1Mechanisms of traditional Chinese medicines in the treatment of allergic rhinitis using a network biology approach显示文摘Objective:To systematically explore the effect and mechanism of traditional Chinese herbs in the treatment of allergic rhinitis.Methods:Magnolia flower,xanthium fruit,astragalus root,paniculate cynanchum,scutellaria root,cicada molting,stephania root,small centipeda herb,and licorice root as traditional Chinese herbs are widely used to treat allergic rhinitis.With multiple database integration and entity grammar systems,a molecular interaction network of traditional Chinese herbs for treating allergic rhinitis was established.Bioinformatics approaches were adopted to integrate relevant data and biological information by multiple databases.Molecular interaction network of Chinese herbs for treating allergic rhinitis was constructed based on entity grammar systems model.The network was then used for elucidating the antiallergic rhinitis mechanism of the Chinese herbs on the molecular level.The subnetworks were also extracted to explicitly display the pathway and targets where effective components acted on.Results:Traditional Chinese herbs could influence various pathological aspects in allergic rhinitis including the production of pro-inflammatory substances,such as histamine and cytokine interleukin-4(IL-4),interleukin-13(IL-13),lowering immunoglobulin E(IgE)level or blocking antigen binding,altering the biological processes of IgE,modulating the balance of T helper(Th)or other cells by cell proliferation and differentiation,mediating cellecell signaling and second-messenger-mediated signaling,stabilizing the cell membrane,and affecting regulation of cellular defense response.Conclusion:The research theoretically confirms the mechanism of anti-allergic rhinitis by traditional Chinese herbs,provides important fundamental research information for treatment of allergic rhinitis,and may serve as a reference for new drug development and effective ingredients of compatibility.Li Du Xiaotong Ye Manman Li Huihui Wang Baixia Zhang Rao Zheng Yun Wang 2021Journal of Traditional Chinese Medical Sciences2021,8,1:3
2BLM helicase inhibition synergizes with PARP inhibition to improve the radiosensitivity of olaparib resistant non-small cell lung cancer cells by inhibiting homologous recombination repair显示文摘Objective:We aimed to investigate the radiosensitizing efficacy of the poly-ADP-ribose polymerase(PARP)inhibitor,olaparib,and the Bloom syndrome protein(BLM)helicase inhibitor,ML216,in non-small cell lung cancer(NSCLC)cells.Methods:Radiosensitization of NSCLC cells was assessed by colony formation and tumor growth assays.Mechanistically,the effects of ML216,olaparib,and radiation on cell and tumor proliferation,DNA damage,cell cycle,apoptosis,homologous recombination(HR)repair,and non-homologous end joining(NHEJ)repair activity were determined.Results:Both olaparib and ML216 enhanced the radiosensitivities of olaparib-sensitive H460 and H1299 cells,which was seen as decreased surviving fractions and Rad51 foci,increased total DNA damage,andγH2AX and 53BP1 foci(P<0.05).The expressions of HR repair proteins were remarkably decreased in olaparib-treated H460 and H1299 cells after irradiation(P<0.05),while olaparib combined with ML216 exerted a synergistic radiosensitization effect on olaparib-resistant A549 cells.In addition to increases of double strand break(DSB)damage and decreases of Rad51 foci,olaparib combined with ML216 also increased pDNA-PKcs(S2056)foci,abrogated G2 cell cycle arrest,and induced apoptosis in A549 lung cancer after irradiation in vitro and in vivo(P<0.05).Moreover,Western blot showed that olaparib combined with ML216 and irradiation inhibited HR repair,promoted NHEJ repair,and inactivated cell cycle checkpoint signals both in vitro and in vivo(P<0.05).Conclusions:Taken together,these results showed the efficacy of PARP and BLM helicase inhibitors for radiosensitizing NSCLC cells,and supported the model that BLM inhibition sensitizes cells to PARP inhibitor-mediated radiosensitization,as well as providing the basis for the potential clinical development of this combination for tumors intrinsically resistant to PARP inhibitors and radiotherapy.Yangyang Kong Chang Xu Xiaohui Sun Hao Sun Xiaotong Zhao Ningning He Kaihua Ji Qin Wang Liqing Du Jinhan Wang Manman Zhang Yang Liu Yan Wang Qiang Liu 2022Cancer Biology & Medicine2022,19,8:1

4A FLASH pipeline for arrayed CRISPR library construction and the gene function discovery of rice receptor-like kinases显示文摘Clustered regularly interspaced short palindromic repeats(CRISPR)-CRISPR-associated protein 9(Cas9)-mediated gene editing is revolutionizing plant research and crop breeding.Here,we present an effective and streamlined pipeline for arrayed CRISPR library construction and demonstrate it is suitable for small-to large-scale genome editing in plants.This pipeline introduces artificial PCR fragment-length markers for distinguishing guide RNAs(gRNAs)(FLASH),and a group of 12 constructs harboring different FLASH tags are co-transformed into plants each time.The identities of gRNAs in Agrobacterium mixtures and trans-genic plants can therefore be read out by detecting the FLASH tags,a process that requires only conven-tional PCR and gel electrophoresis rather than sequencing.We generated an arrayed CRISPR library target-ing all 1,072 members of the receptor-like kinase(RLK)family in rice.One-shot transformation generated a mutant population that covers gRNAs targeting 955 RLKs,and 74.3%(710/955)of the target genes had three or more independent T0 lines.Our results indicate that the FLASH tags act as bona fide surrogates for the gRNAs and are tightly(92.1%)associated with frameshift mutations in the target genes.In addition,the FLASH pipeline allows for rapid identification of unintended editing events without corresponding T-DNA integrations and generates high-order mutants of closely related RLK genes.Furthermore,we showed that the RLK mutant library enables rapid discovery of defense-related RLK genes.This study introduces an effective pipeline for arrayed CRISPR library construction and provides genome-wide rice RLK mutant resources for functional genomics.Kaiyuan Chen Runnan Ke Manman Du Yuqing Yi Yache Chen Xiaochun Wang Lu Yao Hao Liu Xin Hou Lizhong Xiong Yinong Yang Kabin Xie 2022Molecular Plant2022,15,2:0
5Advances in airborne microorganisms detection using biosensors:A critical review显示文摘Humanity has been facing the threat of a variety of ifectious diseases.Airborne microorganisms can cause airbome infectious diseases,which spread rapidly and extensively,causing huge losscs to human society on a global scale.In recent years,the detection technology for airbome microorganisms has developed rapidly;it can be roughly divided into biochemical,immune,and molecular technologies.However,these technologies still have some shortcomings;they are time consuming and have low sensitivity and poor stability.Most of them need to be used in the ideal environment of a laboratory,which limits their applications.A biosensor is a device that converts biological signals into detectable signals.As an interdisciplinary feld,biosensors have successfully introduced a variety of technologies for bio-detection.Given their fast analysis speed,high sensitivity good portability,strong specifcity,and low cost,biosensors have been widely uised in cnvironmental monitoring,medical research,food and agricultural safety,military.medicine and other fields.In recent years,the performance of biosensors has greatly improved,becoming.a promising techmology for airborne microorganism detection.This review introduces the detection principle of biosensors from the three aspects of component identification,energy conversion principle,and signal amplification.It also summarizes its research and application in airborne microorganism detection.The new progress and future development trend of the biosensor detection of airbormne microorganisms are analyzed.Jinbiao Ma Manman Du Can Wang Xinwu Xie Hao Wang Qian Zhang 2021Frontiers of Environmental Science & Engineering2021,15,3:0
