1Efficacy and safety of low-dose interleukin-2 in combination with methotrexate in patients with active rheumatoid arthritis:a randomized,double-blind,placebo-controlled phase 2 trial显示文摘Rheumatoid arthritis(RA)is an aggressive autoimmune arthritis,and current therapies remain unsatisfactory due to low remission rate and substantially adverse effects.Low-dose interleukin-2(Ld-IL2)is potentially a therapeutic approach to further improve the disease.This randomized,double-blind,placebo-controlled trial was undertaken to evaluate the efficacy and safety of Ld-IL2 in patients with active RA.Patients were randomly assigned(1:1)to receive Ld-IL2,defined as a dose of 1 million IU,or placebo in a 12-week trial with a 12-week follow-up.Three cycles of Ld-IL2 or placebo were administered subcutaneously every other day for 2 weeks(a total of 7 doses),followed by a 2-week break.All patients received a stable dose of methotrexate(MTX).The primary outcomes were the proportion of patients achieving the ACR20,DAS28-ESR<2.6,and the change from baseline in CDAI or SDAI at week 24.Secondary endpoints included other clinical responses and safety.The primary outcomes were achieved in the perprotocol population.The improvements from baseline in CDAI and SDAI were significantly greater across time points for the LdIL2+MTX group(n=17)than for the placebo+MTX group(n=23)(P=0.018 and P=0.015,respectively).More patients achieved ACR20 response in the Ld-IL2+MTX group than those in the placebo+MTX group at week 12(70.6%vs 43.5%)and at week 24(76.5%vs 56.5%)(P=0.014).In addition,low Treg and high IL-21 were associated with good responses to Ld-IL2.Ld-IL-2 treatment was well-tolerated in this study.These results suggested that Ld-IL2 was effective and safe in RA.ClinicalTrials.gov number:NCT 02467504.Xiaoying Zhang Miao Miao Ruijun Zhang Xu Liu Xiaozhen Zhao Miao Shao Tian Liu Yuebo Jin Jiali Chen Huixin Liu Xia Zhang Yun Li Yunshan Zhou Yue Yang Ru Li Haihong Yao Yanying Liu Chun Li Yuhui Li Limin Ren Yin Su Xiaolin Sun Jing He Zhanguo Li 2022Signal Transduction and Targeted Therapy2022,7,4:7
2Timing Advanced Estimation Algorithm of Low Complexity Based on DFT Spectrum Analysis for Satellite System显示文摘In satellite mobile communication system, relative movement of the satellite and the terminal will cause a large Doppler offset. Timing advanced estimation with Zadoff-Chu sequence is sensitive to the frequency offset. When the frequency offset is larger than one times subcarrier spacing, the value of peak cannot be detected at the receiving end. To suppress the larger Doppler frequency shift, this paper proposes a novel timing advanced estimation scheme(TAE-MCD) for satellite communication system. In this algorithm, t r a n s m i t t e d s i g n a l i s d i v i d e d i n t o Z C sequence and its conjugate sequence. Using multiplication and DFT operation to find the estimated peak at the receiving end, and make subtraction with the obtained sequences at last. The scheme can not only inhibit the adverse effects of large Doppler frequency shift in timing estimation effectively, but also reduce the computational complexity at the receiving end and improve the work efficiency of the hardware. 3Three-dimensional biofabrication of an aragoniteenriched self-hardening bone graft substitute and assessment of its osteogenicity in vitro and in vivo显示文摘A self-hardening three-dimensional(3D)-porous composite bone graft consisting of 65 wt%hydroxyapatite(HA)and 35 wt%aragonite was fabricated using a 3D-Bioplotter®.New tetracalcium phosphate and dicalcium phosphate anhydrous/aragonite/gelatine paste formulae were developed to overcome the phase separation of the liquid and solid components.The mechanical properties,porosity,height and width stability of the end products were optimised through a systematic analysis of the fabrication processing parameters including printing pressure,printing speed and distance between strands.The resulting 3D-printed bone graft was confirmed to be a mixture of HA and aragonite by X-ray diffraction,Fourier transform infrared spectroscopy and energy dispersive X-ray spectroscopy.The compression strength of HA/aragonite was between 0.56 and 2.49 MPa.Cytotoxicity was assessed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay in vitro.The osteogenicity of HA/aragonite was evaluated in vitro by alkaline phosphatase assay using human umbilical cord matrix mesenchymal stem cells,and in vivo by juxtapositional implantation between the tibia and the anterior tibialis muscle in rats.The results showed that the scaffold was not toxic and supported osteogenic differentiation in vitro.HA/aragonite stimulated new bone formation that bridged host bone and intramuscular implants in vivo.We conclude that HA/aragonite is a biodegradable and conductive bone formation biomaterial that stimulates bone regeneration.Since this material is formed near 37°C,it will have great potential for incorporating bioactive molecules to suit personalised application;however,further study of its biodegradation and osteogenic capacity is warranted.The study was approved by the Animal Ethical Committee at Tongji Medical School,Huazhong University of Science and Technology(IACUC No.738)on October 1,2017.Yunsong Shi Ruijun He Xiangyu Deng Zengwu Shao Davide Deganello Chunze Yan Zhidao Xia 2020Biomaterials Translational2020,1,1:2
4Facile fabrication of amphoteric semi-interpenetrating network membranes for vanadium flow battery applications显示文摘For improvement of vanadium redox flow battery(VRB) performance, novel amphoteric semiinterpenetrating membranes(ASIPN) were prepared using poly(ether ether ketone)(PEEK) and polysulfone(PSf), the former bearing sulfonic groups and the latter imidazolium. These two groups form ionic crosslinks between PEEK and PSf; meanwhile, covalent cross links were built between PSf chains with addition of N-(3-aminopropyl)-imidazole. The amphoteric nature of the membrane allows facile proton and anion transport; the IPN structure and the presence of imidazolium cation effectively suppress vanadium ion crossover through the membrane. Therefore, the ASIPN based VRBs show higher Coulombic efficiency and energy efficiency than that assembled with pristine SPEEK and Nafion 212 membranes. 5Random access preamble design based on time pre-compensation for LTE-satellite system显示文摘Because of the integration of long term evolution(LTE) technology and mobile satellite communication systems, uplink access technology for LTE-based geo-stationary earth orbit(GEO) satellite has become a popular research topic for satellite system. In order to solve the problem of unreasonable design for physical random access channel(PRACH) signal structure and reduce the effect of time uncertainty, this paper proposes a novel random access preamble based on time pre-compensation(TPC) for LTE-Satellite(LTE-S) system. In this scheme, by applying the method of non-linear least squares, the user terminal(UT) can use the receiving power to estimate the communication round trip delay(RTD) and based on the transmission delay of the beam center and the satellite, RTD can be compensated before transmission. Therefore, the preamble length and duration can be reduced without related to the maximum of RTD. In order to verify the performance of the scheme, the MATLAB is used to build a test system.
