1Establishment and application of distant hybridization technology in fish显示文摘Hybridization is widely used. However, for a long time, systematic theories and technologies related to hybridization in fish have been lacking. In this study, through long-term systematic research, we investigated and obtained the main rules regarding inheritance and reproduction related to fish distant hybridization. Furthermore, we established one-step and multistep breeding technologies that were suitable for interspecific hybridization and intraspecific hybridization. Simultaneously, we used these two breeding technologies to produce a batch of diploid fish lineages and tetraploid fish lineages and improved fishes. In addition, we widely discuss the methods, technologies and results of hybridization breeding, referring to the domestic and foreign literature on fish hybridization. 2The chimeric genes in the hybrid lineage of Carassius auratus cuvieri(♀)×Carassius auratus red var.(♂)显示文摘Hybridization can combine the genomes of different strains or species, which leads to changes of genotype and phenotype in the hybrids. In this study, we aimed to investigate the genetic variations of hybrids(WR-F1 and WR-F2) derived from the intraspecific hybridization of white crucian carp(Carassius auratus cuvieri, WCC, ♀) and red crucian carp(Carassius auratus red var., RCC, ♂). Here, we compared the orthologous genes in the liver transcriptomes of hybrids with those of WCC and RCC, and classified the orthologous genes into eight gene patterns within three categories(chimera, mutant, and biparental origin genes).The results revealed 19.04%, 4.17% chimeric genes and 6.90%, 5.05% mutations of orthologous genes in WR-F1 and WR-F2 respectively. Seventeen of twenty-three characterized genes(77%) were confirmed to be the chimeras at the genomic DNA level.The GO classification discovered that some chimeric and mutant genes were related to metabolic process, immune system and developmental process in WR-F1. Our results provide the new evidence that hybridization can combine the parental genomes,leading to changes in the genotype of the resultant hybrids. 3Comparison of diploid and triploid Carassius auratus provides insights into adaptation to environmental change显示文摘Focusing on adaptation of aquatic organisms, especially fish, can help elucidate complex dynamics in freshwater ecology. The differences in genetic and epigenetic regulation between diploid and triploid Carassius auratus affect survival under eutrophication. To identify the underlying mechanisms that lead to better adaption of triploids than diploids, we compared mRNA and microRNA(miRNA) expressions in liver tissue of diploid and triploid individuals obtained from the Dongting lake water system in central China. Differential expression analysis revealed that 566 transcripts were significantly up-regulated, whereas758 were down-regulated in triploids; of these differentially expressed transcripts, 33 transcripts including cacna1 d, nfkb2, hspa1 and fgfr4 were involved in the MAPK signaling pathway, and eight transcripts were determined to be regulated by seven miRNAs. Additionally, four of 25 differential expressed(DE) transcripts(mhc1, irf7, nfkb2 and pik3 c) involving the viral carcinogenesis pathway were regulated by four miRNAs. Furthermore, genetic polymorphisms analysis showed that more heterozygous mutations were detected in triploids than diploids. The d N/d S results revealed that 21 genes were under positive selection(d N/d S>1) in C. auratus complex. 4Evidence for the paternal mitochondrial DNA in the crucian carp-like fish lineage with hybrid origin显示文摘In terms of taxonomic status,common carp(Cyprinus carpio,Cyprininae)and crucian carp(Carassius auratus,Cyprininae)are different species;however,in this study,a newborn homodiploid crucian carp-like fish(2n=100)(2nNCRC)lineage(F1–F3)was established from the interspecific hybridization of female common carp(2n=100)×male blunt snout bream(Megalobrama amblycephala,Cultrinae,2n=48).The phenotypes and genotypes of 2 n NCRC differed from those of its parents but were closely related to those of the existing diploid crucian carp.We further sequenced the whole mitochondrial(mt)genomes of the 2n NCRC lineage from F1 to F3.The paternal mt DNA fragments were stably embedded in the mt-genomes of F1–F3 generations of 2n NCRC to form chimeric DNA fragments.Along with this chimeric process,numerous base sites of F1–F3 generations of 2 n NCRC underwent mutations.Most of these mutation sites were consistent with the existing diploid crucian carp.Moreover,the mt DNA organization and nucleotide composition of 2n NCRC were more similar to those of the existing diploid crucian carp than those of the parents.The inheritable chimeric DNA fragments and mutant loci in the mt-genomes of different generations of 2nNCRC provided important evidence of the mt DNA change process in the newborn lineage derived from hybridization of different species.Our findings demonstrated for the first time that the paternal mt DNA were transmitted into the mt-genomes of homodiploid lineage,which provided new insights into the existence of paternal mt DNA in the mt DNA inheritance.Shi Wang Ni Jiao Lu Zhao Meiwen Zhang Pei Zhou Xuexue Huang Fangzhou Hu Conghui Yang Yuqin Shu Wuhui Li Chun Zhang Min Tao Bo Chen Ming Ma Shaojun Liu 2020Science China(Life Sciences)2020,63,1:2
5Advances of nanoparticles as drug delivery systems for disease diagnosis and treatment显示文摘Decades have passed since the first nanoparticles-base medicine was approved for human cancer treatment, and the research and development of nanoparticles for drug delivery are always undergoing.Nowadays, the significant advances complicate nanoparticles’ branches, including liposomes, solid lipid nanoparticles, inorganic nanoparticles, micelles, nanovaccines and nano-antibodies, etc. These nanoparticles show numerous capabilities in treatment and diagnosis of stubborn diseases like cancer and neurodegenerative diseases, emerging as novel drug carriers or therapeutic agents in future. In this review, the complicated branches of nanoparticles are classified and summarized, with their property and functions concluded. Besides, there are also some delivery strategies that make nanoparticles smarter and more efficient in drug delivery, and frontiers in these strategies are also summarized in this review. 6Efficacy of Cigu Xiaozhi pill(慈菇消脂丸) on non-alcoholic steatohepatitis-associated lipoapoptosis through stress-activated c-Jun N-terminal kinase signalling pathway显示文摘OBJECTIVE: To investigate the efficacy of Cigu Xiaozhi pill(慈菇消脂丸, CGXZ) on non-alcoholic steatohepatitis(NASH)-associated lipoapoptosis through the stress-activated c-Jun N-terminal kinase(JNK)/stress-activated protein kinase signalling pathway.METHODS: Sixty male Sprague-Dawley rats were randomly divided into the following groups(10 rats each): blank control, model, low-dose CGXZ,medium-dose CGXZ, high-dose CGXZ, and positive control(treated with SP600125, a JNK inhibitor).The NASH model was established and the histomor-phological characteristics of haematoxylin and eosin-stained liver tissues were examined under a light microscope. Cell apoptosis in liver tissues was assessed via terminal deoxynucleotidyl transferase d UTP nick-end labelling assay. In addition, the m RNA and protein expression levels of p-JNK,p-c-Jun, caspase-8, Fas, and Fas-L were determined via fluorescence-based quantitative real-time PCR,immunohistochemical and Western blot assays.RESULTS: Histopathological examination of the liver showed that the model rats had moderate-to-severe steatosis with infiltration of inflammatory cells as well as significantly higher expression levels of the p-JNK, p-c-Jun, caspase-8, Fas, and Fas-L proteins, compared with those in the blank control group(P < 0.01). Hepatic lobules of the rats in the treatment groups showed significantly reduced vacuolar degeneration and steatosis as well as alleviated inflammatory cell infiltration. The high and medium-dose CGXZ groups exhibited significantly lower m RNA and protein expression levels of p-JNK, p-c-Jun, caspase-8, Fas, and Fas-L, compared with those in the model group(P < 0.05 or P <0.01).CONCLUSION: CGXZ pill inhibited the onset of hepatocyte apoptosis by regulating the expression of p-JNK, p-c-Jun, caspase-8, Fas, and Fas-L, thereby exerting therapeutic effects against NASH.MA Yanhua YU Chengzu WU Yan HAN Tao YANG Shaojun SHI Xia 2021Journal of Traditional Chinese Medicine2021,41,1:2
7Further evidence for paternal DNA transmission in gynogenetic grass carp显示文摘Gynogenesis is an important breeding method in aquaculture and has been widely applied to many fish species.If gynogenetic progenies are to inherit paternal partial genomic DNA,this will increase genetic variation and will provide a useful outcome for breeding.In this study,we investigated the genetic variation in homeobox(Hox)gene clusters(Hox A4 a,Hox A9 a,Hox A11 b,Hox B1 b,Hox C4 a,Hox C6 b,and Hox D10 a)among koi carp(Cyprinus carpio haematopterus,KOC;the stimulation sperm source),grass carp(Ctenopharyngodon idellus),and gynogenetic grass carp(GGC).We found paternal DNA(a special DNA fragment and Hox C6 b)derived from KOC and a recombinant gene belonging to Hox C6 b in GGC.We are the first to report the recombinant Hox C6 b in GGC.Our study provides further evidence for paternal DNA transmission to gynogenetic progenies,which is a finding with great significance for the genetic breeding of fish.Zhuangwen Mao Yeqing Fu Shi Wang Yude Wang Kaikun Luo Chun Zhang Min Tao Shaojun Liu 2020Science China(Life Sciences)2020,63,9:2

14Rapid sterilisation and diabetic cutaneous regeneration using cascade bio-heterojunctions through glucose oxidase-primed therapy显示文摘The cutaneous wound in diabetic patients frequently encounters intractable pathogenic infections due to the hyperglycemia micromilieu which is conducive to bacterial growth and multiplication.Despite the extensive clinical use of antibiotics to treat bacterial infections,the emergence of drug-resistant and super pathogens as well as the potential side effects of antibiotics have elicited alarming challenges to public health.To address this daunting concern,we devise and develop a photo-activated cascade bio-heterojunctions(C-bio-HJs)for rapid sterilization and diabetic cutaneous regeneration.In the designed C-bio-HJs,photo-generated electron-hole pairs of graphite-phase carbon nitride(g-C_(3)N_(4))are effectively separated with the marriage of molybdenum disulfide(MoS_(2)),which achieves the augmented photodynamic antibacterial effect.Moreover,glucose oxidase(GOx)tethered on the bio-HJs catalyzes glucose into hydrogen peroxide(H_(2)O_(2))in diabetic wounds for starvation therapy.Furthermore,Mo4+enables the catalysis of H_(2)O_(2)into a highly effective hydroxyl radical(⋅OH)for chemodynamic-photothermal combined antibacterial therapy.Both in vitro and in vivo results authenticate the cascading antibacterial properties and skin regeneration-promoting effects of the C-bio-HJs,which provide a facile strategy to combat diabetic wound healing through the synergistic GOx-primed dynamic therapies.Yi Deng Xumei Ouyang Jiyu Sun Xiuyuan Shi Yunfei Li Yau Kei Chan Weizhong Yang Shaojun Peng 2023Bioactive Materials2023,,7:1
15Adsorption of Nin(n=1-4)clusters on perfect and O-defective CuAl2O4 surfaces:A DFT study显示文摘The density functional theory was employed to investigate the adsorption of Nin (n=1–4) on the perfect and O-defect CuAl2O4 surfaces. The computational results show that for single Ni atom on the perfect spinel (100) surface, the adsorption energy is-5.30 eV, much larger than Ni on other CuAl2O4 surfaces. The adsorption of Nin (n=1–4) absorbed on the O-defect CuAl2O4 (100) surface is less stable than on the perfect CuAl2O4 (100) surface. However, the adsorption energy for Nin (n=1–4) on the O-defect CuAl2O4 (110) surface is close to on the perfect CuAl2O4 (110) surface. Bader charge and partial density of states (PDOS) analysis revel that the adsorption of Ni on the CuAl2O4 spinel surface is accompanied by charge transfer from the metal to the support. The growth and aggregations analysis show that the general growth and aggregation ability for Nin clusters follow the order:gas phase>γ-Al2O3 (110)>CuAl2O4 (110)>CuAl2O4 (100). 17Cytoreductive Surgery and Hyperthermic Intraperitoneal Chemotherapy for Pseudomyxoma Peritonei of Appendiceal Origin-801 Cases from a Single Institution in China显示文摘Aim:As more and more centers has published their treatment results of pseudomyxoma peritonei(PMP)with cytoreductive surgery(CRS)and hyperthermic intraperitoneal chemotherapy(HIPEC),the data from China is missing.Myxoma Department of Aerospace Hospital is the biggest center treating PMP in China.The purpose of this study is to report the early and long-term outcomes for PMP from this single center.Methods:801 appendix-derived PMP out of 1008 consecutive patients treated in Myxoma Department of Aerospace Hospital between 2008 and 2019 were retrospectively analyzed.Results:Complete cytoreductive surgery(CCRS)was achieved in 240(30%)patients with median PCI of 14(1~39),and the rest had maximal tumor debulking(MTD),HIPEC was implemented in 96.3%of CCRS and 78.6%of MTD.The major morbidity(grade III/IV)was 11.4%and the 30-day operative mortality is 0.7%.The 5-and 10-year OS of CCRS was 76.9%and 64.1%,which is significantly higher than MTD(5-,10-year OS as 36.1%,27.1%;p<0.001).On the univariate analysis,all prognostic factors(gender,PSS,interval time,prior chemotherapy,prior HIPEC,Peritoneal Cancer Index(PCI),completeness of cytoreduction(CC),HIPEC,pathology,present of serous ascites)were found to be associated with overall survival except for age.On multivariate analysis,only PCI>20,MTD,high pathologic grade and without HIPEC were independent factors predicting poorer prognosis.Conclusions:CCRS+HIPEC can benefit PMP well with controllable risks.MTD+HIPEC may benefit PMP as well when CCRS cannot be achieved after fully asscessment by an experienced peritoneal maglignacy center,but the surgery should be performed as limited as possible.Ao Xia Xichao Zhai Lubiao An Bing Wang Guanjun Shi Ying Cai Yiyan Lu Shaojun Pang Feng Chen Hongbin Xu Ruiqing Ma 2020Journal of Oncology Research2020,2,2:0
18Ti_(3)C_(2)T_(x)-TiSe_(2) analogous heterostructure for flexible zinc ion battery显示文摘Exploring zinc-free anode materials is one of the effective strategies to get the zinc dendrites problem of flexible zinc ion battery(ZIB)solved.In this work,an analogous heterostructure(AHS)is constructed from the excellent MXene(Ti_(3)C_(2)T_(x))and TiSe_(2) nanosheets.The AHS not only possesses numerous diffu-sion paths and Zn^(2+)storage sites but also possesses a stable conductive network to accelerate charge transfer in the electrode.As a collaborative advantage,electrochemical measurement results show that MXene/TiSe_(2) electrodes display an excellent specific capacity of 177.9 mAh g^(-1) at 0.10 A g^(-1) and a long-term cycling stability of 77.4%capacity retention after 400 cycles.DFT computations further demon-strate the excellent performance of MXene/TiSe_(2) electrodes including desirable electronic conductivity and low Zn^(2+)migration barriers.The assembled flexible ZIB not only delivers a good specific capacity of 42.2μAh cm^(-2) at 0.20 mA cm^(-2) and a competitive energy density of 37.4μWh cm^(-2) but also exhibits excellent flexibility and thermostability.Furthermore,after 400 cycles at 0.60 A g^(-1),flexible ZIB shows a capacity retention of 73.8%.This work gives a successful attempt to design 2D layered materials as Zn metal-free anode for flexible ZIB.Li Wen Junjie Shi Qixiang Zhang Fei Wang Siliang Wang Shaojun Zhang Qiang Wang Ke Mao Fei Long Yihua Gao 2023Journal of Materials Science & Technology2023,,19:0
19An improved hybrid bream derived from a hybrid lineage of Megalobrama amblycephala(♀)×Culter alburnus(♂)显示文摘Distant hybridization is an important technique in fish genetic breeding.In this study,based on the establishment of an allodiploid fish lineage(BT,2n=48,F_(1)–F_(6))derived from distant hybridization between female Megalobrama amblycephala(BSB,2n=48)and male Culter alburnus(TC,2n=48),and the backcross progeny(BTB,2n=48)derived by backcrossing female F_(1) of BT to male BSB,an improved hybrid bream(BTBB,2n=48)was obtained by backcrossing BTB(♀)to BSB(♂).Moreover,the morphological and genetic characteristics of BTBB individuals were investigated;BTBB was similar to BSB in appearance but had a higher body height than BSB.The study results regarding chromosome numbers and DNA content indicated that BTBB is a diploid hybrid fish.The 5S rDNA and Hox gene of BTBB were inherited from the original parents.Gonadal development in BTBB was normal.On the other hand,BTBB had a faster growth rate,higher muscle protein level,and lower muscle carbohydrate level than BSB.Hence,bisexual fertile BTBB is promoted and can be applied as a high-quality fish,and it can also be used as a new fish germplasm resource to develop high-quality fish further.Thus,this study is of great significance for fish genetic breeding.Dingbin Gong Min Tao Lihui Xu Fangzhou Hu Zehong Wei Shi Wang Yude Wang Qingfeng Liu Chang Wu Kaikun Luo Chenchen Tang Rong Zhou Chun Zhang Yuequn Wang Shaojun Liu 2022Science China(Life Sciences)2022,65,6:0
20Microstructure and mechanical properties of Mg-3Sn-1Ca reinforced with AlN nano-particles显示文摘Microstructural evolution and strengthening mechanisms of Mg-3Sn-1Ca based alloys with additions of different amounts of Al N nanoparticles were investigated.It was found that with increasing the amount of AlN nano-particles the grain size decreases obviously.The existence of AlN nano-particles could refine the primary crystal phases CaMgSn,which provided more heterogeneous nucleation sites for the formation of magnesium.Moreover,such nano-particles could also restrict the grain growth during solidification.After adding AlN nano-particles,both the tensile properties at room temperature and high temperature 250℃and the hardness are largely improved.The improvement of strength is attributed to grain refinement and second phase refinement.Shaozhu Wang Yuanding Huang Lixiang Yang Ying Zeng Yaozeng Hu Xiao Zhang Qiang Sun Shaojun Shi Guangyao Meng Norbert Hort 2023Journal of Magnesium and Alloys2023,11,1:0
