| 1 | Middle–Upper Ordovician(Darriwilian–Early Katian) Positive Carbon Isotope Excursions in the Northern Tarim Basin, Northwest China: Implications for Stratigraphic Correlation and Paleoclimate显示文摘Three positive carbon isotope excursions are reported from Middle–Upper Ordovician in Tahe oil-gas field, northern Tarim Basin. Based on conodont biostratigraphy, the Middle Darriwilian Isotope Carbon Excursion(MDICE) and the Guttenberg Carbon Isotope Excursion(GICE) are identified from Darriwilian to Early Katian by the aid of whole-rock carbon isotope data from two well cores. Positive excursion within conodont Pygodus anserinus zone is developed in Early Sandbian, and the fluctuation range is no less than MDICE. Because the range of this excursion in the generalized global carbon isotope curve is short, previous studies paid little attention to it, and named Early Sandbian Isotope Carbon Excursion(ESICE) in this paper. Furthermore, these positive excursions are not directly related to sea level fluctuations and the MDICE and GICE identified in northern Tarim can be globally correlated to that in southern China, North America, South America, and Europe. The Saergan Fm. source rocks of Middle-Upper Ordovician in Kalpin Dawangou outcrop are in accord with the geologic time of MDICE and ESICE, and GICE have strong ties to the source rock of Lianglitag Fm. in basin. Abundant organic carbon burial is an important factor in genesis of positive isotope carbon excursions. | 2 | Primarily screening and analyzing ESTs differentially expressed in rats' primary liver cancer显示文摘Objective: To screen and analyze key express sequence tags (ESTs) which were differentially displayed in every period of SD rats' primary hepatic carcinoma and reveal the molecular mechanism of carcinogenesis. Methods: Using diethylnitrosamine (DENA) as a cancerigenic agent, animal models with different phases of primary hepatic cancer were constructed in SD rats. Rats were respectively sacrificed at d 14, d 28, d 56, d 77, d 105 and d 112 after the rats received DENA by gavage, then the livers were harvested. One part of the livers was classified according to their pathological changes, while the other was reserved for molecular mechanism studies on hepatocarcinogenesis. The differentially expressed genes were isolated from both normal and morbid tissues by mRNA differential display technique (DDRT-PCR). After the fragments were sequenced, bioinformatics were used to analyze the results. Results: Twelve differentially expressed cDNA fragments were obtained. Nine fragments had the homology with known cDNA clones, especially EST-7 was similar to BN/SsNHsdMCW mitochondrion gene and the identity was 100% which suggested EST-7 may be the part of BN/SsNHsdMCW mitochondrion gene. In contrast, other three fragments (EST-1, EST-3 and EST-5) had extremely low identity to any genes registered in GENBANK databases. Conclusions: BN/SsNHsdMCW mitochondrion gene was expressed in different periods of hepatocarcinogenesis. Furthermore, the induction period of various temperatures and supersaturation ratios was also measured. | 8 | Quantitative analysis of Pb adsorption on sulfhydryl-modified biochar显示文摘Biochar is an effective absorbent for remediating heavy metal contaminated soil,but functional optimization is still needed to improve its performance in field application.Here,we characterized the physical structures and surface chemical properties of raw wood biochar and palm biochar(WB and PB)and the corresponding sulfhydryl-modified biochar(SWB and SPB).Their adsorption capacity for Pb was evaluated by combining thermodynamic and kinetic adsorption at 0.01 mol/L KCl and corresponding model simulation.The results demonstrated successful grafting of sulfhydryl groups onto the biochar,which dramatically reduced the specific surface area(SSA)and pore volume of biochar.The pKa in the surface complexation model(SCM)indicated similar proton affinity between sulfhydryl groups and original functional groups on the biochar.SCM could satisfactorily fit the Pb adsorption behaviors,and model analysis revealed that Pb tended to be adsorbed on low-proton affin-ity sites at low pH,but high-proton affinity sites became dominant in Pb adsorption with increasing pH and adsorbed almost all Pb ions at pH>7.0.Besides,the Pb adsorption density of SWB and SPB was improved by 8.86 and 3.64 folds relative to that of WB and PB,respectively.Over 90% of initially added Pb ions were removed in 1440 and 720 min by raw and sulfhydryl-modified biochar,respectively,indicating that sulfhydryl modification accelerated the Pb adsorption of biochar.These results suggest that site density,SSA and pore structure of biochar play crucial roles in heavy metal adsorption,and sulfhydryl modification may improve the performance of biochar in remediating heavy metal contaminated soil. | Juan Xiong Mengge Zhou Chenchen Qu Daohai Yu Chang Chen Mingxia Wang Wenfeng Tan | 2021 | Biochar2021,3,1: | 5 |

| 10 | Waste activated sludge treatment based on temperature staged and biologically phased anaerobic digestion system显示文摘The concept of temperature staged and biological phased(TSBP) was proposed to enhance the performance of waste-activated sludge anaerobic digestion. Semi-continuous experiments were used to investigate the efect of temperature(35 to 70°C) as well as the hydraulic retention time(HRT)(2, 4 and 6 days) on the acidogenic phase. The results showed that the solubilization degree of wasteactivated sludge increased from 14.7% to 30.1% with temperature increasing from 35 to 70°C, while the acidification degree was highest at 45°C(17.6%), and this was quite diferent from the temperature impact on hydrolysis. Compared with HRT of 2 and 6 days,4 days was chosen as the appropriate HRT because of its relatively high solubilization degree(24.6%) and acidification degree(20.1%)at 45°C. The TSBP system combined the acidogenic reactor(45°C, 4 days) with the methanogenic reactor(35°C, 16 days) and the results showed 84.8% and 11.4% higher methane yield and volatile solid reduction, respectively, compared with that of the single-stage anaerobic digestion system with HRT of 20 days at 35°C. Moreover, diferent microbial morphologies were observed in the acidogenicand methanogenic-phase reactors, which resulted from the temperature control and HRT adjustment. All the above results indicated that45°C was the optimum temperature to inhibit the activity of methanogenic bacteria in the acidogenic phase, and temperature staging and phase separation was thus accomplished. Various characterization techniques such as XRD, AES, nano indenter and profiler were employed to investigate the influence of modulation period and bombarding energy on microstructure and mechanical properties of the coatings. The results showed that all superlattice coatings had better mechanical properties than the monolithic ZrN and W coatings. At an optimal condition with 300 eV N+ beam bombarding energy and 8―9 nm modulation period, XRD pattern possessed a sig- nificantly structural mixture of strong ZrN (111), W (110), as well as weak ZrN (220) textures in the multilayered coating. | 14 | Highly active multivalent multielement catalysts derived from hierarchical porous TiNb_(2)O_(7) nanospheres for the reversible hydrogen storage of MgH_(2)显示文摘Critical limitations in applying MgH_(2) as a hydrogen-storage medium include the high H_(2) desorption temperature and slow reaction kinetics.In this study,we synthesized hierarchical porous TiNb_(2)O_(7) spheres in micrometer scale built with 20-50 nm nanospheres,which showed stable activity to catalyze hydrogen storage in MgH_(2) as precursors.The addition of 7 wt.%TiNb_(2)O_(7) in MgH_(2) reduced the dehydrogenation onset temperature from 300 to 177℃.At 250℃,approximately 5.5 wt.%H_(2) was rapidly released in 10 min.Hydrogen uptake was detected even at room temperature under 50 bar hydrogen;4.5 wt.%H_(2) was absorbed in 3 min at 150℃,exhibiting a superior low-temperature hydrogenation performance.Moreover,nearly constant capacity was observed from the second cycle onward,demonstrating stable cyclability.During the ball milling and initial de/hydrogenation process,the high-valent Ti and Nb of TiNb_(2)O_(7) were reduced to the lower-valent species or even zero-valent metal,which in situ created multivalent multielement catalytic surroundings.A strong synergistic effect was obtained for hybrid oxides of Nb and Ti by density functional theory(DFT)calculations,which largely weakens the Mg-H bonding and results in a large reduction in kinetic barriers for hydrogen storage reactions of MgH_(2).Our findings may guide the further design and development of high-performance complex catalysts for the reversible hydrogen storage of hydrides. | Lingchao Zhang Ke Wang Yongfeng Liu Xin Zhang Jianjiang Hu Mingxia Gao Hongge Pan | 2021 | Nano Research2021,14,1: | 2 |
| 15 | Nuclear factor κB represses the expression of latent membrane protein 1 in Epstein-Barr virus transformed cells显示文摘AIM: To investigate the role of nuclear factor κB(NF-κB) in the regulation of Epstein-Barr virus(EBV) latent membrane protein 1(LMP1) in EBV transformed cells. METHODS: LMP1 expression was examined in EBV transformed human B lymphocytes with modulation of NF-κB activity. RESULTS: EBV infection is associated with several human cancers. EBV LMP1 is required for efficient transformation of adult primary B cells in vitro, and is expressed in several pathogenic stages of EBVassociated cancers. Regulation of EBV LMP1 involves both viral and cellular factors. LMP1 activates NF-κB signaling pathway that is a part of the EBV transformation program. However, the relation between NF-κB and LMP1 expression is not well established yet. In this report, we found that blocking the NF-κB activity by Inhibitor of κB stimulated LMP1 expression, while the overexpression of NF-κB repressed LMP1 expression in EBV-transformed IB4 cells. In addition, LMP1 repressed its own promoter activities in reporter assays, and the repression was associated with the activation of NF-κB. Moreover, NF-κB alone is sufficient to repress LMP1 promoter activities. | 19 | CRISPR-Cas adaptive immune systems in Sulfolobales:genetic studies and molecular mechanisms显示文摘CRISPR-Cas systems provide the small RNA-based adaptive immunity to defend against invasive genetic elements in archaea and bacteria.Organisms of Sulfolobales,an order of thermophilic acidophiles belonging to the Crenarchaeotal Phylum,usually contain both type I and typeⅢCRISPR-Cas systems.Two species,Saccharolobus solfataricus and Sulfolobus islandicus,have been important models for CRISPR study in archaea,and knowledge obtained from these studies has greatly expanded our understanding of molecular mechanisms of antiviral defense in all three steps:adaptation,expression and crRNA processing,and interference.Four subtypes of CRISPR-Cas systems are common in these organisms,including I-A,I-D,Ⅲ-B,andⅢ-D.These cas genes form functional modules,e.g.,all genes required for adaptation and for interference in the I-A immune system are clustered together to form aCas and i Cas modules.Genetic assays have been developed to study mechanisms of adaptation and interference by different CRISPR-Cas systems in these model archaea,and these methodologies are useful in demonstration of the protospacer-adjacent motif(PAM)-dependent DNA interference by I-A interference modules and multiple interference activities byⅢ-B Cmr systems.Ribonucleoprotein effector complexes have been isolated for SulfolobalesⅢ-B andⅢ-D systems,and their biochemical characterization has greatly enriched the knowledge of molecular mechanisms of these novel antiviral immune responses. | Zhenxiao Yu Suping Jiang Yuan Wang Xuhui Tian Pengpeng Zhao Jianan Xu Mingxia Feng Qunxin She | 2021 | Science China(Life Sciences)2021,64,5: | 2 |
