| 1 | 一个新的水稻逆境响应基因OsMsr1的表达与克隆显示文摘为深入了解植物的逆境反应机理和发现新的水稻耐逆基因,采用Affymetrix水稻表达芯片(含51279个转录本)分析了超级稻两优培九母本培矮64S不同生长发育时期、不同组织器官全基因组在低温、干旱、高温逆境胁迫下的表达水平,筛选出众多表达水平显著升高与降低的基因。OsMsr1(Oryza sativa L.multiple stresses responsive gene 1,Gen Bank登录号为EU284112)是其中一个受高温、低温与干旱诱导、在各生长发育时期与组织器官均显著上调的基因,用实时定量PCR方法对其表达水平进行了进一步的分析,所得结果与基因芯片结果基本吻合,因此,此基因为一多逆境响应基因。用RT-PCR方法扩增获得了包含其完整ORF(open reading frame)的cDNA克隆。根据其ORF序列进行预测,此基因编码一个包含89个氨基酸残基的小分子蛋白,分子量约为10kD,pI约为5。搜索有关数据库,在水稻、玉米、小麦与拟南芥中找到有高相似性的基因,但功能未知,也未发现相同与类似的已知功能的基因保守结构域。对其可能的启动子序列分析,发现5个与逆境反应有关的顺式作用元件。因此,我们认为该基因为一新的水稻耐逆候选基因,进一步的研究正在进行。 | 徐孟亮 陈荣军 ROCHA Pedro 李落叶 王曼玲 徐国云 夏新界 | 2008 | 作物学报2008,34,10: | 17 |

| 9 | Elevated nitric oxide and 3',5' cyclic guanosine monophosphate levels in patients with alcoholic cirrhosis显示文摘AIM: To evaluate whether serum levels of nitric oxide (NO ·) and plasma levels of cyclic guanosine monophosphate (cGMP) and total glutathione (GSH) are altered in patients with alcoholic cirrhosis and to examine their correlation with the severity of liver disease.METHODS: Twenty-six patients with alcoholic liver cirrhosis were studied. Serum levels of NO · and plasma levels of cGMP and GSH were measured in 7 patients with compensated alcoholic cirrhosis (Child-Pugh A) and 19 patients with advanced cirrhosis (Child-Pugh B and C). The model for end-stage liver disease (MELD) score was evaluated. Sixteen healthy volunteers served as controls. Liver enzymes and creatinine levels were also tested.RESULTS: NO · and cGMP levels were higher in patients with Child-Pugh B and C cirrhosis than in Child-Pugh A cirrhosis or controls (NO ·: 21.70 ± 8.07 vs 11.70 ± 2.74; 21.70 ± 8.07 vs 7.26 ± 2.47 μmol/L, respectively; P < 0.001) and (cGMP: 20.12 ± 6.62 vs 10.14 ± 2.78; 20.12 ± 6.62 vs 4.95 ± 1.21 pmol/L, respectively; P < 0.001). Total glutathione levels were lower in patients with Child-Pugh B and C cirrhosis than in patients with Child-Pugh A cirrhosis or controls (16.04 ± 6.06 vs 23.01 ± 4.38 or 16.04 ± 6.06 vs 66.57 ± 26.23 μmol/L, respectively; P < 0.001). There was a significant correlation between NO · and cGMP levels in all patients with alcoholic cirrhosis. A significant negative correlation between reduced glutathione/glutathione disulfide and the MELD score was found in all cirrhotic patients.CONCLUSION: Our results suggest a role for oxidative stress in alcoholic liver cirrhosis, which is more significant in decompensated patients with higher levels of NO ·and cGMP and lower GSH levels than in compensated and control patients. | 13 | Usefulness of serum C-reactive protein and calprotectin for the early detection of colorectal anastomotic leakage:A prospective observational study显示文摘BACKGROUND Colorectal anastomotic leakage(CAL)is one of the most dreaded complications after colorectal surgery,with an incidence that can be as high as 27%.This event is associated with increased morbidity and mortality;therefore,its early diagnosis is crucial to reduce clinical consequences and costs.Some biomarkers have been suggested as laboratory tools for the diagnosis of CAL.AIM To assess the usefulness of plasma C-reactive protein(CRP)and calprotectin(CLP)as early predictors of CAL.METHODS A prospective monocentric observational study was conducted including patients who underwent colorectal resection with anastomosis,from March 2017 to August 2019.Patients were divided into three groups:G1–no complications;G2–complications not related to CAL;and G3–CAL.Five biomarkers were measured and analyzed in the first 5 postoperative days(PODs),namely white blood cell(WBC)count,eosinophil cell count(ECC),CRP,CLP,and procalcitonin(PCT).Clinical criteria,such as abdominal pain and clinical condition,were also assessed.The correlation between biomarkers and CAL was evaluated.Receiver operating characteristic(ROC)curve analysis was used to compare the accuracy of these biomarkers as predictors of CAL,and the area under the ROC curve(AUROC),specificity,sensitivity,positive predictive value,and negative predictive value(NPV)during this period were estimated.RESULTS In total,25 of 396 patients developed CAL(6.3%),and the mean time for this diagnosis was 9.0±6.8 d.Some operative characteristics,such as surgical approach,blood loss,intraoperative complications,and duration of the procedure,were notably related to the development of CAL.The length of hospital stay was markedly higher in the group that developed CAL compared with the group with complications other than CAL and the group with no complications(median of 21 d vs 13 d and 7 d respectively;P<0.001).For abdominal pain,the best predictive performance was on POD4 and POD5,with the largest AUROC of 0.84 on POD4.Worsening of the clinical condition was associated with the diagnosis of CAL,presenting a higher predictive effect on POD5,with an AUROC of 0.9.WBC and ECC showed better predictive effects on POD5(AUROC=0.62 and 0.7,respectively).Those markers also presented a high NPV(94%-98%).PCT had the best predictive effect on POD5(AUROC=0.61),although it presented low accuracy.However,this biomarker revealed a high NPV on POD3,POD4,and POD5(96%,95%,and 96%,respectively).The mean CRP value on POD5 was significantly higher in the group that developed CAL compared with the group without complications(195.5±139.9 mg/L vs 59.5±43.4 mg/L;P<0.00001).On POD5,CRP had a NPV of 98%.The mean CLP value on POD3 was significantly higher in G3 compared with G1(5.26±3.58μg/mL vs 11.52±6.81μg/mL;P<0.00005).On POD3,the combination of CLP and CRP values showed a high diagnostic accuracy(AUROC=0.82),providing a 5.2 d reduction in the time to CAL diagnosis.CONCLUSION CRP and CLP are moderate predictors of CAL.However,the combination of these biomarkers presents an increased diagnostic accuracy,potentially decreasing the time to CAL diagnosis. | Nuno J G Rama Marlene C C Lages Maria Pedro S Guarino Óscar Lourenço Patrícia C Motta Lima Diana Parente Cândida S G Silva Ricardo Castro Ana Bento Anabela Rocha Fernando Castro-Pocas João Pimente | 2022 | World Journal of Gastroenterology2022,28,24: | 1 |
