2A Novel Strategy to Fabricate Core-Sheath Structure Piezoelectric Yarns for Wearable Energy Harvesters显示文摘Wearable and portable electronic devices based on textile structure have been widely used owing to their wearability and comfortableness.However,yarn fineness and the comfort of the fabric cannot satisfy the requirements of smart wearable devices.This work presents a novel strategy to prepare highly integrated PVDF/conductive nylon core-sheath structure piezoelectric yarns for wearable which is fabricated by combining electrospinning strategy with 2D braiding technology.The fineness of single yarns as well as strength are both improved significantly compared to previous works.The piezoelectric outputs of the yarn are still stable after 800 s fatigue test at a frequency of 4 Hz,and the cycle stability can maintain more than 3200 cycles.Furthermore,the piezoelectric yarns are further woven into piezoelectric plain fabric.According to the electrical performance,the length of the piezoelectric yarn and the thickness of the piezoelectric layer would both affect the output electrical performance.The yarn of the 10 cm in length and 600μm in fineness can produce an output voltage of 120 mV.Meanwhile,Both the piezoelectric yarn and the fabric could generate piezoelectric output signals through human movement,such as bending,walking.Therefore,the electrical and mechanical performance of the piezoelectric yarns prepared in our work could be improved significantly,and the comfortableness and durability performance of the piezoelectric fabric can satisfy most wearing requirements,which would provide some help in the field of piezoelectric wearable devices based on yarns and fabrics.Lili Xue Wei Fan Yang Yu Kai Dong Chengkun Liu Yanli Sun Cong Zhang Weichun Chen Ruixin Lei Kai Rong Qi Wang 2021Advanced Fiber Materials2021,3,4:3
3SAMD4 family members suppress human hepatitis B virus by directly binding to the Smaug recognition region of viral RNA显示文摘HBV infection initiates hepatitis B and promotes liver cirrhosis and hepatocellular carcinoma.IFN-α is commonly used in hepatitis B therapy,but how it inhibits HBV is not fully understood.We screened 285 human interferon-stimulated genes(ISGs)for anti-HBV activity using a cell-based assay,which revealed several anti-HBV ISGs.Among these ISGs,SAMD4A was the strongest suppressor of HBV replication.We found the binding site of SAMD4A in HBV RNA,which was a previously unidentified Smaug recognition region(SRE)sequence conserved in HBV variants.SAMD4A binds to the SRE site in viral RNA to trigger its degradation.The SAM domain in SAMD4A is critical for RNA binding and the C-terminal domain of SAMD4A is required for SAMD4A anti-HBV function.Human SAMD4B is a homolog of human SAMD4A but is not an ISG,and the murine genome encodes SAMD4.All these SAMD4 proteins suppressed HBV replication when overexpressed in vitro and in vivo.We also showed that knocking out the Samd4 gene in hepatocytes led to a higher level of HBV replication in mice and AAV-delivered SAMD4A expression reduced the virus titer in HBVproducing transgenic mice.In addition,a database analysis revealed a negative correlation between the levels of SAMD4A/B and HBV in patients.Our data suggest that SAMD4A is an important anti-HBV ISG for use in IFN therapy of hepatitis B and that the levels of SAMD4A/B expression are related to HBV sensitivity in humans.Yuze Wang Xinrui Fan Yunlong Song Yifei Liu Ruixin Liu Jianfeng Wu Xiaoling Li Quan Yuan Guo Fu Ningshao Xia Jiahuai Han 2021Cellular & Molecular Immunology2021,18,4:1
4A novel numerical simulation method for predicting compressive properties of 3D multiaxial braided composites considering various defects显示文摘This paper reports a novel manufacturing process of preparing the Three-Dimensional Multiaxial Braided Composites(3DMBCs) and the effects of various defects generated in the manufacturing process on the compressive mechanical properties of 3DMBCs through the experimental and numerical methods. The five-step fabrication process of the 3D multiaxial braided preform was firstly introduced in detail. Then, the influences of various defects such as voids, waviness, and fiber breakage defects on the compressive properties of 3DMBCs were discussed. It is found that the fiber breakage defect and waviness defects were the two primary factors on the decrease of compressive properties of 3DMBCs. However, void defects in the resin and interface had little effect on the composites. When the fiber breakage defect content was 25% and the waviness was 7°, the composite compressive modulus decreased by 51%. 6A bifunctional MXene-modified scaffold for photothermal therapy and maxillofacial tissue regeneration显示文摘Oral squamous cell carcinoma is one of the most common malignant tumours in the oral and maxillofacial regions and is highly malignant and prone to recur despite the development of various effective treatments,including surgery and chemoradiotherapy.Actually,it is difficult to ensure the complete elimination of tumour cells,and maxillofacial bone defects caused by surgery are hard to heal by themselves.In addition,chemoradiotherapy can bring serious side-effects.Therefore,it is imperative to develop a postoperative therapy to kill residual squamous cancer cells and repair bone defects without any side-effects.Here,we prepared a 3D scaffold by a 3D printing technique and freeze-drying method,which contained collagen,silk and hydroxyapatite(CSH)and was functionalized with MXene nanosheets(M-CSH).The considerable photothermal effect with long-term stability can significantly kill squamous CAL-27 cancer cells in vitro and inhibit tumour growth in vivo,increasing the probability of the M-CSH scaffold being applied in the photothermal therapy of oral squamous cell carcinoma.Moreover,the cell proliferation-and osteogenic-related protein expression of mouse embryonic osteogenic precursors(MC3T3-E1)indicated excellent biocompatibility and osteogenic activity of M-CSH scaffolds.The good compression modulus(52.8362.25 kPa)and in vivo bone formation performance made it possible to be used as reconstructive materials for bone defects.This scaffold is likely promising in future tissue engineering,especially for the multifunctional treatment of maxillofacial tumours.Fengji Li Yanling Yan Yanan Wang Yaru Fan Huiru Zou Han Liu Rui Luo Ruixin Li Hao Liu 2021Regenerative Biomaterials2021,8,6:0
7Archaea, the tree of life, and cellular evolution in eukaryotes显示文摘The early history of life harbours many unresolved evolutionary questions, none more important than the genomic origin and cellular evolution of eukaryotes. An issue central to eukaryote origin concerns the position of eukaryotes in the tree of life and the relationship of the host lineage that acquired the mitochondrion some two billion years ago to lineages of modern-day archaea. Recent analyses indicate that the host lineage branches within the Archaea, prompting the search for novel archaeal lineages that can improve our understanding of the cellular evolution of eukaryotes. Here we give a brief review of the studies on Archaea, the tree of life and the cellular evolution of eukaryotes, which is followed by an overview of recent progress fueled by new genomic technologies and recent status of archaeal research in China. 8Characterization on Modification and Biocompatibility of PCL Scaffold Prepared with Near-field Direct-writing Melt Electrospinning显示文摘In this study,orthogonal experiments were designed to explore the optimal process parameters for preparing polycaprolactone(PCL)scaffolds by the near-field direct-writing melt electrospinning(NFDWMES)technology.Based on the optimal process parameters,the PCL scaffolds with different thicknesses,gaps and structures were manufactured and the corresponding hydrophilicities were characterized.The PCL scaffolds were modified by chitosan(CS)and hyaluronic acid(HA)to improve biocompatibility and hydrophilicity.Both Fourier transform infrared spectroscopy(FTIR)analysis and antibacterial experimental results show that the chitosan and hyaluronic acid adhere to the surface of PCL scaffolds,suggesting that the modification plays a positive role in biocompatibility and antibacterial effect.The PCL scaffolds were then employed as a carrier to culture cells.The morphology and distribution of the cells observed by a fluorescence microscope demonstrate that the modified PCL scaffolds have good biocompatibility,and the porous structure of the scaffolds is conducive to adhesion and deep growth of cells.CHEN Zhijun HAO Ming QIAN Xiaoming CHEN Wenyang ZENG Ming HUANG Juan LI Ruixin FAN Jintu LIU Yanbo 2021Chemical Research in Chinese Universities2021,37,3:0

10Intratumoral CD103+CD8+T cells predict response to neoadjuvant chemoimmunotherapy in advanced head and neck squamous cell carcinoma显示文摘Background Immune cell heterogenicity is known to determine the therapeutic response to cancer progression.Neoadjuvant chemoimmunotherapy(NACI)has shown clinical benefits in some patients with advanced head and neck squamous cell carcinoma(HNSCC),but the underlying mechanism behind this clinical response is unknown.The efficacy of NACI needs to be potentiated by identifying accurate biomarkers to predict clinical responses.Here,we attempted to identify molecules predicting NACI response in advanced HNSCC.Methods We performed combined single-cell RNA sequencing(scRNA-seq)and multiplex immunofluorescence(mIHC)staining with tumor samples derived from NACI-treated HNSCC patients to identify a new tumor-infiltrating cell(TIL)subtype,CD103+CD8+TILs,associated with clinical response,while both in vitro and in vivo assays were carried out to determine its antitumor efficiency.The regulatory mechanism of the CD103+CD8+TILs population was examined by performing cell-cell interaction analysis of the scRNA-seq data and spatial analysis of the mIHC images.Results We established intratumoral CD103+CD8+TILs density as a determinant of NACI efficacy in cancers.Our scRNA-seq results indicated that the population of CD103+CD8+TILs was dramatically increased in the responders of NACI-treated HNSCC patients,while mIHC analysis confirmed the correlation between intratumoral CD103+CD8+TILs density and NACI efficacy in HNSCC patients.Further receiver operating characteristic curve analysis defined this TIL subset as a potent marker to predict patient response to NACI.Functional assays showed that CD103+CD8+TILs were tumor-reactive T cells,while programmed cell death protein-1(PD-1)blockade enhanced CD103+CD8+TILs cytotoxicity against tumor growth in vivo.Mechanistically,targeting the triggering receptor expressed on myeloid cells 2-positive(TREM2+)macrophages might enhance the population of CD103+CD8+TILs and facilitate antitumor immunity during NACI treatment.Conclusions Our study highlights the impact of intratumoral CD103+CD8+TILs density on NACI efficacy in different cancers,while the efforts to elevate its population warrant further clinical investigation.Siqi Ren Tianjun Lan Fan Wu Suling Chen Xue Jiang Chuying Huo Zitian Li Shule Xie Donghui Wu Ruixin Wang Yanyan Li Lin Qiu Guoxin Huang Shurui Li Xiaojuan Wang Meifeng Cen Tingting Cai Zhaoyu Lin Jinsong Li Bowen Li 2023Cancer Communications2023,43,10:0

12Involvement of sulfur assimilation in the low β subunit content of soybean seed storage protein revealed by comparative transcriptome analysis显示文摘The β subunit of soybean [Glycine max (L.) Merr.] seed storage protein is of great significance in sulfur-containing amino acid balance and soybean processing properties. The objective of this study was to elucidate the relationship between the β subunit and sulfur-containing amino acid composition, and the potential regulatory mechanism. The β subunit was independently accumulated in comparison with other major subunits (α/α′, acidic, basic, and A3) during seed filling, and a low level of β subunit content (BSC) was formed during the accumulation process. In low-BSC mature seeds, crude protein, oil content, and fatty acid composition were not changed, but sulfur-containing amino acids (Cys + Met) in the low- BSC seeds increased significantly (by 31.5%), suggesting that an internal regulatory mechanism within seed might be responsible for the rebalance of seed protein composition and that sulfur assimilation might be deeply involved in β subunit accumulation. Transcriptomic analysis revealed that genes involved in anabolism of cysteine, methionine, and glutathione were up-regulated but those involved in the catabolism of these compounds were down-regulated, suggesting a relationship between the elevation of methionine and glutathione and low BSC.
