Kupffer cell restoration after partial hepatectomy is mainly driven by local cell proliferation in IL-6-dependent autocrine and paracrine manners显示文摘Kupffer cells(KCs),which are liver-resident macrophages, originate from the fetal yolk sac and represent one of the largest macrophage populations in the body. However, the current data on the origin of the cells that restore macrophages during liver injury and regeneration remain controversial. Here,we address the question of whether liver macrophage restoration results from circulating monocyte infiltration or local KC proliferation in regenerating livers after partial hepatectomy(PHx)and uncover the underlying mechanisms.By using several strains of genetically modified mice and performing immunohistochemical analyses,we demonstrated that local KC proliferation mainly contributed to the restoration of liver macrophages after PHx.Peak KC proliferation was impaired in Il6-knockout(KO)mice and restored after the administration of IL-6 protein, whereas KC proliferation was not affected in Il4-KO or Csf2-KO mice. The source of IL-6 was identified using hepatocyte-and myeloid-specific Il6-KO mice and the results revealed that both hepatocytes and myeloid cells contribute to IL-6 production after PHx.Moreover, peak KC proliferation was also impaired in myeloid-specific Il6 receptor-KO mice after PHx,suggesting that IL-6 signaling directly promotes KC proliferation.Studies using several inhibitors to block the IL-6 signaling pathway revealed that sirtuin 1(SIRT1)contributed to IL-6-mediated KC proliferation in vitro.Genetic deletion of the Sirt1 gene in myeloid cells, including KCs,impaired KC proliferation after PHx.In conclusion, our data suggest that KC repopulation after PHx is mainly driven by local KC proliferation, which is dependent on IL-6 and SIRT1 activation in KCs. | Yeni Ait Ahmed Yaojie Fu Robim M.Rodrigues Yong He Yukun Guan Adrien Guillot Ruixue Ren Dechun Feng Juan Hidalgo Cynthia Ju Fouad Lafdil Bin Gao | 2021 | Cellular & Molecular Immunology2021,18,9: | 2
Prediction of adolescent subjective well being: A machine learning approach显示文摘Background Subjective well-being(SWB),also known as happiness, plays an important role in evaluating both mental and physical health.Adolescents deserve specific attention because they are under a great variety of stresses and are at risk for mental disorders during adulthood.Aim The present paper aims to predict undergraduate students1 SWB by machine learning method. Methods Gradient Boosting Classifier which was an innovative yet validated machine learning approach was used to analyse data from 10518 Chinese adolescents. The online survey included 298 factors such as depression and personality.Quality control procedure was used to minimise biases due to online survey reports. We applied feature selection to achieve the balance between optimal prediction and result interpretation. Results The top 20 happiness risks and protective factors were finally brought into the predicting model.Approximately 90%individuals'SWB can be predicted correctly, and the sensitivity and specificity were about 92%and 90%,respectively.Conclusions This result identifies at-risk individuals according to new characteristics and established the foundation for adolescent prevention strategies. | Naixin Zhang Chuanxin Liu Zhixuan Chen Lin An Decheng Ren Fan Yuan Ruixue Yuan Lei Ji Yan Bi Zhenming Guo Gaini Ma Fei Xu Fengping Yang Liping Zhu Gabirel Robert Yifeng Xu Lin He Bo Bai Tao Yu Guang He | 2019 | General Psychiatry2019,32,5: | 1
Hollow Fe_(2)O_(3)/Co_(3)O_(4)microcubes derived from metal-organic framework for enhanced sensing performance towards acetone显示文摘In this work, hollow Fe_(2)O_(3)/Co_(3)O_(4)microcubes have been successfully synthesized through a hydrothermal method followed by an annealing process using metal-organic framework of Prussian blue as a soft template. The morphologies, microstructures, surface area and element compositions have been carefully characterized by a series of techniques.Meanwhile, compared with that of pure Fe_(2)O_(3)and Co_(3)O_(4),the gas sensor based on the hollow microcubes exhibits enhanced sensing performances towards acetone,e.g.,a higher response of 21.2 and a shorter response time of 5 s towards 20 ppm acetone at a relatively low working temperature of 200℃.Moreover,the hollow microcubes-based gas sensor still shows perfect long-term stability, excellent repeatability and the ability of sub-ppm level detection, which provides a possibility for its application in real life. The enhanced gas sensing performances can be attributed to the hollow structure with a high surface area and the formed p-n heterojunctions within the microcubes. | Ruixue Mo Dongqiang Han Zeng Ren Dazhuang Yang Fei Wang Caolong Li | 2021 | Chinese Chemical Letters2021,32,11: | 1

Excitonic devices based on two-dimensional transition metal dichalcogenides van der Waals heterostructures显示文摘Excitonic devices are an emerging class of technology that utilizes excitons as carriers for encoding, transmitting, and storing information. Van der Waals heterostructures based on transition metal dichalcogenides often exhibit a type II band alignment, which facilitates the generation of interlayer excitons. As a bonded pair of electrons and holes in the separation layer, interlayer excitons offer the chance to investigate exciton transport due to their intrinsic out-of-plane dipole moment and extended exciton lifetime. Furthermore, interlayer excitons can potentially analyze other encoding strategies for information processing beyond the conventional utilization of spin and charge. The review provided valuable insights and recommendations for researchers studying interlayer excitonic devices within van der Waals heterostructures based on transition metal dichalcogenides. Firstly, we provide an overview of the essential attributes of transition metal dichalcogenide materials, focusing on their fundamental properties, excitonic effects, and the distinctive features exhibited by interlayer excitons in van der Waals heterostructures. Subsequently, this discourse emphasizes the recent advancements in interlayer excitonic devices founded on van der Waals heterostructures, with specific attention is given to the utilization of valley electronics for information processing, employing the valley index. In conclusion, this paper examines the potential and current challenges associated with excitonic devices. | Yulun Liu Yaojie Zhu Zuowei Yan Ruixue Bai Xilin Zhang Yanbo Ren Xiaoyu Cheng Hui Ma Chongyun Jiang | 2024 | Frontiers of Chemical Science and Engineering2024,18,2: | 0
G protein-coupled receptor 35 attenuates nonalcoholic steatohepatitis by reprogramming cholesterol homeostasis in hepatocytes显示文摘Nonalcoholic fatty liver disease(NAFLD)is the most common chronic liver disease worldwide.Fat accumulation“sensitizes”the liver to insult and leads to nonalcoholic steatohepatitis(NASH).G protein-coupled receptor 35(GPR35)is involved in metabolic stresses, but its role in NAFLD is unknown. We report that hepatocyte GPR35 mitigates NASH by regulating hepatic cholesterol homeostasis.Specifically,we found that GPR35 overexpression in hepatocytes protected against high-fat/cholesterol/fructose(HFCF)diet-induced steatohepatitis, whereas loss of GPR35 had the opposite effect.Administration of the GPR35 agonist kynurenic acid(Kyna)suppressed HFCF diet-induced steatohepatitis in mice.Kyna/GPR35 induced expression of StAR-related lipid transfer protein 4(STARD4)through the ERK1/2 signaling pathway, ultimately resulting in hepatic cholesterol esterification and bile acid synthesis(BAS).The overexpression of STARD4 increased the expression of the BAS rate-limiting enzymes cytochrome P450 family 7 subfamily A member 1(CYP7A1)and CYP8B1,promoting the conversion of cholesterol to bile acid. The protective effect induced by GPR35 overexpression in hepatocytes disappeared in hepatocyte STARD4-knockdown mice.STARD4 overexpression in hepatocytes reversed the aggravation of HFCF diet-induced steatohepatitis caused by the loss of GPR35 expression in hepatocytes in mice. Our findings indicate that the GPR35–STARD4 axis is a promising therapeutic target for NAFLD. | Xiaoli Wei Fan Yin Miaomiao Wu Qianqian Xie Xueqin Zhao Cheng Zhu Ruiqian Xie Chongqing Chen Menghua Liu Xueying Wang Ruixue Ren Guijie Kang Chenwen Zhu Jingjing Cong Hua Wang Xuefu Wang | 2023 | Acta Pharmaceutica Sinica B2023,13,3: | 0
