3Structures of EV71 RNA-dependent RNA polymerase in complex with substrate and analogue provide a drug target against the hand-foot-and-mouth disease pandemic in China显示文摘Enterovirus 71(EV71),one of the major causative agents for hand-foot-and-mouth disease(HFMD),has caused more than 100 deaths among Chinese children since March 2008.The EV71 genome encodes an RNAdependent RNA polymerase(RdRp),denoted 3D^(pol),which is central for viral genome replication and is a key target for the discovery of specific antiviral therapeutics.Here we report the crystal structures of EV71 RdRp(3D^(pol))and in complex with substrate guanosine-5'-triphosphate and analog 5-bromouridine-5'-triphosphate best to 2.4Åresolution.The structure of EV71 RdRp(3D^(pol))has a wider open thumb domain compared with the most closely related crystal structure of poliovirus RdRp.And the EV71 RdRp(3D^(pol))complex with GTP or Br-UTP bounded shows two distinct movements of the polymerase by substrate or analogue binding.The model of the complex with the template:primer derived by superimposition with foot-and-mouth disease virus(FMDV)3D/RNA complex reveals the likely recognition and binding of template:primer RNA by the polymerase.These results together provide a molecular basis for EV71 RNA replication and reveal a potential target for anti-EV71 drug discovery.Yang Wu Zhiyong Lou Yi Miao Yue Yu Hui Dong Wei Peng Mark Bartlam Xuemei Li Zihe Rao 2010Protein & Cell2010,1,5:30

5On the timing of the East Asian summer monsoon maximum during the Holocene Does the speleothem oxygen isotope record reflect monsoon rainfall variability?显示文摘The evolution of the East Asian summer monsoon(EASM) during the Holocene has long been of significant interest.Knowledge of past EASM variability not only increases our understanding of monsoon dynamics on a long timescale,but it also provides an environmental and climatic background for research into Chinese cultural development.However,the timing of the EASM maximum remains controversial.The popular concept of an 'early Holocene maximum' is mainly based on speleothemδ^(18)O(δ^(18)O_c) records from caves in southern China;however,the interpretation of δ^(18)O_C as a reliable proxy for EASM intensity is being increasingly challenged.The present paper is a critical review of the climatic significance of the δ^(18)O_C record from China.Firstly,we suggest that precipitation in northern China is an appropriate index of EASM intensity,the variation of which clearly indicates a mid-Holocene monsoon maximum.Secondly,an interregional comparison demonstrates that the precipitation record in northern China is quite different from that in southern China on a range of timescales,and is inconsistent with the spatial similarity exhibited by speleothem oxygen isotope records.Furthermore,both modeling and observational data show that the δ^(18)O_C records from southern China indeed reflect changes in precipitation δ^(18)O(δ^(18)O_P) rather than precipitation amount,and therefore that their use as an EASM proxy is inappropriate.Finally,we address several significant monsoon-related issues-including the driving mechanism of the EASM on an orbital timescale,the climatic significance of speleothem oxygen isotopes,and the relationship between atmospheric circulation and precipitation in monsoonal regions.CHEN JianHui RAO ZhiGuo LIU JianBao HUANG Wei FENG Song DONG GuangHui HU Yu XU QingHai CHEN FaHu 2016Science China(Information Sciences)2016,59,12:18

7Quantification of Panax notoginseng saponins metabolites in rat plasma with in vivo gut microbiota-mediated biotransformation by HPLC-MS/MS显示文摘Panax notoginseng saponins(PNS) are the major components of Panax notoginseng, with multiple pharmacological activities but poor oral bioavailability. PNS could be metabolized by gut microbiota in vitro, while the exact role of gut microbiota of PNS metabolism in vivo remains poorly understood. In this study, pseudo germ-free rat models were constructed by using broad-spectrum antibiotics to validate the gut microbiota-mediated transformation of PNS in vivo. Moreover, a high performance liquid chromatography-electrospray ionization tandem mass spectrometry(HPLC-ESI-MS/MS) was developed for quantitative analysis of four metabolites of PNS, including ginsenoside F1(GF1), ginsenoside Rh2(GRh2), ginsenoside compound K(GCK) and protopanaxatriol(PPT). The results showed that the four metabolites could be detected in the control rat plasma, while they could not be determined in pseudo germ-free rat plasma. The results implied that PNS could not be biotransformed effectively when gut microbiota was disrupted. 8Extracellular vesicles from human urine-derived stem cells prevent osteoporosis by transferring CTHRC1 and OPG显示文摘Osteoporosis is a debilitating bone disease affecting millions of people. Here, we used human urine-derived stem cells(USCs),which were noninvasively harvested from unlimited and easily available urine, as a 'factory' to obtain extracellular vesicles(USCEVs) and demonstrated that the systemic injection of USC-EVs effectively alleviates bone loss and maintains bone strength in osteoporotic mice by enhancing osteoblastic bone formation and suppressing osteoclastic bone resorption. More importantly, the anti-osteoporotic properties of USC-EVs are not notably disrupted by the age, gender, or health condition(with or without osteoporosis) of the USC donor. Mechanistic studies determined that collagen triple-helix repeat containing 1(CTHRC1) and osteoprotegerin(OPG) proteins are enriched in USC-EVs and required for USC-EV-induced pro-osteogenic and anti-osteoclastic effects. 10Panax notoginseng saponins prevent colitis-associated colorectal cancer development: the role of gut microbiota显示文摘Gut microbiota dysbiosis is a risk factor for colorectal cancer(CRC) in inflammatory bowel disease(IBD).In this study, the effects of Panax notoginseng saponins(PNS) on colitis-associated CRC progression were evaluated on an azoxymethane(AOM)/dextran sulfate sodium(DSS) mouse model.In vivo, PNS significantly relieved AOM/DSS-induced colon tumorigenesis and development by reducing the disease activity index(DAI) scores and colon tumor load.The 16S rRNA data of fecal samples showed that the microbiome community was obviously destructed, while PNS could recover the richness and diversity of gut microbiota.Especially, PNS could increase the abundance of Akkermansia spp.which was significantly decreased in model group and negatively correlated with the progression of CRC.Moreover, ginsenoside compound K(GC-K) was evaluated on the effects of human CRC cells,which was the main bio-transformed metabolite of PNS by gut microbiota.Our data showed that PNS played important role in the prevention of the progression of CRC, due to their regulation on the microbiome balance and microbial bio-converted product with antiCRC activity.CHEN Ling CHEN Man-Yun SHAO Li ZHANG Wei RAO Tai ZHOU Hong-Hao HUANG Wei-Hua 2020Chinese Journal of Natural Medicines2020,18,7:12
11Spatial transmission and meteorological determinants of tuberculosis incidence in Qinghai Province,China:a spatial clustering panel analysis显示文摘Background:Tuberculosis(TB)is the notifiable infectious disease with the second highest incidence in the Qinghai province,a province with poor primary health care infrastructure.Understanding the spatial distribution of TB and related environmental factors is necessary for developing effective strategies to control and further eliminate TB.Methods:Our TB incidence data and meteorological data were extracted from the China Information System of Disease Control and Prevention and statistical yearbooks,respectively.We calculated the global and local Moran’s I by using spatial autocorrelation analysis to detect the spatial clustering of TB incidence each year.A spatial panel data model was applied to examine the associations of meteorological factors with TB incidence after adjustment of spatial individual effects and spatial autocorrelation.Results:The Local Moran’s I method detected 11 counties with a significantly high-high spatial clustering(average annual incidence:294/100000)and 17 counties with a significantly low-low spatial clustering(average annual incidence:68/100000)of TB annual incidence within the examined five-year period;the global Moran’s I values ranged from 0.40 to 0.58(all P-values<0.05).The TB incidence was positively associated with the temperature,precipitation,and wind speed(all P-values<0.05),which were confirmed by the spatial panel data model.Each 10°C,2 cm,and 1 m/s increase in temperature,precipitation,and wind speed associated with 9%and 3%decrements and a 7%increment in the TB incidence,respectively.Conclusions:High TB incidence areas were mainly concentrated in south-western Qinghai,while low TB incidence areas clustered in eastern and north-western Qinghai.Areas with low temperature and precipitation and with strong wind speeds tended to have higher TB incidences.Hua-Xiang Rao Xi Zhang Lei Zhao Juan Yu Wen Ren Xue-Lei Zhang Yong-Cheng Ma Yan Shi Bin-Zhong Ma Xiang Wang Zhen Wei Hua-Fang Wang Li-Xia Qiu 2016Infectious Diseases of Poverty2016,5,1:10
12Impact of hepatitis C virus genotype 3 on liver disease progression in a Chinese national cohort显示文摘Background:Hepatitis C virus(HCV)genotype 3,particularly subtype 3b,is increasing in prevalence and distribution in China.This study evaluated the prevalence,regional distribution,clinical characteristics,host factors,treatment outcomes,and disease progression of patients with HCV genotype 3 in China.Methods:A 5-year follow-up was preceded by a cross-sectional study.Treatment choices were at the discretion of treating physicians.Estimated infection time to overall-disease-progression(defined by≥1 of:newly diagnosed cirrhosis;cirrhosis at baseline,Child-Turcotte-Pugh score increased 2 points or more;progression from compensated cirrhosis to decompensated cirrhosis;hepatocellular carcinoma;liver transplantation;or death)was calculated using the Kaplan-Meier method.Cox regression analyses were conducted to evaluate the risk factors for disease progression.Results:The cross-sectional study enrolled 997 patients,including 91 with HCV genotype 3 infection.Among them,subtype 3b(57.1%)was more dominant than subtype 3a(38.5%).Five hundred and twelve patients were included into the follow-up phase.Among patients analyzed for estimated infection time to overall-disease-progression,52/304(17.1%)patients with HCV genotype 1 and 4/41(9.8%)with HCV genotype 3(4/26 with genotype 3b,0/13 with genotype 3a,and 0/2 with undefined subtype of genotype 3)experienced overall-disease-progression.Patients with HCV genotype 3 were younger than those with genotype 1(mean age:39.5±8.7 vs.46.9±13.6 years)and demonstrated more rapid disease progression(mean estimated infection time to overall-disease-progression 27.1 vs.35.6 years).Conclusions:HCV genotype 3,specifically subtype 3b,is associated with more rapid progression of liver disease.Further analysis to compare HCV subtype 3a and 3b is needed in high prevalence regions.Nan Wu Hui-Ying Rao Wei-Bo Yang Zhi-Liang Gao Rui-Feng Yang Ran Fei Ying-Hui Gao Qian Jin Lai Wei 2020Chinese Medical Journal2020,,3:10

14Preparation of cross-linked magnetic chitosan with quaternary ammonium and its application for Cr(Ⅵ) and P(Ⅴ) removal显示文摘Pollutants that exist in anionic species are issues of concern in water treatment.Compared to cationic pollutants,the removal of anionic pollutants by adsorption is more difficult because most adsorbents carry predominantly negative charges in neutral and alkaline environments.In this study,a cross-linked chitosan derivative with quaternary ammonium and magnetic properties(QM-chitosan)was prepared and employed to remove chromium(VI)and phosphorus(V)(Cr(VI)and P(V))from aqueous environments.The QM-chitosan was characterized by Fourier transform infrared spectrometry(FT-IR),thermogravimetric analysis(TGA),energy dispersive X-ray(SEM-EDX)and zeta potential.Batch experiments show that QM-chitosan can effectively remove Cr(VI)and P(V),and the main mechanism was believed to be electrostatic interaction.A pseudosecond-order model was fitted to describe the kinetic processes of Cr(VI)and P(V)removal.The adsorption isotherms of both Cr(VI)and P(V)on the QM-chitosan were well fitted by the Langmuir isotherm equation.The saturated adsorption capacity of P(V)(2.783 mmol/g)was found to be higher than that of Cr(VI)(2.323 mmol/g),resulting from the size of the H2PO-4ions being smaller than that of the HCr O-4ions.However,the theoretical calculation and experimental results showed that QM-chitosan had a stronger affinity for Cr(VI)than P(V).The adsorption–desorption of the QM-chitosan was evaluated,and high regeneration rates were demonstrated.Wei Yao Pinhua Rao Irene M.C.Lo Wenqi Zhang Wenrui Zheng 2014Journal of Environmental Sciences2014,26,12:9
15Outcomes in children with biliary atresia following liver transplantation显示文摘BACKGROUND: Congenital biliary atresia is a rare condition characterized by idiopathic dysgenesis of the bile ducts. If untreated, congenital biliary atresia leads to liver cirrhosis, liver failure and premature death. The present study aimed to evaluate the outcomes of orthotopic liver transplantation in children with biliary atresia. METHOD: We retrospectively analyzed 45 patients with biliary atresia who had undergone orthotopic liver transplantation from September 2006 to August 2012. RESULTS: The median age of the patients was 11.0 months (5-102). Of the 45 patients, 41 were younger than 3 years old. Their median weight was 9.0 kg (4.5-29.0), 34 of the 45 patients were less than 10 kg. Thirty-one patients had undergone Kasai portoenterostomy prior to orthotopic liver transplantation. We performed 30 living donor liver transplants and 15 split liver transplants. Six patients died during a follow-up. The median follow-up time of surviving patients was 11.4 months (1.4-73.7). The overall 1-, 2- and 3-year survival rates were 88.9%, 84.4% and 84.4%, respectively. 16Multifunctional polymeric micelle-based chemo-immunotherapy with immune checkpoint blockade for efficient treatment of orthotopic and metastatic breast cancer显示文摘Immunotherapy has become a highly promising paradigm for cancer treatment. Herein, a chemo-immunotherapy was developed by encapsulating chemotherapeutic drug doxorubicin(DOX) and Toll-like receptor 7 agonist imiquimod(IMQ) in low molecular weight heparin(LMWH)-D-α-tocopheryl succinate(TOS) micelles(LT). In this process, LMWH and TOS were conjugated by ester bond and they were not only served as the hydrophilic and hydrophobic segments of the carrier, but also exhibited strong anti-metastasis effect. The direct killing of tumor cells mediated by DOX-loaded micelles(LT-DOX)generated tumor-associated antigens, initiating tumor-specific immune responses in combination with IMQ-loaded micelles(LT-IMQ). Furthermore, the blockade of immune checkpoint with programmed cell death ligand 1(PD-L1) antibody further elevated the immune responses by up-regulating the maturation of DCs as well as the ratios of CD8+ CTLs/Treg and CD4+ Teff/Treg. 18The Chinese Hα Solar Explorer(CHASE) mission: An overview显示文摘The Chinese Hα Solar Explorer(CHASE), dubbed “Xihe”—Goddess of the Sun, was launched on October 14, 2021 as the first solar space mission of China National Space Administration(CNSA). The CHASE mission is designed to test a newly developed satellite platform and to acquire the spectroscopic observations in the Hα waveband. The Hα Imaging Spectrograph(HIS)is the scientific payload of the CHASE satellite. It consists of two observational modes: raster scanning mode and continuum imaging mode. The raster scanning mode obtains full-Sun or region-of-interest spectral images from 6559.7 to 6565.9 ? and from 6567.8 to 6570.6 ? with 0.024 ? pixel spectral resolution and 1 min temporal resolution. The continuum imaging mode obtains photospheric images in continuum around 6689 ? with the full width at half maximum of 13.4 ?. The CHASE mission will advance our understanding of the dynamics of solar activity in the photosphere and chromosphere. 19Liver retransplantation for ischemic-type biliary lesions after orthotopic liver transplantation:a clinical report of 66 cases显示文摘BACKGROUND:Ischemic-type biliary lesions(ITBLs)play an extremely important role in influencing the long-term survival and quality of life of recipients after orthotopic liver transplantation(OLT).Some patients can be cured by interventional therapies,however others lose their grafts at last and receive liver retransplantation(re-OLT).The aim of this study was to analyze the data of 66 patients who had received re-OLT at our center because of ITBL and to discuss the treatment of ITBL after OLT. METHODS:We retrospectively analyzed 66 re-OLT cases due to ITBL from September 2001 to February 2007 at our center.The Kaplan-Meier method and the Cox-Mantel test were used to identify factors associated with mortality for univariate analysis and multivariate analysis,respectively. RESULTS:Fifty-five of 66 ITBL cases underwent interven- tional therapies before re-OLT.The actuarial survival at 1 month and 1 year for these patients was 83%and 74%, respectively.Prognostic factors for mortality in univariate analysis were model of end-stage liver disease score (MELD)>16.5(χ2=5.856,P=0.016),cold ischemia time>8 hours(χ2=6.539,P=0.011),infections(χ2=5.550,P=0.018) and complications(χ2=12.168,P=0.002)after re-OLT.In the multivariate analysis(Cox regression),the risk factors independently associated with mortality were MELD score>16.5(RR:3.140;P=0.035),cold ischemia time>8.2 hours(RR:0.192;P=0.016)and complications(RR:3.896, P=0.003).CONCLUSIONS:The incidence of ITBL in China is higher than in other countries.Based on our experience,MELD score,cold ischemia time and complications after re-OLT are risk factors independently associated with mortality in retransplanted ITBL patients.Zhu, Zhi-Jun Rao, Wei Sun, Ji-San Cai, Jin-Zhen Deng, Yong-Lin Zheng, Hong Zhang, Ya-Min Jiang, Wen-Tao Zhang, Jian-Jun Gao, Wei Shen, Zhong-Yang 2008Hepatobiliary & Pancreatic Diseases International2008,7,5:7
20Developing potent PROTACs tools for selective degradation of HDAC6 protein显示文摘Dear Editor, Histone deacetylases (HDACs) are a family of enzymes that remove acetyl groups on histone and non-histone proteins, thereby playing a vital role in the modulation of gene expression and protein activity. Eighteen HDACs have been identified in human and subdivided into four classes including I, II (Ila, lib), III and IV (Seto et al., 2014). Among them, HDAC6 is a unique lib HD AC with dominant cytoplasmic localization and two functional catalytic domains. Besides the functions for deacetylation of histone, and modulation of a-tubulin, HSP90 and cortactin, HDAC6 also participates in protein trafficking and degradation, cell shape and migration (Valenzuela-Fernandez et al., 2008). The deregulation of HDAC6 is related to various diseases, such as neurodegenerative diseases, cancer and pathological autoimmune response (Batchu et al., 2016). Hence, it is especially important for directly controlling cellular HDAC6 protein levels to achieve therapeutic purposes. The traditional approaches of red u ci ng cellular protein levels mainly rely on genetic modifications, such as RNA interference, transcription activator-like effector nucleases, recombination-based gene knockout and clustered regularly interspaced short palindromic repeats (CRISPR-Cas9)(Boettcher et al., 2015). However, these approaches have failed to a certain degree to achieve acute and reversible changes of gene function. Furthermore, the complications of potential genetic compensation and/or spontaneous mutations arising in geneknockout models may lead to misinterpretations (Davisson etal., 2012;El-Brolosy etal., 2017).
