1Over-expression of fibroblast growth factor receptor 3 in human hepatocellular carcinoma显示文摘AIM: To describe the significant over-expression of fibroblast growth factor receptor 3 (FGFR3), which is a signal transduction and cell proliferation related gene in hepatocellular carcinoma (HCC).METHODS: Following DNA microarray, Northern blot and quantitative real-time PCR were employed to confirm FGFR3 expression difference in HCC tissues and surrounding non-neoplastic liver tissue. FGFR3 expression levels were further determined by immunohistochemical study in 43 cases of HCC.RESULTS: Northern blot results showed the significant over-expression of FGFR3 in HCC tissues, which was consistent with that from DNA microarray. Quantitative real-time PCR demonstrated that the mean ratio of FGFR3 mRNA to glyceraldehyde-3-phosphate dehydrogenase (GADPH) mRNA in HCC tissue was 0.250, whereas the ratio in non-neoplastic liver tissue was 0.014. Statistical analyses of 43 cases of HCC revealed that HCC scored higher than the matched non-neoplastic liver tissues.Examination of clinicopathological features revealed a strong correlation of over-expression of FGFR3 with poor tumor differentiation and high nuclear grade.CONCLUSION: Over-expression of FGFR3 may play an important role in liver carcinogenesis. 3GADD45β基因表达诱导对不同p53状态的肝癌细胞作用显示文摘目的:体外合成GADD45β基因全序列表达质粒后,研究GADD45β基因表达诱导对呈不同p53状态的肝癌细胞的作用及可能机制。方法:采用RT-PCR法获得GADD45β基因全序列,插入pDrive穿梭克隆载体和pIRES2-EGFP荧光表达载体后大量扩增获得DNA,结合p53全基因表达质粒pp53-EGFP转染HepG2、Hep3B细胞后,以[3H]胸腺嘧啶脱氧核苷掺入法(3H-Tdr)和细胞克隆形成法分析DNA合成变化及细胞生长能力;以双抗体夹心ELISA法测定TGF-β1表达变化。结果:成功合成GADD45β基因全序列和表达质粒,通过流式细胞仪收集转染阳性的荧光表达细胞能显著提高转染效率;转染GADD45β后,具有野生型p53基因的HepG2细胞的细胞克隆形成能力和DNA合成能力明显受到抑制,细胞凋亡明显增加,TGF-β1的表达亦明显受抑。与之相反,缺失p53基因的Hep3B需要同时共转染p53基因后,方出现抑制效应。结论:GADD45β基因能够有效抑制肝癌细胞的生长,其功能需要完整p53基因的辅助和(或)调控。GADD45表达和(或)功能异常,导致p53介导的DNA损伤修复途径异常或阻断,是肝脏细胞恶性转化及形成肿瘤的可能机制。邱伟华 Dana Darwish Peiguo G.Chu Frank Luh 陈皓 邓侠兴 沈柏用 杨卫平 李宏为 Yun Yen 2005外科理论与实践2005,10,2:6
4STAT3在SAMe抑制HepG2细胞VEGF表达中的作用显示文摘目的研究肝癌细胞(HepG2)和正常胎肝细胞(CL-48)血管内皮生长因子(VEGF)和信号转导与转录激活因子3(STAT3)的表达状态,以及S-腺苷甲硫氨酸(SAM e)对其的干预作用,为HepG2和CL-48的不同调控途径提供功能性依据。方法在对数生长期的CL-48和HepG2细胞中加入不同浓度的SAM e(0、0.5、1.0 mmol/L)作用72 h,分别采用Northern印迹法、实时荧光定量PCR、W estern印迹法以及[3H]胸腺嘧啶脱氧核苷掺入法(3H-Tdr)和细胞克隆形成法,观察VEGF和STAT3表达的变化趋势,分析DNA合成及细胞生长能力的变化。结果SAM e作用前(SAM e 0 mmol/L),HepG2和CL-48均呈现VEGF和STAT3高表达状态。SAM e作用后:HepG2的VEGF和STAT3表达明显降低,并呈现剂量-效应关系,DNA合成和细胞生长能力明显受限;CL-48的STAT3表达虽然受到抑制,但VEGF的高表达状态无明显降低趋势,且DNA合成和细胞生长能力无显著变化。结论虽然HepG2和CL-48均呈现VEGF和STAT3的高表达,但二者具有不同的调控途径;SAM e通过抑制STAT3的组成性活化而降低VEGF的高表达状态,可能是SAM e的肝脏保护机制之一。邱伟华 Bingsen Zhou Dana Darwish Peiguo G.Chu Frank Luh 陈皓 杨卫平 李宏为 Yun Yen 2006上海交通大学学报(医学版)2006,26,6:6
5Effects of Panax notoginseng saponins on apoptosis induced by hydrogen peroxide in cultured rabbit bone marrow stromal cells via altering the oxidative stress level and down-regulating caspase-3显示文摘Objective: To investigate the effects of Panax notoginseng saponins (PNS) on hydrogen peroxide (H2O2)-induced apoptosis in cultured rabbit bone marrow stromal cells (BMSCs). Methods: The effects of different concentrations of PNS on proliferation and early osteoblast differentiation of BMSCs were determined by the MTT assay and an alkaline phosphatase (ALP) assay. An optimal effective concentration of PNS was determined and used in subsequent experiments. The cultured BMSCs were divided into three groups: untreated control, H2O2 treated, and PNS pretreatment of H2O2 treated. The oxidative stress level was assessed by superoxide dismutase (SOD) and malondialdehyde (MDA) assays. Flow cytometry was used to determine BMSC apoptosis by staining with annexinV-FITC/propidium iodide (PI). The activity of caspase-3 enzyme was measured by spectrofluorometry. Results: PNS (0.1g/L) significantly increased both BMSC proliferation rate and ALP activity, while it decreased the indicators of oxidative stress, caspase-3 activity, and the apoptosis rate of BMSCs induced by H2O2.. 6羟基脲对肝癌细胞HepG2中GADD45β表达影响及其可能机制显示文摘目的 初步明确肝癌细胞中GADD45β基因近端启动子序列,探索羟基脲对人肝癌细胞HepG2的GADD45β表达影响及可能机制.方法 体外合成GADD45β近端启动子序列群(-618~-314),构建荧光素表达质粒,转染肝癌细胞株HepG2,根据启动子活性强度结合数据库分析存在的转录调节因子结合位点;以实时荧光定量PCR比较羟基脲作用前后HepG2细胞GADD45β表达,并进一步比较羟基脲对GADD45β启动子活性的调控作用、分析羟基脲对HepG2的抑制效应,并通过Caspase-8、Caspase-9和Caspase-3的表达变化测定凋亡的发生和发展.结果 GADD4518近端启动子中含有3个NF-кB(-602/-593、-581/-572、-537/-528)和1个E2F-1(-470/-436)转录调节因子与启动子结合位点;羟基脲能明显诱导HepG2中GADD45β的表达,并呈现出剂量-效应的正相关关系,同时NF-кB和E2F-1启动子均明显增强.羟基脲能够明显抑制HepG2的DNA合成能力和细胞克隆形成能力,同时羟基脲能迅速启动HepG2凋亡的发生和发展.结论 羟基脲能明显诱导肝癌细胞中特异性缺失的GADD45β基因表达,增强转录调节因子的表达水平是其可能的作用机制.邱伟华 Bingsen Zhou Peiguo G.Chu 陈皓 施敏敏 Yun Yen 2010中华肝胆外科杂志2010,16,4:3
