2Energy-level dependent H_2_O_2 production on metal-free,carbon-content tunable carbon nitride photocatalysts显示文摘Light-driven production of H_2O_2 from water and molecular oxygen could be a promising way for obtaining both solar fuels and fundamental chemicals. During that process, the H_2O_2 yield is strongly dependent on the reaction pathway associated with two-electron reduction of dioxygen by the photo-generated electrons. Herein, we synthesized a series of metal-free, carbon-content tunable carbon nitride photocatalysts(named C_3N_4-Carbon) by a facile hydrothermal reaction and subsequent thermal treatment at appropriate temperatures. The energy levels of the C_3N_4-Carbon catalysts vary with the carbon doping level, which is conveniently tuned by changing the initial glucose concentration during the hydrothermal reaction. The surface carbon species evolve with the carbon content and the nitrogen atoms in the structure of carbon nitride are partially substituted by foreign carbon atoms based on XPS measurements. The optimal catalyst leads to the highest H_2O_2 yield of 1271 μmol L^(-1) in an acidic aqueous solution(pH 3) after a reaction period of 4 h, twice higher than the pristine C_3N_4. In addition, the largest formation rate constant and the smallest decomposition rate constant of H_2O_2 are obtained on the optimal one according to the kinetics analyses. The decomposition tests of H_2O_2 indicate that the formation rate could be a dominant factor impacting the H_2O_2 yield. The conduction band position of the optimal catalyst is positively shifted to 0.06 V versus RHE, which is more favorable to the reduction of dioxygen to H_2O_2(O2/H_2O_2 at 0.69 V versus RHE). 3Effect of Gubi prescription on caveolin-1 expression and phosphoinositide 3 kinase/protein kinase B and Fas signal pathways in rats with knee osteoarthritis显示文摘OBJECTIVE: To investigate the effects of Gubi prescription on the expression of caveolin-1, and the phosphoinositide 3 kinase/protein kinase B(PI3 K/Akt) and Fas signal pathways in rats with knee osteoarthritis(KOA).METHODS: Forty KOA model rats were established using a modification of Hulth's method. Rats were divided into five groups by the random number method: model, positive drug(Vicolli group), and high-, medium-, and low-dose Gubi prescription groups(n = 8/group). In the sham surgery group(n = 8), only anterior and posterior cruciate ligaments of rats were exposed during surgery. A normal group(n = 8) consisted of rats with no treatment. Rats were intragastrically administered corresponding drugs once every day for eight consecutive weeks. Then, rat synovial membranes were extracted and histomorphological changes were recorded. m RNA expression was measured by q-PCR.Serum superoxide dismutase(SOD), malondialdehyde(MDA), nitric oxide(NO), and interleukin 1β(IL-1β) levels were measured. Western blotting determined the effects of Gubi prescription on protein expressions of caveolin-1, Bax, Bcl-2, Fas, and caspase-3 in chondrocytes from KOA rats. The knee cartilage of rats was excised and cultured under aseptic conditions. After coincubation of chondrocytes with Gubi prescription-containing serum, IL-1β, and si RNA, Western blotting was used to determine the protein expressions of caveolin-1, Bax, Bcl-2, Fas, and caspase-3.RESULTS: The morphological score of the articular synovium in the model group was significantly higher than in the normal group(P < 0.01). The morphological score in the high-and mediumdose Gubi prescription groups was lower than in the model group(P < 0.05). Chondrocytes from the decoction-containing serum group had a lower expression of Bax(P < 0.05), and higher expressions of Bcl-2(P < 0.05) and caspase-3(P < 0.05) compared with the model group. Chondrocytes in the decoction-containing serum group had higher expressions of Bax and Bcl-2(P < 0.01) and lower expressions of caveolin-1 and Fas(P < 0.05) compared with the model group. Compared with the model group, Bax and caspase-3 expressions were reduced in the chondrocytes of all three Gubi prescription groups(P < 0.05) whereas Bcl-2 expression was increased(P < 0.05). Compared with the model group, the expressions of caveolin-1 and Fas(P < 0.05) were reduced in groups that received high-and medium-doses of Gubi prescription. 6Effects of stochastic food deprivation on energy budget,body mass and activity in Swiss mice显示文摘When small animals are faced with an unpredictable food supply,they can adapt by altering different components of their energy budget such as energy intake,metabolic rate,rate of non-shivering thermogenesis(NST)or behaviour.The present study examined the effect of stochastic food deprivation(FD)on body mass,food intake,resting metabolic rate(RMR),NST and behaviour in male Swiss mice.During a period of 4 weeks' FD,animals were fed ad libitum for a randomly assigned 4 days each week,but were deprived of food for the other 3 days.The results showed that body mass significantly dropped on FD days compared to controls.Food intake of FD mice increased significantly on ad libitum days,ensuring cumulative food intake,final body mass,fat mass,RMR and NST did not differ significantly from controls.Moreover,gastrointestinal tract mass increased in FD mice,but digestibility decreased.In general,activity was higher on deprived days,and feeding behaviour was higher on ad libitum days suggesting that Swiss mice are able to compensate for stochastic FD primarily by increasing food intake on ad libitum days,and not by reducing energy expenditure related to RMR or NST.Zhijun ZHAO Jing CAO Ye TIAN Ruirui WANG Guiying WANG 2009Current Zoology2009,55,4:1
7Pharmacological inhibition of BAP1 recruits HERC2 to competitively dissociate BRCA1-BARD1, suppresses DNA repair and sensitizes CRC to radiotherapy显示文摘Radiotherapy is widely used in the management of advanced colorectal cancer(CRC).However,the clinical efficacy is limited by the safe irradiated dose.Sensitizing tumor cells to radiotherapy via interrupting DNA repair is a promising approach to conquering the limitation.The BRCA1-BARD1 complex has been demonstrated to play a critical role in homologous recombination(HR)DSB repair,and its functions may be affected by HERC2 or BAP1.Accumulated evidence illustrates that the ubiquitination-deubiquitination balance is involved in these processes;however,the precise mechanism for the cross-talk among these proteins in HR repair following radiation hasn’t been defined.Through activity-based profiling,we identified PT33 as an active entity for HR repair suppression.Subsequently,we revealed that BAP1 serves as a novel molecular target of PT33 via a CRISPR-based deubiquitinase screen.Mechanistically,pharmacological covalent inhibition of BAP1 with PT33 recruits HERC2 to compete with BARD1 for BRCA1 interaction,interrupting HR repair.Consequently,PT33 treatment can substantially enhance the sensitivity of CRC cells to radiotherapy in vitro and in vivo.Overall,these findings provide a mechanistic basis for PT33-induced HR suppression and may guide an effective strategy to improve therapeutic gain.Xin Yue Tingyu Liu Xuecen Wang Weijian Wu Gesi Wen Yang Yi Jiaxin Wu Ziyang Wang Weixiang Zhan Ruirui Wu Yuan Meng Zhirui Cao Liyuan Le Wenyan Qiu Xiaoyue Zhang Zhenyu Li Yong Chen Guohui Wan Xianzhang Bu Zhenwei Peng Ran-yi Liu 2023Acta Pharmaceutica Sinica B2023,13,8:1
8Effect of caffeic acid derivatives on polychlorinated biphenyls induced hepatotoxicity in male mice显示文摘Chronic exposure to coplanar polychlorinated biphenyls(PCBs),a potent inducer of toxic reactive oxygen species(ROS),in the environment and food can cause liver diseases.It remains unknown whether caffeic acid derivatives(CADs) exerted protective effect on PCB-induced hepatotoxicity.We sought to evaluate the activities of 3CADs on PCB169-induced oxidative stress and DNA damage in the liver.Male ICR mice were administered with1 μmol/mL PCB169 at 5 mL/kg body weight for 2 weeks.The mice were given CADs by gastric gavage for 3weeks.We found that PCB169 decreased the growth rate and reduced the levels of superoxide dismutase(SOD),glutathione(GSH) and GSH peroxidase(GPx).It increased the liver weight,malondialdehyde(MDA)and 8-hydroxy-2'-deoxyguanosine(8-OHdG) levels and CYPlAl activity in the liver tissues and plasma of mice(P<0.05).Pretreatment of mice with CADs restored the above parameters to normal levels.There was a synergistic protective effect between CADs in preventing MDA and 8-OHdG formation and inducing CYPlAl and phase II metabolism enzyme(SOD,GPx) activities(P<0.05).In conclusion,PCB169 induced hepatotoxicity and pretreatment with CADs had synergistic protective effects on liver damage.Ruirui Li Shuyuan Cao Jinfeng Dai Li Wang Lei Li Yubang Wang Wenqin Yin Yuting Ye 2014The Journal of Biomedical Research2014,28,5:1
9Dynamic recrystallization-induced temperature insensitivity of yield stress in single-crystal Al1.2CrFeCoNi micropillars显示文摘High-entropy alloys, a new class of metallic materials, exhibit excellent mechanical properties at high temperatures. In spite of the worldwide interest, the underlying mechanisms for temperature dependence of mechanical properties of these alloys remain poorly understood. Here, we systemically investigate the mechanical behaviors and properties of Al_(1.2)CrFeCoNi(comprising a body-centered cubic phase) and Al_(0.3)CrFeCoNi(comprising a face-centered cubic phase) single-crystal micropillars with three orientations([100], [110], and [111]) at temperatures varying from 300 to 675 K by using in situ compression of micropillars inside a scanning electron microscope. The results show that the yield stresses of Al_(1.2)CrFeCoNi micropillars are insensitive to temperature changes, and their flow stresses and work hardening rates increase slightly with increasing temperature from 300 to550 K, which differs from the typical temperature dependence of yield/flow stresses in metals and alloys. In contrast,Al_(0.3)CrFeCoNi micropillars exhibit typical thermal softening. Furthermore, it is found that the Al_(1.2)CrFeCoNi micropillars exhibit a transition from homogenous deformation to localized deformation at a critical temperature, while the Al_(0.3)CrFeCoNi micropillars always maintain a well-distributed and fine slip deformation. Detailed transmission electron microscopy analyses reveal that dynamic recrystallization(involving dislocation tangles, and formation of dislocation cell structures and sub-grains)plays a key role in the observed temperature insensitivity of the yield stress and increasing flow stress(and work hardening rate)with increasing temperature in the Al_(1.2)CrFeCoNi micropillars, and that thermally activated dislocation slip leads to thermal softening of the Al_(0.3)CrFeCoNi micropillars. The differences in deformation modes and temperature dependence of the mechanical properties between Al_(1.2)CrFeCoNi and Al_(0.3)CrFeCoNi essentially originate from the differences in dislocation activities and slip systems since the two alloys adopt different phases. 16Nitrogen addition reduced carbon mineralization of aggregates in forest soils but enhanced in paddy soils in South China显示文摘Background:Despite the crucial role of nitrogen(N)availability in carbon(C)cycling in terrestrial ecosystems,soil organic C(SOC)mineralization in different sizes of soil aggregates under various land use types and their responses to N addition is not well understood.To investigate the responses of soil C mineralization in different sized aggregates and land use types to N addition,an incubation experiment was conducted with three aggregate-size classes(2000,250,and 53μm)and two land use types(a Chinese fir plantation and a paddy land).Results:Cumulative C mineralization of the<53-μm fractions was the highest and that of microaggregates was the lowest in both forest and paddy soils,indicating that soil aggregates enhanced soil C stability and reduced the loss of soil C.Cumulative C mineralization in all sizes of aggregates treated with N addition decreased in forest soils,but that in microaggregates and the<53-μm fraction increased in paddy soils treated with 100μgNg−1.Moreover,the effect sizes of N addition on C mineralization of forest soils were below zero,but those of paddy soils were above zero.These data indicated that N addition decreased SOC mineralization of forest soils but increased that of paddy soils.Conclusions:Soil aggregates play an important role in soil C sequestration,and decrease soil C loss through the increase of soil C stability,regardless of land use types.N addition has different effects on soil C mineralization in different land use types.These results highlight the importance of soil aggregates and land use types in the effects of N deposition on the global terrestrial ecosystem C cycle.Ruirui Cao Longchi Chen Xincun Hou Xiaotao Lü Haimei Li 2021Ecological Processes2021,10,1:0
