| 1 | Elevated plasma levels of selective cytokines in COVID-19 patients reflect viral load and lung injury显示文摘A recent outbreak of pneumonia in Wuhan,China was found to be caused by a 2019 novel coronavirus(2019-nCoV or SARS-CoV-2 or HCoV-19).We previously reported the clinical features of 12 patients with2019-n Co V infections in Shenzhen,China.To further understand the pathogenesis of COVID-19 and find better ways to monitor and treat the disease caused by 2019-n Co V,we measured the levels of 48 cytokines in the blood plasma of those 12 COVID-19 patients.Thirty-eight out of the 48 measured cytokines in the plasma of 2019-n Co V-infected patients were significantly elevated compared to healthy individuals.Seventeen cytokines were linked to 2019-nCoV loads.Fifteen cytokines,namely M-CSF,IL-10,IFN-α2,IL-17,IL-4,IP-10,IL-7,IL-1 ra,G-CSF,IL-12,IFN-γ,IL-1α,IL-2,HGF and PDGF-BB,were strongly associated with the lung-injury Murray score and could be used to predict the disease severity of2019-n Co V infections by calculating the area under the curve of the receiver-operating characteristics.Our results suggest that 2019-nCoV infections trigger extensive changes in a wide array of cytokines,some of which could be potential biomarkers of disease severity of 2019-n Co V infections.These findings will likely improve our understanding of the immunopathologic mechanisms of this emerging disease.Our results also suggest that modulators of cytokine responses may play a therapeutic role in combating the disease once the functions of these elevated cytokines have been characterized. | Yingxia Liu Cong Zhang Fengming Huang Yang Yang Fuxiang Wang Jing Yuan Zheng Zhang Yuhao Qin Xiaoyun Li Dandan Zhao Shunwang Li Shuguang Tan Zhaoqin Wang Jinxiu Li Chenguang Shen Jianming Li Ling Peng Weibo Wu Mengli Cao Li Xing Zhixiang Xu Li Chen Congzhao Zhou William J Liu Lei Liu Chengyu Jiang | 2020 | National Science Review2020,7,6: | 6 |
| 2 | Tool Orientation Optimization and Path Planning for 5-Axis Machining显示文摘Tool path generation is a fundamental problem in 5-axis CNC machining, which consists of tool orientation planning and cutter-contact(CC) point planning. The planning strategy highly depends on the type of tool cutters. For ball-end cutters, the tool orientation and CC point location can be planned separately;while for flat end cutters, the two are highly dependent on each other. This paper generates a smooth tool path of workpiece surfaces for flat end mills from two stages: Computing smooth tool orientations on the surface without gouging and collisions and then designing the CC point path. By solving the tool posture optimization problem the authors achieve both the path smoothness and the machining efficiency. | 5 | Carbonization fabrication of a piezoresistive sensor with improved sensitivity via Ni decoration of carbonized cotton fibers显示文摘The development of wearable electronics urgently requires the cost-effective and scalable fabrication of high-performance pressure sensors.This work aims to develop a simple carbonization strategy to facilitate sensor sensitivity by decorating discrete nickel nanoparticles on carbonized cotton fibers(CCFs).The increased air gap between the fibers at the unloading state,as well as the enlargement of the deformation distance and the contact area between the conductive materials at the loading state,contribute to a more significant resistance change.Therefore,the sensitivity of the piezoresistive sensor is improved more than 5 times within 1 N by introducing Ni nanoparticles,and it is characterized by a rapid response(~160 ms)and recovery(~100 ms),wide detection range(~20 N/~130 kPa),and good durability(~4000 cycles).The flexible sensor has been successfully demonstrated to monitor human movements,physical stimuli,and pressure distribution.Furthermore,the proposed device can control temperature accurately as a uniform and large-scale heater.This work reveals that the Ni@CCFs-based sensor is prospective in wearable electronics,artificial intelligence,health monitoring,medical diagnosis and treatment. | HE Xiang WANG FengMing LIANG YiYing WU JiaWei LIANG JiongHong SHEN GengZhe LI BaiJun SU DaoJian ZHANG Chi SONG WeiDong HE Xin | 2022 | Science China(Technological Sciences)2022,65,12: | 0 |

| 7 | Predicting and overcoming resistance to CDK9 inhibitors for cancer therapy显示文摘Abnormally activated CDK9 participates in the super-enhancer mediated transcription of short-lived proteins required for cancer cell survival.Targeting CDK9 has shown potent anti-tumor activity in clinical trials among different cancers.However,the study and knowledge on drug resistance to CDK9 inhibitors are very limited.In this study,we established an AML cell line with acquired resistance to a highly selective CDK9 inhibitor BAY1251152.Through genomic sequencing,we identified in the kinase domain of CDK9 a mutation L156F,which is also a coding SNP in the CDK9 gene.By knocking in L156F into cancer cells using CRISPR/Cas9,we found that single CDK9 L156F could drive the resistance to CDK9 inhibitors,not only ATP competitive inhibitor but also PROTAC degrader.Mechanistically,CDK9 L156F disrupts the binding with inhibitors due to steric hindrance,further,the mutation affects the thermal stability and catalytic activity of CDK9 protein.To overcome the drug resistance mediated by the CDK9-L156F mutation,we discovered a compound,IHMT-CDK9-36 which showed potent inhibition activity both for CDK9 WT and L156F mutant.Together,we report a novel resistance mechanism for CDK9 inhibitors and provide a novel chemical scaffold for the future development of CDK9 inhibitors. | Chen Hu Lijuan Shen Fengming Zou Yun Wu Beilei Wang Aoli Wang Chao Wu Li Wang Jing Liu Wenchao Wang Qingsong Liu | 2023 | Acta Pharmaceutica Sinica B2023,13,9: | 0 |

| 9 | A Neural Circuit Controlling Virgin Female Aggression Induced by Mating-related Cues in Drosophila显示文摘Females increase aggression for mating opportunities and for acquiring reproductive resources.Although the close relationship between female aggression and mating status is widely appreciated,whether and how female aggression is regulated by mating-related cues remains poorly understood.Here we report an interesting observation that Drosophila virgin females initiate high-frequency attacks toward mated females.We identify 11-cis-vaccenyl acetate(cVA),a male-derived pheromone transferred to females during mating,which promotes virgin female aggression.We subsequently reveal a cVA-responsive neural circuit consisting of four orders of neurons,including Or67d,DA1,aSP-g,and pC1 neurons,that mediate cVA-induced virgin female aggression.We also determine that aSP-g neurons release acetylcholine(ACh)to excite pC1 neurons via the nicotinic ACh receptor nAChRα7.Together,beyond revealing cVA as a mating-related inducer of virgin female aggression,our results identify a neural circuit linking the chemosensory perception of mating-related cues to aggressive behavior in Drosophila females. | Xiaolu Wan Peng Shen Kai Shi Jing Li Fengming Wu Chuan Zhou | 2023 | Neuroscience Bulletin2023,39,9: | 0 |
