| 2 | The active principle region of Buyang Huanwu decoction induced differentiation of bone marrow-derived mesenchymal stem cells into neural-like cells Superior effects over original formula of Buyang Huanwu decoction显示文摘The present study induced in vitro-cultured passage 4 bone marrow-derived mesenchymal stem cells to differentiate into neural-like cells with a mixture of alkaloid, polysaccharide, aglycone, glycoside, essential oils, and effective components of Buyang Huanwu decoction (active principle region of decoction for invigorating yang for recuperation). After 28 days, nestin and neuron-specific enolase were expressed in the cytoplasm. Reverse transcription-PCR and western blot analyses showed that nestin and neuron-specific enolase mRNA and protein expression was greater in the active principle region group compared with the original formula group. | 3 | Oxymatrine reduces neuroinflammation in rat brain A signaling pathway显示文摘Cerebral neuroinflammation models were established by injecting 10 μg lipopolysaccharide into the hippocampus of male Sprague-Dawley rats. The rats were treated with an intraperitoneal injection of 120, 90, or 60 mg/kg oxymatrine daily for three days prior to the lipopolysaccharide injection. Twenty-four hours after model induction, the hippocampus was analyzed by real-time quantitative PCR, and the cerebral cortex was analyzed by enzyme-linked immunosorbent assay and western blot assay. The results of the enzyme-linked immunosorbent assay and the real-time quantitative PCR showed that the secretion and mRNA expression of the pro-inflammatory cytokines interleukin-1β and tumor necrosis factor-α were significantly decreased in the hippocampus and cerebral cortex of model rats treated with oxymatrine. Western blot assay and real-time quantitative PCR analysis indicated that toll-like receptor 4 mRNA and protein expression were significantly decreased in the groups receiving different doses of oxymatrine. Additionally, 120 and 90 mg/kg oxymatrine were shown to reduce protein levels of nuclear factor-κB p65 in the nucleus and of phosphorylated IκBα in the cytoplasm of brain cells, as detected by western blot assay. | 4 | Altitudinal variation of ecosystem CO_(2) fluxes in an alpine grassland from 3600 to 4200 m显示文摘Aims Recent studies have recognized the alpine grasslands on the Qinghai-Tibetan plateau as a significant sink for atmospheric CO_(2).The carbon-sink strength may differ among grassland ecosystems at various altitudes because of contrasting biotic and physical environments.This study aims(i)to clarify the altitudinal pattern of ecosystem CO_(2) fluxes,including gross primary production(GPP),daytime ecosystem respiration(Redaytime)and net ecosystem production(NEP),during the period with peak above-ground biomass;and(ii)to elucidate the effects of biotic and abiotic factors on the altitudinal variation of ecosystem CO_(2) fluxes.Methods Ecosystem CO_(2) fluxes and abiotic and biotic environmental factors were measured in an alpine grassland at four altitudes from 3600 to 4200 m along a slope of the Qilian Mountains on the northwestern Qinghai-Tibetan Plateau during the growing season of 2007.We used a closed-chamber method combined with shade screens and an opaque cloth to measure several carbon fluxes,GPP,Redaytime and NEP,and factors,light-response curve for GPP and temperature sensitivity of Redaytime.Above-and below-ground biomasses and soil C and N contents at each measurement point were also measured.Important Findings(i)Altitudinal pattern of ecosystem CO_(2) fluxes:The maximum net ecosystem CO_(2) flux(NEPmax),i.e.the potential ecosystem CO_(2) sink strength,was markedly different among the four altitudes.NEPmax was higher at the highest and lowest sites,ap proximately7.460.9 and6.760.6 lmol CO_(2) m^(-2)s^(-1)(mean 6 standard error),respectively,but smaller at the intermediate altitude sites(3800 and 4000 m).The altitudinal pattern of maximum gross primary production was similar to that of NEPmax.The Redaytime,however,was significantly higher at the lowest altitude(3.460.3 lmol CO_(2) m^(-2)s^(-1))than at the other three altitudes.(ii)Altitudinal variation of vegetation biomass:The aboveground biomass was higher at the highest altitude(154627 g DW m2)than at the other altitudes,which we attribute mainly to the large biomass in cushion plants at the highest altitude.The small above-ground biomass at the lower altitudes was probably due to heavy grazing during the growing season.(iii)Features of ecosystem CO_(2) fluxes:Redaytime and GPP were positively correlated with above-ground biomass.The low ratio of Redaytime to GPP at either the measurement point or the site level suggests that CO_(2) uptake efficiency tends to be higher at higher altitudes,which indicates a high potential sink strength for atmospheric CO_(2) despite the low temperature at high altitudes.The results suggest that the effect of grazing intensity on ecosystem carbon dynamics,partly by decreasing vegetation biomass,should be clarified further. | Mitsuru Hirota Pengcheng Zhang Song Gu Mingyuan Du Ayako Shimono Haihua Shen Yingnian Li Yanhong Tang | 2009 | Journal of Plant Ecology2009,2,4: | 6 |
| 5 | Genomic Epidemiology of ST34 Monophasic Salmonella enterica Serovar Typhimurium from Clinical Patients from 2008 to 2017 in Henan,China显示文摘Salmonella enterica serovar 4,[5],12:i:-(S.4,[5],12:i:-)is a monophasic variant of Salmonella enterica serovar Typhimurium that has emerged as a global serovar causing public health concern.To date,the epidemiology and genomic characterization of this pathogen in China have not been well described.We investigated the prevalence,antimicrobial resistance(AMR)phenotypes,and population genomics of sequence type 34(ST34)S.4,[5],12:i:-among cases of human salmonellosis in Henan Province,China.A total of 100 ST34 S.4,[5],12:i:-isolates were studied from 2008 to 2017 and found mostly resistant to ampicillin(AMP),streptomycin(STR),sulfonamides(SUL),and tetracycline(TET)(ASSu T).Bayesian phylogenetic analysis demonstrated that isolates identified in China were mostly related to the European lineage and evolved into two major clades with different resistance genes and plasmid profiles.Notably,clade 1 showed a significantly higher rate of mutations in gyr A and plasmid-mediated quinolone resistance genes.The carrying of the resistance-containing region(encoding R-type ASSu T),including bla(conferring resistance to AMP),str AB(STR),sul2(SUL),and tet(B)(TET)inserted into the flj BA operon,was responsible for most of the monophasic variants in clade 2.Inc HI2 plasmids were the dominant multi-drug resistance mobile genetic elements accounting for the transmission of acquired resistance genes in this serovar,and these were more prevalent in clade 1.Our findings highlighted the increasing prevalence of multi-drug resistant S.4,[5],12:i:-in China,along with the differential characteristics of resistance gene acquisition among various lineages.Based on our data,control measures are required to address the spread of this zoonotic pathogen.Further owing to its potential origin in food-producing animals,a'One Health'approach,should be implemented to support surveillance whilst informing interventional strategies. | Yujiao Mu Ruichao Li Pengcheng Du Pei Zhang Yan Li Shenghui Cui Séamus Fanning Li Bai | 2022 | Engineering2022,8,8: | 2 |

| 7 | Validation of a novel mixing-plane method for multistage turbomachinery steady flow analysis显示文摘The steady calculation based on the mixing-plane method is still the most widely-used three-dimensional flow analysis tool for multistage turbomachines. For modern turbomachines,the trend of design is to reach higher aerodynamic loading but with still further compact size. In such a case, the traditional mixing-plane method has to be revised to give a more physically meaningful prediction. In this paper, a novel mixing-plane method was proposed, and three representative test cases including a transonic compressor, a highly-loaded centrifugal compressor and a highpressure axial turbine were performed for validation purpose. This novel mixing-plane method can satisfy the flux conservation perfectly. Reverse flow across the mixing-plane interface can be resolved naturally, thus making this method numerically robust. Artificial reflection at the mixing-plane interface is almost eliminated, and then its detrimental impact on the flow field is minimized. | 15 | GPA: A Microbial Genetic Polymorphisms Assignments Tool in Metagenomic Analysis by Bayesian Estimation显示文摘Identifying antimicrobial resistant(AMR) bacteria in metagenomics samples is essential for public health and food safety. Next-generation sequencing(NGS) technology has provided a powerful tool in identifying the genetic variation and constructing the correlations between genotype and phenotype in humans and other species. However, for complex bacterial samples, there lacks a powerful bioinformatic tool to identify genetic polymorphisms or copy number variations(CNVs) for given genes. Here we provide a Bayesian framework for genotype estimation for mixtures of multiple bacteria, named as Genetic Polymorphisms Assignments(GPA). Simulation results showed that GPA has reduced the false discovery rate(FDR) and mean absolute error(MAE) in CNV and single nucleotide variant(SNV) identification. This framework was validated by whole-genome sequencing and Pool-seq data from Klebsiella pneumoniae with multiple bacteria mixture models, and showed the high accuracy in the allele fraction detections of CNVs and SNVs in AMR genes between two populations. The quantitative study on the changes of AMR genes fraction between two samples showed a good consistency with the AMR pattern observed in the individual strains. Also, the framework together with the genome annotation and population comparison tools has been integrated into an application, which could provide a complete solution for AMR gene identification and quantification in unculturable clinical samples. | 18 | Antioxidant procyanidin B2 protects oocytes against cryoinjuries via mitochondria regulated cortical tension显示文摘Background:Irreversible cryodamage caused by oocyte vitrification limited its wild application in female fertility preservation.Antioxidants were always used to antagonist the oxidative stress caused by vitrification.However,the comprehensive mechanism underlying the protective role of antioxidants has not been studied.Procyanidin B2(PCB2)is a potent natural antioxidant and its functions in response to vitrification are still unknown.In this study,the effects of PCB2 on vitrified-thawed oocytes and subsequent embryo development were explored,and the mechanisms underlying the protective role of PCB2 were systematically elucidated.Results:Vitrification induced a marked decline in oocyte quality,while PCB2 could improve oocyte viability and further development after parthenogenetic activation.A subsequent study indicated that PCB2 effectively attenuated vitrification-induced oxidative stress,rescued mitochondrial dysfunction,and improved cell viability.Moreover,PCB2 also acts as a cortical tension regulator apart from strong antioxidant properties.Increased cortical tension caused by PCB2 would maintain normal spindle morphology and promote migration,ensure correct meiosis progression and finally reduce the aneuploidy rate in vitrified oocytes.Further study reveals that ATP biosynthesis plays a crucial role in cortical tension regulation,and PCB2 effectively increased the cortical tension through the electron transfer chain pathway.Additionally,PCB2 would elevate the cortical tension in embryo cells at morula and blastocyst stages and further improve blastocyst quality.What's more,targeted metabolomics shows that PCB2 has a beneficial effect on blastocyst formation by mediating saccharides and amino acids metabolism.Conclusions:Antioxidant PCB2 exhibits multi-protective roles in response to vitrification stimuli through mitochondria-mediated cortical tension regulation. | Qingrui Zhuan Jun Li Xingzhu Du Luyao Zhang Lin Meng Yuwen Luo Dan Zhou Hongyu Liu Pengcheng Wan Yunpeng Hou Xiangwei Fu | 2023 | Journal of Animal Science and Biotechnology2023,14,1: | 0 |
| 19 | Uncertainty analysis of measured geometric variations in turbine blades and impact on aerodynamic performance显示文摘Inevitable geometric variations significantly affect the performance of turbines or even that of entire engines;thus,it is necessary to determine their actual characteristics and accurately estimate their impact on performance.In this study,based on 1781 measured profiles of a typical turbine blade,the statistical characteristics of the geometric variations and the uncertainty impact are analyzed,and some commonly used uncertainty modelling methods based on Principal-Component Analysis(PCA)are verified.The geometric variations are found to be evident,asymmetric,and non-uniform,and the non-normality of the random distributions is non-negligible.The performance is notably affected,which is manifested as an overall offset,a notable scattering,and significant deterioration in several extreme cases.Additionally,it is demonstrated that the PCA reconstruction model is effective in characterizing major uncertainty characteristics of the geometric variations and their impact on the performance with almost the first 10 PCA modes.Based on a reasonable profile error and mean geometric deviation,the Gaussian assumption and stochasticprocess-based model are also found to be effective in predicting the mean values and standard deviations of the performance variations.However,they fail to predict the probability of some extreme cases with high loss.Finally,a Chi-square-based correction model is proposed to compensate for this deficiency.The present work can provide a useful reference for uncertainty analysis of the impact of geometric variations,and the corresponding uncertainty design of turbine blades. | Xiaojing WANG Pengcheng DU Lichao YAO Zhengping ZOU Fei ZENG | 2023 | Chinese Journal of Aeronautics2023,36,6: | 0 |
| 20 | Low molecular weight protein phosphatase APH mediates tyrosine dephosphorylation and ABA response in Arabidopsis.显示文摘Low molecular weight protein tyrosine phosphatase(LWM-PTP),also known as acid phosphatase,is a highly conserved tyrosine phosphatase in living organisms.However,the function of LWM-PTP homolog has not been reported yet in plants.Here,we revealed a homolog of acid phosphatase,APH,in Arabidopsis plants,is a functional protein tyrosine phosphatase.The aph mutants are hyposensitive to ABA in post-germination growth.We performed an anti-phosphotyrosine antibody-based quantitative phosphoproteomics in wild-type and aph mutant and identified hundreds of putative targets of APH,including multiple splicing factors and other transcriptional regulators.Consistently,RNA-seq analysis revealed that the expression of ABA-highly-responsive genes is suppressed in aph mutants.Thus,APH regulates the ABA-responsive gene expressions by regulating the tyrosine phosphorylation of multiple splicing factors and other post-transcriptional regulators.We also revealed that Tyr383 in RAF9,a member of B2 and B3 RAF kinases that phosphorylate and activate SnRK2s in the ABA signaling pathway,is a direct target site of APH.Phosphorylation of Tyr383 is essential for RAF9 activity.Our results uncovered a crucial function of APH in ABA-induced tyrosine phosphorylation in Arabidopsis. | Yanyan Du Shaojun Xie Yubei Wang Yu Ma Bei Jia Xue Liu Jingkai Rong Rongxia Li Xiaohong Zhu Chun-Peng Song WAndy Tao Pengcheng Wang | 2022 | Stress Biology2022,2,1: | 0 |
