| 1 | RAB31 marks and controls an ESCRT-independent exosome pathway显示文摘Exosomes are generated within the multivesicular endosomes(MVEs)as intraluminal vesicles(ILVs)and secreted during the fusion of MVEs with the cell membrane.The mechanisms of exosome biogenesis remain poorly explored.Here we identify that RAB31 marks and controls an ESCRT-independent exosome pathway.Active RAB31,phosphorylated by epidermal growth factor receptor(EGFR),engages flotillin proteins in lipid raft microdomains to drive EGFR entry into MVEs to form ILVs,which is independent of the ESCRT(endosomal sorting complex required for transport)machinery.Active RAB31 interacts with the SPFH domain and drives ILV formation via the Flotillin domain of flotillin proteins.Meanwhile,RAB31 recruits GTPase-activating protein TBC1D2B to inactivate RAB7,thereby preventing the fusion of MVEs with lysosomes and enabling the secretion of ILVs as exosomes.These findings establish that RAB31 has dual functions in the biogenesis of exosomes:driving ILVs formation and suppressing MVEs degradation,providing an exquisite framework to better understand exosome biogenesis. | Denghui Wei Weixiang Zhan Ying Gao Liyan Huang Run Gong Wen Wang Ruhua Zhang Yuanzhong Wu Song Gao Tiebang Kang | 2021 | Cell Research2021,31,2: | 12 |

| 3 | PHYTOCHROME-INTERACTING FACTORS Interact with the ABA Receptors PYL8 and PYL9 to Orchestrate ABA Signaling in Darkness显示文摘PHYTOCHROME-INTERACTING FACTORS(PIFs)are a group of basic helix-loop-helix transcription factors that can physically interact with photoreceptors,including phytochromes and cryptochromes.It was previously demonstrated that PIFs accumulated in darkness and repressed seedling photomorphogenesis,and that PIFs linked different photosensory and hormonal pathways to control plant growth and development.In this study,we show that PIFs positively regulate the ABA signaling pathway during the seedling stage specifically in darkness.We found that PIFs positively regulate ABI5 transcript and protein levels in darkness in response to exogenous ABA treatment by binding directly to the G-box motifs in the ABI5 promoter.Consistently,PIFs and the G-box motifs in the ABI5 promoter determine ABI5 expression in darkness,and overexpression of ABI5 could rescue the ABA-insensitive phenotypes of pifq mutants in the dark.Moreover,we discovered that PIFs can physically interact with the ABA receptors PYL8 and PYL9,and that this interaction is not regulated by ABA.Further analyses showed that PYL8 and PYL9 promote PIF4 protein accumulation in the dark and enhance PIF4 binding to the ABI5 promoter,but negatively regulate PIF4-mediated ABI5 activation.Taken together,our data demonstrate that PIFs interact with ABA receptors to orchestrate ABA signaling in darkness by controlling ABI5 expression,providing new insights into the pivotal roles of PIFs as signal integrators in regulating plant growth and development. | Lijuan Qi Shan Liu Cong Li Jingying Fu Yanjun Jing Jinkui Cheng Hong Li Dun Zhang Xiaoji Wang Xiaojing Dong Run Han Bosheng Li Yu Zhang Zhen Li William Terzaghi Chun-Peng Song Rongcheng Lin Zhizhong Gong Jigang Li | 2020 | Molecular Plant2020,13,3: | 3 |
| 4 | Machine learning for detecting mesial temporal lobe epilepsy by structural and functional neuroimaging显示文摘Mesial temporal lobe epilepsy(mTLE),the most common type of focal epilepsy,is associated with functional and structural brain alterations.Machine learning(ML)techniques have been successfully used in discriminating mTLE from healthy controls.However,either functional or structural neuroimaging data are mostly used separately as input,and the opportunity to combine both has not been exploited yet.We conducted a multimodal ML study based on functional and structural neuroimaging measures.We enrolled 37 patients with left mTLE,37 patients with right mTLE,and 74 healthy controls and trained a support vector ML model to distinguish them by using each measure and the combinations of the measures.For each single measure,we obtained a mean accuracy of 74%and 69%for discriminating left mTLE and right mTLE from controls,respectively,and 64%when all patients were combined.We achieved an accuracy of 78%by integrating functional data and 79%by integrating structural data for left mTLE,and the highest accuracy of 84%was obtained when all functional and structural measures were combined.These findings suggest that combining multimodal measures within a single model is a promising direction for improving the classification of individual patients with mTLE. | Baiwan Zhou Dongmei An Fenglai Xiao Running Niu Wenbin Li Wei Li Xin Tong Graham J Kemp Dong Zhou Qiyong Gong Du Lei | 2020 | Frontiers of Medicine2020,14,5: | 2 |
