1Cytokine- and chemokine-induced inflammatory colorectal tumor microenvironment: Emerging avenue for targeted therapy显示文摘Colorectal cancer(CRC)is a predominant life-threatening cancer,with liver and peritoneal metastases as the primary causes of death.Intestinal inflammation,a known CRC risk factor,nurtures a local inflammatory environment enriched with tumor cells,endothelial cells,immune cells,cancer-associated fibroblasts,immunosuppressive cells,and secretory growth factors.The complex interactions of aberrantly expressed cytokines,chemokines,growth factors,and matrix-remodeling enzymes promote CRC pathogenesis and evoke systemic responses that affect disease outcomes.Mounting evidence suggests that these cytokines and chemokines play a role in the progression of CRC through immunosuppression and modulation of the tumor microenvironment,which is partly achieved by the recruitment of immunosuppressive cells.These cells impart features such as cancer stem cell-like properties,drug resistance,invasion,and formation of the premetastatic niche in distant organs,promoting metastasis and aggressive CRC growth.A deeper understanding of the cytokineand chemokine-mediated signaling networks that link tumor progression and metastasis will provide insights into the mechanistic details of disease aggressiveness and facilitate the development of novel therapeutics for CRC.Here,we summarized the current knowledge of cytokine-and chemokine-mediated crosstalk in the inflammatory tumor microenvironment,which drives immunosuppression,resistance to therapeutics,and metastasis during CRC progression.We also outlined the potential of this crosstalk as a novel therapeutic target for CRC.The major cytokine/chemokine pathways involved in cancer immunotherapy are also discussed in this review.Ajaz A.Bhat Sabah Nisar Mayank Singh Bazella Ashraf Tariq Masoodi Chandra P.Prasad Atul Sharma Selma Maacha Thasni Karedath Sheema Hashem Syed Besina Yasin Puneet Bagga Ravinder Reddy Michael P.Frennaux Shahab Uddin Punita Dhawan Mohammad Haris Muzafar A.Macha 2022Cancer Communications2022,42,8:8
2Prevalence of visual impairment and related factors in Nangarhar Province of Afghanistan:a cross sectional study显示文摘AIM: To determine the prevalence, main causes, and related factors of visual impairment(VI) among people aged 50 y and over in Jalalabad City and four surrounding districts of Nangarhar Province of Afghanistan. METHODS: The data for the population based crosssectional study was collected in 2015. The calculated sample size was 1353, allocated to urban-rural strata using probability proportion to size method. At the end of the study, 1281 people participated in to the study. VI was defined as presenting visual acuity(VA) of less than 6/18 and blindness as VA less than 3/60 in the better eye by using Snellen chart only. Data was analyzed using IBM SPSS 21.0 software.RESULTS: The prevalence of VI was 22.6%(95%CI, 20%-25%) of which 13.9%(95%CI, 12%-16%) was low vision and 8.7%(95%CI, 7%-10%) was blindness. The most common causes of the VI were cataract(52.8%), followed by uncorrected refractive error(URE)(26.9%) and glaucoma(8.6%). Number one cause of the low vision was URE(42%), followed by cataract, glaucoma, age-related macular degeneration(AMD) and diabetic retinopathy(DR), while for blindness they are cataract(72%), other posterior segment disorders, glaucoma, URE and AMD. Illiteracy, bad economic status, hypertension and overweight were factors independently associated with both VI and low vision, whereas, age, illiteracy, bad economic status, hypertension and using of sunglasses were independently associated with blindness.CONCLUSION: Cataract, URE, glaucoma, AMD and DR are the leading causes of VI and blindness in the study area. They are mostly avoidable. 3A study on textural characteristics, heavy mineral distribution and grain-microtextures of recent sediment in the coastal area between the Sarada and Gosthani rivers, east coast of India显示文摘The current study aimed to describe textural characteristics,heavy mineral composition,and grain microtextures of the sediment from three micro-environments(foreshore,berm,and dune).A total of forty-one(41)representative surficial sediment samples have been collected from fifteen(15)locations along the beach area between the Sarada and Gosthani rivers on the east coast of India,where the length of the stretch is more than 100 km.The study reveals that most of the coastal sediment is medium to fine sand with relatively high ratios of coarse sand at Yarada beach,and the nature of the sediment is moderately to well sorted.These characteristics indicate a high energy environment.The heavy mineral analysis of the sediment in the current study was done for coarse(t60#)and fine(t230#)size fractions.Studying the weight percentage(WT%)reveals that a high percentage of heavy minerals is associated with fine fractions.Ilmenite,sillimanite,garnet,zircon,and rutile are the major heavy minerals identified in the current investigation.The concentrations of these heavy minerals show great variations from south to north of the study area.From an economic point of view,a considerable amount of heavy minerals(average 48.41%)are present on both sides(north and south)of the Gosthani River mouth.In the Sarada Estuary,the concentration of the economic heavy minerals was found under the minimum economic range.The grain microtextures of the major heavy minerals from the different locations along the study area demonstrate the variation in grain microtextures,which is controlled by the chemical and mechanical processes.These microtextures reflect moderate to high wave energy on the beach area,in addition to high mechanical impact on the grains from the estuary point.Ali Mohammad Parvathaneni Bhanu Murthy Edupuganti Naga Dhanamjaya Rao Hari Prasad 2020International Journal of Sediment Research2020,35,5:2
4bla_(CTX-M),bla_(TEM),and bla_(SHV) in Enterobacteriaceae from North-Indian tertiary hospital:high occurrence of combination genes显示文摘Objective:To delineate the frequency of occurrence of blaCTX-M,blaTEM,and blaSHV in Enterobacteriaceae from North-Indian tertiary hospital.Methods:A random collection of a subset of 45 Escherichia coli(E.coli) and 28 Klebsiella pneumoniae(K.pneumoniae) that was resistant to a third generation cephalosporin and obtained during 2007-2008 was selected for detailed screening for blaCTX-M,blaTEM,and blaSHV by monoplex PCRs.The isolates demonstrating the presence of blaCTX-M alleles were characterized for the specific CTX-M-genogroup by using a multiplex PCR.Results:Resistance to cefoperazone,ceftazidime,ceftriaxone,cefotaxime, cefoxitin and piperacillin was 100%each in K.pneumoniae isolates,whereas these resistancerates for E.coli isolates were 93.1%,83.8%,91.9%,93.6%,97.3%and 97.1%,respectively. Concomitant resistance to aminoglycosides,quinolones and aztreonam was also noticed.Presence of any of the bla genes(blaCTX-M,blaTEM,and blaSHV) was noticed in a total of 28(38.4%) isolates of the 73 isolates studied.Many isolates demonstrated occurrence of these genes in various combinations.blaCTX-M,blaTEM,and blaSHV were noticed in 28.8%,10.9%and 13.7%isolates, respectively.Multiplex PCR in blaCTX-M harboring isolates demonstrated the presence of CTX-MGenogroup -1 and sequencing for the specific CTX-M-type revealed presence of CTX-M-15 type. 5Changing trends in prevalence of different Plasmodium species with dominance of Plasmodium falciparum malaria infection in Aligarh(India)显示文摘Objective:To determine the prevalence of malaria in Aligarh and analyze species dominance in different years over a decade.Methods:Diagnosis of malaria was done using microscopy as gold standard,rapid antigen detection assays and quantitative buffy coat(QBC) assays.Giemsa stained blood smear examination was done,thick and thin films were examined for presence of different Plasmodium spp.Rapid antigen detection assays employing detection of HRP-2 and parasite lactate dehydrogenase antigen(pLDH) by immunochromatography was done in patients whose blood smear found to be negative by conventional Giemsa slide examination.QBC was done in cases where there is strong clinical suspicion of malaria with blood smear negative,in patients with chronic malaria,splenomegaly,or in those patients who had inadequate treatment and for post-treatment follow up.Results:Plasmodium vivax and Plasmodium falciparum were only species detected in our hospital.Overall prevalence of malaria in Aligarh was found to be 8.8%.The maximum prevalence of 20.1%was observed in year 2008 and lowest 2.3%in 2002. 9In vivo immunomodulatory profile of histamine receptors(H1,H2,H3 and H4):a comparative antagonists study显示文摘Objective:To delineate the comparative immunomodulatory roles of H1R-H4R in antibody generation profile in rabbit model.Methods:The cohort comprised of eight groups containing 18(9 male and 9 female) rabbits in each group.GroupⅠremained non-immunized and received only vehicle(sterile distilled water,1 mL/kg×b.i.d.) intramuscularly.GroupⅡreceived vehicle (1 mL/kg×b.i.d.) while GroupsⅢ-Ⅶ(drugs-treated) received subcutaneous histamine (100μg/kg×b.i.d.),and intramuscular H1R-antagonist(pheniramine,10 mg/kg×b.i.d.), H2R-antagonist(ranitidine,10 mg/kg×b.i.d.),H3R-antagonist(iodophenpropit,1μg/kg×b.i.d.) and H4R-antagonist(JNJ 7777120,10μg/kg×b.i.d.),and GroupⅧDMSO(1 mL/kg×b.i.d.),respectively for 10 days(starting from day 1).They were subsequendy immunized with intravenous injection of sheep red blood cells(SRBC) at day 3.The estimation of serum Igs,IgM and IgG were done by ELISA,and observed at day 0(pre-immunization),and 7,14,21,28 and 58(post-immunization).Results:It was shown that histamine and HRs-antagonists could influence a detectable antibody response to SRBC as early as day 7-post-immunization(post-Ⅰ), which lasted until day 58 post-Ⅰ.The results were found statistically significant(P<0.05,). 10Detection of circulating tumour cells in colorectal cancer:Emerging techniques and clinical implications显示文摘Despite several advances in oncological management of colorectal cancer,morbidity and mortality are still high and devastating.The diagnostic evaluation by endoscopy is cumbersome,which is uncomfortable to many.Because of the intra-and inter-tumour heterogeneity and changing tumour dynamics,which is continuous in nature,the diagnostic biopsy and assessment of the pathological sample are difficult and also not adequate.Late manifestation of the disease and delayed diagnosis may lead to relapse or metastases.One of the keys to improving the outcome is early detection of cancer,ease of technology to detect with uniformity,and its therapeutic implications,which are yet to come.'Liquid biopsy'is currently the most recent area of interest in oncology,which may provide important tools regarding the characterization of the primary tumour and its metastasis as cancer cells shed into the bloodstream even at the early stages of the disease.By using this approach,clinicians may be able to find out information about the tumour at a given time.Any of the following three types of sampling of biological material can be used in the'liquid biopsy'.These are circulating tumour cells(CTCs),circulating tumour DNA,and exosomes.The most commonly studied amongst the three is CTCs.CTCs with their different applications and prognostic value has been found useful in colorectal cancer detection and therapeutics.In this review,we will discuss various markers for CTCs,the core tools/techniques for detection,and also important findings of clinical studies in colorectal cancer and its clinical implications.Alka Yadav Ashok Kumar Mohammad Haris Siddiqui 2021World Journal of Clinical Oncology2021,12,12:0
11Multifaceted Strategies Used by Root-Knot Nematodes to Parasitize Plants-A Review显示文摘Root-knot nematodes being omnipresent in agricultural and horticultural soils are tallied among the most important economic pathogens around the world.For successful parasitism,these nematodes use various strategies to control and manipulate the host plant’s cell machinery.These strategies include the mole-cular mimicry of some host genes by some nematode secreted effector proteins,secretion of cell wall digesting enzymes and other effector proteins that are responsible for the suppression of defence by the host plant.All these secretions which are released through the stylet,contribute to the formation of specialized feeding sites or giant cells.The effector proteins interfere with the normal physiology,cytology and biochemistry of the host plant.The present review brings novel insights by summarizing some novel effectors that have been discovered recently like MgPDI,MiMIF,MiIDL1,MiISE6,Mg16820,etc.It also discusses some novel mechanisms through which these effector proteins target different pathways of host plants and thus facilitate nematode parasitism.Adnan Shakeel Abrar Ahmad Khan Mohammad Haris 2020Phyton-International Journal of Experimental Botany2020,89,2:0
