New strategy for designing orangish-redemitting phosphor via oxygen-vacancyinduced electronic localization显示文摘Phosphor-converted white-light-emitting diodes(pc-WLED)have been extensively employed as solid-state lighting sources, which have a very important role in people’s daily lives. However, due to the scarcity of the red component,it is difficult to realize warm white light efficiently.Hence,red-emitting phosphors are urgently required for improving the illumination quality. In this work,we develop a novel orangish-red La_(4)GeO_(8):Bi^(3+) phosphor, the emission peak of which is located at 600 nm under near-ultraviolet(n-UV)light excitation. The full width at half maximum(fwhm)is 103 nm,the internal quantum efficiency(IQE)exceeds 88%,and the external quantum efficiency(EQE)is 69%.According to Rietveld refinement analysis and density functional theory(DFT)calculations,Bi^(3+) ions randomly occupy all La sites in orthorhombic La_(4)GeO_(8).Importantly,the oxygen-vacancy-induced electronic localization around the Bi3+ions is the main reason for the highly efficient orangish-red luminescence. These results provide a new perspective and insight from the local electron structure for designing inorganic phosphor materials that realize the unique luminescence performance of Bi^(3+) ions. | Yi Wei Gongcheng Xing Kang Liu Guogang Li Peipei Dang Sisi Liang Min Liu Ziyong Cheng Dayong Jin Jun Lin | 2019 | Light(Science & Applications)2019,8,1: | 12
A dimeric structure of PD-L1: functional units or evolutionary relics?显示文摘PD-L1 is a member of the B7 protein family, most of whose members so far were identified as dimers in a solution and crystalline state, either complexed or uncomplexed with their ligand(s).The binding of PD-L1 with its receptor PD-1(CD279)delivers an inhibitory signal regulating the T cell function.Simultaneously with the Garboczi group,we successfully solved another structure of human PD-L1(hPD-L1).Our protein crystallized in the space group of C222_(1) with two hPD-L1 molecules per asymmetric unit. After comparison of reported B7 structures,we have found some intrinsic factors involved in the interaction of these two molecules.Based on these results,we tend to believe this uncomplexed hPD-L1 structure demonstrated its potential dimeric state in solution, althougt it could just be an evolutionary relic, too weak to be detected under present technology,or still a functional unit deserved our attentions. | Yong Chen Peipei Liu Feng Gao Hao Cheng Jianxun Qi George F.Gao | 2010 | Protein & Cell2010,1,2: | 10
Influence of surface modification of SrFe12O19 particles with oleic acid on magnetic microsphere preparation显示文摘Oleic acid was used as surface modification agent to improve the hydrophobicity of magnetic strontium hexaferrite particles. The structure and properties of treated magnetic particles were characterized by scanning electronic microscopy(SEM),Fourier transform infrared spectra(FTIR),powder X-ray diffraction(XRD) and magnetic property measurement system(MPMS).The results show that oleic acid is chemically enwrapped on the surface of SrFe12O19 particles.Magnetic particles modified by oleic acid are highly dispersible and strongly responsive to magnetism but with slight decrease in saturated magnetization. The affinity between magnetic particles and monomers is improved by surface modification, resulting in increased particle incorporation in magnetic polymeric microspheres. The surface modification mechanism of magnetic particles by oleic acid is addressed in this work. | Sifang Kong Peipei Zhang Xiufang Wen Pihui Pi Jiang Cheng Zhuoru Yang Jing Hai | 2008 | Particuology2008,6,3: | 7

A novel PDZ domain-containing gene is essential for male sex differentiation and maintenance in yellow catfish(Pelteobagrus fulvidraco)显示文摘The sex-determining genes are found to be variable among different fish species. Yellow catfish(Pelteobagrus fulvidraco) is an important aquaculture fish species in China with XX/XY sex-determining type. Recently, YY super-male yellow catfish has been successfully produced by combining hormonalinduced sex reversal method with sex chromosome-linked markers. Here, we identified a novel PDZ domain-containing gene in yellow catfish designated as pfpdz1, in whose intron the sex-linked marker was located. The coding sequence of pfpdz1 in Y chromosome was identical to that in X chromosome except a missense SNP(A/T) that changes an amino acid(E8V) in the N-terminal region. Pfpdz1 displayed male-specific expression during sex differentiation. Overexpression of pfpdz1 using additive transgenesis induces XX ovary to differentiate into testis-like tissue, while the targeted inactivation of pfpdz1 in Y chromosome using CRISPR/Cas9-mediated mutagenesis triggers ovarian differentiation. Furthermore,we demonstrated that pfpdz1 initiates testicular differentiation through upregulating expression of amh, dmrt1 and sox9a1, as well as downregulating expression of cyp19a1, foxl2 and wnt4. Our data provide functional evidence that pfpdz1 is significant for male differentiation and maintenance in yellow catfish. | Cheng Dan Qiaohong Lin Gaorui Gong Tianyi Yang Shuting Xiong Yang Xiong Peipei Huang Jian-Fang Gui Jie Mei | 2018 | Science Bulletin2018,63,21: | 4
Genomic characterizations of H4 subtype avian influenza viruses from live poultry markets in Sichuan province of China, 2014–2015显示文摘Dear Editor,Avian influenza viruses (AIVs) have posed a serious threat to poultry production and public health. To date, more than fourteen AIV subtypes that are able to infect human beings have been documented. Also, it is suggested that new subtypes may be reported in the future, owing to the migration of wild birds and live poultry transportation (Gao, 2018).Poultry may act as a potential incubator for novel subtypes of avian influenza virus (Bi et al., 2016a; Bi et al., 2016b; Liu et al., 2014a; Su et al., 2017). Up to date, the H7N9 AIV emerged in February 2013 has caused 1,567 human cases, with a fatality rate of 39.2%(http://gffzz36b398b81f8a4a4ch9uook9kquw0566qw.ffgz.tsg.suse.edu.cn/influenza/ | Chuansong Quan Ting Huang Xiuwei Chen Jie Zhang Qianli Wang Cheng Zhang Tiyan Zhang Lijun Zhou Liumei Shu Chunrong Long Lei Yang Xunbo Du Yingze Zhao Peipei Liu Hao Song Weifeng Shi Yuhai Bi Qiang Lv William J.Liu George F.Gao | 2018 | Science China(Life Sciences)2018,61,9: | 4
Parathyroid hormone increases alveolar bone homoeostasis during orthodontic tooth movement in rats with periodontitis via crosstalk between STAT3 and β-catenin显示文摘Periodontitis patients are at risk of alveolar bone loss during orthodontic treatment. The aim of this study was to investigate whether intermittent parathyroid hormone(1–34)treatment(iPTH)could reduce alveolar bone loss during orthodontic tooth movement(OTM)in individuals with periodontitis and the underlying mechanism.A rat model of OTM in the context of periodontitis was established and alveolar bone loss was observed. The control,iPTH and iPTH+stattic groups received injections of vehicle,PTH and vehicle,or PTH and the signal transducer and activator of transcription 3(STAT3)inhibitor stattic, respectively.iPTH prevented alveolar bone loss by enhancing osteogenesis and suppressing bone resorption in the alveolar bone during OTM in rats with periodontitis. This effect of iPTH was along with STAT3 activation and reduced by a local injection of stattic.iPTH promoted osteoblastic differentiation and might further regulate the Wnt/β-catenin pathway in a STAT3-dependent manner. The findings of this study suggest that iPTH might reduce alveolar bone loss during OTM in rats with periodontitis through STAT3/β-catenin crosstalk. | Cheng Zhang Tiancheng Li Chenchen Zhou Li Huang Yuyu Li Han Wang Peipei Duan Shujuan Zou Li Mei | 2020 | International Journal of Oral Science2020,12,4: | 4
Growth and characterization ofβ-Ga_(2)O_(3)thin films grown on off-angled Al_(2)O_(3)substrates by metal-organic chemical vapor deposition显示文摘Beta-gallium oxide(β-Ga_(2)O_(3))thin films were deposited on c-plane(0001)sapphire substrates with different mis-cut angles along<>by metal-organic chemical vapor deposition(MOCVD).The structural properties and surface morphology of as-grownβ-Ga_(2)O_(3)thin films were investigated in detail. It was found that by using thin buffer layer and mis-cut substrate technology, the full width at half maximum(FWHM)of the()diffraction peak of theβ-Ga_(2)O_(3)film is decreased from 2°on c-plane(0001)Al_(2)O_(3)substrate to 0.64°on an 8°off-angled c-plane(0001)Al_(2)O_(3)substrate.The surface root-mean-square(RMS)roughness can also be improved greatly and the value is 1.27 nm for 8°off-angled c-plane(0001)Al_(2)O_(3)substrate.Room temper-ature photoluminescence(PL)was observed, which was attributed to the self-trapped excitons formed by oxygen and gallium vacancies in the film. The ultraviolet-blue PL intensity related with oxygen and gallium vacancies is decreased with the increas-ing mis-cut angle, which is in agreement with the improved crystal quality measured by high resolution X-ray diffraction(HR-XRD).The present results provide a route for growing high qualityβ-Ga_(2)O_(3)film on Al_(2)O_(3)substrate. | Yabao Zhang Jun Zheng Peipei Ma Xueyi Zheng Zhi Liu Yuhua Zuo Chuanbo Li Buwen Cheng | 2022 | Journal of Semiconductors2022,43,9: | 3

A Variational Binary Level Set Method for Structural Topology Optimization显示文摘This paper proposes a variational binary level set method for shape and topology optimization of structural.First,a topology optimization problem is presented based on the level set method and an algorithm based on binary level set method is proposed to solve such problem.Considering the difficulties of coordination between the various parameters and efficient implementation of the proposed method,we present a fast algorithm by reducing several parameters to only one parameter, which would substantially reduce the complexity of computation and make it easily and quickly to get the optimal solution. The algorithm we constructed does not need to re-initialize and can produce many new holes automatically.Furthermore, the fast algorithm allows us to avoid the update of Lagrange multiplier and easily deal with constraints, such as piecewise constant, volume and length of the interfaces. Finally,we show several optimum design examples to confirm the validity and efficiency of our method. | Xiaoxia Dai Peipei Tang Xiaoliang Cheng Minghui Wu | 2013 | Communications in Computational Physics2013,13,5: | 1

Research Article Core/hybrid-shell structures boost thermoelectric performance of flexible inorganic/organic nanowire films显示文摘Interface control in inorganic/organic composites has always been regarded as one of the effective means to optimize their thermoelectric(TE)performance, and the past few years have witnessed its development, including carrier-energy filtering and phonon scattering. However, the energy barrier created by the band alignment at the composite interface depends on the Fermi level difference between the organic and inorganic components, which is difficult to be controlled by the common means.Herein,a core/hybrid-shell strategy aiming for efficient interface control is proposed to tune the energy barrier of the inorganic/organic core/shell nanowire interface. The Fermi level of hybrid-shell can be effectively controlled by separating the charge carriers compared to the single-shell composites. The energy barrier of the core/hybrid-shell interface is tuned to an appropriate position, and the energy filtering effect is utilized, resulting in a substantial improvement in power factor and reduction in thermal conductivity for the prepared core/hybrid-shell composites with good air-stability and flexibility. Moreover, both the flexible p type and p-n type TE devices based on the prepared core/hybrid-shell films yield excellent output properties with the maximum power densities of 41 and 45μW·cm^(−2)at a temperature difference of ca.30 K,respectively. This study provides a novel strategy to improve the TE performance of the inorganic/organic composites, displaying great potential for low-power wearable electronics. | Changcun Li Xiaoqi Lan Peipei Liu Jingkun Xu Qinglin Jiang Congcong Liu Cheng Liu Fengxing Jiang | 2023 | Nano Research2023,16,4: | 1
High-throughput screening of SARS-CoV-2 main and papain-like protease inhibitors显示文摘The global COVID-19 coronavirus pandemic has infected over 109 million people, leading to over 2 million deaths up to date and still lacking of effective drugs for patient treatment. Here,we screened about 1.8 million small molecules against the main protease(M^(pro))and papain like protease(PL^(pro)),two major proteases in severe acute respiratory syndrome-coronavirus 2 genome, and identified 1851M^(pro)inhibitors and 205 PL^(pro)inhibitors with low nmol/l activity of the best hits.Among these inhibitors, eight small molecules showed dual inhibition effects on both M^(pro)and PL^(pro),exhibiting potential as better candidates for COVID-19 treatment. The best inhibitors of each protease were tested in antiviral assay, with over 40%of M^(pro)inhibitors and over 20%of PL^(pro)inhibitors showing high potency in viral inhibition with low cytotoxicity. The X-ray crystal structure of SARS-CoV-2 M^(pro)in complex with its potent inhibitor 4a was determined at 1.8Åresolution.Together with docking assays, our results provide a comprehensive resource for future research on anti-SARS-CoV-2 drug development. | Yi Zang Mingbo Su Qingxing Wang Xi Cheng Wenru Zhang Yao Zhao Tong Chen Yingyan Jiang Qiang Shen Juan Du Qiuxiang Tan Peipei Wang Lixin Gao Zhenming Jin Mengmeng Zhang Cong Li Ya Zhu Bo Feng Bixi Tang Han Xie Ming-Wei Wang Mingyue Zheng Xiaoyan Pan Haitao Yang Yechun Xu Beili Wu Leike Zhang Zihe Rao Xiuna Yang Hualiang Jiang Gengfu Xiao Qiang Zhao Jia Li | 2023 | Protein & Cell2023,14,1: | 0
Identification of de novo Mutations in the Chinese Autism Spectrum Disorder Cohort via Whole-Exome Sequencing Unveils Brain Regions Implicated in Autism显示文摘Autism spectrum disorder(ASD)is a highly heritable neurodevelopmental disorder characterized by deficits in social interactions and repetitive behaviors.Although hundreds of ASD risk genes, implicated in synaptic formation and transcriptional regulation, have been identified through human genetic studies, the East Asian ASD cohorts are still under-represented in genome-wide genetic studies. Here,we applied whole-exome sequencing to 369 ASD trios including probands and unaffected parents of Chinese origin.Using a joint-calling analytical pipeline based on GATK toolkits,we identified numerous de novo mutations including 55 high-impact variants and 165 moderate-impact variants,as well as de novo copy number variations containing known ASD-related genes.Importantly, combined with single-cell sequencing data from the developing human brain,we found that the expression of genes with de novo mutations was specifically enriched in the pre-,post-central gyrus(PRC,PC)and banks of the superior temporal(BST)regions in the human brain.By further analyzing the brain imaging data with ASD and healthy controls,we found that the gray volume of the right BST in ASD patients was significantly decreased compared to healthy controls, suggesting the potential structural deficits associated with ASD.Finally,we found a decrease in the seed-based functional connectivity between BST/PC/PRC and sensory areas, the insula,as well as the frontal lobes in ASD patients. This work indicated that combinatorial analysis with genome-wide screening,single-cell sequencing, and brain imaging data reveal the brain regions contributing to the etiology of ASD. | Bo Yuan Mengdi Wang Xinran Wu Peipei Cheng Ran Zhang Ran Zhang Shunying Yu Jie Zhang Yasong Du Xiaoqun Wang Zilong Qiu | 2023 | Neuroscience Bulletin2023,39,10: | 0
Origin and chromatin remodeling of young X/Y sex chromosomes in catfish with sexual plasticity显示文摘Assembly of a complete Y chromosome is a significant challenge in animals with an XX/XY sex-determination system.Re cently,we created YY-supermale yellow catfish by crossing XY males with sex-reversed XY females, providing a valuable model for Y-chromosome assembly and evolution. Here,we assembled highly homomorphic Y and X chromosomes by sequencing genomes of the YY supermale and XX female in yellow catfish, reve aling their nucleotide divergences with only less than 1%and with the same gene compositions. The sex-determining region(SDR)was identified to locate within a physical distance of0.3 Mb by FSTscanning.Strikingly, the incipient sex chromosomes were revealed to originate via autosome-autosome fusion and were characterized by a highly rearranged region with an SDR downstream of the fusion site.We found that the Y chromosome was at a very early stage of differentiation,as no clear evidence of evolutionary strata and classical structure features of recombination suppression for a rather late stage of Y-chromosome evolution were observed.Significantly,a number of sex-antagonistic mutations and the accumulation of repetitive elements were discovered in the SDR,which might be the main driver of the initial establishment of recombination suppression between young X and Y chromosomes. Moreover, distinct three-dimensional chromatin organizations of the Y and X chromosomes were identified in the YY supermales and XX females,as the X chromosome exhibited denser chromatin structure than the Y chromosome, while they respectively have significantly spatial interactions with female-and male-related genes compared with other autosomes. The chromatin configuration of the sex chromosomes as well as the nucleus spatial organization of the XX neomale were remodeled after sex reversal and similar to those in YY supermales, and a male-specific loop containing the SDR was found in the open chromatin region. Our results elucidate the origin of young sex chromosomes and the chromatin remodeling configuration in the catfish sexual plasticity. | Gaorui Gong Yang Xiong Shijun Xiao Xi-Yin Li Peipei Huang Qian Liao Qingqing Han Qiaohong Lin Cheng Dan Li Zhou Fan Ren Qi Zhou Jian-Fang Gui Jie Mei | 2023 | National Science Review2023,10,2: | 0
The heterochronic gene Oryza sativa LIKE HETEROCHROMATIN PROTEIN 1 modulates miR156b/c/i/e levels显示文摘The juvenile-to-adult transition in plants involves changes in vegetative growth and plant architecture;the timing of this transition has important implications for agriculture. The microRNA miR156 regulates this transition and shoot maturation in plants. In Arabidopsis thaliana, deposition of histone H3 trimethylation on lysine 27(H3K27me3,a repressive mark)at the MIR156A/C loci is regulated by Polycomb Repressive Complex 1(PRC1)or PRC2,depending on the developmental stage. The levels of miR156 progressively decline during shoot maturation. The amount of H3K27me3 at MIR156A/C loci affects miR156 levels;however, whether this epigenetic regulation is conserved remains unclear. Here,we found that in rice(Oryza sativa),the putative PRC1 subunit LIKE HETEROCHROMATIN PROTEIN 1(OsLHP1),with the miR156–SQUAMOSA PROMOTER BINDING PROTEIN-LIKE(SPL)module,affects developmental phase transitions.Loss of OsLHP1 function results in ectopic expression of MIR156B/C/I/E,phenocopy of miR156 overexpression, and reduced H3k27me3 levels at MIR156B/C/I/E.This indicates that OsLHP1 has functionally diverged from Arabidopsis LHP1.Genetic and transcriptome analyses of wild-type,miR156b/c-overexpression,and Oslhp1-2 mutant plants suggest that OsLHP1 acts upstream of miR156 and SPL during the juvenile-to-adult transition. Therefore, modifying the OsLHP1–miR156–SPL pathway may enable alteration of the vegetative period and plant architecture. | Yongtao Cui Jingfei Cheng Shuang Ruan Peipei Qi Wei Liu Hongwu Bian Luhuan Ye Yuping Zhang Jiang Hu Guojun Dong Longbiao Guo Yijing Zhang Qian Qian d Xingming Hu | 2020 | Journal of Integrative Plant Biology2020,62,12: | 0

Preventive and therapeutic benefits of nelfinavir in rhesus macaques and human beings infected with SARS-CoV-2显示文摘Effective drugs with broad spectrum safety profile to all people are highly expected to combat COVID-19 caused by SARS-CoV-2.Here we report that nelfinavir,an FDA approved drug for the treatment of HIV infection,is effective against SARS-CoV-2 and COVID-19.Preincubation of nelfinavir could inhibit the activity of the main protease of the SARS-CoV-2(IC50=8.26μM),while its antiviral activity in Vero E6 cells against a clinical isolate of SARS-CoV-2 was determined to be 2.93μM(EC50).In comparison with vehicle-treated animals, rhesus macaque prophylactically treated with nelfinavir had significantly lower temperature and significantly reduced virus loads in the nasal and anal swabs of the animals. At necropsy,nelfinavir-treated animals had a significant reduction of the viral replication in the lungs by nearly three orders of magnitude.A prospective clinic study with 37 enrolled treatment-naive patients at Shanghai Public Health Clinical Center, which were randomized(1:1)to nelfinavir and control groups, showed that the nelfinavir treatment could shorten the duration of viral shedding by 5.5 days(9.0 vs.14.5 days,P=0.055)and the duration of fever time by 3.8 days(2.8 vs.6.6 days,P=0.014)in mild/moderate COVID-19 patients. The antiviral efficiency and clinical benefits in rhesus macaque model and in COVID-19 patients, together with its well-established good safety profile in almost all ages and during pregnancy, indicated that nelfinavir is a highly promising medication with the potential of preventative effect for the treatment of COVID-19. | Zhijian Xu Danrong Shi Jian-Bao Han Yun Ling Xiangrui Jiang Xiangyun Lu Chuan Li Likun Gong Guangbo Ge Yani Zhang Yi Zang Tian-Zhang Song Xiao-Li Feng Ren-Rong Tian Jia Ji Miaojin Zhu Nanping Wu Chunhui Wu Zhen Wang Yechun Xu Cheng Peng Min Zheng Junling Yang Feifei Du Junliang Wu Peipei Wang Jingshan Shen Jianliang Zhang Yong-Tang Zheng Hangping Yao Weiliang Zhu | 2023 | Signal Transduction and Targeted Therapy2023,8,5: | 0
MECOM promotes supporting cell proliferation and differentiation in cochlea显示文摘Permanent damage to hair cells(HCs)is the leading cause of sensory deafness.Supporting cells(SCs)are essential in the restoration of hearing in mammals because they can proliferate and differentiate to HCs.MDS1 and EVI1 complex locus(MECOM)is vital in early development and cell differentiation and regulates the TGF-βsignaling pathway to adapt to pathophysiological events, such as hematopoietic proliferation, differentiation and cells death. In addition,MECOM plays an essential role in neurogenesis and craniofacial development. However, the role of MECOM in the development of cochlea and its way to regulate related signaling are not fully understood. To address this problem, this study examined the expression of MECOM during the development of cochlea and observed a significant increase of MECOM at the key point of auditory epithelial morphogenesis, indicating that MECOM may have a vital function in the formation of cochlea and regeneration of HCs.Meanwhile,we tried to explore the possible effect and potential mechanism of MECOM in SC proliferation and HC regeneration.Findings from this study indicate that overexpression of MECOM markedly increases the proliferation of SCs in the inner ear,and the expression of Smad3 and Cdkn2b related to TGF signaling is significantly down-regulated,corresponding to the overexpression of MECOM.Collectively, these data may provide an explanation of the vital function of MECOM in SC proliferation and trans-differentiation into HCs,as well as its regulation. The interaction between MECOM,Wnt,Notch and the TGF-βsignaling may provide a feasible approach to induce the regeneration of HCs. | Peipei Chen Na Zuo Cheng Wu Jun Ma Yao Li Junfei Gu Wen Li Shaofeng Liu | 2022 | Journal of Otology2022,17,2: | 0
