| 1 | 3-year Treatment of Tenofovir Alafenamide vs.Tenofovir Disoproxil Fumarate for Chronic HBV Infection in China显示文摘Background and Aims:Tenofovir alafenamide(TAF)has similar efficacy to tenofovir disoproxil fumarate(TDF)but with improved renal and bone safety in chronic hepatitis B patients studied outside of China.We report 3-year results from two phase 3 studies with TAF in China(Clinicaltrials.gov:NCT02836249 and NCT02836236).Methods:Chinese hepatitis B e antigen(HBeAg)-positive and-negative chronic hepatitis B patients with viremia and elevated alanine aminotransferase were randomized 2:1 to TAF or TDF treatment groups and treated in a double-blind fashion for 144 weeks(3 years).Efficacy responses were assessed by individual study while safety was assessed by a pooled analysis.Results:Of the 334 patients(180 HBeAg-positive and 154 HBeAg-negative)randomized and treated,baseline characteristics were similar between groups.The overall mean age was 38 years and 73%were male.The mean HBV DNA was 6.4 log10 IU/mL.The median alanine aminotransferase was 88 U/L,and 37%had a history of antiviral use.At week 144,the proportion with HBV DNA<29 IU/mL was similar among the two groups,with TAF at 83%vs.TDF at 79%,and TAF at 93%vs.TDF at 92%for the HBeAg-positive and-negative patients,respectively.In each study,higher proportions of TAF than TDF patients showed normalized alanine aminotransferase(via the American Association for the Study of Liver Diseases and the China criteria)and showed loss of HBsAg;meanwhile,the HBeAg seroconversion rates were similar.Treatment was well-tolerated among the TAF patients,who showed a smaller median decline in creatinine clearance(−0.4 vs.−3.2 mL/min;p=0.014)and less percentage change in bone mineral density vs.TDF at hip(−0.95%vs.−1.93%)and spine(+0.35%vs.−1.40%).Conclusions:In chronic hepatitis B patients from China,TAF treatment provided efficacy similar to TDF but with better renal and bone safety at 3 years. | Jinlin Hou Qin Ning Zhongping Duan You Chen Qing Xie Fu-Sheng Wang Lunli Zhang Shanming Wu Hong Tang Jun Li Feng Lin Yongfeng Yang Guozhong Gong John FFlaherty Anuj Gaggar Shuyuan Mo Cong Cheng Gregory Camus Chengwei Chen Yan Huang Jidong Jia Mingxiang Zhang GS-US-320-0110 and GS-US-320-0108 China Investigators | 2021 | Journal of Clinical and Translational Hepatology2021,9,3: | 7 |

| 3 | Endogenous adult neurogenesis and cognitive function recovery following traumatic brain injury in the rat hippocampus显示文摘BACKGROUND:Endogenous neural progenitor cells play a beneficial role for cognitive recovery following traumatic brain injury.However,there are few classification-control studies aimed at varying graded brain trauma.OBJECTIVE:To observe the effects of adult endogenous neurogenesis on cognitive function repair and regeneration of neural progenitor cells following varying graded traumatic hippocampal injury to determine the significance of endogenous neurogenesis in the repair of brain injury.DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed at the Key Laboratory of Injuries,Variations and Regeneration of Nervous System,Tianjin Medical University General Hospital,from February to October 2009.MATERIALS:Mouse anti-rat 5-bromodeoxyuridine (BrdU) and neuronal nuclei (NeuN) monoclonal antibodies were purchased from Millipore Corporation,USA.METHODS:A total of 45 Wistar rats were randomly assigned to three groups.Mild and severe injury groups were respectively subjected to (182 ± 2) kPa and (284 ± 4) kPa lateral fluid percussion to establish models of brain injury,and the control group was subjected to surgery with no lateral fluid percussion.MAIN OUTCOME MEASURES:Cognitive function was estimated using the Morris water maze.Proliferation,survival,and differentiation of newly generated cells in the injured hippocampus were observed through the use of immunofluorescent staining.RESULTS:At 7 days post-injury,the number of BrdU+ cells in the hippocampal dentate gyrus significantly increased in the mild and severe injury groups compared with the control group (P < 0.01).At 61 days post-injury,the number of BrdU+/NeuN+ cells in the hippocampal dentate gyrus was significantly greater in the mild injury group compared with the severe injury and control groups (P < 0.01).In addition,the control group exhibited the greatest proportion of surviving cells that differentiated into mature neurons compared with the injury groups (P < 0.01).Moreover,at 61 days post-injury,cognitive function in rats with mild injury recovered to normal levels,whereas the severe injury group exhibited cognitive deficits (P < 0.01).CONCLUSION:Traumatic brain injury may be a stimulation factor for proliferation of neural progenitor cells in the adult hippocampus but severe brain trauma does not lead to an increased number of newly generated cells.Endogenous adult neurogenesis repairs neurological functions to an extent.However,recovery of neurological function remains limited following severe traumatic brain injury. | Wangmiao Zhao Linchun Huan Yan Zhao Jie Zhao Qi Zhang Lin Zhang Rong Yan Shuyuan Yang Xinyu Yang | 2010 | Neural Regeneration Research2010,5,9: | 2 |

| 12 | The Dof transcription factor COG1 acts as a key regulator of plant biomass by promoting photosynthesis and starch accumulation显示文摘Photosynthetic efficiency is the primary determinant of crop yield,including vegetative biomass and grain yield.Manipulation of key transcription factors known to directly control photosynthetic machinery can be an effective strategy to improve photosynthetic traits.In this study,we identified an Arabidopsis gain-of-function mutant,cogwheel1-3D,that shows a significantly enlarged rosette and increased biomass compared with wild-type plants.Overexpression of COG1,a Dof transcription factor,recapitulated the phenotype of cogwheel1-3D,whereas knocking out COG1 and its six paralogs resulted in a reduced rosette size and decreased biomass.Transcriptomic and quantitative reverse transcription polymerase chain reaction analyses demonstrated that COG1 and its paralogs were required for light-induced expression of genes involved in photosynthesis.Further chromatin immunoprecipitation and electrophoretic mobility shift assays indicated that COG1 can directly bind to the promoter regions of multiple genes encoding light-harvesting antenna proteins.Physiological,biochemical,and microscopy analyses revealed that COG1 enhances photosynthetic capacity and starch accumulation in Arabidopsis rosette leaves.Furthermore,combined results of bioinformatic,genetic,and molecular experiments suggested that the functions of COG1 in increasing biomass are conserved in different plant species.These results collectively demonstrated that COG1 acts as a key regulator of plant biomass by promoting photosynthesis and starch accumulation.Manipulating COG1 to optimize photosynthetic capacity would create new strategies for future crop yield improvement. | Zhuoyun Wei Haoyong Zhang Meng Fang Shuyuan Lin Mingsong Zhu Yuxiu Li Limin Jiang Tianliang Cui Yanwei Cui Hong Kui Liang Peng Xiaoping Gou Jia Li | 2023 | Molecular Plant2023,16,11: | 0 |
| 13 | MBD2 promotes Th2 differentiation in ovalbumin-induced CD4+T cells显示文摘Introduction:Allergen-specific CD4+T cells play a central role in autoimmune disorders,allergies and asthma,with Th2-type immunity being the typical functional response of CD4+T cells.This study aimed to investigate the role of MBD2 in regulating Th2 cell differentiation.Methods:Splenic mononuclear cells were extracted from C57BL/6 mice,and CD4+T cells were isolated using magnetic beads and confirmed through flow cytometry.Lentivirus was employed to construct MBD2-silenced CD4+T cells.In vitro experiments were performed to treat splenogenic mononuclear cells and CD4+T cells with Ovalbumin(OVA),and Th2 cell ratios and IL-4 levels were assessed using flow cytometry and ELISA.Results:The purity of the isolated CD4+T cells was 95.73%,confirming successful isolation of primary CD4+T cells.Compared to the control group,the Th2 cell ratio exhibited an increase in the Th2-induced group.Treatment with 5-Aza(concentrations,1-100μM)promoted Th2 cell differentiation and increased IL-4 levels.Notably,when combined with Th2 induction and 10μM 5-Aza treatment,silencing MBD2 further amplified Th2 cell ratios and elevated IL-4 levels in cell supernatants.Furthermore,OVA(concentration,200μg/mL)induced the differentiation of CD4+T cells into Th2 cells and increased IL-4 secretion.Interestingly,silencing MBD2 significantly increased the Th2 cell ratio and IL-4 levels in OVA-treated CD4+T cells.Conclusion:In summary,OVA promoted CD4+T cell differentiation into Th2 cells and enhanced IL-4 levels.MBD2 was identified as a mediator of Th2 cell differentiation in splenic-derived CD4+T cells,influenced by OVA or 5-Aza treatment. | QILU PAN YAN JIANG LINQIAO LI XIAOJING DU QIAN HAN FEIXIANG LING ROU LI SHUYUAN CHU LIN MAI JIANWEI HUANG LIBING MA | 2023 | BIOCELL2023,47,11: | 0 |
| 14 | The Antidiabetic Effect and Mechanism of JinXiaoXiaoKe Decoction in Type 2 Diabetic Goto-Kakizaki Rats显示文摘Background:Modern research and clinical practice have proved that Traditional Chinese Medicine(TCM)has unique advantages in the treatment of diabetes.JinXiaoXiaoKe decoction(JXXKD)is a prescription for treating diabetes used in ancient China and still has a good clinical effect today.However,the mechanism of JXXKD against T2DM is unclear.Objective:The purpose of this study is to screen the targets through network pharmacology,and to explore the therapeutic effect and mechanism of JXXKD on diabetic rats.Methods:The JXXKD’s active components,related targets and T2DM targets were obtained from the public database,and Venny 2.1 was used to determine the common targets of JXXKD in the treatment of T2DM,and PPI,GO and KEGG analysis were performed.The core components and targets are verified by molecular docking.The diabetic Goto-Kakizaki rat model was established by high-sugar and high-fat diet,Wistar rats were used as a blank control group.The diabetic rats were randomized into three groups and administered saline(MO),metformin(MET),or JXXKD once a day for 4 weeks.Fasting blood glucose(FBG),Fasting serum insulin(FINS),and HOMA-IR were detected,hematoxylin-eosin(HE)staining was used to observe the pathological changes of liver tissue;and qPCR was used to detect the expression of relevant genes screened by network pharmacology.Results:93 active components and 296 targets of JXXKD were identified,of which 156 overlapped with T2DM-related targets.PPI network showed that APP,AKT1,ANXA1,RXRA,C3,EGFR,ESR1,RELA,IL6,and MAPK8 were the top 10 relevant targets.GO analysis showed the common targets are mainly involved in oxidative stress,lipid metabolism,and nutrient levels,while KEGG analysis showed these targets may regulate lipids and atheroscle-rosis,AGE-RAGE signaling pathway,and TNF signaling pathway.Molecular docking suggested a satisfactory potential for key components to bind to these significant targets.The animal experiments showed that JXXKD significantly improved the symptoms of polyuria,decreased the protein levels of FBG and HOMA-IR,improved liver fat deposition,and decreased the gene expressions of Foxo 1,Pparg and Akt in diabetic rats.Conclusion:The mechanism of JXXKD treat T2DM may be achieved by modulating the expression of FOXO1,PPARG,and AKT,regulating the glucose and lipids metabolism. | Miao Lin Lei Wang Lihua Wan Jiadong Xu Yu Li Lingyong Cao Shuyuan Lin | 2023 | Clinical Complementary Medicine and Pharmacology2023,3,1: | 0 |
| 15 | Quantitative morphometric analysis of a deep-water channel in the Taranaki Basin, New Zealand显示文摘The morphological changes of deep-water channels have an important influence on the distributions of channel sand reservoirs,so it is important to explore the morphological change process of deep-water channel for the exploration and development of deep-water oil and gas.Based on a typical sinuous Quaternary channel(Channel I)in the Taranaki Basin,New Zealand,a variety of seismic interpretation techniques were applied to quantitatively characterize the morphological characteristics of the Channel I,and the relationships between the quantitative parameters and the morphological changes of the Channel I,as well as the controlling factors affecting those morphological changes,were discussed.The results are as follows:(1)in the quantitative analysis,six parameters were selected:the channel depth,width,sinuosity,and aspect ratio(width/depth),the channel swing amplitude(λ)and the channel bend frequency(ω);(2)according to the quantitative morphological parameters of the channel(mainly including three parameters such as channel sinuosity,ωandλ),the Channel I was divided into three types:the low-sinuous channel(LSC),the high-sinuous channel(HSC),the moderate-sinuous channel(MSC).U-shaped channel cross-sections developed in the LSC,V-shaped channel cross-sections developed in the HSC,including inclined-V and symmetric-V cross-sections,and dish-shaped channel cross-sections developed in the MSC;(3)the morphological characteristics of the LSC and MSC were related to their widths and depths,while the morphology of the HSC was greatly affected by the channel width,a change in depth did not affect the HSC morphology;(4)the morphological changes of the Channel I were controlled mainly by the slope gradient,the restricted capacity of the channel and the differential in fluid properties. | Wei Wu Guangxu Wang Changsong Lin Weiqing Liu Quan Li Zhendong Feng Shuyuan Ning | 2023 | Acta Oceanologica Sinica2023,42,5: | 0 |

| 17 | Quaternary integrative stratigraphy,biotas,and paleogeographical evolution of the Qinghai-Tibetan Plateau and its surrounding areas显示文摘The Quaternary strata on the Qinghai-Tibet Plateau contain rich information about the paleoclimate and environmental evolution,record the evolution process of the Quaternary regional tectonics,paleogeography,and geomorphology of the plateau,and are extremely important areas for studying the Quaternary geological events and regional environmental evolution.According to a comprehensive analysis of the regional stratigraphic data and the development and evolution characteristics of the biota,based on the differences in the lithostratigraphic units,sedimentary characteristics,landforms,and drainage systems,the Quaternary strata on the Qinghai-Tibet Plateau and its surrounding areas are divided into five stratigraphic regions:the Tarim region,Loess Plateau region,Qinghai-Tibet Plateau region,Yunnan-Guizhou Plateau region,and India-Ganges region.The Qinghai-Tibet Plateau stratigraphic region is divided into seven stratigraphic sub-regions:the West Kunlun-Karakorum,AltunQilian Mountains,Qaidam-Hehuang,East Kunlun-Bayan Har,Qiangtang,East Xizang-West Yunnan-West Sichuan,and Gangdise-Himalayan sub-regions.This paper briefly describes the lithostratigraphic units of the seven stratigraphic sub-regions of the Qinghai-Tibet Plateau.According to the lithostratigraphic sequence and its sedimentary characteristics,stratigraphic contact relationship,formation age,and evolution of the biota in each stratigraphic sub-region,the Quaternary tectonic paleogeographic evolution of the Qinghai-Tibet Plateau is divided into four stages.(1)The inherited differential uplift stage since the Pliocene(2.6-1.8/1.5 Ma):the regional sedimentary differences were significant,and the stratigraphic distribution was limited,the alluvial-proluvial sandy conglomerate was widely developed along the piedmont,and fluvial and lacustrine deposits were developed in the low-lying areas between the mountains.(2)The mountain range flattening stage(1.8/1.5-1.2/0.8 Ma):the erosion unconformity surfaces around the plateau were widely distributed,large rivers were formed,and lake sediments developed in the intermountain basins and the hinterland of the plateau.(3)1.2/0.8-0.128 Ma:the plateau continued to rise in a large range,with significant topographic differences and intensified mountain erosion.At about 0.8 Ma,the plateau uplifted above the snow line and entered the cryosphere,mountain glaciers developed,and the alpine arid environment gradually formed.(4)0.128 Ma-:the mountains rose and erosion intensified,and intermountain basins and lakes were widely distributed.There were significant differences in the regional sedimentary characteristics,and the sedimentary types developed toward diversification.The modern plateau landform pattern was basically formed. | Yong WANG Mianping ZHENG Yuan LING Shuyuan XIANG Zhaogang SHAO Kexin ZHANG Xue KE Xiao LIN Fang HAN Jian’en HAN | 2024 | Science China Earth Sciences2024,67,4: | 0 |
