| 1 | Modeling CADASIL vascular pathologies with patient-derived induced pluripotent stem cells显示文摘Cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy(CADASIL)is a rare hereditary cerebrovascular disease caused by a NOTCH3 mutation.However,the underlying cellular and molecular mechanisms remain unidentified.Here,we generated non-integrative induced pluripotent stem cells(iPSCs)from fibroblasts of a CADASIL patient harboring a heterozygous NOTCH3 mutation(c.3226C>T,p.R1076C).Vascular smooth muscle cells(VSMCs)differentiated from CADASIL-specific iPSCs showed gene expression changes associated with disease phenotypes,including activation of the NOTCH and NF-kB signaling pathway,cytoskeleton disorganization,and excessive cell proliferation.In comparison,these abnormalities were not observed in vascular endothelial cells(VECs)derived from the patients iPSCs.Importantly,the abnormal upregulation of NF-kB target genes in CADASIL VSMCs was diminished by a NOTCH pathway inhibitor,providing a potential therapeutic strategy for CADASIL.Overall,using this iPSCbased disease model,our study identified clues for studying the pathogenic mechanisms of CADASIL and developing treatment strategies for this disease. | Chen Ling Zunpeng Liu Moshi Song Weiqi Zhang Si Wang Xiaoqian Liu Shuai Ma Shuhui Sun Lina Fu Qun Chu Juan Carlos Izpisua Belmonte Zhaoxia Wang Jing Qu Yun Yuan Guang-Hui Liu | 2019 | Protein & Cell2019,10,4: | 7 |
| 2 | Genome and transcriptome of Papaver somniferum Chinese landrace CHM indicates that massive genome expansion contributes to high benzylisoquinoline alkaloid biosynthesis显示文摘Opium poppy(Papaver somniferum)is a source of morphine,codeine,and semisynthetic derivatives,including oxycodone and naltrexone.Here,we report the de novo assembly and genomic analysis of P.somniferum traditional landrace‘Chinese Herbal Medicine’.Variations between the 2.62 Gb CHM genome and that of the previously sequenced high noscapine 1(HN1)variety were also explored.Among 79,668 protein-coding genes,we functionally annotated 88.9%,compared to 68.8%reported in the HN1 genome.Gene family and 4DTv comparative analyses with three other Papaveraceae species revealed that opium poppy underwent two whole-genome duplication(WGD)events.The first of these,in ancestral Ranunculales,expanded gene families related to characteristic secondary metabolite production and disease resistance.The more recent species-specific WGD mediated by transposable elements resulted in massive genome expansion.Genes carrying structural variations and large-effect variants associated with agronomically different phenotypes between CHM and HN1 that were identified through our transcriptomic comparison of multiple organs and developmental stages can enable the development of new varieties.These genomic and transcriptomic analyses will provide a valuable resource that informs future basic and agricultural studies of the opium poppy. | Li Pei Baishi Wang Jian Ye Xiaodi Hu Lihong Fu Kui Li Zhiyu Ni Zhenlong Wang Yujie Wei Luye Shi Ying Zhang Xue Bai Mengwan Jiang Shuhui Wang Chunling Ma Shujin Li Kaihui Liu Wanshui Li Bin Cong | 2021 | Horticulture Research2021,8,1: | 4 |
| 3 | Effect of heat treatment on microstructures and properties of a novel Al-Zn-Mg-Cu alloy for oil drilling显示文摘In view of the special requirements for strength, heat resistance and corrosion resistance of Al-Zn-Mg-Cu alloy for oil drilling, the Al-6.2 Zn-2.5 Mg-1.6 Cu alloy was prepared by increasing Cu content on basis of Russian Series 1953 alloy. The effect of heat treatment on the microstructures and properties of the alloy was characterized by optical microscope(OM), scanning electron microscope(SEM) and transmission electron microscope(TEM), and investigated by tensile test at room temperature, thermal exposure test and corrosion test. The results show that the strength after T6 aging treatment exhibit a decrease trend as an increase of the solution temperature from465 ℃ to 480 ℃. After the solution treated by the rate of 470 ℃/1 h, second phases dissolve into the matrix very well and the strength property reaches optimum. The alloy has better comprehensive properties treated by a solution treatment of 470 ℃/1 h and then followed by an aging treatment of 120 ℃/24 h + 170 ℃/1 h + 120 ℃/24 h. Under the aging state, the precipitated phases inside the grains are suitable in size, while on the grain boundary distribute discontinuously and the precipitate-free zone is obvious. Besides, the alloy still maintain high tensile properties. The yield strength, tensile strength and elongation are 650 MPa, 686 MPa,12.0%, respectively. The yield strength retention after heat exposure is 92%. The alloy has good corrosion resistance and the exfoliation corrosion degree. The average corrosion rate in the H2S and CO2 environment is 0.0024 mm/a, which is far less than the required 0.12 mm/a. | 4 | Potential applications of synchrotron radiation to the treatment of cancer显示文摘Although conventional radiotherapy remains to be one of the most useful treatments for cancer, it is not the best strategy to maximize the effects on the tumors and minimize the damage to the surrounding tissues due to its physical and biological characteristics. Synchrotron radiation (SR) with uniquely physical and biological advantages may represent an innovative approach for cancer treatment. In recent years, SR-based photon activation therapy, stereotactic synchrotron radiation therapy and micro-beam radiation treatment have been developed, and the results of in vitro and in vivo experiments are very promising. It is necessary to understand the physical and radiobiological principle of those novel strategies before the approach is applied to the clinic. In this paper, we summarize the advances of SR in terms of physical, radiobiological advantages and its potential clinical applications. | 6 | Discriminating hydrocarbon generation potential of coaly source rocks and their contribution:a case study from the Upper Paleozoic of Bohai Bay Basin,China显示文摘Although various coaly source rocks widely developed in the Carboniferous-Permian(C-P)of the Bohai Bay Basin,their geochemical characteristics and hydrocarbon generation potential are poorly understood.This study aims to discriminate the contribution of hydrocarbon generation from different C-P coaly source rocks and clarify the differences within generated oils using organic geochemistry,organic petrology,and thermal simulation experiments.The coaly source rocks containt coal clarain and durain,carbonaceous shale,and shale deposited in deltaic and lagoonal environment.The results indicated that clarain,durain,and carbonaceous shale exhibited higher hydrogen index and liquid-gas hydrocarbon yields than lagoonal and deltaic shales,which was mainly associated with the concentrations of sporinite,cutinite,and hydrogen-rich collodetrinite.Aliphatic hydro-carbons originated firom coal and carbonaceous shale presented lower Ts/(Ts+Tm),Ga/17α(H)21β(H)-C_(30)hopane,18α(H)-oleanane/17α(H)21β(H)-C_(30)hopane ratios,and higher 17β3(H)21α(H)-C_(30)Morane/17α(H)21β(H)-C_(30)hopane than deltaic lagoonal shales.Parameters of aromatic hydrocarbons generated from five lithologies of coaly source rocks trended as clear group distribution,e.g.,clarain and durain showing lower MNR,DBT/Fluorene(F)ratios and higher DBF/F ratio than coaly shales.The distinct descending trend of hydrocarbon potential is obtained from clarain,durain,carbonaceous shale to lagoonal and deltaic shales,implying dominated the petroleum and natural gas supplement from coal and carbonaceous shale.The difference between aliphatic and aromatic hydrocarbons provides a significant contribution to analyze the generic relationship between coaly source rock and lacustrine shale.Our results illustrate the importance of coaly source rocks for the in-depth oil-gas exploration of the Bohai Bay Basin and understanding hydrocarbon generation potential of source rocks in coal bearing strata. | Jinjun XU Da LOU Qiang JIN Lixin FU Fuqi CHENG Shuhui ZHOU Xiuhong WANG Chao LIANG Fulai LI | 2021 | Frontiers of Earth Science2021,15,4: | 1 |

| 10 | Effect of periodic wide atmospheric pressure change on CO emission in closed goaf显示文摘Atmospheric pressure fuctuation is one of the most important factors afecting the climate environment and gas emission in the fre area.To obtain the infuence rule of the surface atmospheric pressure change on the gas sampling and abnormal emission in the mine closed goaf,the No.1 coal mine in Dananhu was taken as the research object.Using Fourier transform and Fisher harmonic analysis and other statistical methods,the infuence of the periodic variation of atmospheric pressure on the gas leakage and outfow in the closed goaf was studied.The results showed that there were three atmospheric pressure periods of 15.2 d,1 d and 182.2 d,and the probability was greater than 95%.The time period with the highest number of atmospheric pressure peaks was 7:00–8:00,which accounted for 20.2%of total occurrence number in a day.And the time periods with the highest number of atmospheric pressure trough were 2:00,15:00 and 16:00,accounting for 27.4%.The peak-to-trough transition time was mainly concentrated around 6 h,and the diurnal variation curve of atmospheric pressure was mainly bimodal.The atmospheric pressure change rate was mostly concentrated in 10–50 Pa/h.It was determined that the distance that the gas sampling pipe was pre-laid into the inner side of the closed wall should be greater than 44.4 m,and the CO concentration and atmospheric pressure in the closed goaf were both periodic and negative with atmospheric pressure.The research results have important guiding signifcance for the monitoring,early warning and environmental protection of the goaf. | Zhuangzhuang Shao Bo Tan Ruili Hu Xiaozhen Song Yan Guo Tianze Li Shuhui Fu Chaohuan Chen Haiyan Wang | 2023 | International Journal of Coal Science & Technology2023,10,4: | 0 |
| 11 | 改性煤基腐黑物对Cd(Ⅱ)的吸附效果及其机理研究显示文摘针对山西煤基腐黑物(SCH)碱性高和对重金属吸附率低的问题,以山西省灵石县风化煤为原料,提取SCH,采用Ca(OH)2对SCH进行改性制备改性山西煤基腐黑物(Ca-SCH),并通过表征分析和静态吸附实验,探究SCH和Ca-SCH对Cd(Ⅱ)的吸附动力学和热力学特性。研究结果表明,通过扫描电子显微镜(SEM)、比表面积(BET)法和X射线光电子能谱(XPS)证实Ca(Ⅱ)可以成功负载到SCH表面,且改性后Ca-SCH表面粗糙,孔径增大,含氧官能团含量增加,利于对Cd(Ⅱ)的吸附。SCH和Ca-SCH在120 min内达到吸附平衡,符合准二级动力学、Langmuir和Freundlich模型。Ca-SCH对Cd(Ⅱ)的最大吸附量(Qm)为89.29 mg/g,较SCH增加了23.23%。化学改性后,Ca(Ⅱ)负载到SCH表面,通过Ca(Ⅱ)和Cd(Ⅱ)的离子交换可提高SCH的吸附性能,对土壤中Cd(Ⅱ)的钝化具有重要意义。 | Ping Wang Fangjun Ding Zhanbin Huang Zhanyong Fu Peng Zhao Shuhui Men 王平(译) 付战勇(译) 黄占斌(译) 董政(译) | 2023 | 腐植酸2023,,3: | 0 |
