| 1 | Preparation and evaluation of nattokinaseloaded self-double-emulsifying drug delivery system显示文摘In the present study,we prepared nattokinase-loaded self-double-emulsifying drug delivery system(SDEDDS)and investigated its preliminary pharmacodynamics.The type and concentration of oil phase,inner aqueous phase and emulsifier were screened to prepare optimum nattokinase-loaded SDEDDS.Next,the optimum formulations were characterized based on microstructure,volume-weighted mean droplet size,self-emulsifying rate,yield,storage stability,in vitro release and in vivo pharmacodynamics studies.The water/oil/watermultiple emulsions exhibited typicalmultiple structure,with relatively small volumeweighted mean droplet size 6.0±0.7μm and high self-emulsifying ability(self-emulsifying time<2 min).Encapsulation of nattokinase was up to 86.8±8.2%.The cumulative release of nattokinase within 8 h was about 30%,exhibiting a sustained release effect.The pharmacodynamics study indicated that nattokinase-loaded SDEDDS could significantly prolong the whole blood clotting time in mouse and effectively improve the carrageenan-induced tail thrombosis compared with nattokinase solution.Moreover,we showed that SDEDDS could successfully self-emulsify into water/oil/water multiple emulsions upon dilution in dispersion medium with gentle stirring and effectively protect nattokinase activity in gastric environment.Our findings suggested that SDEDDS could be a promising strategy for peptide and protein drugs by oral administration. | Xiaona Wang Sifan Jiang Xinyue Wang Jie Liao Zongning Yin | 2015 | Asian Journal of Pharmaceutical Sciences2015,10,5: | 9 |

| 3 | A novel aldo-keto reductase gene, IbAKR, from sweet potato confers higher tolerance to cadmium stress in tobacco显示文摘High concentrations of Cd can inhibit growth and reduce the activity of the photosynthetic apparatus in plants. In several plant species, aldo-keto reductases(AKRs) have been shown to enhance tolerance to various abiotic stresses by scavenging cytotoxic aldehydes;however, few AKRs have been reported to enhance Cd stress tolerance. In this study, the gene Ib AKR was isolated from sweet potato. The relative expression levels of Ib AKR increased significantly(approximately 3-fold) after exposure to 200 mmol$L–1 Cd Cl2 or 10 mmol$L–1 H2 O2. A subcellular localization assay showed that Ib AKR is predominantly located in the nucleus and cytoplasm.Ib AKR-overexpressing tobacco plants showed higher tolerance to Cd stress than wild-type(WT). Transgenic lines showed a significant ability to scavenge malondialdehyde(MDA) and methylglyoxal(MG). In addition,proline content and superoxide dismutase activity were significantly higher and H2 O2 levels were significantly lower in the transgenic plants than in the WT. Quantitative real-time PCR analysis showed that the reactive oxygen species(ROS) scavenging genes encoding guaiacol peroxidase(GPX), ascorbate peroxidase(APX), monodehydroascorbate reductase(MDHAR) and peroxidase(POD) were significantly upregulated in transgenic plants compared to WT under Cd stress. | 6 | Identification, through transcriptome analysis, of transcription factors that regulate anthocyanin biosynthesis in different parts of red-fleshed apple 'May' fruit显示文摘Anthocyanins, a class of flavonoid compounds that provide pigmentation to plant organs, are beneficial components of the human diet and there is an interest in understanding the regulation of their formation in crop plants. The apple cultivar ‘May’ represents an excellent model for the study of anthocyanin metabolism due to the spatial color pattern of its flesh, which transitions sequentially from red to white to red again,from the outside to the inside of the apple fruit. To screen the transcription factors involved in the anthocyanin pathway, transcriptomes from differently colored sections of ‘May’ fruits were analyzed by RNA sequencing. Kyoto Encyclopedia of Genes and Genomes(KEGG) database was used to analyse the gene expression data. The results suggested differentially expressed genes(DEGs) that are related to phytohormones are involved in regulating anthocyanin biosynthesis. K-means clustering analysis revealed 167 common DEGs between different sections of fruit with the same expression pattern as candidates for regulating anthocyanin synthesis. Further analysis showed that nine of the 167 DEGs were annotated as transcription factors and quantitative real time PCR(qRT-PCR) confirmed that their expression was obviously higher in red regions of the fruit, consistent with their roles as hub genes that regulate anthocyanin synthesis. | 9 | Tandem Molecular Self-Assembly Selectively Inhibits Lung Cancer Cells by Inducing Endoplasmic Reticulum Stress显示文摘The selective formation of nanomaterials in cancer cells and tumors holds great promise for cancer diagnostics and therapy.Until now,most strategies rely on a single trigger to control the formation of nanomaterials in situ.The combination of two or more triggers may provide for more sophisticated means of manipulation.In this study,we rationally designed a molecule(Comp.1)capable of responding to two enzymes,alkaline phosphatase(ALP),and reductase.Since the A549 lung cancer cell line showed elevated levels of extracellular ALP and intracellular reductase,we demonstrated that Comp.1 responded in a stepwise fashion to those two enzymes and displayed a tandem molecular self-assembly behavior.The selective formation of nanofibers in the mitochondria of the lung cancer cells led to the disruption of the mitochondrial membrane,resulting in an increased level of reactive oxygen species(ROS)and the release of cytochrome C(Cyt C).ROS can react with proteins,resulting in endoplasmic reticulum(ER)stress and the unfolded protein response(UPR).This severe ER stress led to disruption of the ER,formation of vacuoles,and ultimately,apoptosis of the A549 cells.Therefore,Comp.1 could selectively inhibit lung cancer cells in vitro and A549 xenograft tumors in vivo.Our study provides a novel strategy for the selective formation of nanomaterials in lung cancer cells,which is powerful and promising for the diagnosis and treatment of lung cancer. | Debin Zheng Yumiao Chen Sifan Ai Renshu Zhang Zhengfeng Gao Chunhui Liang Li Cao Yaoxia Chen Zhangyong Hong Yang Shi Ling Wang Xingyi Li Zhimou Yang | 2019 | Research2019,,1: | 1 |

| 13 | Drug discovery of sclerostin inhibitors显示文摘Sclerostin, a protein secreted from osteocytes, negatively regulates the WNT signaling pathway by binding to the LRP5/6 co-receptors and further inhibits bone formation and promotes bone resorption. Sclerostin contributes to musculoskeletal system-related diseases, making it a promising therapeutic target for the treatment of WNT-related bone diseases. Additionally, emerging evidence indicates that sclerostin contributes to the development of cancers, obesity, and diabetes, suggesting that it may be a promising therapeutic target for these diseases. Notably, cardiovascular diseases are related to the protective role of sclerostin. In this review, we summarize three distinct types of inhibitors targeting sclerostin, monoclonal antibodies, aptamers, and small-molecule inhibitors, from which monoclonal antibodies have been developed. As the first-in-class sclerostin inhibitor approved by the U.S. FDA,the monoclonal antibody romosozumab has demonstrated excellent effectiveness in the treatment of postmenopausal osteoporosis;however, it conferred high cardiovascular risk in clinical trials. Furthermore,romosozumab could only be administered by injection, which may cause compliance issues for patients who prefer oral therapy. | 14 | Local engineering of topological phase in monolayer MoS2显示文摘Monolayer transition metal dichalcogenides(TMDCs) with the 1 T0 structure are a new class of large-gap two-dimensional(2 D) topological insulators, hosting topologically protected conduction channels on the edges. However, the 1 T0 phase is metastable compared to the 2 H phase for most of 2 D TMDCs, among which the 1 T0 phase is least favored in monolayer MoS2. Here we report a clean and controllable technique to locally induce nanometer-sized 1 T0 phase in monolayer 2 H-MoS2 via a weak Argon-plasma treatment,resulting in topological phase boundaries of high density. We found that the stabilization of 1 T0 phase arises from the concerted effects of S vacancies and the tensile strain. | 15 | Relaxor behavior and superior ferroelectricity of Y_(2)O_(3)-doped(Ba_(0.98)Ca_(0.02))(Ti_(0.94)Sn_(0.04)Zr_(0.02))O_(3) lead-free ceramics显示文摘To upgrade the electric properties of lead-free piezoceramics,(1-x)(Ba_(0.98)Ca_(0.02)Ti_(0.94)Sn_(0.04)Zr_(0.02))O_(3)-xY_(2)O_(3)(abbreviated as(1-x)BCTSZ-xY,x=0 mol%,0.02 mol%,0.04 mol%,0.06 mol%,0.08 mol%and 0.1 mol%)ceramics were successfully synthesized by traditional solid-state sintering method.The phase structure and microstructure of ceramics were investigated by X-ray diffraction(XRD),scanning electron microscopy(SEM)and piezoresponse force microscopyeramics(PFM).The electric properties of ceramics were researched through piezoelectric,dielectric and ferroelectric test instruments.The results show that all samples have pure perovskite structure and favorable electric properties.The optimal electric properties which especially include superior ferroelectric properties are gained when Y_(2)O_(3)content is 0.06 mol%(d_(33)=419 pC/N,k_(p)=52%,T_(c)=89.5℃,ε_(r)=26900,tanδ=2.86%,P_(r)=14.41μC/cm^(2),Ec=1.8 kV/cm).Moreover,the temperature-dependent dielectricity of samples shows apparent relaxor behavior under different frequencies.The Curie-Weiss law further proves that all samples are typical relaxor ferroelectrics,and the relaxor degree of samples decreases with increase of Y_(2)O_(3)content.In conclusion,Y_(2)O_(3)plays a significant role in enhancing electric properties of BCTSZ ceramics. | Sifan Wang Qibin Liu Enpei Cai Fanghui Mou An Xue | 2022 | Journal of Rare Earths2022,40,6: | 1 |

Through a combination of experiments and numerical simulations, we here propose a method to regulate the wrinkling patterns in a film-substrate system by introducing periodic surface stiffness, which is generated through sequential specified ultraviolet-ozone(UVO) treatments. Both experiments and numerical simulations demonstrate that the proposed technique can produce various patterns with wide, tunable geometrical features and anisotropy.
