1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2Genomic structure of metabotropic glutamate receptor 7 and comparison of genomic structures of extracellular domains of mGluR family显示文摘Metabotropic glutamate receptor 7, coupled with a chemical neurotransmitter L-glutamate, plays an important role in the development of many psychiatric and neurological disorders. To study the biological and genetic mechanism of the mGluR7-related diseases, a physical map covering the full-length mGluR7 genomic sequence has been constructed through seed clone screening and fingerprinting database searching. These BAC clones in the physical map have been sequenced with shotgun strategy and assembled by Phred-Phrap-Consed software; the error rate of the final genoniic sequence is less than 0.01%. mGluR7 spans 880 kb genoniic region, the GC content and repeat content of mGluR7 genoniic sequence are 38% and 37.5% respectively. mGluR7 has a typical 'house-keeping' promoter and consists of 11 exons, with introns ranging from 6 kb to 285 kb. mGluR7a and mGluR7b are two known alternatively splicing variants. 6Automatic extraction method of force chain information and its application in the flow photoelastic experiment of granular matter显示文摘The force chain is the core of the multi-scale analysis of granular matter.Accurately extracting the force chain information among particles is of great significance to the study of particle mechanics and geological hazards caused by particle flow.However,in the photoelastic experiment,the precise identification of the branching points of force chains has not been effectively realized.Therefore,this study proposes an automatic extraction method of force chain key information.First,based on the Hough transform and the Euclidean distance,a particle geometric information identification model is established and geometric information such as particle circle center coordinates,radius,contact point location,and contact angle is extracted.Then,a particle contact force information identification model is established following the color gradient mean square method.The model realizes the rapid calibration and extraction of a large number of particle media contact force information.Next,combined with the force chain composition criterion and its quasilinear feature,an automatic extraction method of force chain information is established,which solves the problem of the accurate identification of the force chain branch points.Finally,in the photoelastic experiment of ore drawing from a single drawpoint,the automatic extraction method of force chain information is verified.The results show that the macroscopic distribution of force chains during ore drawing from a single drawpoint is left–right symmetrical.Strong force chains are mostly located on the two sides of the model but in small numbers and they mainly develop vertically.Additionally,the ends are mostly in a combination of Y and inverted Y shapes,while the middle is mostly quasilinear.Weak force chains are abundant and mostly distributed in the middle of the model,and develop in different directions.The proposed extraction method accurately extracts the force chain network from the photoelastic experiment images and dynamically characterizes the force chains of granular matter,which has significant advantages in particle geometry information extraction,force chain branch point discrimination,force chain retrieval,and force chain distribution and its azimuthal characterization.The results provide a scientific basis for studying the macroscopic and microscopic mechanical parameters of granular matter.Qingfa Chen Enlin Long Wenjing Niu Jun Liu Wenyu Fan Hangyu Li 2023Particuology2023,,12:0
7Modulation of fasting blood glucose by raw banana powder in alloxan-induced diabetic rats显示文摘This study aimed to observe the influence of raw banana powder(RBP)on fasting blood glucose(FBG),blood lipid and other biochemical indicators in type-2 diabetic rats and therefore to provide experimental evidences for developing suitable food from banana powder for diabetic patients.Eight Sprague-Dawley rats were selected randomly as the normal control group(NCG)before the experiment.After establishing type-2 diabetic rat models(11.1-16.7 mmoL/L)by alloxan,32 rats were divided into four groups:the diabetic control group(DCG,n=8),low-dose group(LDG,n=8),middle-dose group(MDG,n=8)and high-dose group(HDG,n=8).The LDG,MDG and HDG rats received gastric perfusion of RBP at the doses of 2 g/kg,4 g/kg and 6 g/kg per day,respectively.After four weeks,oral glucose tolerance test was carried out in each group,and then the FBG level,blood lipid,insulin,short chain fatty acids content,pH value of colon content and other biochemical indicators of rats in each group were determined and compared among the groups.Results showed that the levels of FBG significantly decreased in the LDG(11.97±0.83),MDG(8.95±0.45)and HDG(9.28±1.45),compared with their initial values(13.00±1.25,13.68±0.75 and 13.91±0.80,respectively).The FBG levels in these three groups were obviously lower than that in the DCG.However,there were no dramatic FBG changes in the NCG and DCG(5.77±0.59,14.14±0.72)compared with the initial stage(5.55±0.23,13.93±0.47).The RBP intervention increased insulin-sensitivity index and regulated postprandial blood glucose.Besides,RBP showed the positive effects on symptoms of type 2 diabetic rats,such as the reduction of weight gain and total cholesterol.Bai Yongliang Yu Ming Du Bing Chen Qingfa Yang Gongming 2013International Journal of Agricultural and Biological Engineering2013,6,3:0
