Chinese deserts and sand fields in Last Glacial Maximum and Holocene Optimum显示文摘The Last Glacial Maximum (LGM, c. 26-16 ka) and the Holocene Optimum (HO, c. 9-5 ka) were characterized by cold-dry and warm-wet climates respectively in the recently geological Earth. How Chinese deserts and sand fields responded to these distinctive climatic changes is still not clear, however. To reconstruct environments of the deserts and sand fields during the LGM and HO is helpful to understand the forcing mechanisms of environment change in this arid region, and to test paleoclimatic modeling results. Through our long-term field and laboratory investigations, 400 optically stimulated luminescence (OSL) ages and more than 100 depositional records in the Chinese deserts and sand fields were obtained; on the basis of these data, we reconstruct spatial distributions of the deserts and sand fields during the LGM and HO. Our results show that the sand fields of Mu Us, Hunshandake, Horqin and Hulun Buir in northern and northeastern China had expanded 25%, 37%, 38% and 270%, respectively, during the LGM; the sand fields of Gonghe in the northeastern Qinghai-Tibetan Plateau had expanded 20%, and the deserts of Badain Jaran, Tengger in central northern China had expanded 39% and 29% separately during the LGM; the deserts of Taklimakan, Gurbantünggüt and Kumtag in northwestern China had expanded 10%-20% respectively, compared to their modern areas. On the other hand, all of the sand fields were nearly completely covered by vegetation during the HO; the deserts in northwestern and central northern China were reduced by around 5%-20% in area during this time. Lakes in this arid region were probably expanded during the HO but this conclusion needs more investigation. Compared with the geological distributions of deserts and sand fields, human activity has clearly changed (expanded) the area of active sand dunes at the present time. Our observations show that environmental conditions of Chinese deserts and sand fields are controlled by regional climate together with human activity. | LU HuaYu YI ShuangWen XU ZhiWei ZHOU YaLi ZENG Lin ZHU FangYing FENG Han DONG LiNa ZHUO HaiXin YU KaiFeng MASON Joseph WANG XiaoYong CHEN YingYong LU Qi WU Bo DONG ZhiBao QU JianJun WANG XunMing GUO ZhengTang | 2013 | Chinese Science Bulletin2013,58,23: | 40
Microbial treatment in chronic constipation显示文摘Chronic functional constipation is a kind of common intestinal disease that occurs in children, adults and elderly people. This disease not only causes great influence to physiological function, but also results in varying degrees of psychological barriers. At present, constipation treatments continue to rely on traditional methods such as purgative therapy and surgery. However, these approaches can disrupt intestinal function. Recent research between intestinal diseases and gut microbiota has gradually revealed a connection between constipation and intestinal flora disturbance, providing a theoretical basis for microbial treatment in chronic constipation. Microbial treatment mainly includes probiotic preparations such as probiotics, prebiotics, synbiotics and fecal microbiota transplantation(FMT). Due to its safety, convenience and curative effect, probiotic preparations have been widely accepted, especially gradually developed FMT with higher curative effects. Microbial treatment improves clinical symptoms, promotes the recovery of intestinal flora, and has no complications during the treatment process. Compared with traditional treatments, microbial treatment in chronic constipation has advantages, and is worthy of further promotion from clinical research to clinical application. | Linsheng Huang Qi Zhu Xiao Qu Huanlong Qin | 2018 | Science China(Life Sciences)2018,61,7: | 40
Re-Os isotope dating of molybdenites in the Huang-shaping Pb-Zn-W-Mo polymetallic deposit, Hunan Province, South China and its geological significance显示文摘The large-scale Huangshaping Pb-Zn-W-Mo polymetallic deposit is located in the central Nanling min- eralization zone, South China. Six molybdenite samples from the Huangshaping deposit were selected for Re-Os isotope measurement in order to define the mineralization age of the deposit. It yields a Re-Os isochron age of 154.8±1.9 Ma (2σ ), which is in accordance with the Re-Os model ages of 150.9― 156.9 Ma. This age is about 7 Ma younger than their host granite porphyry, which was dated as 161.6±1.1 Ma by zircon U-Pb method using LA-ICPMS. All these ages demonstrate that the Huang- shaping granite and related Pb-Zn-W-Mo deposit occurred in the middle Yanshanian period, when many other granitoid and related ore deposits emplaced and formed, e.g. the Qitianling granite and Furong tin deposit, the Qianlishan granite and giant Shizhuyuan W-Sn-Mo-Bi deposit and Jinchuantang Sn-Bi deposit in the nearby area. They constitute the main part of the magmatic-metallogenic belt of southern Hunan, and represent the large-scale metallogeny in middle Yanshanian in the area. The lower rhenium content in molybdenite of Huangshaping deposit suggests that the ore-forming material was mainly of crust origin. | YAO JunMing HUA RenMin QU WenJun QI HuaWen LIN JinFu DU AnDao | 2007 | Science China Earth Sciences2007,50,4: | 29
The distribution rule and seepage effect of the fractures in the ultra-low permeability sandstone reservoir in east Gansu Province,Ordos Basin显示文摘To study the impact of the fractures on development in the ultra-low permeability sandstone reservoir of the Yangchang Formation of the Upper Triassic in the Ordos Basin, data on outcrops, cores, slices, well logging and experiments are utilized to analyze the cause of the formation of the fractures, their distribution rules and the control factors and discuss the seepage flow effect of the fractures. In the studied area developed chiefly high-angle tectonic fractures and horizontal bedding fractures,inter-granular fractures and grain boundary fractures as well. Grain boundary fractures and intragranular fractures serve as vital channels linking intragranular pores and intergranular solution pores in the reservoir matrix, thus providing a good connectivity between the pores in the ultra-low perme-ability sandstone reservoir. The formation of fractures and their distribution are influenced by such external and internal factors as the palaeo-tectonic stress field, the reservoir lithological character, the thickness of the rock layer and the anisotropy of a layer. The present-day stress field influences the preservative state of fractures and their seepage flow effect. Under the tec-tonic effect of both the Yanshan and Himalayan periods,in this region four sets of fractures are distributed, respectively assuming the NE-SW,NW-SE,nearly E-W and nearly S-N orientations, but, due to the effect of the rock anisotropy of the rock formation,in some part of it two groups of nearly orthogonal fractures are chiefly distributed. Under the effect of the present-day stress field, the fractures that assume the NE-SW direction have a good connectivity, big apertures,a high permeability and a minimum starting pressure, all of which are main advantages of the seepage fractures in this region. With the development of oilfields, the permeability of the fractures of dif-ferent directions will have a dynamic change. | ZENG LianBo1,GAO ChunYu2,QI JiaFu1,WANG YongKang2,LI Liang2 & QU XueFeng2 1 State Key Laboratory of Petroleum Resource and Prospecting,China University of Petroleum,Beijing 102249,China 2 Changqing Oilfield Branch,PetroChina,Xi’an 163517,China | 2008 | Science China Earth Sciences2008,51,S2: | 22
Age-related rhesus macaque models of COVID-19显示文摘Background:Since December 2019,an outbreak of the Corona Virus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus(SARS-CoV-2)in Wuhan,China, has become a public health emergency of international concern. The high fatality of aged cases caused by SARS-CoV-2 was a need to explore the possible age-related phenomena with non-human primate models. Methods:Three 3-5 years old and two 15 years old rhesus macaques were intratracheally infected with SARS-CoV-2,and then analyzed by clinical signs, viral replication, chest X-ray,histopathological changes and immune response. Results:Viral replication of nasopharyngeal swabs, anal swabs and lung in old monkeys was more active than that in young monkeys for 14 days after SARS-CoV-2 challenge.Monkeys developed typical interstitial pneumonia characterized by thickened alveolar septum accompanied with inflammation and edema, notably, old monkeys exhibited diffuse severe interstitial pneumonia.Viral antigens were detected mainly in alveolar epithelial cells and macrophages.Conclusion:SARS-CoV-2 caused more severe interstitial pneumonia in old monkeys than that in young monkeys.Rhesus macaque models infected with SARS-CoV-2 provided insight into the pathogenic mechanism and facilitated the development of vaccines and therapeutics against SARS-CoV-2 infection. | Pin Yu Feifei Qi Yanfeng Xu Fengdi Li Peipei Liu Jiayi Liu Linlin Bao Wei Deng Hong Gao Zhiguang Xiang Chong Xiao Qi Lv Shuran Gong Jiangning Liu Zhiqi Song Yajin Qu Jing Xue Qiang Wei Mingya Liu Guanpeng Wang Shunyi Wang Haisheng Yu Xing Liu Baoying Huang Wenling Wang Li Zhao Huijuan Wang Fei Ye Weimin Zhou Wei Zhen Jun Han Guizhen Wu Qi Jin Jianwei Wang Wenjie Tan Chuan Qin | 2020 | Animal Models and Experimental Medicine2020,3,1: | 22
Acupuncture at Waiguan(SJ5) and sham points influences activation of functional brain areas of ischemic stroke patients: a functional magnetic resonance imaging study显示文摘Most studies addressing the specificity of meridians and acupuncture points have focused mainly on the different neural effects of acupuncture at different points in healthy individuals. This study examined the effects of acupuncture on brain function in a pathological context. Sixteen patients with ischemic stroke were randomly assigned to true point group(true acupuncture at right Waiguan(SJ5)) and sham point group(sham acupuncture). Results of functional magnetic resonance imaging revealed activation in right parietal lobe(Brodmann areas 7 and 19), the right temporal lobe(Brodmann area 39), the right limbic lobe(Brodmann area 23) and bilateral occipital lobes(Brodmann area 18). Furthermore, inhibition of bilateral frontal lobes(Brodmann area 4, 6, and 45), right parietal lobe(Brodmann areas 1 and 5) and left temporal lobe(Brodmann area 21) were observed in the true point group. Activation in the precuneus of right parietal lobe(Brodmann area 7) and inhibition of the left superior frontal gyrus(Brodmann area 10) was observed in the sham group. Compared with sham acupuncture, acupuncture at Waiguan in stroke patients inhibited Brodmann area 5 on the healthy side. Results indicated that the altered specificity of sensation-associated cortex(Brodmann area 5) is possibly associated with a central mechanism of acupuncture at Waiguan for stroke patients. | Ji Qi Junqi Chen Yong Huang Xinsheng Lai Chunzhi Tang Junjun Yang Hua Chen Shanshan Qu | 2014 | Neural Regeneration Research2014,9,3: | 19

Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. Given that the H5N8 viruses are likely to continue to spread in wild birds, vaccination of poultry is highly recommended in high-risk countries to prevent H5N8 avian influenza. | Pengfei Cui Xianying Zeng Xuyong Li Yanbing Li Jianzhong Shi Conghui Zhao Zhiyuan Qu Yanwen Wang Jing Guo Wenli Gu Qi Ma Yuancheng Zhang Weipeng Lin Minghui Li Jingman Tian Dongxue Wang Xin Xing Yanjing Liu Shuxin Pan Yaping Zhang Hongmei Bao Liling Liu Guobin Tian Chengjun Li Guohua Deng Hualan Chen | 2022 | Science China(Life Sciences)2022,65,4: | 12
Single-cell transcriptomic atlas of primate cardiopulmonary aging显示文摘Aging is a major risk factor for many diseases, especially in highly prevalent cardiopulmonary comorbidities and infectious diseases including Coronavirus Disease 2019(COVID-19).Resolving cellular and molecular mechanisms associated with aging in higher mammals is therefore urgently needed. Here,we created young and old non-human primate single-nucleus/cell transcriptomic atlases of lung, heart and artery, the top tissues targeted by SARS-CoV-2.Analysis of cell type-specific aging-associated transcriptional changes revealed increased systemic inflammation and compromised virus defense as a hallmark of cardiopulmonary aging. With age, expression of the SARS-CoV-2 receptor angiotensin-converting enzyme 2(ACE2)was increased in the pulmonary alveolar epithelial barrier, cardiomyocytes, and vascular endothelial cells. We found that interleukin 7(IL7)accumulated in aged cardiopulmonary tissues and induced ACE2 expression in human vascular endothelial cells in an NF-κB-dependent manner.Furthermore, treatment with vitamin C blocked IL7-induced ACE2 expression.Altogether, our findings depict the first transcriptomic atlas of the aged primate cardiopulmonary system and provide vital insights into age-linked susceptibility to SARS-CoV-2,suggesting that geroprotective strategies may reduce COVID-19 severity in the elderly. | Shuai Ma Shuhui Sun Jiaming Li Yanling Fan Jing Qu Liang Sun Si Wang Yiyuan Zhang Shanshan Yang Zunpeng Liu Zeming Wu Sheng Zhang Qiaoran Wang Aihua Zheng Shuguang Duo Yang Yu Juan Carlos Izpisua Belmonte Piu Chan Qi Zhou Moshi Song Weiqi Zhang Guang-Hui Liu | 2021 | Cell Research2021,31,4: | 10

Management of Cesarean Scar Pregnancy: A Case Series显示文摘Objective To survey effective treatment strategies for cesarean scar pregnancy(CSP). Methods The clinical data of 78 patients diagnosed with CSP from January 2010 to December 2013 were reviewed. Results Among these patients, 17 patients were first treated at our hospital; of them, 2 were misdiagnosed. The other 61 patients were referred from other hospitals; of them, 21 were initially misdiagnosed. There were 9 patients who were treated with laparotomy, 50 patients with curettage after uterine artery embolization(UAE) with or without local methotrexate(MTX) infusion, 10 patients with dilatation and curettage, 6 patients with transvaginal sonographic guided local intragestational MTX injection, and 3 patients with systemic MTX injection. All patients finally recovered. Patients with excessive vaginal hemorrhage underwent either emergency UAE treatment or laparotomy. These two treatments had similar success rates(81.82% vs. 100%, χ2 =0.289, P>0.05). Conclusions The accurate diagnosis of CSP is important. Curettage after UAE with or without local MTX infusion is a safe and effective method. | Min-hui Guo Mei-fen Wang Man-man Liu Feng Qi Fan Qu Jian-hong Zhou | 2015 | Chinese Medical Sciences Journal2015,30,4: | 9
Chemical characteristics of size-resolved aerosols in winter in Beijing显示文摘Size-resolved aerosols were continuously collected by a Nano Sampler for 13 days at an urban site in Beijing during winter 2012 to measure the chemical composition of ambient aerosol particles. Data collected by the Nano Sampler and an ACSM(Aerodyne Aerosol Chemical Speciation Monitor) were compared. Between the data sets, similar trends and strong correlations were observed, demonstrating the validity of the Nano Sampler. PM10 and PM2.5concentrations during the measurement were 150.5 ± 96.0 μg/m3(mean ± standard variation)and 106.9 ± 71.6 μg/m3,respectively. The PM2.5/PM10 ratio was 0.70 ± 0.10,indicating that PM2.5dominated PM10. The aerosol size distributions showed that three size bins of 0.5–1,1–2.5 and 2.5–10 μm contributed 21.8%,23.3% and 26.0% to the total mass concentration(TMC),respectively. OM(organic matter) and SIA(secondary ionic aerosol, mainly SO42-,NO3-and NH4+) were major components of PM2.5. Secondary compounds(SIA and secondary organic carbon) accounted for half of TMC(about 49.8%) in PM2.5,and suggested that secondary aerosols significantly contributed to the serious particulate matter pollution observed in winter. Coal burning, biomass combustion, vehicle emissions and SIA were found to be the main sources of PM2.5. Mass concentrations of water-soluble ions and undetected materials,as well as their fractions in TMC,strikingly increased with deteriorating particle pollution conditions, while OM and EC(elemental carbon) exhibited different variations, with mass concentrations slightly increasing but fractions in TMC decreasing. | Kang Sun Yu Qu Qiong Wu Tingting Han Jianwei Gu Jingjing Zhao Yele Sun Qi Jiang Ziqi Gao Min Hu Yuanhang Zhang Keding Lu Stephan Nordmann Yafang Cheng Li Hou Hui Ge Masami Furuuchi Mitsuhiko Hata Xingang Liu | 2014 | Journal of Environmental Sciences2014,26,8: | 9
Effect of proteolytic starter culture isolated from Chinese Dong fermented pork(Nanx Wudl)on microbiological, biochemical and organoleptic attributes in dry fermented sausages显示文摘The effect of a proteolytic starter culture isolated from Nanx Wudl,on microbiological, biochemical and organoleptic attributes of dry fermented sausages was investigated during processing.Based on preliminary screening, the combination of Staphylococcus xylosus SX16 and Lactobacillus plantarum CMRC6,showing excellent proteolytic activity in vitro, was selected as the multi-strain starter(starter LS).For comparison, the single-strain starter culture of L.plantarum CMRC6(starter LB)and non-inoculated control were also tested.During fermentation, lactic acid bacteria and staphylococci dominated the microbiota and suppressed the Enterobacteriaceae growth in LS-inoculated sausages. The addition of LS starter accelerated acidification and proteolysis during ripening, improving the contents of total free amino acids and several essential free amino acids(Phe,Ile and Leu).Volatile compounds analysis revealed that LS-fermented sausage showed the highest abundance of 3-methyl-1-butanol,probably due to the inoculated S.xylosus.The inoculation of LS starter improved the sensory properties of sausages, especially the flavor attribute.Therefore,S.xylosus SX16 and L.plantarum CMRC6 are promising candidates for inclusion as multi-strain starters in the manufacture of gourmet fermented dry sausage. | Xi Chen Ruifang Mi Biao Qi Suyue Xiong Jiapeng Li Chao Qu Xiaoling Qiao Wenhua Chen Shouwei Wang | 2021 | Food Science and Human Wellness2021,10,1: | 9
Surges of hospital-based rhinovirus infection during the 2020 coronavirus disease-19(COVID-19)pandemic in Beijing,China显示文摘Background A series of public health preventive measures has been widely implemented in Beijing to control the corona-virus disease-19(COVID-19)pandemic since January 2020.An evaluation of the effects of these preventive measures on the spread of other respiratory viruses is necessary. Methods Respiratory specimens collected from children with acute respiratory infections were tested by NxTAGTN,respiratory pathogen panel assays during January 2017 and December 2020.Specimens characterized as rhinoviruses(RVs)were sequenced to identify the RV species and types. Then, the epidemiology results of respiratory pathogens in 2020 were compared with those from 2017 to 2019 using SPSS statistics 22.0.Results The positive rates of adenovirus(ADV),influenza virus(flu),RVs,and respiratory syncytial virus(RSV)dropped abruptly by 86.31%,94.679c,94.59%,and 92.17%,respectively,from February to May 2020,compared with the average level in the same period during 2017-2019.Positive rates of RVs then steeply increased from June 2020(13.77%),to an apex(37.25%)in August 2020,significantly higher than the average rates(22.51%)in August 2017-2019(7^=0.005).The increase, especially in group>3 years, was accompanied by the reopening of schools and kindergartens after the 23rd and 24th week of 2020 in Beijing. Conclusions Whereas the abrupt drop in viral pathogen positive rates from February to May 2020 revealed the remarkable effects of the COVID-19 preventive measures, the sharp increase in positive rates of RVs from the 23rd week of 2020 might be explained by the reopening of schools and kindergartens in Beijing. | Ruo-Xi Zhang Dong-Mei Chen Yuan Qian Yu Sun Ru-Nan Zhu Fang Wang Ya-Xin Ding Qi Guo Yu-Tong Zhou Dong Qu Ling Cao Chun-Mei Zhu Lin-Qing Zhao | 2021 | World Journal of Pediatrics2021,17,6: | 9
Stabilization of heterochromatin by CLOCK promotes stem cell rejuvenation and cartilage regeneration显示文摘Accumulating evidence indicates an association between the circadian clock and the aging process. However,it remains elusive whether the deregulation of circadian clock proteins underlies stem cell aging and whether they are targetable for the alleviation of aging-associated syndromes. Here,we identified a transcription factor-independent role of CLOCK,a core component of the molecular circadian clock machinery,in counteracting human mesenchymal stem cell(hMSC)decay.CLOCK expression was decreased during hMSC aging. In addition,CLOCK deficiency accelerated hMSC senescence, whereas the overexpression of CLOCK,even as a transcriptionally inactive form, rejuvenated physiologically and pathologically aged hMSCs.Mechanistic studies revealed that CLOCK formed complexes with nuclear lamina proteins and KAP1,thus maintaining heterochromatin architecture and stabilizing repetitive genomic sequences. Finally, gene therapy with lentiviral vectors encoding CLOCK promoted cartilage regeneration and attenuated age-related articular degeneration in mice. These findings demonstrate a noncanonical role of CLOCK in stabilizing heterochromatin, promoting tissue regeneration, and mitigating aging-associated chronic diseases. | Chuqian Liang Zunpeng Liu Moshi Song Wei Li Zeming Wu Zehua Wang Qiaoran Wang Si Wang Kaowen Yan Liang Sun Tomoaki Hishida Yanning Cai Juan Carlos lzpisua Belmonte Pedro Guillen Piu Chan Qi Zhou Weiqi Zhang Jing Qu Guang-Hui Liu | 2021 | Cell Research2021,31,2: | 8
Endostatin enhances antitumor effect of tumor antigen-pulsed dendritic cell therapy in mouse xenograft model of lung carcinoma显示文摘Objective: To investigate the antitumor effect of endostatin combined with tumor antigen-pulsed dendritic cell(DC)-T cell therapy on lung cancer. Methods: Transplanted Lewis lung cancer(LLC) models of C57BL/6 mice were established by subcutaneous injection of LLC cells in left extremity axillary. Tumor antigen-pulsed DC-T cells from spleen cells and bone of mice were cultured in vitro. Tumor-bearing mice were randomly divided into three groups, including DCT+endostatin group, DC-T group, and phosphate-buffered saline(PBS) control group. Microvessel density(MVD) of tumor tissue in tumor-bearing mice was determined by immunohistochemistry(IHC). The expressions of vascular endothelial growth factor(VEGF) and hypoxia-inducible factor-1α(HIF-1α) were determined by Western blotting and IHC staining. The proportions of CD8+ T cells, mature dendritic cells(m DC), tumor-associated macrophages [TAM(M1/M2)], and myeloid-derived suppressor cells(MDSC) in suspended cells of tumor tissue were determined by flow cytometry. The expressions of interleukin(IL)-6, IL-10, IL-17, transforming growth factor-β(TGF-β) and interferon-γ(IFN-γ) in suspended cells of tumor tissue were detected by enzyme-linked immune sorbent assay(ELISA).Results: DC-T cells combined with endostatin remarkably suppressed tumor growth. MVD of mice in DCT+endostatin group was significantly lower than that of the control group and DC-T monotherapy group. The expressions of VEGF, IL-6 and IL-17 in tumors were markedly decreased, but IFN-γ and HIF-1α increased after treating with DC-T cells combined with endostatin, compared to control group and DC-T group. In the DCT+endostatin group, the proportions of MDSC and TAM(M2 type) were significantly decreased, m DC and TAM(M1 type) were up-regulated, and CD8+ T cells were recruited to infiltrate tumors, in contrast to PBS control and DC-T monotherapy. DC-T cells combined with endostatin potently reduced the expressions of IL-6, IL-10, TGF-β and IL-17 in tumor tissue, and enhanced the expression of IFN-γ.Conclusions: The study indicated the synergic antitumor effects between endostatin and tumor antigen-pulsed DC-T cells, which may be a prospective therapy strategy to achieve potent antitumor effects on lung cancer. | ring Liang Xiaolin Liu Qi Xie Guoling Chen Xingyu Li Yanrui Jia Beibei Yin Xun Qu Yan Li | 2016 | Chinese Journal of Cancer Research2016,28,4: | 8
Development of mouse preimplantation embryos in space显示文摘The development o f life beyond planet Earth is a long-standing quest of the human race, but whether normal mammalian embryonic development can occur in space is still unclear. Here,we show unequivocally that preimplantation mouse embryos can develop in space;but the rate of blastocyst formation and blastocyst quality are compromised.Additionally, the cells in the embryo contain severe DNA damage, while the genome of the blastocysts developed in space is globally hypomethylated with a unique set of differentially methylated regions. The developmental defects,DNA damage and epigenetic abnormalities can be largely mimicked by the treatment with ground-based low-dose radiation. However, the exposure to simulated microgravity alone does not cause major disruptions of embryonic development, indicating that radiation is the main cause for the developmental defects. This work advances the understanding of embryonic development in space and reveals long-term extreme low-dose radiation as a hazardous factor for mammalian reproduction. | Xiaohua Lei Yujing Cao Baohua Ma Yunfang Zhang Lina Ning Jingjing Qian Liwen Zhang Yongcun Qu Tao Zhang Dehong Li Qi Chen Junchao Shi Xudong Zhang Chiyuan Ma Ying Zhang Enkui Duan | 2020 | National Science Review2020,7,9: | 8
Hierarchical activation of compartmentalized pools of AMPK depends on severity of nutrient or energy stress显示文摘AMPK, a master regulator of metabolic homeostasis, is activated by both AMP-dependent and AMP-independent mechanisms. The conditions under which these different mechanisms operate, and their biological implications are unclear. Here, we show that, depending on the degree of elevation of cellular AMP, distinct compartmentalized pools of AMPK are activated, phosphorylating different sets of targets. Low glucose activates AMPK exclusively through the AMP-independent, AXIN-based pathway in lysosomes to phosphorylate targets such as ACC1 and SREBP1c, exerting early anti-anabolic and pro-catabolic roles. Moderate increases in AMP expand this to activate cytosolic AMPK also in an AXIN-dependent manner. In contrast, high concentrations of AMP, arising from severe nutrient stress, activate all pools of AMPK independently of AXIN. Surprisingly, mitochondrion-localized AMPK is activated to phosphorylate ACC2 and mitochondrial fission factor (MFF) only during severe nutrient stress. Our findings reveal a spatiotemporal basis for hierarchical activation of different pools of AMPK during differing degrees of stress severity. | Yue Zong Chen-Song Zhang Mengqi Li Wen Wang Zhichao Wang Simon A. | 20 | No association between low-dose reserpine use and depression in older hypertensive patient: result of a multicenter, cross-sectional study显示文摘Background Reserpine is currently used by millions of Chinese hypertensive patients, in spite of the continued concern of its depressogenic effect, even when used in low dose. This study aimed to investigate the association between low-dose reserpine use and depression in older Chinese hypertensive patient. Methods In this cross-sectional, case-control study, we recruited patient aged 60 years or over who had regularly taken one or two tables of “compound reserpine and triamterene tablets (CRTTs)” for more than one year (reserpine user) from 26 community health centers located in 10 provinces in China. For each patient who took CRTTs, we selected an age (within five years) and sex matched hypertensive patient who had never taken any drugs containing reserpine (non-reserpine user) as control. Depressive symptoms were evaluated using a Chinese depression scale adapted from the Zung Self-Rating Depression Scale. Demographic, clinical data and laboratory examination results within six months were collected. Results From August 2018 to December 2018, 787 reserpine user and 787 non-reserpine user were recruited. The mean age of all study subjects was 70.3 years, with about equal numbers of males and females. The mean depression score was 40.4 in reserpine users and 40.6 in non-reserpine users (P = 0.7). The majority of study subject had a depression score < 53 (87.6% in reserpine users and 88.2% in non-reserpine users, respectively). There were no significant differences in the prevalence of mild, moderate or severe depression in reserpine users and non-reserpine users. Conclusions There is no association between low-dose reserpine use and depression in older hypertensive patient. The role of reserpine in the treatment and control of hypertension should be reconsidered;and further studies, especially randomized, controlled clinical trials to compare efficacy and safety of reserpine and other widely recommended anti-hypertensive agents are needed. | Guo-Hua ZHU Xi-Peng SUN Jing LI Lin PI Hai-Qin TANG Hai-Qing GAO Hong-Liang CONG Peng QU Xin-Zheng LU Xin-Jun ZHANG Luo-Sha ZHAO Yi-Fang GUO Dong-Xia LIU Liang-Qing ZHANG Hua TANG Yi-Xin HU Li FAN Qi HUA | 2019 | Journal of Geriatric Cardiology2019,16,8: | 8
