| 1 | 8-bromo-7-methoxychrysin inhibits properties of liver cancer stem cells via downregulation of β-catenin显示文摘AIM:To evaluate whether 8-bromo-7-methoxychrysin(BrMC),a synthetic analogue of chrysin,inhibits the properties of cancer stem cells derived from the human liver cancer MHCC97 cell line and to determine the potential mechanisms.METHODS:CD133+cells were sorted from the MHCC97 cell line by magnetic activated cell sorting,and amplified in stem cell-conditioned medium to obtain the enriched CD133+sphere forming cells(SFCs).The stem cell properties of CD133+SFCs were validated by the tumorsphere formation assay in vitro and the xenograft nude mouse model in vivo,and termed liver cancer stem cells(LCSCs).The effects of BrMC on LCSCs in vitro were evaluated by MTT assay,tumorsphere formation assay and transwell chamber assay.The effects of BrMC on LCSCs in vivo were determined using a primary and secondary xenograft model in Balb/c-nu mice.Expressions of the stem cell markers,epithelialmesenchymal transition(EMT)markers andβ-catenin protein were analyzed by western blotting or immunohistochemical analysis.RESULTS:CD133+SFCs exhibited stem-like cell properties of tumorsphere formation and tumorigenesis capacity in contrast to the parental MHCC97 cells.We found that BrMC preferentially inhibited proliferation and self-renewal of LCSCs(P<0.05).Furthermore,BrMC significantly suppressed EMT and invasion of LCSCs.Moreover,BrMC could efficaciously eliminate LCSCs in vivo.Interestingly,we showed that BrMC decreased the expression ofβ-catenin in LCSCs.Silencing ofβ-catenin by small interfering RNA could synergize the inhibition of self-renewal of LCSCs induced by BrMC,while Wnt3a treatment antagonized the inhibitory effects of BrMC.CONCLUSION:BrMC can inhibit the functions and characteristics of LCSCs derived from the liver cancer MHCC97 cell line through downregulation ofβ-catenin expression. | Mei-Fang Quan Li-Hong Xiao Zhi-Hong Liu Hui Guo Kai-Qun Ren Fei Liu Jian-Guo Cao Xi-Yun Deng | 2013 | World Journal of Gastroenterology2013,19,43: | 22 |

| 3 | Experimental investigations on the heat transfer characteristics of micro heat pipe array applied to flat plate solar collector显示文摘This paper introduces a novel flat plate solar collector(FPC) using micro heat pipe array(MHPA) as a key element.To analyze the thermal transfer behavior of flat plate solar collector with micro heat pipe array(MHPA-FPC),an indoor experiment for thermal transfer characteristic of MHPA applied to FPC was conducted by using an electrical heating film to simulate the solar radiation.Different cooling water flow rates,cooling water temperatures,slopes,and contact thermal resistances between the condenser of MHPA and the heat exchanger were tested at different heating powers.The experimental results indicate that MHPA-FPC exhibits the enhanced heat transfer capability with increased cooling water flow rate and temperature.Total thermal resistance has a maximum decline of approximately 10% when the flow rate increases from 180 to 360 L h 1 and 38% when the cooling water temperature increases from 20℃ to 40℃.When the inclination angle of MHPA-FPC exceeds 30°,the slope change has a negligible effect on the heat transfer performance of MHPA-FPC.In addition,contact thermal resistance significantly affects the heat transfer capability of MHPA-FPC.The total thermal resistances lowers to nearly half of the original level when contact material between the condenser of MHPA and the heat exchanger changes from conductive silicone to conductive grease.These results could provide useful information for the optimal design and operation of MHPA-FPC. | DENG YueChao QUAN ZhenHua ZHAO YaoHua WANG LinCheng | 2013 | Science China(Technological Sciences)2013,56,5: | 17 |
| 4 | Impact of dissolved organic matter on the photolysis of the ionizable antibiotic norfloxacin显示文摘Norfloxacin(NOR), an ionizable antibiotic frequently used in the aquaculture industry, has aroused public concern due to its persistence, bacterial resistance, and environmental ubiquity.Therefore, we investigated the photolysis of different species of NOR and the impact of a ubiquitous component of natural water — dissolved organic matter(DOM), which has a special photochemical activity and normally acts as a sensitizer or inhibiter in the photolysis of diverse organics; furthermore, scavenging experiments combined with electron paramagnetic resonance(EPR) were performed to evaluate the transformation of NOR in water. The results demonstated that NOR underwent direct photolysis and self-sensitized photolysis via hydroxyl radical(U OH) and singlet oxygen(1O2) based on the scavenging experiments. In addition, DOM was found to influence the photolysis of different NOR species, and its impact was related to the concentration of DOM and type of NOR species. Photolysis of cationic NOR was photosensitized by DOM at low concentration, while zwitterionic and anionic NOR were photoinhibited by DOM, where quenching of U OH predominated according to EPR experiments, accompanied by possible participation of excited triplet-state NOR and1O2. Photo-intermediate identification of different NOR species in solutions with/without DOM indicated that NOR underwent different photodegradation pathways including dechlorination, cleavage of the piperazine side chain and photooxidation, and DOM had little impact on the distribution but influenced the concentration evolution of photolysis intermediates. | 5 | Development of a Drilling and Coring Test-bed for Lunar Subsurface Exploration and Preliminary Experiments显示文摘Drill sampling has been widely employed as an effective way to acquire deep samples in extraterrestrial exploration. A novel sampling method, namely, flexible-tube coring, was adopted for the Chang'e mission to acquire drilling cores without damaging stratification information. Since the extraterrestrial environment is uncertain and different from the terrestrial environment, automated drill sampling missions are at risk of failure. The principles of drilling and coring for the lunar subsurface should be fully tested and verified on earth before launch. This paper proposes a test-bed for conducting the aforementioned experiments on earth. The test-bed comprises a rotary-percussive drilling mechanism, penetrating mechanism, drilling medium container, and signal acquisition and control system. For granular soil, coring experiments indicate that the sampling method has a high coring rate greater than 80%. For hard rock, drilling experiments indicate that the percussive frequency greatly affects the drilling efficiency. A multi-layered simulant composed of granular soil and hard rock is built to test the adaptability of drilling and coring. To tackle complex drilling media, an intelligent drilling strategy based on online recognition is proposed to improve the adaptability of the sampling drill. | 6 | Drilling Power Consumption and Soil Conveying Volume Performances of Lunar Sampling Auger显示文摘The sampling auger used in lunar sampling and return mission is to transmit power and convey soil, and its performance is the key factor of the whole mission. However, there is currently a lack of the optimization research on soil conveying volume and power consumption models in auger structure design. To provide the drilled object, the simulation lunar soil, whose physical and mechanical property is the same as the real soil, is made by reducing soil void ratio. The models are formulated to analyze the influence of auger structure parameters on power consumption and soil conveying volume. To obtain the optimized structure parameters of auger, the multi-objective optimization functions of the maximum soil conveying volume and minimum power consumption are developed. To verify the correctness of the models, the performances of different augers drilling simulation soil are tested. The test results demonstrate that the power consumption of optimized auger is the lowest both in theory and test, and the experimental results of soil conveying volume are in agreement with theoretical analysis. Consequently, a new method for designing a lunar sampling auger is proposed which includes the models of soil conveying volume and transportation power consumption, the optimization of structure parameters and the comparison tests. | 9 | Drilling load modeling and validation based on the filling rate of auger flute in planetary sampling显示文摘Some type of penetration into a subsurface is required in planetary sampling. Drilling and coring, due to its efficient penetrating and cuttings removal characteristics, has been widely applied in previous sampling missions. Given the complicated mechanical properties of a planetary regolith, suitable drilling parameters should be matched with different drilling formations properly.Otherwise, drilling faults caused by overloads could easily happen. Hence, it is necessary to establish a drilling load model, which is able to reveal the relationships among drilling loads, an auger's structural parameters, soil's mechanical properties, and relevant drilling parameters. A concept for the filling rate of auger flute(FRAF) is proposed to describe drilling conditions. If the FRAF index under one group of drilling parameters is less than 1, this means that the auger flute currently removes cuttings smoothly. Otherwise, the auger will be choked with compressed cuttings. In drilling operations, the drilling loads on the auger mainly come from the conveyance action, while the drilling loads on the drill bit primarily come from the cutting action. Experiments in one typical lunar regolith simulant indicate that the estimated drilling loads based on the FRAF coincide with the test results quite well. | 11 | AFF1 and AFF4 differentially regulate the osteogenic differentiation of human MSCs显示文摘AFF1 and AFF4 belong to the AFF(AF4/FMR2) family of proteins, which function as scaffolding proteins linking two different transcription elongation factors, positive elongation factor b(P-TEFb) and ELL1/2, in super elongation complexes(SECs). Both AFF1 and AFF4 regulate gene transcription through elongation and chromatin remodeling. However, their function in the osteogenic differentiation of mesenchymal stem cells(MSCs) is unknown. In this study, we show that small interfering RNA(si RNA)-mediated depletion of AFF1 in human MSCs leads to increased alkaline phosphatase(ALP) activity, enhanced mineralization and upregulated expression of osteogenic-related genes. On the contrary, depletion of AFF4 significantly inhibits the osteogenic potential of MSCs. In addition, we confirm that overexpression of AFF1 and AFF4 differentially affects osteogenic differentiation in vitro and MSC-mediated bone formation in vivo.Mechanistically, we find that AFF1 regulates the expression of DKK1 via binding to its promoter region.Depletion of DKK1 in HA-AFF1-overexpressing MSCs abrogates the impairment of osteogenic differentiation. Moreover, we detect that AFF4 is enriched in the promoter region of ID1. AFF4 knockdown blunts the BRE luciferase activity, SP7 expression and ALP activity induced by BMP2 treatment. Compound K-treated liver cancer cells were harvested for analysis of transcriptional levels of both enoyl-Co A hydratase and 3-hydroxyacyl Co A dehydrogenase(EHHADH)-AS1 and ENTPD5.RESULTS: The results revealed that 11 Encyclopedia of DNA Elements annotated lnc RNAs were differentially expressed in the process of HBV-associated HCG. Among these lnc RNAs, 95 potential ce RNA networks with highly positively correlated expression profiles between the interacting lnc RNAs and m RNAs(Pearson correlation coefficient ≥ 0.7) were constructed. Of note, two HBV-associated ce RNA networks, EHHADH-AS1-hsa-mi R-4459-ectonucleoside triphosphate diphosphohydrolase 5 and LINC01018-hsa-mi RNA-574-5p-glucose-6-phosphatase catalytic subunit, with Pearson correlation coefficient ≥ 0.9, may play a critical role in hepatocellular carcinoma development, which was supported by experimental evidence. | 17 | Multi-state autonomous drilling for lunar exploration显示文摘Due to the lack of information of subsurface lunar regolith stratification which varies along depth, the drilling device may encounter lunar soil and lunar rock randomly in the drilling process. To meet the load safety requirements of unmanned sampling mission under limited orbital resources, the control strategy of autonomous drilling should adapt to the indeterminable lunar environments. Based on the analysis of two types of typical drilling media(i.e., lunar soil and lunar rock), this paper proposes a multi-state control strategy for autonomous lunar drilling. To represent the working circumstances in the lunar subsurface and reduce the complexity of the control algorithm, lunar drilling process was categorized into three drilling states: the interface detection, initiation of drilling parameters for recognition and drilling medium recognition. Support vector machine(SVM) and continuous wavelet transform were employed for the online recognition of drilling media and interface, respectively. Finite state machine was utilized to control the transition among different drilling states. To verify the effectiveness of the multi-state control strategy, drilling experiments were implemented with multi-layered drilling media constructed by lunar soil simulant and lunar rock simulant. | 20 | Impact of human leukocyte antigen mismatching on outcomes of liver transplantation:A meta-analysis显示文摘AIM:To assess the effect of human leukocyte antigen(HLA) mismatching on liver graft outcome and acute rejection from a meta-analysis of available cohort studies.METHODS:Articles in PubMed/MEDLINE,EMBASE and the Cochrane database from January 1970 to June 2009,including non-English literature identified in these databases,were searched.Only studies comparing HLA or sub-phenotype matching with mismatching were extracted.The percentage of graft survival was extracted by 'Engauge Digitizer' from survival curves if the raw data were not displayed.A meta-analysis was performed when at least 3 studies provided data.RESULTS:Sixteen studies met the inclusion criteria.A lower number of HLA mismatches(0-2 vs 3-6) did reduce the incidence of acute rejection(relative risk:0.77,P = 0.03).The degree of HLA mismatching(0-2 vs 3-6) had no significant effect on 1-year [hazard ratio(HR):1.04,P = 0.68] and 5-year(HR:1.09,P = 0.38) graft survival.In sub-phenotype analysis,the degree of HLA-A,B and DR mismatching(0 vs 1-2) had no significant effect on 1-year and 5-year graft survival,either.The HRs and P-values were 0.95,0.71(HLA-A,1-year);1.06,0.60(HLA-A,5-year);0.77,0.16(HLA-B,1-year);1.07,0.56(HLA-DR,1-year);1.18,0.23(HLADR,5-year),respectively.CONCLUSION:The results of this systematic review imply that good HLA compatibility can reduce the incidence of acute rejection in spite of having no influence on graft outcomes.To obtain a short recovery time and minimize rejection post transplantation,HLA matching studies should be considered before the operation. | Xiang Lan Ming-Man Zhang Cong-Lun Pu Chun-Bao Guo Quan Kang Ying-Chun Li Xiao-Ke Dai Yu-Hua Deng Qiang Xiong Zhi-Mei Ren | 2010 | World Journal of Gastroenterology2010,16,27: | 2 |
