| 1 | Suppression of bcl-2 Gene by RNA Interference Increases Chemosensitivity to Cisplatin in Nasopharyngeal Carcinoma Cell Line CNE1显示文摘To explore the effect of suppressing BCL-2 expression using RNA interference (RNAi) technique in nasopharyngeal carcinoma cell line CNE 1. CNE 1 cell lines stably expressing shRNAs targeted bcl-2 and GL3 gene were established and gene expression inhibition was assessed by Western blotting analysis. The effect of suppressing bcl-2 by RNAi on cell growth was studied, the apoptosis induction and the sensitization of CNE 1 cells to cisplatin were quantified by MTT assay and flow cytometry. The results showed that: stable transfection of CNE 1 cells with vectors expressing shRNAs against bcl-2 decreased the expression of BCL-2 protein; suppression of BCL-2 expression did not affect cell proliferation but could increase the chemosensitivity to cisplatin in CNE 1 cells. | 2 | Deterministic Approach to Achieve Full-Polarization Cloak显示文摘Achieving full-polarization(σ)invisibility on an arbitrary three-dimensional(3D)platform is a long-held knotty issue yet extremely promising in real-world stealth applications.However,state-of-the-art invisibility cloaks typically work under a specific polarization because the anisotropy and orientation-selective resonant nature of artificial materials made theσ-immune operation elusive and terribly challenging.Here,we report a deterministic approach to engineer a metasurface skin cloak working under an arbitrary polarization state by theoretically synergizing two cloaking phase patterns required,respectively,at spin-up(σ+)and spin-down(σ−)states.Therein,the wavefront of any light impinging on the cloak can be well preserved since it is a superposition ofσ+andσ−wave.To demonstrate the effectiveness and applicability,several proof-of-concept metasurface cloaks are designed to wrap over a 3D triangle platform at microwave frequency.Results show that our cloaks are essentially capable of restoring the amplitude and phase of reflected beams as if light was incident on a flat mirror or an arbitrarily predesigned shape under full polarization states with a desirable bandwidth of~17.9%,conceiving or deceiving an arbitrary object placed inside.Our approach,deterministic and robust in terms of accurate theoretical design,reconciles the milestone dilemma in stealth discipline and opens up an avenue for the extreme capability of ultrathin 3D cloaking of an arbitrary shape,paving up the road for real-world applications. | He-Xiu Xu Yanzhao Wang Chaohui Wang Mingzhao Wang Shaojie Wang Fei Ding Yongjun Huang Xiaokuan Zhang Haiwen Liu Xiaohui Ling Wei Huang | 2021 | Research2021,,1: | 5 |

| 10 | RelB intrinsically regulates the development and function of medullary thymic epithelial cells显示文摘Medullary thymic epithelial cells(mTECs) act as one of the major stromal components in the thymus for selection and maturation of both conventional T cells and non-conventional T cells. Extensive efforts have been spent to understand how mTEC development and function are regulated. Although RelB has been well accepted to be a critical transcriptional factor for mTEC development, the underlying mechanisms still remain largely unclear. In this study, by generating thymic epithelial cell specific RelB deficient mice, we found that epithelial intrinsic RelB is required for mTEC homeostatic proliferation. Mechanistically, RelB regulates the expression of genes involved in cell cycle. Functionally, lack of intrinsic RelB in thymic epithelial cells results in dramatically reduced population of mTECs and impaired development of thymic invariant natural killer T(iNKT) cells and intraepithelial lymphocyte precursors(IELPs). | 13 | Establishment of Human Embryonic Stem Cell Line Stably Expressing Epstein-Barr Virus-Encoded Nuclear Antigen 1显示文摘Human embryonic stem (hES) cells have the capability of unlimited undifferentiated proliferation, yet maintain the potential to form perhaps any cell type in the body. Based on the high efficiency of the Epstein-Barr virus-based episomal vector in introducing exogenous genes of interest into mammalian cells, we applied this system to hES cells, expecting that this would resolve the problem of poor transfection efficiency existing in current hES cell research. Therefore, the first step was to establish EBNA1-positive hES cells. Using the Fugene 6 transfection reagent, we transfected hES cells with the EBNA1 expression vector and subsequently generated hES cell clones that stably expressed EBNA1 under drug selection. These clones were confirmed to express EBNA1 mRNA by RT-PCR and to express EBNA1 protein by Western blotting. Furthermore, luciferase reporter gene analysis was performed on the EBNA1 clones and revealed that the expressed EBNA1 protein was functional. When the EBNA1-positive cells were injected into severe combined immunodeficient (SCID) mice, they formed teratoma tissues containing all three embryonic germ layers and EBNA1 protein was detected in these teratoma tissues by Western blotting. | 17 | Anatomical study of sciatic nerve and common peroneal nerve compression显示文摘BACKGROUND: Many diseases of the common peroneal nerve are a result of sciatic nerve injury. The present study addresses whether anatomical positioning of the sciatic nerve is responsible for these injuries. OBJECTIVE: To analyze anatomical causes of sciatic nerve and common peroneal nerve injury by studying the relationship between the sciatic nerve and piriformis. DESIGN, TIME AND SETTING: Observe and measure repeatedly. The experiment was conducted in the Department of Anatomy, Tianjin Medical College between January and June 2005. MATERIALS: Fifty-two adult cadavers 33 males and 19 females, with a total of 104 hemispheres, and fixed with formaldehyde, were provided by Tianjin Medical College and Tianjin Medical University. METHODS: A posterior cut was made from the lumbosacral region to the upper leg, fully exposing the piriformis and path of the sciatic nerve. MAIN OUTCOME MEASURES: (1) Anatomical characteristics of the tibial nerve and common peroneal nerve. (2) According to different areas where the sciatic nerve crosses the piriformis, the study was divided into two types-normal and abnormal. Normal is considered to be when the sciatic nerve passes through the infrapiriform foramen. Remaining pathways are considered to be abnormal. (3) Observe the relationship between the suprapiriform foramen, infrapiriform foramen, as well as the superior and inferior space of piriformis. RESULTS: (1) The nerve tract inside the common peroneal nerve is smaller and thinner, with less connective tissue than the tibial nerve. When pathological changes or variations of the piriformis, or over-abduction of the hip joint, occur, injury to the common peroneal nerve often arises due to blockage and compression. (2) A total of 76 hemispheres (73.08%) were normal, 28 were abnormal (26.92%). The piriformis can be injured, and the sciatic nerve can become compressed, when the hip joint undergoes intorsion, extorsion, or abduction. (3) The structures between the infrapiriform and suprapiriform foramen are where 'the first threshold' sciatic nerve projects. The structures between the infrapiriform and suprapiriform gap were 'the second threshold'. This became the concept of 'double threshold'. The reduced area caused by pathological changes of 'double threshold' may block and compress the sciatic nerve. Because the common peroneal nerve lies on the anterolateral side of the sciatic nerve, injury to the common peroneal nerve is more serious.
