In nature,the chameleon can achieve self-adaptive camouflage by changing its skin color in real time with the change of the background color.To imitate the chameleon skin,a camouflaged film controlled by a color-changing microfluidic system is proposed in this paper.The film with microfluidic channels fabricated by soft materials can achieve dynamic cloaking and camouflage by circulating color liquids through channels inside the film.By sensing and collecting environmental color change information,the control signal of the microfluidic system can be adjusted in real time to imitate chameleon skin.The microstructure of the film and the working principle of the microfluidic color-changing system are introduced.The mechanism to generate the control signal by information processing of background colors is illustrated.“Canny”double-threshold edge detection algorithm and color similarity are used to analyze and evaluate the camouflage.The tested results show that camouflaged images have a relatively high compatibility with environmental backgrounds and the dynamic cloaking eff ect can be achieved.Huanhuan Li Tianhang Yang Lujia Li Sining Lv Songjing Li 2021Journal of Bionic Engineering2021,18,5:0
17Nitrogen addition promotes foliar litterfall and element return in a subtropical forest,southwestern China显示文摘Nitrogen deposition has a considerable impact on biogeochemical cycling in terrestrial ecosystems.However,how litter production and element return respond to N addition remains poorly understood in nitrogen-rich subtropical regions.In this study,a 4-year nitrogen addition experiment explored its eff ects on foliar litter production and carbon,nitrogen and phosphorus in a subtropical Michelia wilsonii forest.A clear seasonal pattern in foliar litterfall was observed,regardless of nitrogen treatments,with a peak in spring and a smaller one in autumn.Foliar litter increased with increasing nitrogen but did not aff ect litter carbon concentrations and often decreased nitrogen and phosphorous concentrations.The eff ect of nitrogen addition was dependent on time(month/year).Carbon,nitrogen and phosphorous return showed similar bimodal seasonal patterns.Nitrogen addition increased carbon and nitrogen return but did not aff ect phosphorous.Our results suggest that the addition of nitrogen stimulates carbon and nutrient return via litterfall.Yue Zhang Shichen Xiong Chengming You Sining Liu Lixia Wang Li Zhang Han Li Bo Tan Yang Liu Zhenfeng Xu 2023Journal of Forestry Research2023,34,4:0
18Negative correlation between acetyl-CoA acyltransferase 2 and cetuximab resistance in colorectal cancer显示文摘The emergence of anti-EGFR therapy has revolutionized the treatment of colorectal cancer(CRC).However,not all patients respond consistently well.Therefore,it is imperative to conduct further research to identify the molecular mechanisms underlying the development of cetuximab resistance in CRC.In this study,we find that the expressions of many metabolism-related genes are downregulated in cetuximab-resistant CRC cells compared to their sensitive counterparts.Specifically,acetyl-CoA acyltransferase 2(ACAA2),a key enzyme in fatty acid metabolism,is downregulated during the development of cetuximab resistance.Silencing of ACAA2 promotes proliferation and increases cetuximab tolerance in CRC cells,while overexpression of ACAA2 exerts the opposite effect.RTK-Kras signaling might contribute to the downregulation of ACAA2 expression in CRC,and ACAA2 predicts CRC prognosis in patients with Kras mutations.Collectively,our data suggest that modulating ACAA2 expression contributes to secondary cetuximab resistance in Kras wild-type CRC patients.ACAA2 expression is related to Kras mutation and demonstrates a prognostic role in CRC patients with Kras mutation.Thus,ACAA2 is a potential target in CRC with Kras mutation.Yitao Yuan Xun Sun Mengling Liu Suyao Li Yu Dong Keshu Hu Jiayu Zhang Bei Xu Sining Ma Hesheng Jiang Pengcong Hou Yufu Lin Lu Gan Tianshu Liu 2023Acta Biochimica et Biophysica Sinica2023,55,9:0
